[Progress on the application of respondent-driven sampling in population size estimation].

Respondent-driven sampling (RDS) is a method of sampling specifically for hidden populations, often applied to transgender women, female sex workers, men who have sex with men, and other groups that are difficult to identify and contact because of stigma and legal constraints. However, this method is gradually applied to the general population. With the continuous improvement of RDS, studies find that the network size of RDS samples can be weighted to estimate the overall situation and population size. This article summarizes the current application progress of RDS in population size estimatio

Empirically-derived synthetic populations to mitigate small sample sizes.

Limited sample sizes can lead to spurious modeling findings in biomedical research. The objective of this work is to present a new method to generate synthetic populations (SPs) from limited samples using matched case-control data (n = 180 pairs), considered as two separate limited samples. SPs were generated with multivariate kernel density estimations (KDEs) with unconstrained bandwidth matrices. We included four continuous variables and one categorical variable for each individual. Bandwidth matrices were determined with Differential Evolution (DE) optimization by covariance comparisons. Fo

Robust Markers and Sample Sizes for Multicenter Trials of Huntington Disease.

OBJECTIVE: The identification of sensitive biomarkers is essential to validate therapeutics for Huntington disease (HD). We directly compare structural imaging markers across the largest collective imaging HD dataset to identify a set of imaging markers robust to multicenter variation and to derive upper estimates on sample sizes for clinical trials in HD. METHODS: We used 1 postprocessing pipeline to retrospectively analyze T1-weighted magnetic resonance imaging (MRI) scans from 624 participants at 3 time points, from the PREDICT-HD, TRACK-HD, and IMAGE-HD studies. We used mixed effects mode

Group Sequential Sample Size for Comparing Two Survival Probabilities at a Specific Time Point.

We propose a novel method that simultaneously determines the sample size for testing two survival probabilities at a pre-specified time while guaranteeing Type I error control in both fixed and group-sequential trial designs. Simulations across varying hypothesized differences, failure distributions, censoring proportions, and nominal powers demonstrate consistent performance, while interim analyses highlight maintained nominal Type I error control and increased power at each look relative to the standard binomial approach, regardless of the underlying failure time distribution or spending fun

The Non-Human Primate Sample Size Calculator: a design tool for longitudinal research projects.

Nonhuman primates (NHP) are crucial models of human health and disease and offer the opportunity to bridge the gap between basic research and the clinic. Given the ethical considerations, high costs, and logistical constraints associated with NHP studies, careful planning is essential to maximize scientific rigor while minimizing animal use. Statistical power calculations, to determine optimal sample size, require knowing the expected number of animals at each timepoint, a challenge for longitudinal studies that must account for natural deaths or disease during a study. Here, we leveraged data

Sample size determination across study designs in health professions education research: a methodology guide.

BACKGROUND: Sample size determination is one of the least reported elements in the methods sections of health professions education (HPE) research. Underpowered studies miss real effects, whereas overpowered studies waste participants and resources. Guidance remains fragmented across statistical textbooks and discipline-specific papers, with no unified reference for HPE researchers. METHODS: We developed this guide as a structured narrative synthesis. Sources were identified through targeted database searching, citation chaining, and standard statistical texts, and selected against pre-specif

Evaluating the Impact of Outcome Delay on the Efficiency of Sample Size Re-Estimation.

Sample size re-estimation (SSR) can be a powerful tool to ensure that a clinical trial meets its pre-specified power requirements when uncertainty regarding a design parameter exists at the planning stage. However, long-term primary endpoints can be harmful to the efficiency of this trial design. If recruitment is continued while treatment outcomes are awaited, long delays can potentially lead to a large number of 'pipeline' participants being recruited in the trial that do not contribute to the interim analysis. This may lead to a larger number of recruited participants than are actually deem

Extended sample size calculations for evaluation of prediction models using a threshold for classification.

When evaluating the performance of a model for individualised risk prediction, the sample size needs to be large enough to precisely estimate the performance measures of interest. Current sample size guidance is based on precisely estimating calibration, discrimination, and net benefit, which should be the first stage of calculating the minimum required sample size. However, when a clinically important threshold is used for classification, other performance measures are also often reported. We extend the previously published guidance to precisely estimate threshold-based performance measures.

Sample Size for Enriched Biomarker Designs With Measurement Error for Time-to-Event Outcomes.

A major emphasis in personalized medicine is to optimally treat subgroups of patients who may benefit from certain therapeutic agents. One relevant study design is the targeted design, in which patients have consented for their specimens to be obtained at baseline and the specimens are sent to a laboratory for assessing the biomarker status prior to randomization. Here, only biomarker-positive patients will be randomized to either an experimental or the standard of care arms. Many biomarkers, however, are derived from patient tissue specimens, which are heterogeneous leading to variability in

Penetration depth and effective sample size characterization of UV/Vis radiation into pharmaceutical tablets.

The pharmaceutical industry is moving from off-line to real-time release testing (RTRT) to enhance quality while reducing costs. UV/Vis spectroscopy has emerged as a promising tool for RTRT given its simplicity, sensitivity and cost-effectiveness. Nevertheless, the effective sample size must be characterized in relation to the penetration depth to justify its representativeness and suitability for RTRT. In this study, bilayer tablets were produced using a hydraulic tablet press. The lower layer contained titanium dioxide and microcrystalline cellulose (MCC), while the upper layer consisted of

Brain-age prediction: Systematic evaluation of site effects, and sample age range and size.

Structural neuroimaging data have been used to compute an estimate of the biological age of the brain (brain-age) which has been associated with other biologically and behaviorally meaningful measures of brain development and aging. The ongoing research interest in brain-age has highlighted the need for robust and publicly available brain-age models pre-trained on data from large samples of healthy individuals. To address this need we have previously released a developmental brain-age model. Here we expand this work to develop, empirically validate, and disseminate a pre-trained brain-age mode

Cardiovascular magnetic resonance feature tracking in pigs: a reproducibility and sample size calculation study.

Cardiovascular magnetic resonance feature tracking (CMR-FT) is a novel technique for non-invasive assessment of myocardial motion and deformation. Although CMR-FT is standardized in humans, literature on comparative analysis from animal models is scarce. In this study, we measured the reproducibility of global strain under various inotropic states and the sample size needed to test its relative changes in pigs. Ten anesthetized healthy Landrace pigs were investigated. After baseline (BL), two further steps were performed: (I) dobutamine-induced hyper-contractility (Dob) and (II) verapamil-indu

Sample Size Estimates for Cluster-Randomized Trials in Hospital Infection Control and Antimicrobial Stewardship.

IMPORTANCE: An important step in designing, executing, and evaluating cluster-randomized trials (CRTs) is understanding the correlation and thus nonindependence that exists among individuals in a cluster. In hospital epidemiology, there is a shortage of CRTs that have published their intraclass correlation coefficient or coefficient of variation (CV), making prospective sample size calculations difficult for investigators. OBJECTIVES: To estimate the number of hospitals needed to power parallel CRTs of interventions to reduce health care-associated infection outcomes and to demonstrate how di

Genotyping as a Key Element of Sample Size Optimization in Bioequivalence of Risperidone Tablets.

BACKGROUND AND OBJECTIVES: Risperidone is a derivative of benzisoxazole and is widely used for schizophrenia and other psychiatric illnesses in both adults and children. Previous studies have confirmed that it is a highly variable drug (within-subject variability ≥ 30%). To reduce the large sample size required for bioequivalence researches on highly variable drugs, a role for genotyping in the design of the bioequivalence study was employed. METHODS: A randomized, open-label, two-period crossover study was adopted: 20 subjects with specific genotypes carrying cytochrome P450 (CYP) 2D6*10 wer

Sample size calculation for agreement between two raters with binary endpoints using exact tests.

In an agreement test between two raters with binary endpoints, existing methods for sample size calculation are always based on asymptotic approaches that use limiting distributions of a test statistic under null and alternative hypotheses. These calculated sample sizes may be not reliable due to the unsatisfactory type I error control of asymptotic approaches. We propose a new sample size calculation based on exact approaches which control for the type I error rate. The two exact approaches are considered: one approach based on maximization and the other based on estimation and maximization.

Sample size and robust marginal methods for cluster-randomized trials with censored event times.

In cluster-randomized trials, intervention effects are often formulated by specifying marginal models, fitting them under a working independence assumption, and using robust variance estimates to address the association in the responses within clusters. We develop sample size criteria within this framework, with analyses based on semiparametric Cox regression models fitted with event times subject to right censoring. At the design stage, copula models are specified to enable derivation of the asymptotic variance of estimators from a marginal Cox regression model and to compute the number of cl

Intravenous immunoglobulin maintenance treatment in myasthenia gravis: a randomized, controlled trial sample size simulation.

INTRODUCTION: In cases of exacerbation or crisis, myasthenia gravis (MG) patients can be treated with intravenous immunoglobulin (IVIg), plasmapheresis, or immunoadsorption. However, IVIg efficacy data in maintenance treatment are sparse. METHODS: We prospectively observed 16 index patients with chronic and insufficiently controlled MG under standard immunosuppressant therapy and symptomatic treatment. The IVIg treatment response was measured using changes in quantitative myasthenia gravis (QMG) score and surrogates. Based on these results, a sample size calculation for a future randomized, c

What is an adequate sample size? Operationalising data saturation for theory-based interview studies.

In interview studies, sample size is often justified by interviewing participants until reaching 'data saturation'. However, there is no agreed method of establishing this. We propose principles for deciding saturation in theory-based interview studies (where conceptual categories are pre-established by existing theory). First, specify a minimum sample size for initial analysis (initial analysis sample). Second, specify how many more interviews will be conducted without new ideas emerging (stopping criterion). We demonstrate these principles in two studies, based on the theory of planned behav

Unbiased distance correlation with sample-size-aware confidence bounds for comparative omics network analysis.

INTRODUCTION: Data-driven determination is a powerful approach for unbiased investigation of the functional relationships in biomolecular networks. Such networks can be inferred from omics data, where correlation analysis is a commonly used method. However, the correlation values depend strongly on sample variability and size of the sample set in the general case, leading to unstable results and possibly highly erroneous conclusions. METHODS: In this work, we show that similar to the Pearson and Spearman correlation approaches, distance correlation as a general non-linear polytonic correlatio
