Thyroid hormones and tendon: current views and future perspectives. Concise review.

Thyroid hormones (THs) T3 and T4, play an essential role in the development and metabolism of many tissues and organs, and have profound metabolic effects in adult life. THs action is mediated mainly by the thyroid hormone receptor (TRs) which seem to be ubiquitous. To-date thyroid-associated disease are not thought to be related in tendinopathies and tendons tears. Recent study demonstrated the presence of TRs in tendons and their possible role in the proliferation and apoptosis of human tenocyte isolated from tendon. We review new discovery that revisit our current thinking on the tendon bio

Ginsenoside Rg1 enhances the healing of injured tendon in achilles tendinitis through the activation of IGF1R signaling mediated by oestrogen receptor.

BACKGROUND: During the pathogenesis of tendinopathy, the chronic inflammation caused by the injury and apoptosis leads to the generation of scars. Ginsenoside Rg1 (Rg1) is extracted from ginseng and has anti-inflammatory effects. Rg1 is a unique phytoestrogen that can activate the estrogen response element. This research aimed to explore whether Rg1 can function in the process of tendon repair through the estrogen receptor. METHODS: In this research, the effects of Rg1 were evaluated in tenocytes and in a rat model of Achilles tendinitis (AT). Protein levels were shown by western blotting. qR

Bone Marrow Mesenchymal Stem Cells Rescue Tendon Injury by Regulating FTO-Mediated m6A Methylation of ELOB.

Bone marrow mesenchymal stem cells (BMSCs) prominent capacity for tissue repair and regenerative applications due to their multipotent differentiation potential and immunomodulatory activities. This study explores the function of BMSCs in tendon repair, with a particular focus on their regulatory effect on fat mass and obesity-associated protein (FTO)-mediated m6A methylation of Elongin B (ELOB). In this study, BMSCs were isolated from mice with experimental tendon injury and characterized by detecting the expression of surface biomarkers (CD90, CD44, CD29, CD34, CD45). Farrerol alleviates collagenase-induced tendinopathy by inhibiting ferroptosis in rats.

Tendinopathy is mainly characterized by local pain, functional limitation and decreased athletic ability, which seriously affects the quality of life of patients and the career of athletes. Farrerol (FA), one of the main active compounds extracted from Rhododendron and plants in the Rhododendron family, has a wide range of pharmacological activities, such as immunomodulatory, anti-inflammatory and antiviral effects. However, the effect of FA on tendinopathy is unclear. Here, we investigated the pharmacological effect and mechanism of FA in tendon injury through collagenase-induced tendinopathy

Characterization of tendon cell cultures of the human rotator cuff.

Rotator cuff tears are common soft tissue injuries of the musculoskeletal system that heal by formation of repair tissue and may lead to high retear rates and joint dysfunction. In particular, tissue from chronic, large tendon tears is of such degenerative nature that it may be prone to retear after surgical repair. Besides several biomechanical approaches, biologically based strategies such as application of growth factors may be promising for increasing cell activity and production of extracellular tendon matrix at the tendon-to-bone unit. As a precondition for subsequent experimental growth

Spatial and temporal expression of molecular markers and cell signals during normal development of the mouse patellar tendon.

Tendon injuries are common clinical problems and are difficult to treat. In particular, the tendon-to-bone insertion site, once damaged, does not regenerate its complex zonal arrangement. A potential treatment for tendon injuries is to replace injured tendons with bioengineered tendons. However, the bioengineering of tendon will require a detailed understanding of the normal development of tendon, which is currently lacking. Here, we use the mouse patellar tendon as a model to describe the spatial and temporal pattern of expression of molecular markers for tendon differentiation from late feta

Moderate- and High-Speed Treadmill Running Exercise Have Minimal Impact on Rat Achilles Tendon.

Exercise influences clinical Achilles tendon health in humans, but animal models of exercise-related Achilles tendon changes are lacking. Moreover, previous investigations of the effects of treadmill running exercise on rat Achilles tendon demonstrate variable outcomes. Our objective was to assess the functional, structural, cellular, and biomechanical impacts of treadmill running exercise on rat Achilles tendon with sensitive in and ex vivo approaches. Three running levels were assessed over the course of 8 weeks: control (cage activity), moderate-speed (treadmill running at 10 m/min, no incl

Prolyl-Hydroxyproline Promotes Structural Matrix Organization in 3D Equine Tendon Proper and Peritenon Post-Injury Model.

Tendinopathy of the superficial digital flexor tendon affects horse health and performance, with slow healing and high re-injury rates due to the formation of mechanically weaker scar tissue. Tendons are rich in Type I collagen and may benefit from supplementation with collagen-derived dipeptide prolyl hydroxyproline (H-Pro-Hyp-OH) which improves collagen regeneration and reduces scarring in murine injury models. However, its effects on equine tenocytes in the tendon proper (TP) and peritenon (PERI) under stress are unknown. Three-dimensional fibrin-based TP and PERI constructs were used to ev

Thrombospondin-4 controls matrix assembly during development and repair of myotendinous junctions.

Tendons are extracellular matrix (ECM)-rich structures that mediate muscle attachments with the skeleton, but surprisingly little is known about molecular mechanisms of attachment. Individual myofibers and tenocytes in Drosophila interact through integrin (Itg) ligands such as Thrombospondin (Tsp), while vertebrate muscles attach to complex ECM fibrils embedded with tenocytes. We show for the first time that a vertebrate thrombospondin, Tsp4b, is essential for muscle attachment and ECM assembly at myotendinous junctions (MTJs). Tsp4b depletion in zebrafish causes muscle detachment upon contrac

Achilles tendinopathy.

BACKGROUND: Achilles Tendinopathy (AT) is essentially a failed healing response with haphazard proliferation of tenocytes, abnormalities in tenocytes with disruption of collagen fibers, and subsequent increase in non-collagenous matrix. METHODS: The diagnosis of Achilles tendinopathy is clinical, and MRI and utrasound imaging can be useful in differential diagnosis. Conservative manegement, open surgery or minimally invasive techniques are available. Injections and physical therapy are also vauable options. RESULTS: Eccentric exercises are useful tools to manage the pathology. If the conditi

Identification of the tendon/ligament stem cell in mice and humans.

Ligamentum flavum (LF) hypertrophy resulting in spinal cord compression is a key cause of lumbar spinal stenosis (LSS). There are no medical therapies for LSS, in part because the identity of the tendon/ligament stem cell (TLSC) mediating LSS remains unclear. Here, we identify the TLSC present in all tendons and ligaments in humans and mice as Lin-Thy1.2-Sca-1-CD73+CD140α- cells that display self-renewal and give rise to all other tenocyte lineage cells. This TLSC also sits at the apex of their differentiation hierarchy, as determined by in vivo somatic-variant-based lineage tracing. LSS induc

Effects of lidocaine on torn rotator cuff tendons.

We determined lidocaine's action on torn rotator cuff tendons in vitro and in vivo. For in vitro experiments, cell proliferation and viability assays were performed using tenocytes derived from human torn rotator cuff tendons. For in vivo experiments, acute rotator cuff tears were made on the supraspinatus tendons in the rats' bilateral shoulders; before closure, lidocaine was injected into the shoulder and saline into the contralateral shoulder (control). After sacrifice, the specimens underwent biomechanical testing or histological analysis at 24 h and at 2, 4, and 8 weeks after surgery. The

Lovastatin-Mediated Changes in Human Tendon Cells.

Statins are among the most widely prescribed drugs worldwide. Numerous studies have shown their beneficial effects in prevention of cardiovascular disease through cholesterol-lowering and anti-atherosclerotic properties. Although some statin patients may experience muscle-related symptoms, severe side effects of statin therapy are rare, primarily due to extensive first-pass metabolism in the liver. Skeletal muscles appear to be the main site of side effects; however, recently some statin-related adverse effects have been described in tendon. The mechanism behind these side effects remains unkn

Recapitulating the Native Tendon Environment in a Synthetic 3D Anisotropic Hydrogel as an Engineered Extracellular Matrix.

The ability of tendons to transmit forces from muscle to bone is fundamentally attributed to the hierarchical anisotropy of the tissue. After injury, disorganized fibrotic scar tissue forms during the natural healing process, resulting in inferior mechanical properties that often lead to reinjury and limited restoration of function. Therefore, intervention is necessary to facilitate regenerative healing of the tendon. Polymeric biomaterials have historically been used to guide cell behavior, showing promise for the use of topological guidance and cell-mediated matrix remodeling as mechanisms f

Corticosteroid versus platelet-rich plasma injection in epicondylitis.

Lateral epicondylitis (LE) is often considered an inflammatory disease characterized by tendon microtears that are followed by an incomplete reparative response that leads to chronic pain and decreased function on the upper extremity. A modality that is commonly used for the treatment of LE is the injection of corticosteroids. Evidence supports corticosteroid injections to be an effective short-term intervention that lacks intermediate and long-term relief as well as having negative effects on tenocyte proliferation, which is essential in the tissue healing process. Platelet-rich plasma (PRP)

Inhibitory effect of taurine on rotator cuff degeneration via mitochondrial protection.

OBJECTIVES: Degenerative rotator cuff tears do not heal spontaneously, necessitating surgical intervention. This makes prevention crucial, but effective prophylactic measures are currently lacking. Oxidative stress has recently been implicated as a cause of degenerative rotator cuff tears, while mitochondrial injury has been reported in the development of age-related rotator cuff degeneration. Taurine, which has antioxidant properties, has been found to be effective in the treatment of various mitochondrial abnormalities. This prompted us to investigate the inhibitory effect of taurine and som

[Fine structural changes in degenerative tendinopathy].

50 tendons were examined by electron microscopy. Although they did not demonstrate any signs of inflammation or lipomatosis marked hypoxic changes in the tenocytes were evident on ultrastructural examination. The most frequent alterations were longitudinal splitting, disintegration, angulation and the so-called "kink deformation" of the collagen fibers.

Ruthenium Oxide Nanozyme for Tendinopathy Treatment via Countering Ferroptosis.

The global prevalence of tendinopathy (TP) is steadily increasing, resulting in functional impairments in tendons across individuals of all ages. Excessive accumulation of reactive oxygen species (ROS) plays a pivotal role in the development of TP, which compromises tendon repair and integrity through oxidative stress. This process is often accompanied by ferroptosis-a newly recognized form of iron-dependent programmed cell death. This study explores ruthenium oxide (RuO2) hollow nanospheres as an effective nanozyme to address oxidative damage and ferroptosis in type I collagenase-induced TP.

Therapeutic Efficacy of Intratendinous Delivery of Dexamethasone Using Porous Microspheres for Amelioration of Inflammation and Tendon Degeneration on Achilles Tendinitis in Rats.

Achilles tendinitis caused by overuse, aging, or gradual wear induces pain, swelling, and stiffness of Achilles tendon and leads to tendon rupture. This study was performed to investigate the suppression of inflammation responses in interleukin-1β- (IL-1β-) stimulated tenocytes in vitro and the suppression of the progression of Achilles tendinitis-induced rat models in vivo using dexamethasone-containing porous microspheres (DEX/PMSs) for a sustained intratendinous DEX delivery. Cellular response of healing tissue to DegraPol tube implantation in rabbit Achilles tendon rupture repair: an in vivo histomorphometric study.

In tendon rupture repair, improvements such as higher primary repair strength, anti-adhesion and accelerated healing are needed. We developed a potential carrier system of an electrospun DegraPol tube, which was tightly implanted around a transected and conventionally sutured rabbit Achilles tendon. Histomorphometric analysis of the tendon tissue 12 weeks postoperation showed that the tenocyte density, tenocyte morphology and number of inflammation zones were statistically equivalent, whether or not DegraPol tube was implanted; only the collagen fibres were slightly less parallelly orientated
