The association between fatty acid index and in vitro fertilization outcomes.

PURPOSE: Fatty acids have been shown to play an important role in oocyte competence and early implantation of the embryo. Our hypothesis-generating study sought to determine if individual fatty acids expressed as a percentage of total erythrocyte fatty acids are associated with embryo quality and other in vitro fertilization (IVF) outcomes. METHODS: This was a prospective cohort study at an academic fertility center. Sixty women undergoing their first IVF cycle were recruited. Serum measurements of 22 fatty acids were obtained. We calculated each fatty acid as a percentage of total fatty acid

Trans-arachidonic acids: new mediators of nitro-oxidative stress.

A reaction of arachidonic acid with the nitrogen dioxide radical (*NO2) or its precursors (peroxynitrite, nitrous acid, nitrogen trioxide) generates a group of nitro lipids named nitroeicosanoids. A distinct feature of this reaction is abundant formation of four trans isomers of arachidonic acid (TAA) via reversible addition of the NO2 radical to the arachidonic acid cis double bonds. This cis-trans isomerization is biologically relevant because many pathologies that involve NO formation such as inflammation, hyperoxia, hypercapnia or exposure to cigarette smoke increase the TAA levels in cell

Incorporation of dietary linoleic and conjugated linoleic acids and related effects on eggs of laying hens.

In the present study, laying hens received 29 g per kg diet of a preparation containing either 70% linoleic acid (LA) or approximately the same amount of conjugated linoleic acid (CLA) in the control and experimental treatments, respectively. The CLA preparation consisted predominantly of cis-9,trans-11 and trans-10,cis-12 fatty acid isomers as free fatty acids in a ratio of 1:1. The diets were fed for 8 wk to determine the effect of dietary CLA on quality characteristics of eggs. In addition, the fatty acid composition of liver and heart was analyzed. Performance parameters (egg weight, feed

Catabolism and Accumulation of Dietary cis- and trans-Epoxy Octadecanoic Acids in Mice.

In vivo metabolism of epoxy fatty acids, the unsaturated fatty acid oxidation products formed upon heating of edible oils, remains unclear. Here, we compared the catabolic rates and accumulation of dietary cis- and trans-epoxy octadecanoic acids (epoxy-C18:0) with those of oleic acid (cis-C18:1) in mice using stable isotope-labeled compounds. Both epoxy isomers were catabolized more rapidly than cis-C18:1, with trans-epoxy-C18:0 exhibiting a slightly faster rate than cis-epoxy-C18:0. The concentrations of cis- and trans-epoxy-C18:0 were significantly lower than those of cis-C18:1 in the plasma

Fatty Acids and Their Roles in Cardiac Physiology and Pathology: Mechanistic and Interventional Studies.

Fatty acids serve dual roles in cardiac physiology: as energy substrates and as precursors of bioactive lipid mediators (prostaglandins, leukotrienes, oxylipins) from n-3/n-6 PUFAs that regulate inflammation, thrombosis, and remodeling. Saturated, monounsaturated, and trans fatty acids modulate metabolism and membrane function, thereby shaping these pathways. Clinically, n-3 long-chain PUFAs (EPA and DHA) reduce cardiovascular mortality and aid postischemic remodeling; however, high doses increase the risk of atrial fibrillation. By contrast, trans and saturated fatty acids promote dyslipidemi

Fatty acid profile, vitamin D, and iodine status in Taiwanese vegans, vegetarians, and omnivores.

OBJECTIVES: Vegan diet - the exclusion of all animal products, even eggs and dairy - ranked the most climate-friendly diet, but concerns persist regarding its nutritional adequacy, particularly for omega-3 fatty acids, vitamin D, and iodine. This study compares the nutritional status of fatty acids, vitamin D, and iodine, among Taiwanese vegans, vegetarians, and omnivores. MATERIALS AND METHODS: This cross-sectional study included 45 omnivores, 46 lacto-ovo vegetarians, and 24 vegans. Blood and urine samples were collected to analyze nutritional biomarkers, including erythrocyte fatty acid pr

EPA and DHA acylcarnitines are less cardiotoxic than are saturated and monounsaturated long-chain acylcarnitines.

Elevated levels of fatty acid-derived long-chain acylcarnitines are detrimental to cardiac health, primarily because of their adverse effects on mitochondrial function and key metabolic pathways in the heart. While trans-fatty acids are considered harmful and omega-3 polyunsaturated fatty acids (PUFAs) are considered beneficial, the specific properties of acylcarnitines derived from these types of fatty acids are not characterized. This study aimed to compare the effects of saturated palmitoylcarnitine (PC), monounsaturated cis-oleoylcarnitine (cis-OC), trans-elaidoylcarnitine (trans-EC), and

Circulating plasma fatty acids and risk of pancreatic cancer: Results from the Golestan Cohort Study.

BACKGROUND & AIMS: Pancreatic cancer (PC) with a dismal prognosis is considered as a fatal malignancy, attracting the scientists' attention to study its causes and pathogenesis pathways. Given the lack of enough evidence and conflicting findings about the association of PC risk with plasma fatty acids, we aimed to explore the associations of circulating plasma fatty acids with the risk of PC in a cohort study. METHODS: From about 50,000 subjects participated in this cohort study in 2004-2008, fifty incident cases of PC were recruited and 150 controls matched by age, sex and residence place (u

alpha-Linolenic acid intake is not beneficially associated with 10-y risk of coronary artery disease incidence: the Zutphen Elderly Study.

BACKGROUND: Data on the relation between alpha-linolenic acid intake and coronary artery disease (CAD) are limited. Other dietary components appear to modify the reported relation between alpha-linolenic acid intake and CAD. OBJECTIVE: We examined whether dietary alpha-linolenic acid intake was inversely associated with risk of CAD. DESIGN: We prospectively studied 667 men aged 64-84 y from the Zutphen Elderly Study who were free of CAD at baseline. Dietary intake was assessed by using a cross-check dietary history method. RESULTS: During the 10-y follow-up, we documented 98 cases of CAD. A

Regulation of stearoyl-CoA desaturase activity by the trans-10,cis-12 isomer of conjugated linoleic acid in HepG2 cells.

Stearoyl-CoA desaturase (SCD) catalyzes the rate-limiting step in the cellular synthesis of monounsaturated fatty acids mainly oleate (C18:1) and palmitoleate (C16:1) which are the major monounsaturated fatty acids of membrane phospholipids, cholesterol esters, waxes, and triglycerides. Several SCD isoforms exist in the mouse whereas the human has one well-characterized SCD gene. The trans-10,cis-12 isomer of conjugated linoleic acid has been previously shown to repress the expression of the mouse SCD1 gene isomer by decreasing SCD gene expression as well as by direct inhibition of SCD enzyme

A commonly used rumen-protected conjugated linoleic acid supplement marginally affects fatty acid distribution of body tissues and gene expression of mammary gland in heifers during early lactation.

BACKGROUND: Conjugated linoleic acids (CLA) in general, and in particular the trans-10,cis-12 (t10,c12-CLA) isomer are potent modulators of milk fat synthesis in dairy cows. Studies in rodents, such as mice, have revealed that t10,c12-CLA is responsible for hepatic lipodystrophy and decreased adipose tissue with subsequent changes in the fatty acid distribution. The present study aimed to investigate the fatty acid distribution of lipids in several body tissues compared to their distribution in milk fat in early lactating cows in response to CLA treatment. Effects in mammary gland are further

Evaluation of the non-catalytic binding function of Ts26GST a glutathione transferase isoform of Taenia solium.

Taenia solium glutathione transferase isoform of 26.5 kDa (Ts26GST) was observed to bind non-catalytically to porphyrins, trans-trans-dienals, bile acids and fatty acids, as assessed by inhibition kinetics, fluorescence spectroscopy and competitive fluorescence assays with 8-anilino-1-naphthalene sulfonate (ANS). The quenching of Ts26GST intrinsic fluorescence allowed for the determination of the dissociation constants (KD) for all ligands. Obtained data indicate that Ts26GST binds to all ligands but with different affinity. Porphyrins and lipid peroxide products inhibited Ts26GST catalytic ac

Prolonged feeding of mice with conjugated linoleic acid increases hepatic fatty acid synthesis relative to oxidation.

Feeding mice conjugated linoleic acid (9 cis,11 trans/9 trans,11 cis-and 10 trans,12 cis-CLA in equal amounts) resulted in triacylglycerol accumulation in the liver. The objective of this study was to examine whether this steatosis is associated with changes in hepatic fatty acid synthesis and oxidation. Therefore, we measured the activities of key enzymes of fatty acid synthesis, i.e., acetyl-CoA carboxylase and fatty acid synthase and of fatty acid oxidation, i.e., 3-hydroxy-acyl-CoA dehydrogenase and citrate synthase in livers of mice fed a diet with 0.5% (w/w) CLA. Location of double bonds in monounsaturated fatty acids of Campylobacter cryaerophila with dimethyl disulfide derivatives and combined gas chromatography-mass spectrometry.

Location of the double-bond position of monounsaturated fatty acids in Campylobacter cryaerophila was accomplished with combined gas chromatography-mass spectrometry analysis of dimethyl disulfide (DMDS) derivatives. The monoenoic fatty acids from whole bacterial cells were converted to methyl esters and then to DMDS adducts and analyzed by capillary gas chromatography-mass spectrometry. The mass spectra of DMDS adducts gave an easily recognizable molecular ion (M+) and two major diagnostic ions attributable to fragmentation between the two CH3S groups located at the original site of unsaturat
