SIRT1, a Class III Histone Deacetylase, Regulates LPS-Induced Inflammation in Human Keratinocytes and Mediates the Anti-Inflammatory Effects of Hinokitiol.

Skin inflammation is a response of the immune system to infection and injury. In this study, we report that hinokitiol, a tropolone-related natural compound that exhibits antioxidant, anti-inflammatory, and anticancer properties in various cell types, can modulate the inflammatory responses of primary human keratinocytes challenged with lipopolysaccharide (LPS). Hinokitiol treatment inhibited LPS-mediated up-regulation of proinflammatory factors including tumor necrosis factor alpha, IL-6, and prostaglandin E2 (PGE2). NF-κB activation and cell migration induced by LPS were blocked in keratinoc

Studies on the antifungal effects of Hinokitiol on Candida albicans: inhibition of germ tube formation and synergistic pharmacological effects of miconazole.

One of the goals of oral healthcare management is to manage dry mouth. Thus, moisturizers containing antimicrobial ingredients, such as hinokitiol (HT), are applied to the oral mucosa after oral care. In this study, we investigated the preventive effect of HT against the growth of Candida albicans (C. al) and its synergistic effect when combined with miconazole (MCZ), an oral treatment for candidiasis. As the concentration of HT increased, the length and percentage of germ tubes (GT) decreased. Larger inhibition circles were observed for MCZ concentrations of 2.0 and 4.0 μg/disc compared to th

CRYSTAL STRUCTURES OF TROPONOID LIQUID-CRYSTALLINE COMPOUNDS: 5-CYANO-2-(4-OCTYLOXYBENZOYLAMINO)-TROPONE AND BIS(5-DODECYLTROPOLONATO)COPPER(II).

Crystal structures of 5‑cyano‑2‑(4‑octyloxybenzoylamino)tropone (1) and bis(5‑dodecyltropolonato)copper(II) (2) were elucidated by single‑crystal X‑ray analysis. The cya-notropone derivative 1 forms centrosymmetric dimers through intermolecular C-H···N interac-tions, and its 2‑(benzoylamino)tropone core adopts an almost planar conformation. Molecules of 1 assemble into a tilted layered structure via C-H···N, C-H···O, and π-π interactions. Complex 2 forms a 2:1 tropolonato-Cu(II) coordination unit with a tetragonally planar geometry, in which the Cu(II) ion lies on an inversion center and is co

Species-specific isotope dilution analysis of mono-, di, and tri-butyltin compounds in sediment using gas chromatography-inductively coupled plasma mass spectrometry with synthesized 118Sn-enriched butyltins.

A species-specific isotope dilution (ID) method is described for the determination of mono-, di, and tri-butyltin compounds in sediment by gas chromatography-inductively coupled plasma mass spectrometry (GC-ICP-MS), where the mixture of 118Sn-enriched butyltin compounds synthesized in our laboratory was used as a spike. A correction method for the mass bias, a quantitative extraction of the butyltins from sediment, and an assay for the concentration of the standard solution for the reverse ID procedure were investigated to achieve a reliable ID analysis. The spike solution was added with tri-p

Self-Assembled Metal-Polyphenol Colchicine Nanoparticles Targeting Oxidative Stress and Inflammation for Treatment of Atherosclerosis.

Anti-inflammatory colchicine therapy has emerged as a new era for atherosclerotic cardiovascular diseases. However, the therapeutic benefit of colchicine has not been clearly defined. Herein, we present a double coordination-driven approach to fabricate a stable metal-organic nano-assembly of colchicine (COL-TA-Zn) by uniting the tropolone ring of colchicine (COL), phenolic groups of tannic acid (TA), and Zn2+ ions. This design leverages the antioxidant and anti-inflammatory properties of COL and TA to create a nanoscale platform capable of scavenging radicals and modulating inflammatory pathw

Zinc thiotropolone combinations as inhibitors of the SARS-CoV-2 main protease.

Numerous organic molecules are known to inhibit the main protease of SARS-CoV-2, (SC2Mpro), a key component in viral replication of the 2019 novel coronavirus. We explore the hypothesis that zinc ions, long used as a medicinal supplement and known to support immune function, bind to the SC2Mpro enzyme in combination with lipophilic tropolone and thiotropolone ligands, L, block substrate docking, and inhibit function. This study combines synthetic inorganic chemistry, in vitro protease activity assays, and computational modeling. While the ligands themselves have half maximal inhibition concent

Eucalyptus globulus Bark Extract and Hinokitiol in Hybrid Anti-Sapstain Formulations with Reduced Chlorothalonil, Carbendazim and Copper-8-quinolinolate Loads.

The general objective of this study was to evaluate Eucalyptus globulus bark extract as a candidate component of wood preservatives with a reduced load of synthetic biocides. Extraction conditions were screened using a 23 full factorial design with a center point, identifying 75 °C, 60% ethanol and 1 h as the highest-yielding conditions (5.94%) within the tested ranges. In four-week laboratory wood-block assays, the extract alone did not exceed 40% inhibition of wood-staining fungi, whereas hinokitiol reached complete inhibition at 0.4%. Combining the extract (5%) with hinokitiol (0.4%) achiev

Interactions of hinokitiol with fungal membrane lipids in model systems.

Hinokitiol (β-thujaplicin) is a naturally occurring substance of antimicrobial properties, which can be used e.g. as a cosmetic preservative. In this work the influence of hinokitiol on the monolayers and bilayers formed from fungal membrane lipids (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine - POPE; 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine - POPC and ergosterol) was investigated. These studies aimed to investigate the effect of hinokitiol on fungal membranes, being indicated as a target for this compound. In this context, the affinity of hinokitiol for ergosterol-containing

Transmission electron microscopy of Schistosoma mansoni cercariae treated with hinokitiol (beta-thujaplicin), a compound for potential skin application against cercarial penetration.

Since skin is the only route of entry of the parasite in schistosomiasis patients, intervention at the level of skin penetration should control the infection. Several compounds were screened for their ability to protect against cercarial penetration. Hinokitiol (beta-thujaplicin) was found to have a significant cercaricidal effect in vitro, although there is no information on its cercaricidal mechanisms. To study the kinetics of morphological changes in Schistosoma mansoni associated with exposure to hinokitiol in vitro, cercariae were incubated in media containing hinokitiol at different conc

Anti-obesity carbonic anhydrase inhibitors: challenges and opportunities.

The mitochondrial isoforms VA/VB of metalloenzyme carbonic anhydrase (CA, EC 4.2.1.1) are involved in metabolic processes, such as de novo lipogenesis and fatty acid biosynthesis. We review the drug design landscape for obtaining CA VA/VB-selective/effective inhibitors, starting from the clinical observations that CA inhibitory drugs, such as the antiepileptics topiramate and zonisamide, or the diuretic acetazolamide induce a significant weight loss. The main approaches for designing such compounds consisted in drug repurposing of already known CA inhibitors (CAIs); screening of synthetic/natu

[Technic and clinical use of radioactive labelling of autologous granulocytes].

Gamma-camera imaging after injection of radiolabelled autologous leucocytes can be very helpful in the diagnosis, localization and further clinical treatment of inflammatory diseases. We present a technique allowing sterile separation of white blood cells and labelling with 99mTc-phytate or -oxine and with 111In-oxine, -oxine sulphate or -tropolone. The method is non-invasive and the radiation dose amounts to less than 80 mrad using 100 microCi 111Indium. The use of radiolabelled granulocytes is of particular diagnostic value in patients with septicaemia of unknown origin. Whole body scanning

Re-examining the iron paradox: the influence of growth medium on regulation and functionality of the tropodithietic acid (TDA) biosynthetic pathway in Phaeobacter piscinae.

Tropodithietic acid (TDA) is a secondary metabolite with antimicrobial and iron-binding properties, produced by several marine Roseobacter group bacteria. Although classified as a siderophore, TDA is also produced under iron-replete conditions. To address this paradox, we fused the tdaCDE promoter region of Phaeobacter piscinae S26 with a promoter-less gfp reporter gene to monitor the transcription of TDA biosynthetic genes under different conditions and validated its accuracy using RT-qPCR. In both complex ($\frac{1}{2}$YTIO) and low-complexity (IOCGH) marine media, iron supplementation repre

Minimizing higher-order aggregation maximizes iron mobilization by small molecules.

The natural product hinokitiol mobilizes iron across lipid bilayers at low concentrations and restores hemoglobinization in iron transporter protein-deficient systems. But hinokitiol fails to similarly mobilize iron at higher concentrations, limiting its uses in chemical biology and medicine. Here we show that at higher concentrations, hinokitiol3:Fe(III) complexes form large, higher-order aggregates, leading to loss of transmembrane iron mobilization. Guided by this understanding and systematic structure-function studies enabled by modular synthesis, we identified FeM-1269, which minimally ag

Discovery of potential components characteristic by conjugated enone from the branches and leaves of Croton lauioides with anti-neuroinflammatory activity via regulating the NF-κB pathway.

In this study, the chemical composition and pharmacological activity of Croton lauioides were investigated for the first time. The bioactive and HPLC-UV guided isolation led to the discovery of twenty-three conjugated enone-type components (1-23), including nine previously unknown sesquiterpenoid derivatives (1-4, 9-10, 12-14). Notably, compounds 1 and 12 are epoxides containing an endoperoxide bridge (1) or a unique dioxaspiro core (12), respectively. Compounds 2-7 are non-benzenoid aromatics featuring a tropone function, while 9-11 possess a rare rearranged scaffold with tropone shift into b

Molecular mechanism of the unusual biphasic effects of the natural compound hinokitiol on iron-induced cellular DNA damage.

Hinokitiol is a natural monoterpene compound found in the heartwood of cupressaceous plants that have anticancer and anti-inflammatory properties. However, few studies have focused on its effect on iron-mediated cellular DNA damage. Here we show that hinokitiol exhibited unusual biphasic effects on iron-induced DNA damage in a molar ratio (hinokitiol/iron) dependent manner in HeLa cells. Under low ratios (<3:1), hinokitiol markedly enhanced DNA damage induced by Fe(II) or Fe(II)-H2O2; However, when the ratios increased over 3:1, the DNA damage was progressively inhibited. We found that the tot

Inhibition of the ANT(2")-Ia resistance enzyme and rescue of aminoglycoside antibiotic activity by synthetic α-hydroxytropolones.

Aminoglycoside-2"-O-nucleotidyltransferase ANT(2")-Ia is an aminoglycoside resistance enzyme prevalent among Gram-negative bacteria, and is one of the most common determinants of enzyme-dependant aminoglycoside-resistance. The following report outlines the use of our recently described oxidopyrylium cycloaddition/ring-opening strategy in the synthesis and profiling of a library of synthetic α-hydroxytropolones against ANT(2")-Ia. In addition, we show that two of these synthetic constructs are capable of rescuing gentamicin activity against ANT-(2")-Ia-expressing bacteria.

Optimizing Analog Identification and Evaluation in the OECD QSAR Toolbox to Enable Robust Read-Across.

Read-across is one of the most widely used New Approach Methods (NAMs) for evaluating the safety of a chemical with insufficient toxicological data. However, the identification of analogs suitable for read-across is not always straightforward as it requires understanding of critical chemical and biological features that drive toxicity. The OECD QSAR Toolbox is recommended as a valuable tool to support analog identification. Here, a case study is presented on Benzenesulfonic acid, comparing two different analog identification approaches within the Toolbox. The suitability of analogs identified

Gas chromatographic determination of inorganic tin in rat urine after a single oral administration of stannous chloride and mono-, di-, and triphenyltin chloride.

A method is described for the determination of inorganic tin by gas chromatography with flame photometric detection. The inorganic tins, stannous and stannic, were extracted with hydrochloric acid and n-hexane-benzene in the presence of 0.05% tropolone, and both inorganic tins were pentylated to tetrapentyltin with a Grignard reagent prior to gas chromatography. The absolute limit of detection for tetrapentyltin was 3 pg as tin. The recovery of stannous chloride added to rat urine samples was 80.2 +/- 2.4% (mean +/- S.D., n = 8). The application of this method to the study of urinary excretion
