Development and Validation of the AI-HeartAge Model in Framingham and UK Biobank.

BACKGROUND: Arterial pressure waveform shape conveys information regarding interactions between the left ventricle and aorta that could provide an estimate of biological heart age and cardiovascular disease (CVD) risk. METHODS: Artificial intelligence heart age (AI-HA) was estimated by averaging results from 2 convolutional neural networks trained to predict mitral annulus tissue Doppler e' and s' peak velocities using an uncalibrated arterial tonometry or photoplethysmography waveform as input. Models were developed using FHS (Framingham Heart Study) participant pressure waveforms and echoca

Sex differences in the experience of pain in the UK Biobank cohort study.

OBJECTIVE: Chronic pain has been shown to be more prevalent among women than men. However, each person's experience of pain is shaped by a complex interplay of biological, psychological and social factors. The aim of this study was to summarise a comprehensive 'experience of pain' questionnaire in the UK Biobank and identify differences in the experience of chronic pain between females and males. METHODS: This was an exploratory analysis of an online self-assessment questionnaire consisting of 128 questions related to UK Biobank participant's experience of pain that was administered in 2019.

Prevalent Stroke, Fatigue, and Incident Dementia Risk in the UK Biobank.

BACKGROUND: Stroke is a major risk factor for dementia, and fatigue is common among stroke survivors. Whether self-reported fatigue is associated with dementia risk among individuals with prevalent stroke remains unclear. METHODS: We conducted a prospective cohort study of 484,749 UK Biobank participants without dementia at baseline. Participants were classified into four groups according to prevalent stroke and self-reported fatigue: neither exposure, fatigue only, prevalent stroke only, and prevalent stroke with fatigue. The primary outcome was incident all-cause dementia. Secondary outcome

Photoreceptor thickness in UK Biobank participants with and without diabetes mellitus.

OBJECTIVE: Photoreceptors can be impacted early in diabetes mellitus (DM). The primary goal of this study was to determine if having DM and no/mild diabetic retinopathy (DR) affected photoreceptor layer thickness. METHODS AND ANALYSIS: Participants from the UK Biobank database who underwent fundus photographs and macula-centred spectral domain-optical coherence tomography were considered for inclusion in the study. Multivariable linear regression models were used to compare photoreceptor thickness between participants with DM (no/mild DR) and without DM after adjusting for confounders. Second

Interactions with polygenic background impact quantitative traits in the UK Biobank.

Association studies have linked many genetic variants to a variety of phenotypes but understanding the biological mechanisms underlying these signals remains a major challenge. Since genes operate within complex networks, statistical interactions between genetic mutations that reflect biological pathways are expected to exist. However, their discovery has been hampered by the vast search space of variant combinations and the multiplicatively small expected effect sizes of interactions. To increase power, we created a test for interaction between single-nucleotide polymorphisms (SNPs) and group

Loneliness and Risk of Incident Hearing Loss: The UK Biobank Study.

Background: Hearing loss (HL) is one major cause of disability and can lead to social impairments. However, the relationship between loneliness and the risk of incident HL remains unclear. Our study aimed to investigate this association among adults in the UK. Methods: This cohort study was based on data from the UK Biobank study. Loneliness was assessed by asking participants if they often felt lonely. Incident HL was defined as a primary diagnosis, ascertained via linkage to electronic health records. Cox proportional hazard regression models were used to examine the association between lone

Social isolation and the risk of Parkinson disease in the UK biobank study.

Parkinson disease (PD) has become one of the most rapidly growing causes of disability among the older population and social isolation is a major concern in the PD community. However, the relationship between social isolation and future risk of PD remains unclear. This study included 192,340 participants aged 60 or older who were free of dementia and PD at baseline from the UK Biobank study. Social isolation was measured using a composite score derived from three questions on number in household, frequency of friend/family visits, and leisure/social activities. Incident PD cases were identifie

An Integrative Pancreatic Cancer Risk Prediction Model in the UK Biobank.

Pancreatic cancer (PaCa) is a lethal cancer with an increasing incidence, highlighting the need for early prevention strategies. There is a lack of a comprehensive PaCa predictive model derived from large prospective cohorts. Therefore, we have developed an integrated PaCa risk prediction model for PaCa using data from the UK Biobank, incorporating lifestyle-related, genetic-related, and medical history-related variables for application in healthcare settings. We used a machine learning-based random forest approach and a traditional multivariable logistic regression method to develop a PaCa pr

Association of HLA diversity with the risk of 25 cancers in the UK Biobank.

BACKGROUND: The human leukocyte antigen (HLA) is a highly polymorphic region, and HLA diversity may play a role in presenting tumour-associated peptides and inducing immune responses. However, the effect of HLA diversity on cancers has not been fully assessed. We aimed to explore the role of HLA diversity on cancer development. METHODS: A pan-cancer analysis was performed to evaluate the effect of HLA diversity, measured by HLA heterozygosity and HLA evolutionary divergence (HED), on the susceptibility of 25 cancers in the UK Biobank. FINDINGS: We observed that the diversity of HLA class II

Genome-by-Trauma Exposure Interactions in Adults With Depression in the UK Biobank.

IMPORTANCE: Self-reported trauma exposure has consistently been found to be a risk factor for major depressive disorder (MDD), and several studies have reported interactions with genetic liability. To date, most studies have examined gene-environment interactions with trauma exposure using genome-wide variants (single-nucleotide variations [SNVs]) or polygenic scores, both typically capturing less than 3% of phenotypic risk variance. OBJECTIVE: To reexamine genome-by-trauma interaction associations using genetic measures using all available genotyped data and thus, maximizing accounted varian

Exposure to air pollution and risk of incident dementia in the UK Biobank.

BACKGROUND: Air pollution may cause inflammatory and oxidative stress damage to the brain, leading to neurodegenerative disease. The association between air pollution and dementia, and modification by apolipoprotein E genotype 4 (APOE-ε4) has yet to be fully investigated. OBJECTIVES: To examine associations of air pollution with three types of incident dementias (Alzheimer's disease (AD), frontotemporal dementia (FTD), and vascular dementia (VAD)), and their potential modification by APOE-ε4 genotype. METHODS: The UK Biobank enrolled >500,000 participants (2006-2010) with ongoing follow-up.

The genetic and environmental hierarchical structure of anxiety and depression in the UK Biobank.

BACKGROUND: Anxiety and depressive disorders can be classified under a bidimensional model, where depression and generalized anxiety disorder are represented by distress and the other anxiety disorders, by fear. The phenotypic structure of this model has been validated, but twin studies only show partial evidence for genetic and environmental distinctions between distress and fear. Moreover, the effects of genetic variants are mostly shared between anxiety and depression, but the genome-wide genetic distinction between distress and fear remains unexplored. This study aimed to examine the degre

Phenome-wide Burden of Copy-Number Variation in the UK Biobank.

Copy-number variations (CNVs) represent a significant proportion of the genetic differences between individuals and many CNVs associate causally with syndromic disease and clinical outcomes. Here, we characterize the landscape of copy-number variation and their phenome-wide effects in a sample of 472,228 array-genotyped individuals from the UK Biobank. In addition to population-level selection effects against genic loci conferring high mortality, we describe genetic burden from potentially pathogenic and previously uncharacterized CNV loci across more than 3,000 quantitative and dichotomous tr

Association between cardiorespiratory fitness and colorectal cancer in the UK Biobank.

Increased cardiorespiratory fitness is related to decreased risk of major chronic illnesses, including cardiovascular disease, type 2 diabetes, and cancer, but its association with colorectal cancer specifically has received very little attention. We examined the relation of cardiorespiratory fitness to colorectal cancer in 59,191 UK Biobank participants aged 39-70 years without prevalent cancer at baseline, followed from 2009 to 2014. Submaximal bicycle ergometry was conducted at study entry, and cardiorespiratory fitness was defined as physical work capacity at 75% of the maximum heart rate,

A phenome-wide structural variant map from 467,152 UK Biobank genomes.

Structural variants (SVs) are large (>50 base pairs) genomic rearrangements that can significantly influence human phenotypes, yet their contribution to health and disease remains undercharacterized. We conducted a phenome-wide association study (PheWAS) of 1.6 million SVs identified in whole-genome sequencing data from 467,152 UK Biobank participants, across 13,322 binary, 2,018 quantitative, and 2,923 plasma protein traits. Of these, 82.25% of SVs were outside the detectable range of prior copy-number variant studies. Protein quantitative trait loci analysis linked 3,467 SVs to 1,690 protein
