Antibacterial mechanism of octyl gallate against methicillin-resistant staphylococcus aureus and its application in pork preservation: insights from multi-omics and computational simulation.

The widespread emergence of methicillin-resistant Staphylococcus aureus (MRSA) in the food chain, particularly in pork, has become an urgent public health and food safety challenge. However, safe and effective food-grade preservatives capable of both eliminating MRSA and elucidating clear antibacterial mechanisms remain limited. Herein, the food-grade additive octyl gallate (OG) exhibited remarkable activity against MRSA, with a minimum inhibitory concentration (MIC) of only 20 μg/mL and completely eradicating MRSA at 40 μg/mL within 2 h, demonstrating potent efficacy comparable to vancomycin.

Genetic determinants of disinfectant and antibiotic resistance, and virulence factors in Enterococcus faecalis isolated from a pasteurized crab canning environment.

Blue swimming crab (Portunus pelagicus) is highly commercial Indonesian seafood commodity and exported in the form of pasteurized canned crab product. As a ready-to-eat food industry, the effectiveness of sanitization practices and the level of hygiene in the production environment are critical factors in ensuring food safety. These challenges are heightened by bacteria that form biofilms or carry resistance genes. This study aimed to identify resistance bacterial species and strain, identify disinfectants and antibiotic resistance genes, and virulence genes in the pasteurized crab production

Prevalence of quorum sensing luxS gene in multidrug resistant uropathogens.

BACKGROUND: The increasing prevalence of antimicrobial resistance (AMR) among urinary tract infections (UTIs) is making clinical management challenging. Biofilm formation is a key mechanism contributing to antibiotic resistance in uropathogens. Quorum sensing is known to regulate several bacterial processes like bioluminescence, virulence, and biofilm formation which is known to play a role in the development of drug resistance. Out of three major quorum sensing systems, luxS/AI-2 is a universal system used by both Gram-positive and Gram-negative bacteria. METHODS: Community-acquired AMR UTI

Targeting cyclic di-AMP signaling through diadenylate cyclase inhibition reduces methicillin resistance in clinical MRSA isolates.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) remains a major clinical challenge due to persistent β-lactam resistance and biofilm-associated tolerance. While canonical resistance mechanisms are well defined, non-canonical pathways such as cyclic di-adenosine monophosphate (c-di-AMP) signaling remain poorly understood, and no effective inhibitors have yet been shown to reduce MRSA resistance. METHODS: Clinical MRSA isolates were analyzed for c-di-AMP levels, gene expression, biofilm formation, and host immune responses under sub-inhibitory methicillin exposure. Structure-base

KpFhaB/FhaC is a virulence-associated TPS system of the globally-disseminated Klebsiella pneumoniae ST15 high-risk clone.

INTRODUCTION: The global dissemination of Klebsiella pneumoniae sequence type 15 (ST15), a Q9 high-risk clone frequently resistant to carbapenems and third-generation cephalosporins, represents a major public health concern. While chaperone-usher pili (CUP) systems (i.e. Kpi system) are known mediators of K. pneumoniae adhesion and biofilm formation, additional adhesin mechanisms remain poorly characterized in this pathogen. In this study, we identified and functionally characterized a two-partner secretion (TPS) system, KpFhaB/FhaC, in the outbreak strain K. pneumoniae Kp3380 (ST15). METHODS

Search for Potentially Virulent Staphylococcus epidermidis Isolated from Healthy Human Skin.

Genomic studies suggest that the skin microbiota of healthy individuals may harbor Staphylococcus epidermidis strains with putative virulence-associated characteristics, and that contamination of medical devices can originate from the patient's own skin microbiota. However, the virulence potential of these strains and their ability to contaminate medical devices have not been directly validated experimentally. Therefore, this study evaluated whether S. epidermidis isolates recovered from the skin of healthy individuals possess the capacity to colonize implanted catheters and form device-associ

Strategies to Enhance the Efficacy and Clinical Translation of Antimicrobial Photodynamic Therapy.

Background: Antimicrobial resistance represents a growing global health challenge, necessitating the development of effective non-antibiotic antimicrobial approaches. Antimicrobial photodynamic therapy (aPDT) has emerged as a promising localized antimicrobial strategy owing to its broad-spectrum activity, biofilm-targeting capability, and low propensity to induce resistance. However, its clinical translation remains restricted by limited photosensitizer (PS) performance, insufficient light penetration, oxygen dependency, biofilm-associated barriers, and the lack of standardized treatment proto

Graphene Oxide and Methylglyoxal: a combined strategy against chronic wound pathogens.

Chronic wound infections are difficult to treat due to the polymicrobial biofilms that delay wound healing increasing antimicrobial tolerance and limiting patient compliance. To address this challenge, we developed a non-antibiotic combination of Graphene Oxide (GO) and Methylglyoxal (MGO) (patent N. 102022000024408-Composizione per il trattamento delle infezioni a carico delle lesioni cutanee) against clinically relevant chronic wound pathogens. For the tests, clinical antimicrobial resistant Staphylococcus aureus and Pseudomonas aeruginosa were used. MGO MIC was determined and the best GO +

Antimicrobial Properties of a Novel PEGylated Copper Nanoparticle-Embedded Silicone Rubber with Potential for Use in Biomedical Applications.

Background: Healthcare-associated infections (HAIs) significantly increase morbidity, mortality, and healthcare costs. Among HAIs, catheter-associated infections are particularly prevalent due to the susceptibility of catheters to microbial contamination and biofilm formation, especially with prolonged use. Biofilms act as infection reservoirs, complicating treatment and often requiring catheter removal, thus extending hospital stays and increasing costs. Recent technological advances in catheter design have focused on integrating antifouling and antimicrobial coatings to mitigate or prevent b

Fibronectin recognition by the emerging multidrug-resistant opportunistic fungal pathogen Candida haemulonii.

The multidrug-resistant pathogen Candida haemulonii (Candidozyma haemuli) can cause superficial and systemic infections in vulnerable individuals. The ability of C. haemulonii to interact with extracellular matrix (ECM) components remains largely unexplored. Herein, we investigated C. haemulonii's capacity to bind to fibronectin and assessed how this ECM protein influences fungal adhesion and biofilm development. The results revealed that C. haemulonii yeasts can recognize and interact with soluble fibronectin in a dose-dependent and isolate-specific manner, as demonstrated by flow cytometry.
