Generation and characterization of a human induced pluripotent stem cell line (SNUi001-A) harboring the MT-ND4 m.11778G>A mutation.

Leber hereditary optic neuropathy (LHON) is a maternally inherited optic neuropathy caused by mutations in mitochondrial DNA. To facilitate disease modeling and the investigation of pathogenetic mechanisms, we generated an induced pluripotent stem cell (iPSC) line, SNUi001-A, derived from the peripheral blood mononuclear cells (PBMCs) of a patient carrying the m.11778G>A mutation in the MT-ND4 gene. The iPSCs were generated using non-integrating episomal vectors. These reprogrammed cells maintained the patient-specific mutation, expressed key pluripotency markers, and demonstrated the potentia

Morphology and molecular characterisation of two new species of Mymarothecioides (Monopisthocotyla: Dactylogyridae) parasitising the gills of anostomid fishes from Brazil.

Using an integrative taxonomic approach, we describe two new species of Monopisthocotyla, Mymarothecioides leporini n. sp. and Mymarothecioides paranaensis n. sp., parasitising the gills of anostomid fishes from the Jaguaribe River and Paraná River basins, Brazil. These descriptions represent the first record of species of Mymarothecioides in hosts of the family Anostomidae. Mymarothecioides leporini n. sp. is distinguished by a male copulatory organ (MCO) with a spoon-shaped distal portion and a ventral bar with an ornamented anteromedial process. Mymarothecioides paranaensis n. sp. differs f

Comparison of conventional techniques to differentiate between Taenia solium and Taenia saginata and an improved polymerase chain reaction-restriction fragment length polymorphism assay using a mitochondrial 12S rDNA fragment.

Given the constraints of classical diagnostic methods, i.e., morphological and isoenzymatic studies of proglottids, a polymerase chain reaction test complemented with restriction enzyme analysis has been modified by redesigning one of the primers to reduce nonspecific amplifications experienced when using field samples. The use of these new, highly cestode-specific primers and the restriction enzyme Ddel led to the development of a diagnostic assay that clearly distinguishes between Taenia saginata and T. solium proglottids in field samples. This assay confirms the presence of T. saginata in E

Novel SCO2 mutation (G1521A) presenting as a spinal muscular atrophy type I phenotype.

Rare cases of suspected spinal muscular atrophy (SMA) have been found to have cytochrome c oxidase (COX) deficiency. To date, four cases with SMA features have been reported in children with mutations in the synthesis of cytochrome oxidase 2 (SCO2) gene. We report a male neonate who was born hypotonic, with persistent lactic acidosis, spontaneous activity with EMG testing, development of respiratory distress in the first few hours of life, and died at 30 days of age with progressive cardiomyopathy. Testing for survival motor neurone (smn) and NAIP deletions were negative and a skeletal muscle

A new mtDNA mutation in the tRNA(Lys) gene associated with myoclonic epilepsy and ragged-red fibers (MERRF).

Myoclonic epilepsy with ragged-red fibers (MERRF) has been associated with an A--G transition at mtDNA nt 8344, within a conserved region of the tRNA(Lys) gene. Although the 8344 mutation is highly prevalent in patients with MERRF, it is not observed in 10%-20% of the cases, suggesting genetic heterogeneity. We have sequenced the tRNA(Lys) gene of five MERRF patients lacking the common 8344 mutation. One of these showed a novel T-->C transition at nucleotide position 8356, disrupting a highly conserved base pair in the T psi C stem. The mutant mtDNA population was essentially homoplasmic in mu

Mitoepigenetic Alterations in Early-Onset Parkinson's Disease.

There is accumulating evidence that distinct mitochondrial DNA (mtDNA) methylation and hydroxymethylation patterns exist in Parkinson's disease (PD). However, most studies have been limited to the investigation of specific target regions, rather than the entire mtDNA, and have been further hindered by other methodological discrepancies and the lack of non-CpG context investigation. Here, we provide a comprehensive profile of methylation and hydroxymethylation levels across the mitochondrial genome, at global and single-base resolution, in CpG and non-CpG (CHG, CHH) contexts in blood samples fr

T-Cell Immunosenescence in Systemic Lupus Erythematosus: Molecular Mechanisms and Therapeutic Perspectives.

Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disease characterized by chronic immune activation, relapsing-remitting clinical progression, and progressive multi-organ damage. In addition to the classical characteristics of autoantibodies and immune complexes, emerging evidence highlights T-cell immunosenescence as a key factor in SLE pathogenesis. In this review, we elucidate the phenotypic and molecular landscapes of immunosenescent T cells in SLE, highlighting the core cellular programs that reshape their functional identity, including cell cycle arrest, dysregulated apop

Progress in research on the association between mesenchymal stem cell senescence and knee osteoarthritis.

Osteoarthritis (OA) is an age-related disease characterized by cartilage degeneration, subchondral bone remodeling, and chronic low-grade inflammation, with knee osteoarthritis (KOA) being a leading cause of functional impairment and reduced quality of life in middle-aged and elderly individuals. In recent years, advances in stem cell biology and aging research have highlighted the critical role of mesenchymal stem cells (MSCs) in maintaining joint homeostasis, regulating inflammatory responses, and mediating cartilage repair. Accumulating evidence indicates that reductions in MSC quantity and

Exploring mitochondrial genetic features in intervertebral disc degeneration through multi-omics integration analyses.

The mitochondrial genetic basis of intervertebral disc degeneration (IVDD) remains incompletely understood. This study employed multi-omics Mendelian randomization (MR) analysis to investigate the potential mitochondrial-related genetic mechanisms underlying IVDD. Using two-sample MR, we integrated and analyzed multi-omics quantitative trait loci, encompassing methylation, expression, protein abundance, and mitochondrial DNA, from genome-wide association studies (GWAS). Genetic associations with IVDD were obtained from the FinnGen study for discovery and the GWAS Catalog database for validatio

Diazonium-Enabled Post-Synthetic Construction of Enantiomeric Single-Atom Nanoagents Used for Alzheimer's Disease Treatment.

Alzheimer's disease (AD) is driven by a self-amplifying interplay between oxidative stress and neuroinflammation, in which mitochondrial DNA (mtDNA) leakage-induced activation of the cGAS-STING pathway plays a central role. Strategies that solely eliminate reactive oxygen species (ROS) are insufficient to suppress downstream inflammatory cascades. Here, we report a pair of chiral dual-functional single-atom nanoagents that can simultaneously scavenge ROS and sequester leaked mtDNA. A diazonium-enabled post-synthetic modification strategy is employed to graft benzoic acid linkers onto a single-

Tet2 deficiency orchestrates Alzheimer's pathogenesis through oxidative mtDNA-driven cGAS-STING activation.

Emerging evidence underscores the pivotal role of Ten-eleven translocation 2 (Tet2), as an epigenetic regulator with neuroprotective functions, yet its temporal dynamics and pathogenic contributions to Alzheimer’s disease (AD) remain poorly understood. By integrating human longitudinal cohort data with experimental models, we demonstrated that Tet2 deficiency accelerated AD‑like neurodegeneration through oxidative stress-dependent cGAS-STING activation. Clinically, Tet2 loss-of-function carriers among Aβ-positive individuals exhibited significantly accelerated cognitive decline. In mice, Tet2

The Oxidative-Mitochondrial-Inflammatory Axis in Retinitis Pigmentosa: Extracellular mtDNA as Biomarker and Therapeutic Read-Out.

Retinitis pigmentosa (RP) is the most common inherited retinal dystrophy (prevalence ~1:4000) and a leading Mendelian cause of working-age blindness. Despite marked genetic heterogeneity, its progression converges on a common secondary cascade of outer-retinal hyperoxia, increased reactive oxygen species (ROS), and mitochondrial dysfunction that drives cone degeneration and central vision loss. Because this oxidative cascade is largely genotype-independent and pharmacologically tractable, oxidative stress is a cross-cutting therapeutic target. Within it, mitochondrial DNA (mtDNA) is a key elem

Mitochondrial Mechanisms in the Potential Male Reproductive Toxicity of E-Cigarette-Derived Metal-Containing Nanoparticles.

E-cigarette devices emit ultrafine metal-containing particles from heating coils, solder joints, and other wetted components, but their reproductive toxicology remains poorly defined. This review integrates evidence from aerosol chemistry, nanoparticle toxicology, and male reproductive biology to evaluate whether these particles may contribute to male reproductive dysfunction through mitochondrial injury. E-cigarette aerosols can contain nanoscale and ultrafine particles bearing chromium, nickel, iron, copper, zinc, tin, and lead. These particles may deposit in the distal lung and, based on br

Major Traumatic Injury and Exposure to Mitochondrial-Derived Damage-Associated Molecular Patterns Promotes Neutrophil Survival Accompanied by Stabilisation of the Anti-Apoptotic Protein Mcl-1.

Traumatic injury leads to an extension of the half-life of circulating neutrophils. However, how quickly neutrophil apoptosis is delayed post-injury is currently unknown, as are the underlying mechanisms and factors that promote this extension of lifespan. During the ultra-early (≤1 h) and acute (4-12 and 48-72 h) post-injury phases, we collected blood samples from 73 adult trauma patients. Following ex vivo culture, neutrophil apoptosis was measured, alongside caspase-3 activation and expression of the anti-apoptotic protein Mcl-1. To identify factors that may promote neutrophil survival post

HSP90α facilitates the dephosphorylation of DRP1 at the Ser637 residue, thereby promoting NLRP3 inflammasome activation and exacerbating brain injury in sepsis.

Sepsis-associated encephalopathy (SAE) is a common and serious complication of sepsis, characterized by neuroinflammation and cognitive dysfunction, yet its molecular mechanisms remain unclear. This study aimed to investigate the role of heat shock protein 90α (HSP90α) in microglial NLRP3 inflammasome activation and cognitive impairment in SAE, and to explore the therapeutic potential of a small molecule compound, nicotinamide N-oxide (NAMO). SAE models were established using cecal ligation and puncture (CLP) in mice and LPS/ATP stimulated primary microglial cells/BV-2 cells. A combination of

Effect of OmpA of Acinetobacter baumannii on apoptosis in pulmonary epithelial cells and its molecular mechanism.

BACKGROUND: Acinetobacter baumannii (AB) is a key pathogen in hospital-acquired infections, with rising concerns around its virulence and drug resistance. Outer membrane protein A (OmpA) plays a crucial role in AB's pathogenicity, facilitating bacterial adhesion, biofilm formation, toxin release, and immune responses. OmpA consists of two proline-rich regions, with its N-terminus forming a β-barrel transmembrane domain, allowing AB to bind host cells and invade mitochondria and the nucleus. The N-terminus triggers a caspase cascade leading to mitochondrial damage, while the C-terminus causes D

Sulindac prevents increased mitochondrial VDAC1 expression and cell surface mistargeting induced by pathological conditions in retinal cells.

Diabetic retinopathy (DR) continues to be the primary cause of vision loss in poorly controlled diabetic subjects. The molecular mechanisms underlying retinal pigment epithelium (RPE) cell dysfunction in DR still remain elusive. We investigated the role of mitochondrial volt-age-dependent anion channel 1 (VDAC1) in RPE dysfunction under glucotoxic and inflammatory conditions. Our results demonstrate that both glucotoxicity and cytokine treatment reduces cellular viability accompanied by increased VDAC1 and inducible nitric oxide synthase (iNOS) expression, concomitant with decreased expression

RVFV virulence factor NSs triggers the mitochondrial MCL-1-BAK axis to activate pathogenic NLRP3 pyroptosis.

Infection of Rift Valley fever virus (RVFV), a highly pathogenic mosquito-borne zoonotic virus, triggers severe inflammatory pathogenesis but the underlying mechanism of inflammation activation is currently unclear. Here, we report that the non-structural protein NSs of RVFV triggers mitochondrial damage to activate the NLRP3 inflammasome leading to viral pathogenesis in vivo. It is found that the host transcription inhibition effect of NSs causes rapid down-regulation of myeloid cell leukemia-1(MCL-1), a pro-survival member of the Bcl-2 (B-cell lymphoma protein 2) protein family. RNA Sequence Analyses throughout the Course of Mouse Cardiac Laminopathy Identify Differentially Expressed Genes for Cell Cycle Control and Mitochondrial Function.

Lamin A/C (LMNA) gene mutations are a known cause of familial dilated cardiomyopathy, but the precise mechanisms triggering disease progression remain unknown. We hypothesize that analysis of differentially expressed genes (DEGs) throughout the course of Lmna knockout (Lmna-/-)-induced cardiomyopathy may reveal novel Lmna-mediated alterations of signaling pathways leading to dilated cardiomyopathy. Although Lmna was the only DEG down-regulated at 1 week of age, we identified 730 and 1004 DEGs in Lmna-/- mice at 2 weeks and 1 month of age, respectively. At 2 weeks, Lmna-/- mice demonstrated bot

Maternally inherited diabetes and deafness complicated by mesangial galactose-deficient IgA1 deposits: a case report.

BACKGROUND: Maternally inherited diabetes and deafness (MIDD), a mitochondrial genetic disorder, typically affects the kidneys and results in end-stage renal disease. Early diagnosis of MIDD is challenging when renal manifestations precede other key clinical features such as diabetes and deafness and/or when the disease is complicated by other renal pathologies. CASE PRESENTATION: Here, we present the case of a 33-year-old Japanese woman who had initially been diagnosed with IgA nephropathy but was found to have MIDD 6 years later. Two renal biopsies were conducted six years apart. While asse
