Diabetic foot ulcers (DFU) present a severe clinical challenge, marked by poor healing, chronic inflammation, and elevated amputation risk. The M2 macrophage is known to support the wound healing of DFU, and syndecan-4 (SDC4) contributes to this process. However, the upstream regulatory mechanisms involving post-translational modifications and transcriptional networks remain unclear. The related molecule levels were measured by RT-qPCR and Western blot. The M2 macrophage biomarkers (IL-10, Arg-1) were detected by ELISA kits, while the number of F4/80+CD206+ cells was quantified via flow cytome

BACKGROUND: The use of ankle-foot orthoses (AFOs) is a mainstay in the treatment of cavovarus deformity associated with Charcot-Marie-Tooth (CMT) disease, but their ability to reduce cavovarus deformity is poorly characterized. Similarly, associations between AFO design features and degree of deformity reduction are unknown. Weightbearing CT has been used to describe CMT disease-related cavovarus deformity through both two-dimensional (2D) radiographic measures and three-dimensional statistical shape modeling (SSM). However, neither method has been used to evaluate differences in foot posture

BACKGROUND: Diabetic foot ulcers (DFUs) represent a major and growing complication of diabetes in the Gulf and wider Middle East and North Africa (MENA) region, which has the world's highest diabetes prevalence at approximately 17.6% of adults. DFUs carry a high burden of morbidity, mortality and economic cost, and their complex multifactorial aetiology, encompassing peripheral neuropathy, ischaemia, deformity and infection, is compounded by region-specific cultural, climatic and healthcare factors.
AIMS: To develop evidence-based, Gulf-specific clinical guidelines on the assessment, manageme

Stroke is a leading cause of severe long-term disability, and foot drop is a common post-stroke gait impairment that may reduce toe clearance, increasing the risk of toe contacts and, in turn, trip-related falls. This case series evaluated the novel combination of transcranial direct current stimulation (tDCS), functional electrical stimulation (FES) to the tibialis anterior, and physical therapy training to improve minimum toe clearance (MTC) in individuals with chronic stroke. The rationale for this combined approach was that tDCS may prime the motor cortex and enhance neuroplastic responsiv

Predicting early cognitive decline is important for timely interventions. However, existing approaches for early detection remain limited by cost, scalability, or concerns about their validity. This study evaluated the potential of lower-extremity force control capacities, a largely unexplored area, to improve prediction of cognitive status in older adults with and without Mild Cognitive Impairment (MCI). MCI refers to an early stage of cognitive decline that does not meet criteria for dementia. 224 participants aged 80-91 years completed a foot-pedal tracking task in which they continuously a

Scalable tools for Parkinson's disease (PD) detection are needed beyond laboratory-bound assessments. We present and evaluate a deliberately minimalist approach: a single six-axis IMU per shoe coupled with a lightweight 1D-CNN, tested during a six-minute walk test (6MWT) conducted indoors and outdoors under semi-controlled conditions. Eighty-nine participants (44 PD; 45 healthy controls) were instrumented bilaterally at 50 Hz. Signals (accelerometer and gyroscope) were standardized to a common shoe-referenced frame and ingested as raw time series using 0.5-s non-overlapping windows. The networ

BACKGROUND/OBJECTIVES: Diabetic foot infections (DFIs) are a major cause of morbidity in low- and middle-income countries such as Pakistan, where antimicrobial resistance (AMR) and the absence of localized guidelines complicate their management. This study aimed to analyze the microbiological profile of DFIs and develop evidence-based, context-specific empirical treatment recommendations.
METHODS: We conducted a retrospective study at a tertiary care hospital in Pakistan, reviewing all diabetic foot related admissions between January 2022 and December 2023. Patients with clinically suspected

To study the risk factors associated with multidrug-resistant bacteria (MDRB) infection in diabetic foot (DF) wounds and to develop a nomogram model to predict the risk of MDRB infection in patients with DF. A total of 157 patients with DF between January 2013 and December 2023 were included in this study. A retrospective analysis was conducted to examine the characteristics of MDRB infections. Univariate and multivariate logistic regression analyses were performed to identify risk factors associated with MDRB infection. Based on these risk factors, a predictive model was built using R softwar

Diabetic foot ulcers (DFUs) are devastating complications of diabetes, defined by impaired healing, high amputation rates, and substantial mortality, with pathogenesis rooted in interconnected metabolic, immune, and vascular dysregulation. Serum metabolomic profiling identifies elevated branched-chain amino acid (BCAA) levels in DFU patients, yet the metabolic mechanisms linking BCAA dysregulation to DFU remain unexplored. This review systematically presents a causal framework: impairment of the BCAA metabolic process (downregulation of BCKDH in skeletal muscle/fat tissue), dysbiosis of the in

PURPOSE: Since hand-foot skin reaction (HFSR) is a dose-limiting toxicity associated with certain multikinase inhibitors, prophylactic measures to manage this adverse effect require improvement. This study aimed to evaluate the efficacy of depressurization using hydrocolloid dressing by comparing the effects of adding hydrocolloid dressing to standard prophylactic moisturizing versus moisturizing alone.
METHODS: We conducted a phase 3 randomized self-controlled study for patients with unresectable colorectal cancer, gastrointestinal stromal tumors, or hepatocellular carcinoma who planned to r

Diabetic foot ulcers (DFUs) are a serious diabetic complication leading to high rates of morbidity, amputation risks, and economic burdens on the treatment facilities. The present review continues our exploration of the novel paradigm of combining micronutrients with nanoparticles to address diabetes-related DFUs effectively. We performed a detailed examination of published studies to discuss our views on the ever-increasing utilization of essential micronutrients like zinc, selenium, and vitamins A, C, D, and E to modulate oxidative stress levels, stimulate the secretion of vascular endotheli

BACKGROUND: The height of the foot arch is highly related to knee joint function. Knee joint strength is a significant contributing factor to knee joint injuries, while existing research has not yet clearly established the correlation between arch height and knee joint strength.
OBJECTIVE: This study aims to investigate the influence of arch height on isokinetic knee muscle strength at varying angular velocities, thereby providing a potential scientific basis for the prevention and rehabilitation of associated conditions.
DESIGN: Cross-sectional study.

Grapevine black-foot disease is a destructive trunk disease with a complex pathogen composition that often involves mixed and latent infections, making timely field diagnosis challenging. To improve rapid field detection, we developed a rapid, sensitive, and low instrument-dependent nucleic acid assay. The assay integrates recombinase polymerase amplification (RPA) and clustered regularly interspaced short palindromic repeats (CRISPR)-Cas12a for the detection of Ilyonectria and Dactylonectria, two genera associated with grapevine black-foot disease. Conserved regions of the histone H3 and β-tu

Diabetic foot infection (DFI) includes soft tissue infection and osteomyelitis below the ankle and is a leading cause of lower limb amputation and mortality in diabetic patients. Treatment involves prolonged antibiotic therapy with surgical debridement or amputation. Local antimicrobial therapy offers an adjunct or alternative to systemic therapy. This systematic review and meta-analysis assessed the efficacy of systemic antibiotic therapy compared with local and combination (local and systemic) therapy in DFI. A meta-analysis was conducted according to the Preferred Reporting Items for System

Transcatheter arterial microembolization (TAME) is a minimally invasive endovascular procedure initially developed for bleeding control and later applied to the treatment of chronic musculoskeletal conditions characterized by pathological hypervascularization, to inhibit or reduce the release of inflammatory mediators. First introduced in anatomical districts such as the knee, TAME has more recently been proposed as a therapeutic option for chronic inflammatory foot and ankle disorders refractory to conservative treatment. A narrative review of the literature was conducted by searching PubMed

Congenital talipes equinovarus (CTEV) is a common musculoskeletal entity that affects 1-2 per 1000 of live births worldwide. Most are idiopathic with a typical deformity of midfoot cavus, forefoot adductus, hindfoot varus, and equinus. Controversy exists whether CTEV results primarily from in-utero soft-tissue abnormality or the defect is in the osteocartilage anlagen. This fetus study aimed to elucidate the gross anatomical morphology and morphometry of bones, joints, and soft tissue in CTEV compared with the normal foot. This comparative cross-sectional study done on 12 normal feet and four

Objective.Gait and the timed up and go (TUG) time are the two tests that are widely used to assess functional mobility; however, their administration requires space and trained personnel, limiting feasibility for routine assessment. A brief, seated foot-tapping test may offer a practical alternative. This study examined whether accelerometer-derived features from unilateral foot-tapping can predict gait and TUG times across the adult lifespan.Approach.Seventy-eight adults aged 19-95 years (58 older adults: 80.25 ± 8.2 yrs; 20 young-to-midlife: 32.2 ± 14.1 yrs) performed 30-second unilateral fo
