Severe (type III) osteogenesis imperfecta due to glycine substitutions in the central domain of the collagen triple helix.

The molecular defects responsible for three cases of severe (type III) osteogenesis imperfecta (OI) were investigated. The mutation sites were localized in pro alpha 1(I) and pro alpha 2(I) mRNA molecules, respectively, by chemical cleavage of mismatch in heteroduplex nucleic acids. Mutation identification was achieved by reverse transcription-polymerase chain reaction-DNA amplification, followed by cloning and sequencing. Two unrelated patients were demonstrated to bear the same G-A transition at nucleotide 2418 of the pro alpha 1(I) coding region, leading to G589S substitution and resulting

Prevalence of herpesvirus, polyomavirus, and respiratory viruses in saliva from a Quilombola community in the Brazilian Amazon.

INTRODUCTION: Quilombola communities, composed of descendants of enslaved individuals from Brazil's colonial period, often face limited access to healthcare, which may increase their susceptibility to viral infections. Oral transmission routes, particularly through saliva, are relevant for viruses such as those in the Polyomaviridae family. Therefore, here we aimed to investigate the prevalence of HSV-1, JC (JCV), and BK polyomaviruses, and respiratory viruses (Influenza A/B, RSV, SARS-CoV-2), as well as the salivary inflammatory profile in individuals from a Quilombola community. METHODS: A

Cancer Metastasis Blockage by Exosome-Neutrophil Nanocoupler.

Most malignancy patients die of cancer cell metastasis, in which tumor-derived exosomes (TDEs) play a critical role by priming premetastatic niche formation, since most TDEs carry cancer-specific biomolecules such as cytokines, nucleic acids, and lipids through the peripheral blood to distant organs. Thus, blocking the TDEs' migration in the vascular system is an especially promising antimetastasis strategy; unfortunately, however, existing treatments have shown limited efficacy. In this study, we present an exosome-neutrophil binding strategy using a reactive oxygen species-producing nanocoup

Digital Immunoassays for Sensitive Quantification of Blood Biomarkers Using Solid-State Nanopores.

Digital immunoassays enable highly sensitive detection of biomolecules, offering absolute quantification rather than relying on bulk signal intensity. We adapt a digital immunoassay scheme for a nanopore sensor, a versatile platform for single-molecule counting. Current nanopore sensors have demonstrated great progress when counting nucleic acids but struggle with proteins due to variability in translocation behavior and limited recognition strategies. While recent advancements have highlighted the promise of nanopore platforms for protein studies, precise quantification remains a challenge. H

Developing blood extracellular vesicle protein biomarkers for hepatocellular carcinoma: mechanisms, methods, and translation.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality globally, with late-stage diagnoses predominantly driven by the absence of early clinical symptoms and the inadequate sensitivity of current diagnostic tools, highlighting a critical unmet need in liver cancer management. Circulating extracellular vesicles (EVs), which naturally carry proteins and nucleic acids, serve as both mediators of tumor-specific biology and reporters of early pathophysiological changes, offering a transformative avenue for noninvasive liquid biopsy. This review integrates mechanistic adv

Vibrational spectroscopy combined with chemometric approaches in bacterial studies: a review of recent advances and applications.

Bacterial infections remain among the leading causes of global mortality and represent significant challenges to public health. Conventional methods such as cultivation procedures, polymerase chain reaction, and instrumental techniques are routinely used in microbiology laboratories. However, these methods are time-consuming and labor-intensive, and require multi-step sample preparation. Moreover, there is a risk of sample contamination. Recently, vibrational spectroscopic techniques, including near-infrared, mid-infrared, and Raman spectroscopy, have gained considerable attention in microbiol

Extracellular vesicles as paradigm shifters: Transformative roles in diagnosis and therapy for brain disorders.

Extracellular Vesicles (EVs), the nano-sized extracellular membrane-bound vesicles, facilitate cell-to-cell communication by transporting bioactive molecules like proteins, lipids, and nucleic acids. Their unique cargo, determined by the cell of origin, makes them valuable tools for studying disease pathogenesis and potential drug delivery systems. Research suggests that EVs play a role in the pathogenesis of various diseases, including neurodegenerative and neurodevelopmental disorders. They have been implicated in diseases like Parkinson's disease (PD), Alzheimer's disease (AD), autism spect

A new and eco-friendly disinfectant for antimicrobial-resistant bacteria: ozone nano water.

BACKGROUND: Escherichia coli (E. coli) is a major foodborne pathogen that affects 2.8 million people globally. The widespread use of antibiotics and disinfectants in livestock production has contributed to the emergence of multidrug-resistant strains, underscoring the need for resistance surveillance and the development of alternative control strategies, such as ozone nano water (ONW). ONW has emerged as a promising disinfectant, particularly for controlling microbial contamination in food safety and medical applications. In this study, we evaluated the bactericidal activity of ONW against fou

From bench to bedside: stem and progenitor cell-derived extracellular vesicles in cardiac therapy.

Initially regarded as insignificant cellular waste, extracellular vesicles (EVs) are now recognized as key mediators of intercellular communication, capable of transferring bioactive molecules-such as proteins, nucleic acids, and small compounds-between cells. This function has positioned EVs as promising cell-free therapeutic agents with the potential to transform modern medicine. Stem and progenitor cells naturally release EVs that can replicate many of the therapeutic effects of cell transplantation, while avoiding the challenges associated with administering living cells. Hybridization capture increases on-target nanopore sequencing of plant RNA tobamovirus- derived cDNA libraries.

High-throughput sequencing (HTS) can support plant virus surveillance, but host nucleic acids often reduce on-target read recovery. We evaluated a targeted hybridization-capture workflow in which barcoded double-stranded cDNA (ds-cDNA) libraries generated from plant RNA extracts spiked with lyophilized tobamovirus-positive controls were enriched before Oxford Nanopore sequencing. Biotinylated probes targeted conserved regions of cucumber green mottle mosaic virus (CGMMV), species Tobamovirus viridimaculae; pepper mild mottle virus (PMMoV), species Tobamovirus capsici; and tobacco mosaic virus
