Characterization of the Effects of a Humanin Fragment Peptide (HNF14) in Age-Related Macular Degeneration.

Background: Age-related macular degeneration (AMD) is a leading cause of vision loss and is strongly associated with mitochondrial dysfunction in retinal pigment epithelial cells. Mitochondrial-derived peptides, including Humanin and its analogs, have demonstrated cytoprotective effects in AMD-related cellular models. However, the effects of shorter Humanin-derived fragments in disease-specific mitochondrial models remain incompletely characterized. Methods: Transmitochondrial retinal pigment epithelial cybrid cell lines containing mitochondria from AMD patients or age-matched normal donors we

Vagal activity related events and glucagon-like peptide-1 receptor agonists.

BACKGROUND AND AIMS: Glucagon-like peptide-1 receptor agonists (GLP-1 RA) carry side-effects such as dizziness and nausea related to increased vagal tone. This study investigated associations between GLP-1 RA use and the risk of vagal activity related (VAR) events. METHODS: Using the Danish health registers, patients with type 2 diabetes mellitus (T2DM) first-time initiated on GLP-1 RA (exposure) or SGLT-2i (active control) were identified between January 2010 and October 2022. Standardized 1-year absolute risks of VAR events (syncope, fractures, bradyarrhythmia, or cardiac device implantatio

A Natural Peptide from the American Cockroach Suppresses Feline Infectious Peritonitis Virus.

Feline infectious peritonitis virus (FIPV) causes a fatal and immune-mediated disease in cats with limited treatment options. Natural products offer promising antiviral candidates, but animal-derived peptides remain underexplored. Here, we identified PI-8, a Periplaneta americana (P. americana)-derived peptide with validated binding to ISG15, and evaluated its anti-FIPV effect and mechanism. PI-8 showed low cytotoxicity (CC50 > 40 μM) and inhibited FIPV replication with an IC50 of 12.9 μM, acting primarily at the post-entry stage. PI-8 significantly upregulated IFNA1, IFNB, ISG15, and MX1 in i

Assays for Plasma ADAMTS13 Activity and Inhibitors Using Recombinant FRETS-VWF73 Peptide.

Thrombotic thrombocytopenic purpura (TTP) is a life-threatening thrombotic microangiopathy caused by severe deficiency of plasma ADAMTS13 activity. ADAMTS13 cleaves ultra-large von Willebrand factor (ULVWF) multimers, thus preventing the pathological accumulation of platelet-VWF aggregates at sites of vascular injury. The deficiency of ADAMTS13 can be congenital or more commonly immune-mediated, involving the formation of autoantibodies that bind and inactivate plasma ADAMTS13 activity. A simple, rapid, and cost-effective assay is crucial for the diagnosis of TTP and for the conduction of tran

Screening of Small Peptide Against Ischemic Fatty-Acid Binding Protein (FABP).

A low-molecular-weight cytoplasmic protein called heart-type fatty acid-binding protein (H-FABP) is released by cardiac myocytes during an ischemic episode. The normal serum level of FABP is around 3.5 ± 0.4 ng/ml (males), 3.9 ± 0.4 ng/ml (females), while the cutoff value of H-FABP in case of acute myocardial infarction (AMI) is 21.85 ng/ml within 3 hours. In case of angina pectoris, ischemic patients can be distinguished from non-ischemic patients and treated appropriately by detecting the presence of serum H-FABP in a point-of-care (POC) diagnostic approach. As the mortality rate due to coro

Pathology-Targeted Nanoparticles Guided by Peptide Remodel the Periodontal Microenvironment for Periodontitis Therapy.

Periodontitis is an inflammatory and destructive condition, conventional therapies yield inadequate long-term efficacy and possess limited regenerative potential, while the diseased microenvironment driven by inflammation and oxidative stress further hampers tissue repair. Crucially‌, because of the unique nature of the periodontal microenvironment, limitations still exist, such as a short drug retention period and low drug delivery efficiency. To overcome these limitations, we developed a new nanoplatform designed to improve local drug retention and target the core pathological mechanisms of

Discovery and engineering of bispecific TCR-mimic antibodies targeting peptide-HLA complex.

TCR-mimic (TCRm) antibodies represent a promising new frontier in cancer immunotherapy, enabling the targeting of intracellular targets previously considered undruggable with antibodies. We developed an integrated platform for generating human antibodies that mimic T-cell receptors (TCRs) and recognize peptide-HLA complexes with high specificity and affinity. These antibody binders were then engineered into highly potent bispecific antibodies capable of redirecting T cells to kill tumor cells. By leveraging high-quality pHLA immunogens, fully human transgenic mice, advanced single B-cell disco

Glucagon-Like Peptide-1 Receptor Agonists for Obstructive Sleep Apnea: A Review.

IMPORTANCE: Obstructive sleep apnea (OSA) affects nearly 1 billion adults worldwide and is independently associated with cardiovascular disease, stroke, and increased mortality. While continuous positive airway pressure (CPAP) remains the criterion standard treatment, adherence rates of only 30% to 60% limit its population-level effectiveness. Obesity is present in 60% to 70% of patients with OSA and represents the primary modifiable risk factor, yet, until recently, no pharmacological therapies specifically targeted this pathophysiological mechanism. Peptidic Phosphonates as Irreversible Covalent Inhibitors of Plasmodium falciparum Serine Protease PfSUB1.

Malaria, caused by Plasmodium parasites, remains a major global health challenge, exacerbated by the widespread emergence of drug-resistant plasmodium strains. Subtilisin-like serine protease SUB1 triggers escape of the parasite from the red cell via a process called egress, rendering the enzyme a prospective antimalarial drug target. While several SUB1 inhibitors have been developed, irreversible covalent inhibition has not been explored so far. In this work, we report our studies of peptidic inhibitors bearing covalent serine traps such as β-lactam, β-lactone, epoxide, and diaryl phosphonate

Evaluation of a Novel Peptide-Based ELISA for the Serological Diagnosis of Trypanosoma cruzi Infection.

Chagas disease is a neglected tropical disease endemic in most of the Americas, caused by Trypanosoma cruzi. New serological tests with improved performance are urgently needed for accurate diagnosis of T. cruzi infections. We evaluated here the performance of a novel peptide ELISA based on highly conserved parasite antigens, using a large panel of well characterized samples (N = 308) from Mexico, where serological discordance is particularly high. Comparison of peptide ELISA with various serological and molecular assays indicated that it outperformed an FDA-approved commercial ELISA in terms

Effects of novel antimicrobial peptide F1 on quality and storage stability of litchi fruit.

BACKGROUND: Litchi, a fruit of considerable commercial value, is highly susceptible to postharvest deterioration. Chemical preservatives pose potential risks to consumer health and the environment. Therefore, there is an urgent demand for natural preservative alternatives. RESULTS: This study investigates the efficacy of an antimicrobial peptide extract (AMS) as a natural preservative for litchi. Fruit samples were immersed in distilled water (control), sodium hypochlorite, prochloraz, or AMS solutions at varying concentrations (0.5 and 1.0 mg mL-1), then stored at room temperature. CSF peptidomics reveals an IGF2-derived peptide as a candidate biomarker for neurosyphilis diagnosis.

BACKGROUND: Neurosyphilis (NS) diagnosis is challenging due to the cerebrospinal fluid-Venereal Disease Research Laboratory (CSF-VDRL) assay has limited sensitivity, whereas CSF abnormalities lack sufficient specificity. METHODS: To identify novel CSF peptide biomarkers for NS, we performed comparative peptidomic analysis of CSF samples from 51 individuals across four groups: NS, syphilis/non-NS (NNS), infectious brain diseases without syphilis (IBD), and non-infectious brain disorders without syphilis (NIBD). Peptides were profiled by high-resolution liquid chromatography-tandem mass spectro

Host-directed antimicrobial peptide biomaterials for combating Gram-negative bacterial infections.

Antimicrobial resistance (AMR) represents a defining crisis in modern infectious disease medicine. Gram-negative microbes such as carbapenem-resistant Acinetobacter baumannii, Pseudomonas aeruginosa, and Klebsiella pneumoniae are becoming more resistant than ever. Traditional methods of treatment using antibiotics are ineffective because of the presence of several envelope layers, active efflux pumps, and biofilm production. In contrast, bacteriolytic therapy can exacerbate host immunopathology owing to systemic endotoxin release. Host-defense peptides are thus being explored for their ability

AI-assisted design of a VEGFR2 agonistic peptide that promotes angiogenesis and wound repair.

Vascular endothelial growth factor (VEGF) serves as a principal regulator of neovascularization and is essential for vascular regeneration. In this study, we identified and evaluated VEGF Mimetic Peptide3 (VMP3), a novel synthetic peptide developed as a potential VEGFR-2/VEGF-A modulator. VMP3 was generated using an AI-assisted peptide design framework incorporating long short-term memory (LSTM) networks, which enabled learning of sequence-level patterns within known VEGF agonist peptides and assisted the motif-guided and structure-informed design of new candidates. A novel cyclic peptide that blocks human autoimmune antibodies for the treatment of Multiple sclerosis.

Multiple sclerosis (MS) is a chronic autoimmune disease causing demyelination in the central nervous system, with binding of auto-reactive (auto)antibodies to myelin auto-antigens such as GlialCAM protein, myelin basic protein (MBP) and myelin oligodendrocyte glycoprotein (MOG) contributing to myelin destruction. Current MS therapeutics are largely non-specific to the molecular underpinnings of disease, primarily suppressing B- and/or T-cell sub/populations to combat autoimmunity. We hypothesized that selectively blocking disease-specific autoantibodies from binding autoantigen represents a ta

A novel peptide-based strategy to enhance GBA1 expression for treating Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by α-synuclein aggregation and lysosomal dysfunction, with GBA1 mutations representing the most common genetic risk factor. Reduced glucocerebrosidase (GCase) activity is observed in both familial and sporadic PD, promoting α-synuclein accumulation and neuronal toxicity. Here, we developed a GBA1-HiBiT tagged knock-in HEK293T reporter system, providing a highly sensitive, real-time quantitative measurement of GCase dynamics. From a leech-derived peptide library, we identified a cell-penetrating peptide hirunipin

A novel peptide targeting CCR7 inhibits tumor cell lymph node metastasis.

Lymph nodes are the most common metastasis sites for tumor cells, which are intimately linked to patient prognosis. It has been reported that cancer cells can upregulate CC Chemokine Receptor 7 (CCR7) expression and hijack its normal functions, enabling them to migrate along the gradient of CCL19 and CCL21 toward the lymph nodes and colonies as the initial stage of distant metastasis. In tumor patients, the metastatic tumor in the lymph nodes exhibited higher expression of CCR7, as well as inhibitory immune checkpoints PD-1, LAG-3, and TIM-3 compared to the primary tumors with the analysis of

Discovery of conserved peptide-MHC epitopes for directly alloreactive CD8+ T cells.

Mass Spectrometry allied with in-vivo generation of activated alloreactive T cell populations and tetramer screening facilitates the identification of endogenous peptides that are directly recognised in complex with allogeneic Major Histocompatibility class I (MHC I) molecules by alloreactive CD8+ T cells. We had previously used this approach for the discovery of immunogenic self-peptides presented by the allomorph H-2Kb (Kb). In this study, we identified 22 highly immunogenic self-peptides presented by H-2Kd (Kd). Peptide abundance across skin, spleen and liver samples (estimated as the produ

Glucagon-like peptide-1 receptor agonists in neurodegenerative diseases: Promises and challenges.

Glucagon-like peptide-1 (GLP-1) receptor agonists (GRA) belong to a class of compounds that reduce blood glucose and energy intake by simulating actions of endogenous incretin hormone GLP-1 after it is released by the gut following food consumption. They are used to treat type 2 diabetes mellitus (T2DM) and obesity and have systemic effects on various organs, including the brain, liver, pancreas, heart, and the gut. Patients with T2DM have a higher risk of developing neurodegenerative diseases (NDs), including Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (A
