Bone sialoprotein (BSP) is an anionic phosphoprotein in the extracellular matrix of mineralized tissues, and a promoter of biomineralization and osteoblast development. Previous studies on the Bsp-deficient mouse (Bsp(-/-)) have demonstrated a significant bone and periodontal tissue phenotype in adulthood. However, the role of BSP during early long bone development is not known. To address this, early endochondral ossification in the Bsp(-/-) mouse was studied. Embryonic day 15.5 (E15.5) wild-type (WT) tibiae showed early stages of ossification that were absent in Bsp(-/-) mice. At E16.5, mine

Diffuse idiopathic skeletal hyperostosis (DISH) is a noninflammatory spondyloarthropathy, characterized by ectopic calcification of spinal tissues. Symptoms include spine pain and stiffness, and in severe cases dysphagia and spinal cord compression. The etiology of DISH is unknown and there are no specific treatments. Recent studies have suggested a role for purine metabolism in the regulation of biomineralization. Equilibrative nucleoside transporter 1 (ENT1) transfers hydrophilic nucleosides, such as adenosine, across the plasma membrane. In mice lacking ENT1, we observed the development of

Osteoporosis international : a journal established as result of cooperation between the European Foundation for Osteoporosis and the National Osteoporosis Foundation of the USA2010eng
UNLABELLED: The study assessed contribution of genetic factors to variability of osteopontin (OPN) levels. Evidence of association of OPN levels with polymorphisms in its structural gene and integrin-binding sialoprotein gene loci was obtained. The results motivate research of OPN-related proteins and genes with respect to biomineralization and other biological processes.
INTRODUCTION: OPN is a major phosphoprotein in bone, which plays key role in regulation of bone mineralization process. It is considered as a promising biomarker for osteoarthritis and osteoporosis, and various other patholo

Mineralized collagen fibrils are the basic building blocks of bone tissue at the supramolecular level. Several disease states, manipulation of the expression of specific proteins involved in biomineralization, and treatment with different agents alter the extent of mineralization as well as the morphology of mineral crystals which in turn affect the mechanical function of bone tissue. An experimental assessment of mineralized fibers' mechanical properties is challenged by their small size, leaving analytical and computational models as a viable alternative for investigation of the fibril-level

Bone sialoprotein (BSP) is a secreted glycoprotein primarily found in sites of biomineralization. Recently, we demonstrated that BSP is strongly upregulated in osteotropic cancers and particularly those that exhibit microcalcifications. BSP contains an Arg-Gly-Asp (RGD) motif found in other adhesive molecules that interact with cellular integrins. In bone, BSP has been shown to mediate the attachment of osteoblasts and osteoclasts via alpha(v)beta(3) integrin receptors. Ligands for alpha(v)beta(3) integrin are considered to play a central role during angiogenesis. Therefore, we used human umbi

This study investigated the delivery of small interfering RNA (siRNA) silencing prolyl hydroxylase domain 2 (PHD2) via poly(lactic-co-glycolic acid) (PLGA)-bioactive glass scaffold to improve angiogenesis and osteogenesis for biomineralized tissue engineering. Polyethylenimine (PEI) was complexed with PHD2siRNA at 3:1, 6:1, and 8:1 ratios, and the resulting complexes were characterized by transmission electron microscopy, NanoSight analysis, and gel retardation assays. Their biological effects on human umbilical vein endothelial cells (HUVECs) were evaluated to determine the optimal ratio. PLG

Pathological crystallization within soft tissues often yields biominerals with properties differing from those of their geological or synthetic counterparts. Microcalcifications (MCs) are abundant in breast tumors, particularly in non-invasive lesions, such as ductal carcinoma in situ (DCIS). Given the challenge of predicting DCIS progression into invasive cancer, it has been suggested that MCs can be leveraged to inform DCIS prognosis. The predominant type of breast MCs are those containing calcium phosphates (CaP), whose crystal properties are commonly held to correlate with malignancy. Less

Healing of significant segmental bone defects remains a challenge, and various studies attempt to make materials that mimic bone structures and have biocompatibility, bioactivity, biodegradability, and osteoconductivity to native bone tissues. In this work, a nanofiber scaffold membrane of polyvinyl alcohol (PVA)/polyvinylpyrrolidone (PVP)/chitosan (CS) combined with hydroxyapatite (HAp) from sand lobster (SL; Panulirus homarus) shells, as a calcium source, was successfully synthesized to mimic the nanoscale extracellular matrix (ECM) in the native bone. The HAp from SL shells was synthesized

Caries, a major global disease associated with dental enamel demineralization, remains insufficiently understood to devise effective prevention or minimally invasive treatment. Understanding the ultrastructural changes in enamel is hampered by a lack of nanoscale characterization of the chemical spatial distributions within the dental tissue. This leads to the requirement to develop techniques based on various characterization methods. The purpose of the present study is to demonstrate the strength of analytic methods using a correlative technique on a single sample of human dental enamel as a

The transition from normal forage to a highly fermentable diet to achieve rapid weight gain in the cattle industry can induce ruminal acidosis. The molecular host mechanisms that occur in acidosis are largely unknown. Therefore, the histology and transcriptome profiling of rumen epithelium was investigated in normal and acidosis animals to understand the molecular mechanisms involved in the disease. The rumen epithelial transcriptome from acidosis (n=3) and control (n=3) Holstein steers was obtained using RNA-sequencing. The mean values of clean reads were 70,975,460±984,046 and 71,

Engineering scaffolds combining natural biomineral and artificially synthesized material hold promising potential for bone tissue regeneration. We fabricated a bioengineering scaffold, oyster shell (OS) and alpha-calcium sulfate hemihydrate (α-CSH) as scaffold, platelet-rich plasma (PRP) as provider of growth factors and bone mesenchymal stem cells (BMSCs) as seed cells, and determined it could be applied as a new type of bone graft substitutes by rat calvarial defects repairing experiment in vitro and in vivo. SEM showed that the mean diameter of the pores was about 150 μm with a range of 50-

Disease can alter natural ramp-like elastic gradients to steeper step-like profiles at soft-hard tissue interfaces. Prolonged function can further mediate mechanochemical events that alter biomechanical response within diseased organs. In this study, a human bone-tooth fibrous joint was chosen as a model system, in which the effects of bacterial-induced disease, i.e. periodontitis, on natural elastic gradients were investigated. Specifically, the effects of ectopic biomineral, i.e. calculus, on innate chemical and elastic gradients within the cementum-dentin complex, both of which are fundamen

Biosignature detection remains a key challenge in astrobiology, yet robust mineral biosignatures remain limited. Raman spectroscopy is increasingly applied in planetary exploration, but its high-dimensional spectral information has not yet been fully exploited for biosignature discrimination using data-driven approaches. Here, we integrate Raman spectroscopy with interpretable machine learning to distinguish biotic from abiotic apatite, a ubiquitous phosphate mineral in terrestrial and extraterrestrial environments. We compile 331 apatite Raman spectra from abiotic and biotic sources and extra

Magnetotactic bacteria (MTB) are generally thought to navigate as individual cells by using intracellular magnetosome chains that provide a stable magnetic moment for orientation along magnetic field lines. Whether nonclassical magnetosome geometries and particle-scale magnetic states can support robust chain-level magnetic function, and potentially mediate interactions between cells, remains unresolved. Here, to we report a unicellular magnetotactic bacterium, strain WYHV-1, that biomineralizes comparatively large prismatic magnetite magnetosomes with distinctive dimples at both crystal ends

Sustainable alternatives are essential to improving agriculture production to meet the growing world's critical demands. Cyanobacteria and microalgae are considered renewable resources with a wide range of potential uses in the agricultural sector. We aimed to isolate cyanobacteria and microalgae from the mud of a carbon dioxide-rich sulfur pond and to investigate their plant growth-promoting (PGP) and soil bio-consolidating ability. Mud samples were subjected to DNA extraction and 16S rRNA gene sequencing to characterize the prokaryotic community. Phototrophic culturable microbiota was isolat

The essence of bionanotechnology lies in the application of nanotechnology/nanomaterials to solve the biological problems. Quantum dots and nanoparticles hold potential biomedical applications, but their inherent problems such as low solubility and associated toxicity due to their interactions at nonspecific target sites is a major concern. The self-assembled, thermostable, ferritin protein nanocages possessing natural iron scavenging ability have emerged as a potential solution to all the above-mentioned problems by acting as nanoreactor and nanocarrier. Ferritins, the cellular iron repositor

Fibroblast growth factor (FGF) signaling has been implicated in the regulation of osteogenesis in both intramembranous and endochondral ossifications. In the developing palate, the anterior bony palate forms by direct differentiation of cranial neural crest (CNC)-derived mesenchymal cells, but the signals that regulate the osteogenic cell fate in the developing palate remain unclear. In the present study, we investigated the potential role of FGF signaling in osteogenic fate determination of the palatal mesenchymal cells. We showed that locally activated FGF8 signaling in the anterior palate u

Human enamel is a hierarchically organized hydroxyapatite biomineral, but it lacks regenerative capacity, motivating interfacial engineering strategies that control mineral growth at the enamel surface. This work examines an electric-field-assisted biomimetic mineralization approach that forms organized organo-mineral coatings composed of nanocrystalline carbonate-substituted hydroxyapatite (ncHAp), amino acids (AAs), and polydopamine (PDA). The method utilizes isolated electrodes to apply an electrostatic field, eliminating direct current through the substrate and directing the assembly at th

Marine fish play a significant yet understudied role in the oceanic carbon cycle through the production of magnesium-rich calcium carbonate (CaCO3) precipitates known as ichthyocarbonates. These deposits form in the gut of marine teleost fish in response to salinity, serving as part of their osmoregulation strategy. Through this, marine fish may contribute as much as 9.04 Pg of CaCO3 per year in global new carbonate production, being equivalent to or potentially higher than the production by coccolithophores and pelagic foraminifera. Despite their ecological relevance, the biological mechanism
