Oral chromium(VI) does not affect the frequency of micronuclei in hematopoietic cells of adult mice and of transplacentally exposed fetuses.

Chromium(VI) compounds are genotoxic in a variety of cellular systems. Their potential carcinogenicity is affected by toxicokinetic patterns restricting bioavailability to certain targets, and by metabolic pathways affecting interaction of chromate-derived reactive species with DNA. Epidemiological data indicate that chromium(VI) can be carcinogenic to the human respiratory tract following inhalation at doses that are only achieved in certain occupational settings. However, concern has been raised that adverse effects may also result from oral intake. In order to further explore this issue, we

[Speciation of metals in occupational medicine].

The identification and quantification of elements as such (that make up the historical analytical method used in toxicological studies) are necessary but not sufficient, in all cases, to understand and evaluate the mechanism of interaction between elements and biological targets. In fact, in order to approximate the factors that control the concentrations of elements in environmental or biological media, we need to know the occurrence of species of the elements themselves. This is element speciation, i.e. the capability of separating, identifying and determining the species in which an element

The effects of overfeeding and moderate dietary restriction on Sprague-Dawley rat survival, pathology, carcinogenicity, and the toxicity of pharmaceutical agents.

Ad libitum (AL) overfeeding is the most significant uncontrolled variable effecting the rodent bioassay. There is a highly significant correlation between food consumption, the resultant body weight, and two-year survival in laboratory rats. We have studied the effects of AL overfeeding, moderate dietary restriction (DR) and several modified diets on Sprague-Dawley (SD) rat longevity, spontaneous disease, carcinogenesis and the toxicity of pharmaceuticals. AL feeding of diets varying in protein, fiber and metabolizable energy content did not significantly alter two-year rat survival. Moderate

Circadian clock regulates hepatotoxicity of Tripterygium wilfordii through modulation of metabolism.

OBJECTIVES: We aimed to determine the diurnal rhythm of Tripterygium wilfordii (TW) hepatotoxicity and to investigate a potential role of metabolism and pharmacokinetics in generating chronotoxicity. METHODS: Hepatotoxicity was determined based on assessment of liver injury after dosing mice with TW at different circadian time points. Circadian clock control of metabolism, pharmacokinetics and hepatotoxicity was investigated using Clock-deficient (Clock-/- ) mice. KEY FINDINGS: Hepatotoxicity of TW displayed a significant circadian rhythm (the highest level of toxicity was observed at ZT2 an

Static or microfluidic? A comparative pilot study of an in vivo-in vitro validated human iPSC-derived blood-brain barrier model for neurotoxicity hazard assessment.

Blood-brain barrier (BBB) permeation determines human brain exposure to chemicals and is therefore a prerequisite for neurotoxicity. However, the lack of toxicokinetic BBB models remains a critical uncertainty in regulatory in vitro risk assessment, which was highlighted in the new EU roadmap towards phasing out animal testing. Human induced pluripotent stem cell (hiPSC)-derived BBB models combined with clinical positron emission tomography (PET) data provide translational benchmarks for evaluating whether in vitro permeability data predict human brain exposure. Microphysiological systems (MPS

Environmental exposure risk of typical rubber antioxidant 6PPD and its oxidation product 6PPD-Q.

Environmental distribution, transformation pathways, and scenario-specific non-dietary intake of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) and 6PPD-quinone (6PPD-Q) are reviewed. Both compounds occur in water, air, dust, and soil, with distributions governed by traffic emissions, hydrological connectivity, particle properties, and environmental conditions. Runoff-affected waters are major hotspots for 6PPD and 6PPD-Q contamination and potential ecological risk. 6PPD transformation modifies compound-specific mobility, persistence, and hazard but does not consistently increase to

Preclinical Safety Evaluation of Human Lactoferrin Alpha (Effera®).

The preclinical safety of human-equivalent lactoferrin alpha (heqLFα; effera®), produced by Komagataella phaffii, was evaluated to support its use as a food ingredient in infant formula and products for young children. Genotoxicity was assessed using a bacterial reverse mutation (Ames) assay in five strains of Salmonella typhimurium and Escherichia coli, and an in vitro micronucleus assay in TK6 cells. Both studies followed OECD guidelines and were conducted up to the recommended limit concentrations (5000 µg/plate and 2000 µg/mL, respectively). A 14-day non-GLP juvenile dose range-finding stu

The role of mycotoxins in lipid metabolism and homeostasis: a systematic review.

BACKGROUND: Mycotoxins are widespread food contaminants that pose a significant global health concern. While their hepatotoxic, nephrotoxic, and carcinogenic effects are well established and characterized, their effects on adipose tissue biology and systemic lipid regulation remain incompletely understood. Given adipose tissue's central role in lipid homeostasis, endocrine regulation, and as a storage reservoir for adipophilic compounds, understanding these effects is critical. METHODS: This systematic review was conducted in accordance with the PRISMA 2020 guidelines, and prospectively regis

Metabolism and urinary excretion kinetics of 2,4-di-tert-butylphenol after a single oral dose: Characterisation of suitable exposure biomarkers for human biomonitoring.

2,4-Di-tert-butylphenol (DTBP) is a degradation product of the antioxidant and plastic additive tris(2,4-di-tert-butylphenyl)phosphite. DTBP can migrate into food or drinking water from high production volume polyolefins containing this additive, leading to potential human exposures, confirmed by findings of DTBP in blood and urine. Natural sources of DTBP have also been described. We have recently developed a sensitive analytical method for the quantification of urinary DTBP and its postulated oxidised metabolite 2-tert-butyl-4-(2-hydroxy-1,1-dimethylethyl)phenol (DTBP-4-OH) after enzymatic c

Maternal Diabetes Mellitus and Child Neurodevelopmental Disorders: Rethinking Causality in Light of Environmental Pollutants.

Numerous epidemiological studies have linked maternal diabetes mellitus (DM) to neurodevelopmental disorders in offspring, such as autism spectrum disorder and attention-deficit/hyperactivity disorder. However, recent evidence challenges a direct causal relationship: sibling-matched analyses show similar risks regardless of gestational DM status, and glycemic control during pregnancy does not consistently predict neurodevelopmental outcomes. We propose that exposure to environmental pollutants-rather than DM itself-may underlie these associations. Although usually examined separately, many pol

Transport of Perfluoroalkyl Substances (PFAS) by Three Renal Transporters: Implications for PFAS Bioaccumulation Mechanisms.

The long biological half-lives of per- and polyfluoroalkyl substances (PFAS) in humans have been linked to interactions with renal transport proteins and polypeptides. However, only a limited number of kidney transporters have been studied for their ability to transport specific PFAS. Moreover, few studies have investigated whether PFAS alternatives serve as substrates for these transporters. In this study, we focused on one renal influx transporter, organic anion transporter 1 (OAT1) and two efflux transporters (P-glycoprotein (P-g) and breast cancer resistance protein (BCRP)) involved in PFA

Stable isotope-assisted mass spectrometry reveals in vivo distribution, metabolism, and excretion of tire rubber-derived 6PPD-quinone in mice.

6PPD-quinone (6PPD-Q) has been identified as a ubiquitous contaminant in the surrounding locality, including air particles, roadside soils, dust, and water. Recently, the prevalence of 6PPD-Q in human urine has accentuated the urgency for investigating its biological fate. To address this, we conducted a stable isotope-assisted high-resolution mass spectrometry (HRMS) assay to unveil the distribution, metabolism, excretion, and toxicokinetic properties of this contaminant in a mouse model. Mice were fed with a single dose of deuterated 6PPD-Q-d5 at human-relevant exposure levels. Results indic

The environmental pollutant BDE-47 modulates immune responses in invitro and in vivo murine models.

2,2',4,4'-tetra-bromodiphenyl ether (BDE-47) is widespread in the environment and biological samples. Its association with health risks is an increasing concern, yet information on BDE-47 immunotoxicity remains limited. This study investigated the impact of BDE-47 on innate and adaptive immune responses through in vitro and in vivo approaches. BDE-47's capacity to directly induce cell responses and modulate responses induced by known stimuli was studied in vitro using the RAW 264.7 murine macrophage cell line and spleen-derived lymphocytes, and in vivo using keyhole limpet hemocyanin (KLH)-imm

Perimortem Distribution of U-47700, Tramadol and Their Main Metabolites in Pigs Following Intravenous Administration.

In spite of a decreasing number of new releases, new synthetic opioids (NSOs) are gaining increasing importance in postmortem (PM) forensic toxicology. For the interpretation of analytical results, toxicokinetic (TK) data, e.g., on tissue distribution, are helpful. Concerning NSOs, such data are usually not available due to the lack of controlled human studies. Hence, a controlled TK study using pigs was carried out, and the tissue distribution of U-47700 and tramadol as reference was examined. Twelve pigs received an intravenous dose of 100 µg/kg body weight (BW) U-47700 or 1,000 µg/kg BW tra
