Pulpal response to prolonged dentinal exposure to sodium hypochlorite.

AIM: The aim of this study was to examine the effect of undiluted NaOCl on vital pulp tissue when applied to freshly cut dentine. METHODOLOGY: Class V cavities were prepared to a depth of 2 mm in 20 teeth in four Beagle dogs. The cavities on one side of each dog were irrigated continuously for 5 min with 5.25% NaOCl, whilst the cavities on the opposite side in each dog were irrigated with saline for the same length of time. Prior to filling each cavity with Cavit, they were again rinsed with saline and gently dried with an air stream. The dogs were sacrificed to allow for investigation of pul

Epithelial fibroblast growth factor receptor 1 regulates enamel formation.

The interaction between epithelial and mesenchymal tissues plays a critical role in the development of organs such as teeth, lungs, and hair. During tooth development, fibroblast growth factor (FGF) signaling is critical for regulating reciprocal epithelial and mesenchymal interactions. FGF signaling requires FGF ligands and their receptors (FGFRs). In this study, we investigated the role of epithelial FGF signaling in tooth development, using the Cre-loxp system to create tissue-specific inactivation of Fgfr1 in mice. In K14-Cre;Fgfr1(fl/fl) mice, the apical sides of enamel-secreting amelobla

[Three dimensional study of the pulpal vascular changes during inflammation].

In this study, the changes of canine pulpal vascular architecture, at various stage of growth when subjected so acute and chronic inflammation were examined through a scanning electron microscope using vascular corrosive resin casts. The following results were obtained. 1. The sites of leakage of MMA resin out of the blood vessels coincided with the site where microleakage occurred, thus suggesting that resin cast method is a useful technique to study the area where vascular permeability was increased. 2. In an acute inflammation, leakage of resin out of blood vessels was mainly observed at th

A pharmacodynamic investigation into the efficacy of osteoprotegerin during aseptic inflammation.

BACKGROUND: Osteoprotegerin (OPG), as an osteoclast antagonist, limits mineralised tissue resorption under physiological conditions. Previous work investigating OPG in a rat periodontal ligament (PDL) ankylosis model found no inhibitory effect on osteoclasts when OPG was administered at a dosage of 2.5mg/kg. AIMS: The object of this study was to determine whether dosages higher than 2.5 mg/kg of OPG were required to limit osteoclastic activity in an aseptic inflammatory model in rats. MATERIALS AND METHODS: Dry ice was applied for 15 minutes to the upper right first molar crown of eighteen,

Citrate Transporter NaCT and Enamel Mineralization: The Slc13a5R337* Mouse Model.

Solute Carrier Family 13 Member 5 (SLC13A5) encodes the sodium-dependent citrate cotransporter NaCT, which mediates citrate transport across cell membranes. Pathogenic variants in SLC13A5 cause developmental and epileptic encephalopathy 25 with amelogenesis imperfecta, DEE25; OMIM #615905, a debilitating autosomal recessive disorder. To better define the role of NaCT in ameloblast function and enamel mineralization, we used CRISPR/Cas9 genome editing to generate Slc13a5R337* knock-in mice that terminate NaCT translation at the Arg337 codon, which is homologous to the human SLC13A5R333* variant

Pulpal reactions in rat incisors to Caridex.

This study examined the in vivo effects of Caridex, a chemomechanical caries removal system, on rat pulpal tissue. Rat incisors were opened and the pulps exposed to Caridex or physiological saline and sealed with calcium hydroxide. After various time periods, teeth were extracted and examined by light microscopy. Histological evaluation revealed an almost identical response in both test and control teeth which consisted of a transient inflammatory reaction and a limited necrosis in adjacent pulp tissue. Within seven days, formation of hard tissue matrix was seen below the necrotic area and on

GelMA/TCP nanocomposite scaffold for vital pulp therapy.

Pulp capping is a necessary procedure for preserving the vitality and health of the dental pulp, playing a crucial role in preventing the need for root canal treatment or tooth extraction. Here, we developed an electrospun gelatin methacryloyl (GelMA) fibrous scaffold incorporating beta-tricalcium phosphate (TCP) particles for pulp capping. A comprehensive morphological, physical-chemical, and mechanical characterization of the engineered fibrous scaffolds was performed. In vitro bioactivity, cell compatibility, and odontogenic differentiation potential of the scaffolds in dental pulp stem cel

Wnt5a regulates dental follicle stem/progenitor cells of the periodontium.

INTRODUCTION: Dental follicle gives rise to one or several tissues of the periodontium including the periodontal ligament, cementum and/or alveolar bone. Whether Wnt5a is expressed in the postnatal periodontium or regulates dental follicle stem/progenitor cells is unknown. METHODS: Dental follicle stem/progenitor cells were isolated from postnatal day 1 (p1) to p11 from rat mandibular first molars. Immunolocalization mapped Wnt5a expression in the alveolar bone, periodontal ligament, and the developing ameloblast and odontoblast layers. Mononucleated and adherent cells were isolated from p7 d

Widespread expression of tartrate-resistant acid phosphatase (Acp 5) in the mouse embryo.

Tartrate-resistant acid phosphatase (TRAP, Acp 5) is considered to be a marker of the osteoclast and studies using 'knockout' mice have demonstrated that TRAP is critical for normal development of the skeleton. To investigate the distribution of TRAP in the mammalian embryo, cryostat sections of 18 d murine fetuses were examined by in situ hybridisation, immunohistochemistry and histochemical reactions in situ. Abundant expression of TRAP mRNA was observed in the skin and epithelial surfaces of the tongue, oropharynx and gastrointestinal tract including the colon, as well as the thymus, ossify

Correlation between clinical and histologic pulp diagnoses.

INTRODUCTION: Clinicians routinely face conditions in which they have to decide whether the dental pulp can be saved or not. This study evaluated how reliable the clinical diagnosis of normal pulp/reversible pulpitis (savable pulp) or irreversible pulpitis (nonsavable pulp) is when compared with the histologic diagnosis. METHODS: The study material consisted of 95 teeth collected consecutively in a general practice over a 5-year period and extracted for reasons not related to this study. Based on clinical criteria, teeth were categorized as having normal pulps, reversible pulpitis, or irrever

The effect of etching on bacterial microleakage of an adhesive composite restoration.

OBJECTIVES: The incidence of bacterial microleakage, pulp inflammation and necrosis associated with dentine etching treatments prior to restoration are not known. Consequently, to resolve some of the controversy surrounding the effects and importance of vital dentine etching, the authors investigated these factors. METHODS: 110 standardised class V cavities were cut into buccal dentine, without exposing the pulp of teeth scheduled for extraction for orthodontic reasons. Cavities were either left unetched, or etched with the non-equivalent treatments of phosphoric acid gel for 60s or Ethylened

Genetic evidence for key roles of decorin and biglycan in dentin mineralization.

Targeted disruption of the dentin sialophosphoprotein (DSPP) gene in the mice (Dspp(-/-)) results in dentin mineralization defects with enlarged predentin phenotype similar to human dentinogenesis imperfecta type III. Using DSPP/biglycan (Dspp(-/-)Bgn(-/0)) and DSPP/decorin (Dspp(-/-)Dcn(-/-)) double knockout mice, here we determined that the enlarged predentin layer in Dspp(-/-) teeth is rescued in the absence of decorin, but not in the absence of biglycan. However, Fourier transform infrared (FTIR) spectroscopy analysis reveals similar hypomineralization of dentin in both Dspp(-/-)Bgn(-/0) a
