Targeted degradation of proteins has emerged as a powerful method for modulating protein homeostasis. Identification of suitable degraders is essential for achieving effective protein degradation. Here, we present a non-covalent degrader construction strategy, based on a modular supramolecular co-assembly system consisting of two self-assembling peptide ligands that bind cell membrane receptors and the protein of interest simultaneously, resulting in targeted protein degradation. The developed lysosome-targeting co-assemblies (LYTACAs) can induce lysosomal degradation of extracellular protein

Hepatic diseases are turning into one of the few diseases that cannot be effectively cured due to some reasons although various receptors existed in the liver. Currently, several passive targeting delivery systems have been used in the drug/gene delivery for the treatment of hepatic diseases. For example, Zinostatin stimalamer (trade name of Smancs(®)), a drug-polymer conjugate, was launched in Japan in 1994, which treats hepatocellular carcinoma. More improtantly, different measures would be taken in accord to the specified cell that was lesioned or dysfunctioned via interaction between homin

Accurately quantifying protein-protein binding at the single-molecule level is essential for understanding the mechanisms of viral infection and therapeutic targeting. Here, we use solid-state nanopores (SSNs) to detect complex formation between the SARS-CoV-2 Spike receptor-binding domain (Spike RBD) and the alternative host receptors KREMEN1 and Asialoglycoprotein Preceptor 1 (ASGR1). Single-molecule translocation events were analyzed using unsupervised Gaussian mixture modeling and a control-anchored semisupervised classification framework to resolve overlapping free-protein and complex pop

Hepatic fibrosis is characterized by persistent hepatocyte injury and activation of hepatic stellate cells (HSCs), leading to excessive extracellular matrix deposition and impaired liver function. 5-hydroxytryptamine (5-HT) promotes HSCs activation and fibrogenesis via the 5-HT2A receptor (5-HT2AR, a GPCR). Herein, we developed novel bifunctional lysosome-targeting chimeras (LYTACs) by conjugating a potent 5-HT2AR antagonist to a triantennary N-acetylgalactosamine (tri-GalNAc) ligand by using precisely optimized linkers. LY-res-3 induced potent, rapid, and selective degradation of endogenous 5

A cationic, dendrimer-like oligo(aminoamide) carrier with four-arm topology based on succinoyl tetraethylene pentamine and histidines, cysteines, and N-terminal azido-lysines was screened for plasmid DNA delivery on various cell lines. The incorporated azides allow modification with various shielding agents of different polyethylene glycol (PEG) lengths and/or different ligands by copper-free click reaction, either before or after polyplex formation. Prefunctionalization was found to be advantageous over postfunctionalization in terms of nanoparticle formation, stability, and efficacy. A lengt

Many encapsulated bacteria use capsules to cause invasive diseases. However, it remains largely unknown how the capsules enhance bacterial virulence under in vivo infection conditions. Here we show that the capsules primarily target the liver to enhance bacterial survival at the onset of blood-borne infections. In a mouse sepsis model, the capsules enabled human pathogens Streptococcus pneumoniae and Escherichia coli to circumvent the recognition of liver-resident macrophage Kupffer cells (KCs) in a capsular serotype-dependent manner. In contrast to effective capture of acapsular bacteria by K

Hepatocellular carcinoma (HCC) is the fifth most common malignancy worldwide. There is a substantial need for new chemotherapeutic drugs effective against this tumor. Doxorubicin (DOXO), used for chemoembolization of HCCs, is poorly efficacious when administered systemically at conventional doses; dose escalation is hindered by unacceptable toxicity. Here, we review preclinical experiments showing that the efficacy of DOXO against HCCs and its safety increased following conjugation to lactosaminated human albumin (L-HSA). L-HSA-DOXO was initially prepared to improve the anticancer activity of

This study explores a novel strategy to enhance liver-targeted gene delivery, crucial for treating metabolic disorders. While AAV-based gene therapy holds promise, high vector doses raise safety concerns, often requiring the use of corticosteroids and immunosuppression to mitigate immune adverse events. Optimizing capsids to improve hepatocyte transduction efficiency is key to reducing these risks. We employed bioconjugation chemistry to target the Asialoglycoprotein receptor (ASGPR), highly expressed on hepatocytes. Covalent attachment of GalNAc-derived ligands to rAAV2 capsids significantly

The mechanisms involved in regulating von Willebrand factor (VWF) clearance remain poorly understood. However recent studies have shown that macrophages play a critical role in regulating the half-life of VWF, and have identified specific lectin (including asialoglycoprotein, macrophage galactose-type lectin, Sigec-5 and C-type lectin domain family 4 member M) and scavenger receptors (including low-density lipoprotein receptor-related protein-1, scavenger receptor A1 and stabilin-2) that are involved in VWF clearance. Further studies will be required to determine the relative importance of the

BACKGROUND: Chronic infection with the Hepatitis B virus (HBV) is a major contributor to advanced liver diseases. The HBV replication process and the in vivo models available to study this virus are complex. HBV entry into hepatocytes involves a multifaceted interplay of viral particles, host cell receptors, and endocytic pathways. Key host receptors, including sodium taurocholate co-transporting polypeptide (NTCP), epidermal growth factor receptor (EGFR), and asialoglycoprotein receptor (ASGPR), facilitate viral internalization via clathrin-mediated endocytosis.
SUMMARY: Following entry, the

Autoimmune hepatitis (AIH) is a progressive inflammatory liver disease characterized by specific duration of symptoms (above 6 months), 3- to 10-fold elevation in serum aminotransferase activities, 2-fold elevation in gamma-globulin, high titers (above 1:40) of circulating autoantibodies. Antinuclear antibodies (ANA), smooth muscle antibodies (SMA), and antibodies to liver/kidney microsome type 1 (anti-LKM1) are the commonly available markers of AIH. SMA and ANA lack disease specificity and they are present commonly in different liver diseases. Anti-LKM1 usually connote a type 2 form of AIH wh

Immunoglobulin A (IgA) is important in local immunity and is also abundant in the blood. This study aimed to evaluate the effects of serum IgA on cultured lung microvascular endothelial cells (HMVEC-Ls), which are involved in the pathogenesis of inflammatory lung diseases. Serum IgA induced adhesion molecules and inflammatory cytokine production from HMVEC-Ls, and enhanced adhesion of peripheral blood mononuclear cells to HMVEC-Ls. In contrast, migration, proliferation, and tube formation of HMVEC-Ls were significantly suppressed by serum IgA. Experiments with siRNAs and western blotting revea

The molecular mechanisms used by the gonococcus to initiate infection exhibit gender specificity. The clinical presentations of disease are also strikingly different upon comparison of gonococcal urethritis to gonococcal cervicitis. An intimate association occurs between the gonococcus and the urethral epithelium and is mediated by the asialoglycoprotein receptor. Gonococcal interaction with the urethral epithelia cell triggers cytokine release, which promotes neutrophil influx and an inflammatory response. Similarly, gonococcal infection of the upper female genital tract also results in infla

Proper functioning of the endocrine stress axis requires communication between the stress axis and other regulatory mechanisms. We here describe an intimate interplay between the stress axis and recombinant human leptin (rhLeptin) in a teleostean fish, the common carp Cyprinus carpio. Restraint stress (by netting up to 96h) increased plasma cortisol but did not affect hepatic leptin expression. Perifusion of pituitary glands or head kidneys with rhLeptin revealed direct effects of rhLeptin on both tissues. RhLeptin suppresses basal and CRF-induced ACTH-secretion in a rapid and concentration-de

The potential of reconstituted Sendai viral envelopes containing only the fusion protein (F-virosomes) was evaluated for a targeted cytosolic delivery of lysozyme to human hepatoblastoma cells (HepG2) in culture. 125I-Lysozyme loaded into F-virosomes was used to monitor its fusion-mediated transfer to the HepG2 cells. Using fusion assay based on the transfer of water soluble probe, we have demonstrated the existence of aqueous connection between F-virosomes and target cells. Target specificity of the F-virosomes was ensured by the strong interaction between terminal beta-galactose moiety of F

While targeted therapies for hepatocellular carcinoma (HCC) have advanced, their clinical benefits remain constrained by limited efficacy, off-target toxicity, and the frequent emergence of drug resistance and recurrence. To address these challenges, we developed a multivalent DNA nanoflower-based dual-targeting LYTAC (lysosome-targeting chimera) platform tailored for HCC. This NanoLYTAC integrates two key functional elements: (i) a high-density array of bispecific aptamers that simultaneously recognize glypican-3 (GPC-3)-a highly HCC-specific surface marker-and vascular endothelial growth fac

N-glycosylation is one of major post-translational modifications in nature, and it is essential for protein structure and function. As hydrophilic moieties of glycoproteins, N-glycans play important roles in protein stability. They protect the proteins against proteolytic degradation, aggregation, and thermal denaturation through maintaining optimal conformations. There are extensive evidences showing the involvement of N-glycans in the pharmacodynamics and pharmacokinetics of recombinant therapeutic proteins and antibodies. Highly sialylated complex-type glycans enable the longer serum half-l

Two pilus receptors are identified for the pathogenic Neisseria, CD46 and complement receptor 3. An intimate association between the asialoglycoprotein receptor and gonococcal lipooligosaccharide mediates invasion of primary, male urethral epithelial cells (UECs); however, studies to identify pilus receptors on these cells have not been performed. Based on our previous studies we reasoned that the I-domain-containing (IDC), alpha(1)- and alpha(2)-integrins might serve as pilus receptors on UECs and on urethral tissue. Confocal microscopy revealed colocalization of pilus with alpha(1) and alpha

IgA nephropathy (IgAN) is initiated by glomerular deposition of polymeric IgA1(pIgA1). In IgAN pIgA production is reduced in the mucosal immune system, perhaps mediated by a mucosal gamma delta T cell defect, and mucosal IgA responses to immunisation are impaired. But pIgA1 production by the marrow is increased. Human pIgA1 has an O-glycosylated hinge region unique to circulating immunoglobulins and there is reduction of galactosyl residues in the hinge of serum IgA1 in IgAN and Henoch-Schönlein nephritis. This reduced galactosylation may be due to a functional defect in plasma cell beta 1,3-g
