Spectroscopic, Electrochemical, and Biological Assays of Copper-Binding Molecules for Screening of Different Drugs and Plant Extracts against Neurodegenerative Disorders.

Neurodegenerative disorders, caused by prone-to-aggregation proteins, such as Alzheimer disease or Huntington disease, share other traits such as disrupted homeostasis of essential metal ions, like copper. In this context, in an attempt to identify Cu2+ chelating agents, we study several organic compounds (ethylenediaminetetraacetic acid, phenylenediamine, metformin, salicylate, and trehalose) and organic extracts obtained from Bacopa monnieri L., which has been used in Ayurvedic therapies and presented a broad spectrum of biological properties. For this purpose, UV-visible spectroscopy analys

Metabolite and Mechanistic Basis of Antifungal Property Exhibited by Endophytic Bacillus amyloliquefaciens BmB 1.

Plants are ubiquitously colonized by endophytic microorganisms which contribute significantly to plant health through production of plant growth regulators or disease suppression. In the present study, an endophytic bacterial isolate designated as BmB 1 with significant antifungal and plant growth promoting properties was isolated from the stem tissue of Bacopa monnieri (L.) Pennell. The isolate was studied in detail for the molecular and chemical basis of its bioactivity which proved it to have the presence of surfactin, iturin, and type I polyketide synthase (PKS) genes. Investigating the Effects of a Multinutrient Supplement on Cognition, Mood and Biochemical Markers in Middle-Aged Adults with 'Optimal' and 'Sub-Optimal' Diets: A Randomized Double Blind Placebo Controlled Trial.

Background: Previous randomized controlled trials examining cognitive and mood effects of combination multivitamin supplements in healthy, non-clinical adults have reported mixed results. One purported explanation for this is that the dietary status of participants at the start of supplement interventions may influence the magnitude of the effect of supplementation. Methods: In this study, we evaluated the effect of a multinutrient formula containing B group vitamins, Bacopa monniera and Ginkgo biloba on memory, attention, mood and biochemical markers of nutrient status in middle-aged adults (

Identification of Three Cytochrome P450 Enzymes Responsible for the Biosynthesis of Jujubogenin and Its De Novo Production in Yeast.

Jujubosides are the principal bioactive constituents of the traditional Chinese medicine Ziziphi Spinosae Semen and are well-known for their sedative, anxiolytic, and sleep-promoting activities. These compounds are dammarane-type triterpenoid saponins characterized by a unique aglycone, jujubogenin, featuring a 5,6-spiroketal moiety that forms a rigid D/E/F tricyclic scaffold. However, the enzymatic basis underlying the formation of this distinctive spiroketal structure remains elusive. In this study, we identified and functionally characterized three cytochrome P450 enzymes from wild jujubes

In vivo and in vitro perspectives in Parkinson's disease: Mechanisms and the role of phytomedicine.

Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by dopaminergic neuronal loss, oxidative stress, neuroinflammation, and α-synuclein aggregation. Despite advances, current treatments only offer symptomatic relief without altering disease progression. This review aims to evaluate the therapeutic potential of plantderived phytochemicals in PD through evidence from both in vivo and in vitro experimental models. Key findings indicate that rodent models (e.g., MPTP, rotenone) and neuronal cell lines (e.g., SH-SY5Y) help replicate PD pathology and mechanistic insights. Phy

Bacoside-A enhances temozolomide-induced cytotoxicity in U87MG glioblastoma cells via ROS-mediated apoptosis and EGFR/MAPK inhibition.

Temozolomide (TMZ) is the primary chemotherapeutic agent for glioblastoma multiforme (GBM), but its effectiveness is limited by resistance, recurrence, and toxicity. Combining natural compounds with TMZ is a promising approach to overcome these challenges. Bacoside-A, a brain tonic derived from Bacopa monnieri with proven anticancer properties, was evaluated for its chemotherapeutic potential in U87MG cells. Our study demonstrates that the combination of Bacoside-A and TMZ significantly enhanced intracellular TMZ accumulation and promoted ROS-mediated apoptosis through Ca2+ influx, mitochondri

Integrative Phytotherapy for Neurodegenerative Disorders: Bridging Traditional Botanicals, Bioactive Compounds, and Nanotechnology.

Neurodegenerative disorders (NDDs) such as Alzheimer's, Parkinsons etc., are progressive and debilitating conditions that have little therapeutic intervention. Existing pharmacological interventions mainly provide symptomatic relief and do not respond to the pathophysiology of these diseases, which is complex and multifactorial and includes oxidative stress, mitochondrial dysfunction, neuroinflammation, and protein misfolding. The synthesis of emerging evidence regarding the neuroprotective efficacy of phytotherapy is presented in this review, and the focus is on such traditional medicinal pla

Plant-derived nootropics and human cognition: A systematic review.

Substances with modulatory capabilities on certain aspects of human cognition have been revered as nootropics from the dawn of time. The plant kingdom provides most of the currently available nootropics of natural origin. Here, in this systematic review, we aim to provide state-of-the-art information regarding proven and unproven effects of plant-derived nootropics (PDNs) on human cognition in conditions of health and disease. Six independent searches, one for each neurocognitive domain (NCD), were performed in parallel using three independent scientific library databases: PubMed, Cochrane and

The antidepressant-like effect of bacopaside I: possible involvement of the oxidative stress system and the noradrenergic system.

In the present study, the antidepressant-like effect of bacopaside I, a saponin compound present in the Bacopa monniera plant, was evaluated by behavioral and neurochemical methods. Bacopaside I (50, 15 and 5 mg/kg) was given to mice via oral gavage for 7 successive days. The treatment significantly decreased the immobility time in mouse models of despair tests, but it did not influence locomotor activity. Neurochemical assays suggested that treatment by bacopaside I (50, 15 and 5 mg/kg) improved brain antioxidant activity to varying degrees after the behavioral despair test. Bacopaside I (15

Natural products proposed for the management of Huntington's disease (HD): a comprehensive review.

Huntington's disease (HD), a neurodegenerative disease, typically begins in the prime of adulthood, followed by a gradual onset of specific mental abnormalities and cognitive and physical impairment. To the best of our knowledge, no medication exists to totally stop the progression of HD. Among numerous therapy techniques, extensive literature reviews have confirmed the medicinal importance of natural products in HD experimental models. This review provides a literature survey of natural compounds and medicinal plants used as neuroprotective agents against HD. Relevant studies were found in a

Computational Fragment-Based Design of Phytochemical Derivatives as EGFR Inhibitors.

Epidermal growth factor receptor (EGFR) is a potential target with disease modifying benefits against Alzheimer's disease (AD). Repurposing of FDA approved drugs against EGFR have shown beneficial effect against AD but are confined to quinazoline, quinoline and aminopyrimidines. Futuristically, the possibility of acquiring drug resistance mutation as seen in the case of cancer could also hamper AD treatment. To identify novel chemical scaffolds, we rooted on phytochemicals identified from plants such as Acorus calamus, Bacopa monnieri, Convolvulus pluricaulis, Tinospora cordifloia, and Withani

Antiamnesic effect of B. monniera on L-NNA induced amnesia involves calmodulin.

Amnesia may result from ageing, chronic drug abuse or head injury and there are limited therapeutic strategies to such conditions. We have shown that Bacopa monniera, a memory enhancing drug can reverse both diazepam and scopolamine induced amnesia in mice. In order to understand the downstream effects of B. monniera, this study was designed to investigate how B. monniera antagonizes MK801, an NMDA receptor antagonist and N(omega)-Nitro-L-arginine (L-NNA), a nitric oxide synthase inhibitor. We compared the degree of reversal B. monniera imparts on MK801 and L-NNA induced anterograde amnesia in

Biochemical modulators of synaptic plasticity: New horizons in Alzheimer's disease treatment.

Synaptic dysfunction is the earliest and most critical pathological feature of Alzheimer's disease (AD), directly contributing to cognitive decline. This review provides an integrative overview of the molecular and biochemical modulators governing synaptic plasticity and their disruption in AD. We discuss how the collective impairment of Aβ aggregation, tau pathology, calcium imbalance, oxidative stress, and neuroinflammation affects dendritic spine morphology and synaptic connectivity. Particular attention is given to neurotrophins such as brain-derived neurotrophic factor and TrkB signaling,

Cholinesterase Inhibitors from Plants and Their Potential in Alzheimer's Treatment: Systematic Review.

INTRODUCTION: Alzheimer's disease (AD) is a neurodegenerative disorder characterized by memory loss and cognitive decline, primarily due to dysfunction of acetylcholine caused by acetylcholinesterase and butyrylcholinesterase. While synthetic cholinesterase inhibitors like donepezil, rivastigmine, and galantamine are commonly used, they have notable side effects, prompting interest in natural alternatives. Medicinal plants, rich in bioactive compounds like flavonoids and alkaloids, have shown potential as cholinesterase inhibitors with additional antioxidants and anti-inflammatory benefits. Th
