The therapeutic interventions of human hypertrophic scars (HHS) remain puzzle largely due to the lack of accepted models. Current HHS models are limited by their inability to mimic native scar architecture and associated pathological microenvironments. Here, we create a 3D functional HHS model by preformed cellular aggregates (PCA) bioprinting, firstly developing bioink from scar decellularized extracellular matrix (ECM) and alginate-gelatin (Alg-Gel) hydrogel with suitable physical properties to mimic the microenvironmental factors, then pre-culturing patient-derived fibroblasts in this bioin

In esophageal pathologies, such as esophageal atresia, cancers, caustic burns, or post-operative stenosis, esophageal replacement is performed by using parts of the gastrointestinal tract to restore nutritional autonomy. However, this surgical procedure most often does not lead to complete functional recovery and is instead associated with many complications resulting in a decrease in the quality of life and survival rate. Esophageal tissue engineering (ETE) aims at repairing the defective esophagus and is considered as a promising therapeutic alternative. Noteworthy progress has recently been

Bioprinting is an emerging field in regenerative medicine. Producing cell-laden, three-dimensional structures to mimic bodily tissues has an important role not only in tissue engineering, but also in drug delivery and cancer studies. Bioprinting can provide patient-specific spatial geometry, controlled microstructures and the positioning of different cell types for the fabrication of tissue engineering scaffolds. In this brief review, the different fabrication techniques: laser-based, extrusion-based and inkjet-based bioprinting, are defined, elaborated and compared. Advantages and challenges

Organ manufacturing is an attractive high-tech research field which can solve the serious donor shortage problems for allograft organ transplantation, high throughput drug screening, and energy metabolism model establishment. How to integrate heterogeneous cell types along with other biomaterials to form bioartificial organs is one of the kernel issues for organ manufacturing. At present, three-dimensional (3D) bioprinting of adipose-derives stem cell (ADSC) containing hydrogels has shown the most bright futures with respect to overcoming all the difficult problems encountered by tissue engine

Advancements in regenerative medicine are increasingly driven by the convergence of stem cell biology and bioprinting technologies. Stem cells have the potential to be used to engineer functional tissues and organs because of their innate ability to self-renew and differentiate. But conventional methods that use xenografts and decellularized scaffolds have issues with biocompatibility and architectural complexity. This review critically evaluates bioprinting strategies tailored for stem cell applications, including the selection of stem cell sources, bioink formulation, crosslinking methodolog

Three-dimensional (3D) bioprinting is an evolving biofabrication approach in regenerative medicine with the potential to overcome many limitations of conventional reconstructive techniques, including donor-site morbidity, limited tissue availability, and suboptimal restoration of form and function. Recent advances in biofabrication have accelerated the development of patient-specific living constructs for reconstructive applications. This narrative review synthesizes contemporary evidence on the use of 3D bioprinting in reconstructive surgery, emphasizing developments most relevant to plastic

The engineering of functional meniscal grafts remains elusive, largely due to an inability to recapitulate the highly organized collagen architecture of the native tissue which is integral to its biomechanical function. In this study, we investigated whether external geometric confinement can direct collagen alignment in fibrocartilaginous tissues generated by mesenchymal stem/stromal cells (MSCs). First, MSCs were cast within non-adhesive agarose channels, which supported the development of fibrocartilaginous tissues with collagen fibres aligned parallel to the long axis of the confining agar

Progress of breakthrough Roll Porous Scaffold (RPS) 3D Bioprinting technology with great potential for personalized medicine, targeted therapy and overcoming the shortage of organs for implantation achieves first physical confirmation. The efficiency of RPS for the formation of solid organoid can exceed liters per hour with ~10-35 µm cells in 5, 18, and 84 pL drops due to significantly increased print exposure for cell filtration and based on the lines of many usual Kyocera inkjet printheads "KJ4B-1200," "KJ4C-0360," and "KJ4A-0300, KJ4B-0300" with resolution of 1200, 360, and 300 DPI accordin

BACKGROUND: Choriocarcinoma is a rare and highly aggressive trophoblastic tumor. Although in-depth research has been conducted on the treatment options for choriocarcinoma, especially chemotherapy regimens, there are still 10% to 20% of choriocarcinoma patients who develop resistance or experience tumor recurrence. The research on the pathogenesis of choriocarcinoma and its treatment remains a major challenge and an unresolved clinical issue. Accurately replicating the key characteristics of choriocarcinoma in preclinical tumor models is crucial for research into tumor mechanisms and treatment

Tissue engineering offers immense potential for addressing the unmet needs in repairing tissue damage and organ failure. Vascularization, the development of intricate blood vessel networks, is crucial for the survival and functions of engineered tissues. Nevertheless, the persistent challenge of ensuring an ample nutrient supply within implanted tissues remains, primarily due to the inadequate formation of blood vessels. This issue underscores the vital role of the human vascular system in sustaining cellular functions, facilitating nutrient exchange, and removing metabolic waste products. In

The development of perfusable and multiscale vascular networks remains one of the largest challenges in tissue engineering. As such, there is a need for the creation of customizable and facile methods to produce robustly vascularized constructs. In this study, secondarily crosslinkable (clickable) poly(ethylene glycol)-norbornene (PEGNB) microbeads were produced and evaluated for their ability to sequentially support suspension bioprinting and microvascular self-assembly towards the aim of engineering hierarchical vasculature. The clickable PEGNB microbead slurry exhibited mechanical behavior

Biological scaffolds have been widely employed in wound healing applications, while their practical efficiency is compromised by insufficient oxygen delivery to the 3-dimensional constructs and inadequate nutrient supply for the long-term healing process. Here, we present an innovative living Chinese herbal scaffold to provide a sustainable oxygen and nutrient supply for promoting wound healing. Through a facile microfluidic bioprinting strategy, a traditional Chinese herbal medicine (Panax notoginseng saponins [PNS]) and a living autotrophic microorganism (microalgae Chlorella pyrenoidosa [MA

4D bioprinting is promising to build cell-laden constructs (bioconstructs) with complex geometries and functions for tissue/organ regeneration applications. The development of hydrogel-based 4D bioinks, especially those allowing living cell printing, with easy preparation, defined composition, and controlled physical properties is critically important for 4D bioprinting. Here, a single-component jammed micro-flake hydrogel (MFH) system with heterogeneous size distribution, which differs from the conventional granular microgel, has been developed as a new cell-laden bioink for 4D bioprinting. The efficient establishment of in vitro neural models that accurately mimic the structural and functional connectivity of neural networks is critical in neuroscience research. 3D bioprinting shows great potential for constructing sophisticated in vitro models with high freedom of design. However, mature neurons are delicate and susceptible to manipulation. Here, extrusion-based 3D bioprinting is employed to fabricate gelatin methacryloyl (GelMA)-based constructs containing embryonic day 18 (E18) rat cortical neurons, referred to as 3D neuMatrix. 3D neuMatrix displays favorable neuronal viabili
