[Construction, expression and enzymatic activity analysis of AUR1 eukaryotic expression vector of Botrytis cinerea].

In order to study the expression and the activity of inositol phosphorylceramide synthase (BcAUR1 gene) in Botrytis cinerea, we amplified BcAUR1 by RT-PCR from Botrytis cinerea, using the special primers with FLAG and BamH I/Xho I restriction sites. Recombinant pYES2-BcAUR1 was constructed to transform into Saccharomyces cerevisae deltayorl by LiAC. The expression of inositol phosphorylceramide (IPC) synthase and its activity were detected by Western blotting and HPLC, respectively. The results show that pYES2-BcAUR1 could express in uracil mutant deltayorl of Saccharomyces cerevisae. IPC synt

CtChi19: a valuable gene for improving resistance to Botrytis cinerea and Alternaria alternata in Carthamus tinctorius.

Safflower (Carthamus tinctorius L.) is an economically important crop with both medicinal and oil-producing values, However, its production is severely threatened by fungal diseases, including those caused by Alternaria alternata and other pathogenic fungi. To date, molecular investigations into the disease resistance mechanisms of safflower remain largely limited. In this study, proteomic analysis was performed on safflower following infection by A. alternata, identifying 490 and 921 differentially expressed proteins (DEPs) at 48 hours post-infection (hpi) and 72 hpi, respectively, Among thes

Transcription factor VlPAT2 enhances the resistance of grapevine to Botrytis cinerea by promoting ROS accumulation.

The overexpression of VlPAT2 enhances the resistance of grapevine and Arabidopsis thaliana to Botrytis cinerea, and promotes the accumulation of reactive oxygen species, as well as the expression of multiple PR genes and R genes. Grape grey mould caused by the necrotrophic fungus Botrytis cinerea causes severe economic losses to the grape industry. Identifying disease resistance genes and elucidating their mechanisms provide critical insights for molecular breeding. Here, we report a GRAS family transcription factor VlPAT2 from the grapevine cultivar 'Beta' (Vitis labrusca) that exhibits high

Mulberrin inhibits Botrytis cinerea for strawberry storage by interfering with the bioactivity of 14α-demethylase (CYP51).

Currently, chemical agents hold great promise in preventing and combating Botrytis cinerea. However, the antifungal mechanism of some agents for B. cinerea remains rather vague, imposing restrictions on the research and development of novel antifungal inhibitors. In this work, we discovered that mulberrin (MBN), a natural compound from the root bark of Ramulus Mori, with an IC50 of 1.38 μM together, demonstrated marked anti-14α-demethylase (CYP51) activity through high throughput virtual screening and in vitro bioactivity assay. The computational biology results demonstrated that MBN and its d

Evidence for protein degradation by Botrytis cinerea and relationships with alteration of synthetic wine foaming properties.

Botrytis cinerea is an important fungal pathogen particularly dreaded in the cool climate vineyard. It is responsible for important damage, especially the decrease in foamability of sparkling wines, such as Champagne. Different studies have shown that proteins are largely involved in the stabilization of Champagne foam despite their low concentration. Other works demonstrated changes in the electrophoretic characteristics of must proteins originating from botrytized grapes, although the cause of such alterations was never explained. In the first part of this study, results showed the release b

Methods for Evaluating the Efficacy of Microbial Biocontrol Agents Against Botrytis cinerea on Tomato Plant.

Botrytis cinerea, the causal agent of gray mold, is a highly adaptable necrotrophic fungus that causes significant losses in economically important crops such as tomatoes. Increasing resistance to fungicides and environmental concerns have stimulated interest in microbial biocontrol agents as part of integrated pest management strategies. This chapter presents standardized protocols for evaluating the efficacy of microbial biocontrol agents against B. cinerea on tomato plants under controlled conditions. Methods include preparation of pathogen and biocontrol inoculum, production of healthy pla

Perillaldehyde Functions as a Potential Antifungal Agent by Triggering Metacaspase-Independent Apoptosis in Botrytis cinerea.

Botrytis cinerea, the causal agent of gray mold, is an important plant pathogen causing preharvest and postharvest diseases. Due to the extensive use of commercial fungicides, fungicide-resistant strains have emerged. Natural compounds with antifungal properties are widely present in various kinds of organisms. Perillaldehyde (PA), derived from the plant species Perilla frutescens, is generally recognized as a potent antimicrobial substance and to be safe to humans and the environment. In this study, we demonstrated that PA could significantly inhibit the mycelial growth of B. cinerea and redu

A Reductase from the Phytopathogenic Fungus Botrytis cinerea Degrades Plant Antimicrobial Coumarins to Promote Infection.

Phytopathogenic fungi must contend with host immune responses to successfully colonize plants. These responses include the production of heterogenous antimicrobial compounds (phytoalexins), which plants deploy to intoxicate infecting microbes. Among these host defense compounds are the coumarins, a family of aromatic lactones widely present in dicotyledonous plants (dicots). The fungal necrotroph Botrytis cinerea (gray mold) infects a plethora of dicots, causing large losses in the produce, viticulture, and ornamental flower industries. B. cinerea detoxifies numerous host defense compounds to

Trichoderma asperellum Suppresses Gray Mold Caused by Botrytis cinerea and Enhances Disease Resistance of Blueberry.

With growing global recognition of the nutritional value of blueberries, their cultivation has expanded significantly across various regions. However, gray mold disease, caused by Botrytis cinerea, remains a major challenge in postharvest storage and transportation. In this study, two strains-Trichoderma asperellum BBR-A and B. cinerea BC7-1-were isolated and identified from the blueberry rhizosphere and diseased leaves. Co-culture assays demonstrated that BBR-A exhibits potent antagonistic activity, rapidly suppressing the growth of the pathogen BC7-1 through mycoparasitic behaviors, includin

Cross-Resistance Between Pseudomonas chlororaphis Strain AFS009 Metabolites (Howler EVO) and Fludioxonil in Botrytis cinerea.

Howler EVO is a biological fungicide based on metabolites of the bacterium Pseudomonas chlororaphis strain AFS009. One of the metabolites, pyrrolnitrin, is a chemical analogue of the phenylpyrrole fludioxonil used to manage gray mold of fruit crops caused by Botrytis cinerea. Resistance to fludioxonil in B. cinerea is well documented and linked to mutations in the transcription factor mrr1, leading to overexpression of the ATP-binding cassette transporter gene BcatrB. Moderately resistant isolates are designated MDR1 and MDR1h based on the specific variation of mutations in mrr1 and the level

Botrytis cinerea type II inhibitor of apoptosis BcBIR1 enhances the biocontrol capacity of Coniothyrium minitans.

Apoptosis-like programmed cell death is associated with fungal development, ageing, pathogenicity and stress responses. Here, to explore the potential of Botrytis cinerea type II inhibitor of apoptosis (IAP) BcBIR1 in elevating the biocontrol efficacy of Coniothyrium minitans, the BcBIR1 gene was heterologously expressed in C. minitans. Results indicated that the strains expressing BcBIR1 had higher rates of conidiation, mycelial growth and biomass growth than the wild-type strain. Moreover, BcBIR1 was found to inhibit apoptosis, indicating its role as an IAP in C. minitans. Under various abio

Leaf abaxial and adaxial surfaces differentially affect the interaction of Botrytis cinerea across several eudicots.

Eudicot plant species have leaves with two surfaces: the lower abaxial and the upper adaxial surface. Each surface varies in a diversity of components and molecular signals, resulting in potentially different degrees of resistance to pathogens. We tested how Botrytis cinerea, a necrotroph fungal pathogen, interacts with the two different leaf surfaces across 16 crop species and 20 Arabidopsis genotypes. This showed that the abaxial surface is generally more susceptible to the pathogen than the adaxial surface. In Arabidopsis, the differential lesion area between leaf surfaces was associated wi

Action mechanism for 3β-hydroxykaurenoic acid and 4,4-dimethylanthracene-1,9,10(4H)-trione on Botrytis cinerea.

The mechanism of action of the diterpenoid 3β-hydroxykaurenoic acid and the anthraquinone 4,4-dimethylanthracene-1,9,10(4H)-trione on the phytopathogenic fungus Botrytis cinerea was studied. The effect of both compounds on the respiratory process and on the membrane integrity of B. cinerea was evaluated. The results showed that 3β-hydroxykaurenoic acid inhibited the growth of this fungus by disrupting the plasmatic membrane. This compound also partially affected oxygen consumption of B. cinerea germinating conidia. Conversely, 4,4-dimethylanthracene-1,9,10(4H)-trione did not produce membrane d

Botrytis cinerea manipulates the antagonistic effects between immune pathways to promote disease development in tomato.

Plants have evolved sophisticated mechanisms to sense and respond to pathogen attacks. Resistance against necrotrophic pathogens generally requires the activation of the jasmonic acid (JA) signaling pathway, whereas the salicylic acid (SA) signaling pathway is mainly activated against biotrophic pathogens. SA can antagonize JA signaling and vice versa. Here, we report that the necrotrophic pathogen Botrytis cinerea exploits this antagonism as a strategy to cause disease development. We show that B. cinerea produces an exopolysaccharide, which acts as an elicitor of the SA pathway. In turn, the

Botrytis cinerea PMT4 Is Involved in O-Glycosylation, Cell Wall Organization, Membrane Integrity, and Virulence.

Proteins found within the fungal cell wall usually contain both N- and O-oligosaccharides. N-glycosylation is the process where these oligosaccharides (hereinafter: glycans) are attached to asparagine residues, while in O-glycosylation the glycans are covalently bound to serine or threonine residues. The PMT family is grouped into PMT1, PMT2, and PMT4 subfamilies. Using bioinformatics analysis within the Botrytis cinerea genome database, an ortholog to Saccharomyces cerevisiae Pmt4 and other fungal species was identified. The aim of this study was to assess the relevance of the bcpmt4 gene in

Fructan oligosaccharide priming alters apoplastic sugar dynamics and improves resistance against Botrytis cinerea in chicory.

Carbohydrates such as fructans can be involved in priming or defence stimulation, and hence potentially provide new strategies for crop protection against biotic stress. Chicory (Cichorium intybus) is a model plant for fructan research and is a crop with many known health benefits. Using the chicory-Botrytis cinerea pathosystem, we tested the effectiveness of fructan-induced immunity, focussing on different plant and microbial fructans. Sugar dynamics were followed after priming and subsequent pathogen infection. Our results indicated that many higher plants might detect extracellular levan ol

Efficacy of Peroxyacetic Acid for Controlling Gray Mold (Botrytis cinerea) in Stored Table Grapes in California.

Postharvest fruit rot diseases limit the storability of table grapes and can cause significant economic losses if left uncontrolled. Gray mold, caused by Botrytis cinerea, is a major postharvest disease of table grapes. This study evaluated the effectiveness of peroxyacetic acid (PAA) in controlling gray mold and other fruit rots and its effects on fruit quality on two grape cultivars. Four concentrations of PAA, 50, 100, 500, and 1000 µg mL-1 applied either by dipping or spraying were tested on grape berries that were inoculated with B. cinerea prior to the treatment. BcMFSR Is Required for Efficient Uptake of Exogenous dsRNA and Spray-Induced Gene Silencing in Botrytis cinerea.

Spray-induced gene silencing (SIGS) uses exogenous double-stranded RNA (dsRNA) to suppress pathogen virulence or growth-related genes, offering a promising strategy for controlling grey mould caused by Botrytis cinerea at the gene level. In this study, we investigated the molecular mechanisms underlying the efficient uptake of dsRNAs by B. cinerea, making significant progress towards addressing key gaps in our understanding of SIGS. We identified four B. cinerea genes, BcBMP1, BcSOD1, BcGAR2, and BcVELB, as effective targets for grey mould control by employing an Arabidopsis thaliana SIGS scre
