Intestinal hemodynamic effects of dextran-induced hyperviscosity in the cat.

A hyperviscous state with red cell aggregation and increased plasma viscosity was induced in cats by intravenous infusion of high molecular weight dextran. A segment of the small intestine was isolated with intact vascular and nervous supply and placed in a plethysmograph for determination of the intestinal tissue volume and capillary filtration coefficient. The intestinal blood flow was measured by a drop recorder unit. Measurements of intestinal blood flow, flow resistance and capillary filtration coefficient were performed during normotension and regional hypotension before and after infusi

Experimental investigations in adult respiratory distress syndrome (ARDS). Repeated pulmonary lavage in LEWE-mini-pigs. I. Pulmonary mechanics, gas exchange, and pulmonary hemodynamics.

Disturbed water balance at the alveolo-capillary membrane resulting in decreased pulmonary compliance is one of the functional characteristics in the Adult Respiratory Distress Syndrome (ARDS). In artificially ventilated LEWE Mini Pigs repeated pulmonary lavage was performed. The aim was to produce a state like ARDS by affecting the lung alveolar surfactant system which is included into the sum of factors which are responsible for the normal water balance of the alveolo-capillary membrane. This condition was supposed to be maximized if the lung compliance reached its lowest value after pulmona

Effect of calcium bodesilate (doxium) on circulatory disorders of the retina with special emphasis on diabetic retinopathy.

The authors examined the effect of calcium dobesilate (Doxium ¿) on 85 subjects with diabetic retinopathy, occlusion of the central vein of the retina and other retinopathies of vascular origin. Capillary resistance measured on the skin and conjunctiva showed, as compared with 100 healthy subjects, a significant decrease. The efficacy of long-term treatment with Doxium was highly significant in restoring CR to normal. Visual acuity improved in 45.8% of simple DR cases, and became stabilized in 27.5% of cases with malignant DR. Hyperpermeability of retinal vessels decreased in 60% of simple DR

Endothelial survival factors and spatial completion, but not pericyte coverage of retinal capillaries determine vessel plasticity.

Pericyte loss and capillary regression are characteristic for incipient diabetic retinopathy. Pericyte recruitment is involved in vessel maturation, and ligand-receptor systems contributing to pericyte recruitment are survival factors for endothelial cells in pericyte-free in vitro systems. We studied pericyte recruitment in relation to the susceptibility toward hyperoxia-induced vascular remodeling using the pericyte reporter X-LacZ mouse and the mouse model of retinopathy of prematurity (ROP). Pericytes were found in close proximity to vessels, both during formation of the superficial and th

Amyloid β oligomers constrict human capillaries in Alzheimer's disease via signaling to pericytes.

Cerebral blood flow is reduced early in the onset of Alzheimer's disease (AD). Because most of the vascular resistance within the brain is in capillaries, this could reflect dysfunction of contractile pericytes on capillary walls. We used live and rapidly fixed biopsied human tissue to establish disease relevance, and rodent experiments to define mechanism. We found that in humans with cognitive decline, amyloid β (Aβ) constricts brain capillaries at pericyte locations. This was caused by Aβ generating reactive oxygen species, which evoked the release of endothelin-1 (ET) that activated pericy

Urapidil-induced hemodynamic changes in humans.

In hypertensive patients as well as in normal subjects urapidil has a hypotensive action. This is mainly mediated by a peripheral alpha 1-adrenoceptor blockade with a decrease in systemic vascular resistance; in addition, during acute animal experiments a centrally mediated hypotensive action was demonstrated, possibly by 5-hydroxytryptamine1A (5-HT1A)-receptor stimulation. Studies in humans showed an increase in cardiac output, which was not always significant; it did result either from an increased heart rate or an increased stroke volume. Acute changes in pulmonary hemodynamics after admini

Adaptive motility enables neutrophils to rapidly navigate confined capillaries.

To reach inflamed tissues, neutrophils must traverse capillaries as narrow as 2 micrometers. However, how they do so without compromising blood flow or capillary function has remained unclear. By combining intravital live-cell imaging with biomimetic microdevices, we show that neutrophils maintain migration speed in capillaries across increasing levels of confinement. This behavior was not shared by other leukocytes and was independent of nuclear properties. Instead, confinement rapidly engaged Rho-dependent actomyosin contractility at the cell rear, thereby offsetting the increased mechanical

Constitutive androstane receptor-mediated up-regulation of ATP-driven xenobiotic efflux transporters at the blood-brain barrier.

ATP-driven efflux transporters at the blood-brain barrier both protect against neurotoxicants and limit drug delivery to the brain. In other barrier and excretory tissues, efflux transporter expression is regulated by certain ligand-activated nuclear receptors. Here we identified constitutive androstane receptor (CAR) as a positive regulator of P-glycoprotein, multidrug resistance-associated protein 2 (Mrp2), and breast cancer resistance protein (BCRP) expression in rat and mouse brain capillaries. Exposing rat brain capillaries to the CAR activator, phenobarbital (PB), increased the transport

Autoregulation of capillary pressure and filtration in cat skeletal muscle in states of normal and reduced vascular tone.

The controversial hypothesis that capillary pressure (Pc) is autoregulated in response to arterial pressure (PA) alterations was tested in sympathectomized cat skeletal muscle by studying the relation between Pc and PA under conditions of well preserved vascular tone and reactivity, during papaverine-induced maximal vasodilatation (passive vascular bed), and during impaired vascular reactivity caused by preparatory surgery, or by low dose isoproterenol administration. The latter states resembled such under which Pc autoregulation unintentionally seems to have been studied previously. Capillary

Biochemical opening of the blood-brain barrier.

Biochemical agents that can influence the abnormal BBB are reviewed. Vasoactive compounds, including leukotrienes, bradykinin, histamine appear to selectively increase permeability in abnormal brain capillaries. Normal brain capillaries resist the effects of these compounds through an 'enzymatic barrier' that may inactivate vasoactive agents. In contrast, this enzymatic barrier is lost in certain diseased states, allowing vasoactive compounds to selectively increase permeability in abnormal capillaries. Clinically, intracarotid infusion of leukotrienes, bradykinin and other vasoactive agents c

Acute vasoconstriction-induced insulin resistance in rat muscle in vivo.

Insulin-mediated changes in blood flow are associated with altered blood flow distribution and increased capillary recruitment in skeletal muscle. Studies in perfused rat hindlimb have shown that muscle metabolism can be regulated by vasoactive agents that control blood flow distribution within the hindlimb. In the present study, the effects of a vasoconstrictive agent that has no direct effect on skeletal muscle metabolism but that alters perfusion distribution in rat hindlimb was investigated in vivo to determine its effects on insulin-mediated vascular action and glucose uptake. We measured

[Drug treatments of chronic venous insufficiency: pharmaco-clinical evaluation].

There are many methods for the assessment of venotropic drugs. Clinical trials based on randomized comparison with placebo are essential. The different methodes used for the evaluation of the functional effects are chosen on the basis of their performance and pertinence for the study objectives. These include three aims which we feel are essential: increased venous tone, decreased capillary permeability and reversal of microcirculatory impairment. Calf plethysmography is highly adapted for the evaluation of venous tone. The mercury gauge works well, but the future will tell if air plethysmogra

The haemodynamic responses to incremental doses of intravenous nitroglycerin in left ventricular failure.

The effects of incremental dose infusions of nitroglycerin, 10-160 micrograms/min, on pulmonary capillary wedge pressure and other haemodynamic variables have been studied in patients with left ventricular failure. Intravenous (IV) nitroglycerin caused a dose-related improvement in myocardial performance, with significant reductions in the raised pulmonary capillary wedge pressure and systemic vascular resistance. At the lowest dose of infusion only the venodilator action of nitroglycerin was apparent. From 40-160 micrograms/min the associated arteriolar dilatation was associated with a consis

Hemodynamic alterations of heat stroke in the elderly.

Serial hemodynamic alterations were investigated in seven elderly patients with heat stroke. Their mean age was 72 +/- 6 years. The circulatory response to heat stroke was either hyperdynamic of hypodynamic. Two patients had increased cardiac index (4.3 and 4.4 L/min/m2), increased right atrial pressure (10 and 12 mm Hg), normal pulmonary capillary wedge pressure (10 and 12 mm Hg), and decreased systemic vascular resistance (542 and 738 dyne.sec.cm-5). Five patients had decreased cardiac index (mean 2.3 +/- 0.2 L/min/m2), decreased right atrial pressure (mean 2 +/- 1 mm Hg), normal pulmonary c

An analysis of the effect of synovial capillary distribution upon trans-synovial concentration profiles and exchange.

Recent morphometric studies of synovium lining the rabbit knee showed that capillaries extend to greater than 200 microns below the synovial surface; that a proportion of the superficial capillaries are fenestrated; and that the numerical density of the capillaries varies with depth ( Knight & Levick , 1983, 1984). Such observations raised the question 'Down to what depth do capillaries contribute significantly to the diffusional flux of a small extracellular solute (e.g. sulphate) into the joint cavity?' This problem was addressed by a mathematical model of trans-synovial diffusion. The peria

Treatment of essential hypertension does not normalize capillary rarefaction.

OBJECTIVES: To determine if capillary rarefaction persists when hypertension is treated with angiotensin converting enzyme inhibitor, thiazidic diuretic and/or beta-blocker, and to identify which microcirculatory alterations (structural and functional) persist after anti-hypertensive treatment. METHODS: We evaluated 28 well-controlled essential hypertensive patients and 19 normotensive subjects. Nailfold videocapillaroscopy examination of the fourth finger of the left hand was used to determine the functional capillary densities at baseline, during post-occlusive hyperemia, and after venous c

P-gp Protein Expression and Transport Activity in Rodent Seizure Models and Human Epilepsy.

A cure for epilepsy is currently not available, and seizure genesis, seizure recurrence, and resistance to antiseizure drugs remain serious clinical problems. Studies show that the blood-brain barrier is altered in animal models of epilepsy and in epileptic patients. In this regard, seizures increase expression of blood-brain barrier efflux transporters such as P-glycoprotein (P-gp), which is thought to reduce brain uptake of antiseizure drugs, and thus, contribute to antiseizure drug resistance. The goal of the current study was to assess the viability of combining in vivo and ex vivo prepara

Capillary-induced adhesive contact dynamics determines dissipation and flow structure in wetted hydrogel packings.

The bulk response of a granular material is strongly influenced by particle and contact properties, such as friction coefficients, particle softness, lubrication on the contact scale and adhesion between particles. This study explores the bulk flow of wetted hydrogel particles, which are soft but also weakly adhesive due to capillary bridges. This simplified granular material with minimal contact friction reveals key insights in the role of capillary stresses on the macroscopic flow. At the micro-scale, we demonstrate a direct correlation between relative humidity (RH) and liquid bridge size b
