Phenotypical Detection of β-Lactamases in a Multidrug-Resistant and Extensively Drug-Resistant Chryseobacterium indologens: A Rare Human Pathogen With Special References to Risk Factor.

Chryseobacterium indologenes is gram-negative bacteria that cause infection in humans. It is less frequently isolated in the laboratory. The development of drug-resistant and its intrinsic ability to resist a wide range of antimicrobials enables them to cause mortality in an immunocompromised patient with a longer hospital stay. Our study objectives are to investigate antimicrobial-resistant patterns, drug-resistant enzymes, and the risk factor analysis associated with multidrug-resistant (MDR), extensively drug-resistant (XDR), and Pan-drug resistant (PDR) within 2 years. Altogether 53 strain

Genetic-biochemical analysis and distribution of the Ambler class A beta-lactamase CME-2, responsible for extended-spectrum cephalosporin resistance in Chryseobacterium (Flavobacterium) meningosepticum.

In vitro synergy between extended-spectrum cephalosporins and either clavulanic acid or cefoxitin was found for Chryseobacterium meningosepticum isolates during a double-disk assay on an agar plate. An extended-spectrum beta-lactamase (ESBL) gene from a C. meningosepticum clinical isolate was cloned and expressed in Escherichia coli DH10B. Its protein conferred resistance to most beta-lactams including extended-spectrum cephalosporins but not to cephamycins or to imipenem. Its activity was strongly inhibited by clavulanic acid, sulbactam, and tazobactam, as well as by cephamycins and imipenem.

Isolation and Characterization of a New Lytic Phage MA9V-2 Against Chryseobacterium indologenes MA9 and Its Combined Application with MA9V-1 for the Control of Panax notoginseng Root Rot.

Panax notoginseng, a valuable medicinal plant in Yunnan, suffers significant yield losses due to root rot, with Chryseobacterium indologenes MA9 as a major causal agent. Conventional chemical control methods are limited by residues and the development of bacterial resistance, underscoring the need for alternative strategies. In this study, we isolated a new lytic myovirus, vB_CinP_MA9V-2, from wastewater using MA9 as the host. MA9V-2 exhibited high lytic efficiency with 75% adsorption in 25 min and a burst size of ~100 PFU per cell, stability across pH 4 to 11 and temperatures of 4 to 50 °C, a

Molecular and biochemical characterization of a carbapenem-hydrolysing beta-lactamase from Flavobacterium johnsoniae.

Flavobacterium johnsoniae CIP100931 is resistant to most beta-lactam antibiotics and has a decreased susceptibility to carbapenems. A beta-lactamase gene was cloned and expressed in Escherichia coli DH10B. The purified beta-lactamase, JOHN-1, with a pI value of 9.0 and with a determined relative molecular mass of approximately 27 kDa was found to be a monomeric zinc-dependent enzyme that hydrolyses penicillins, narrow- and expanded-spectrum cephalosporins, carbapenems, but not monobactams. Sequence analysis revealed that JOHN-1 is a molecular class B beta-lactamase that is most closely related

Active site loops of membrane-anchored metallo-β-lactamases from environmental bacteria determine cephalosporinase activity.

Antimicrobial resistance is a significant global public health threat that limits treatment options for bacterial infections. This situation is aggravated by the environmental spread of β-lactamase genes. In particular, metallo-β-lactamases (MBLs) hydrolyze almost all available β-lactam antibiotics, including late-generation cephalosporins and carbapenems. Among MBLs, the New Delhi metallo-β-lactamase (NDM-1) of subclass B1 has shown the most ominous dissemination. NDM variants are the only MBLs of clinical importance that are membrane-anchored, a sub-cellular localization that endows them wit

EBR-1, a novel Ambler subclass B1 beta-lactamase from Empedobacter brevis.

Empedobacter brevis (formerly designated Flavobacterium breve) is a gram-negative aerobe involved in nosocomial infections. The Ambler class B beta-lactamase gene bla(EBR-1) was cloned and expressed in Escherichia coli from E. brevis clinical strain ASS-1, which had reduced susceptibility to expanded-spectrum cephalosporins and carbapenems. Purified beta-lactamase EBR-1 hydrolyzed penicillins, cephalosporins, and carbapenems efficiently but not aztreonam. Kinetic parameters of EBR-1 were similar to those of class B enzymes such as BlaB, IND-2, and GOB-1 identified from other Flavobacteriaceae

Effect of plant-derived microbial soil legacy in a grafting system-a turn for the better.

BACKGROUND: Plant-soil feedback arises from microbial legacies left by plants in the soil. Grafting is a common technique used to prevent yield declines in monocultures. Yet, our understanding of how grafting alters the composition of soil microbiota and how these changes affect subsequent crop performance remains limited. Our experiment involved monoculturing ungrafted and grafted watermelons to obtain conditioned soils, followed by growing the watermelons on the conditioned soils to investigate plant-soil feedback effects. RESULTS: Ungrafted plants grew better in soil previously conditioned

Assessing Potential Reservoir of Multidrug-Resistant Bacteria in the Oral Microbiota of Captive Burmese and Royal Pythons.

The oral microbiota has a diversity of microorganisms that together maintain the homeostasis of the oral cavity. Disruptions in the balance of these microbial populations can lead to the development of oral and dental diseases. To characterize the normal oral microbiota of captive ophidians, samples were obtained from the oral cavity of eight Burmese pythons (Python bivittatus) and 11 royal pythons (Python regius), using oral swab, during routine evaluations. In the Laboratory-Antimicrobials, Biocides and Biofilms Unit at University of Trás-os-Montes e Alto Douro, the identification and antimi

Physicochemical and microbiome changes in queso Crema de Chiapas during ripening.

The dynamic changes in the physicochemical, microbiological, and metagenomic profiles of Crema de Chiapas cheese were evaluated across three ripening stages (2, 29, and 58 days). Although the main physicochemical properties -including fat content- remained remarkably stable, salt and protein levels showed noticeable variation throughout ripening. Protein content had the strongest influence on sample differentiation across ripening stages in unsupervised multivariate models, enabling the clustering of microbial diversity according to maturation time. A clear shift in microbial diversity was det
