Crotiglianoids A-M, tigliane-type diterpenoids from the seeds of Croton tiglium and their anti-hepatic fibrosis activity.

The phytochemical investigation of the seeds of Croton tiglium led to the isolation and structural characterization of 20 tigliane-type diterpenoids (1-20), comprising 13 previously undescribed analogues designated as crotiglianoids A-M (1-13). Their structures were elucidated through comprehensive spectroscopic analysis, and the absolute configurations were determined by a combination of chemical transformation and comparative electronic circular dichroism data. Notably, crotiglianoids A (1) and B (2) were characterized as the first examples of 20-nor-tigliane diterpenoids. All isolates were

Novel findings to the biosynthetic pathway of magnoflorine and taspine through transcriptomic and metabolomic analysis of Croton draco (Euphorbiaceae).

BACKGROUND: Croton draco is an arboreal species and its latex as well as some other parts of the plant, are traditionally used in the treatment of a wide range of ailments and diseases. Alkaloids, such as magnoflorine, prevent early atherosclerosis progression while taspine, an abundant constituent of latex, has been described as a wound-healer and antitumor-agent. Despite the great interest for these and other secondary metabolites, no omics resources existed for the species and the biosynthetic pathways of these alkaloids remain largely unknown. RESULTS: To gain insights into the pathways i

Hydroethanolic root extract of Croton membranaceus suppresses angiogenesis in zinc-testosterone-induced prostate adenocarcinoma and chorioallantoic membrane models.

Angiogenesis plays a pivotal role in the growth, progression, and metastatic spread of cancer cells. However, the clinical utility of current antiangiogenic agents has been limited by the emergence of drug resistance, thereby highlighting the need for novel therapeutic candidates, including natural products. Croton membranaceus has previously been shown to suppress prostate cancer cell growth and may represent a potential antiangiogenic agent. In this study, we investigated the effect of Croton membranaceus hydroethanolic root extract (CMERE) on new blood vessel formation. The zinc-testosteron

Antioxidant and Anticancer Activity of Tamarix Indica Aerial Extracts against DMBA/croton Oil-Induced Skin Carcinogenesis in Mice.

BACKGROUND: The largest organ, skin, acts as the body's first line of protection against various environmental threats and harmful substances, which may be carcinogenic. The widespread acceptance of conventional chemotherapy has been hindered by its high cost and severe side effects, which have encouraged the need for alternatives. The phytoconstituents produced by plants promise to prevent skin carcinogenesis by targeting multiple steps. They are widely accepted, safe, and have few side effects. OBJECTIVE: The current study aimed to determine the antitumor potential of the aerial extracts of

Exploring four South African Croton species for potential anti-inflammatory properties: in vitro activity and toxicity risk assessment.

ETHNOPHARMACOLOGICAL RELEVANCE: The African Continent harbours approximately 26 Croton species. Many Croton species are used in traditional medicine in southern Africa to treat a variety of ailments including malaria, tuberculosis, microbial infection and inflammation. Considering the high diversity of the genus Croton, the ethnopharmacological information available on southern African species is rather limited. Furthermore, the potential for novel anti-inflammatory drug scaffolds has not previously been investigated. AIM OF THE STUDY: The aim of the study was to evaluate the potential of fou

Bioassay-guided isolation of an anti-ulcer diterpenoid from Croton reflexifolius: role of nitric oxide, prostaglandins and sulfhydryls.

Croton reflexifolius H. B. K (Euphorbiaceae) is a very common medicinal plant in the Huastecan region of Mexico that, according to local folk medicine, is considered useful in the treatment of gastritis and gastric ulcer. We have aimed to test the validity of this practice by using the experimental model of an ethanol-induced gastric ulcer in male Wistar rats. The results showed that C. reflexifolius had gastroprotector activity, that the hexane extract had the highest protective activity (64.38+/-7.72%), and that polyalthic acid isolated from this extract was the main active gastroprotector a

[Croton oil-induced hemorrhoid model in rat: comparison of anti-inflammatory activity of diflucortolone valerate with other glucocorticoids].

A hemorrhoid model was prepared by means of application of croton oil onto the recto-anus of rats. Cotton swab soaked with the inducer, which consisted of water, pyridine, diethylether and 6% croton oil in diethylether, was inserted into the anus. The following conditions were found to be optimal for preparing the model: cotton swab containing 0.16 ml of the inducer solution was applied to the anus of a 6 week-old rat (body wt. about 140 g) for 10 sec. The edema developed linearly until 7-8 hr after application, and the severity of the edema was sustained almost constantly for more than 24 hr.

Crotonate suppresses breast cancer metastasis and promotes immunotherapy response by inducing ACSS2-mediated EZH2-K348 crotonylation.

Crotonate, a short-chain fatty acid, generates protein crotonylation. However, the role of crotonate in cancer progression is unknown. Here, we present a crotonate-crotonyl-coenzyme A (CoA)-enhancer of zeste homolog 2 (EZH2) crotonylation cascade blocking breast cancer growth and metastasis. We demonstrated that crotonate promotes EZH2 degradation via crotonyl-CoA-mediated crotonylation at Lys348 in EZH2 (EZH2-K348cr). EZH2-K348cr leads to reduced genome-wide H3K27me3 (trimethylation of lysine-27 on histone-3) occupancy. Crotonate metabolizes to crotonyl-CoA by ACSS2 (acyl-CoA synthetase 2), a

[Exploring Japanese Kampo medicine in fighting epidemics: discovering and popularising croton and satou as 'specific drugs' to treat cholera].

Japanese Kampo medicine (medicine with Han Fang) was found effective to treat some epidemic diseases.Historical records show that Namikawa Saimin, a Kampo medicine (Han Fang Medicine) practitioner during the cholera pandemic in the period of Ansei in Japan (1858-1860), discovered that the treatment effect of croton fruit against cholera was remarkable. Another physician in Naniwa also found that satou (Zhǎ Dá, visceral stones of animals and livestock) had the same effect in treating cholera. Subsequently, Wani Tadatane, an official physician in Komatsu Han of Iyo Province, learned about Namika
