Old Data-New Concepts: Integrating "Indirect Effects" Into Radiation Protection.

PURPOSE: To address the following key question, what are the consequences of nontargeted and delayed effects for linear nonthreshold models of radiation risk? This paper considers low-dose "indirect" or nontargeted effects and how they might impact radiation protection, particularly at the level of the environment. Nontargeted effects refer to effects in cells, tissues, or organisms that were not targeted by irradiation and that did not receive direct energy deposition. They include genomic instability and lethal mutations in progeny of irradiated cells and bystander effects in neighboring cel

A sense of danger from radiation.

Tissue damage caused by exposure to pathogens, chemicals and physical agents such as ionizing radiation triggers production of generic "danger" signals that mobilize the innate and acquired immune system to deal with the intrusion and effect tissue repair with the goal of maintaining the integrity of the tissue and the body. Ionizing radiation appears to do the same, but less is known about the role of "danger" signals in tissue responses to this agent. This review deals with the nature of putative "danger" signals that may be generated by exposure to ionizing radiation and their significance.

An evaluation of the ethical principles of the ICRP's radiation protection standards for workers.

After reviewing international recommendations and national standards for occupational radiation exposures, the article summarizes the major ethical theories so as to analyze which theories various ICRP principles presuppose. It also shows how proponents of each ethical theory would support or criticize various radiation principles and practices. Finally the article discusses four ethical issues crucial to the debate over workplace standards for radiation: (1) how to respond to the uncertainty over effects of low-dose exposures; (2) the conditions under which worker consent legitimates higher e

Validation of a High-Throughput Dicentric Chromosome Assay Using Complex Radiation Exposures.

Validation of biodosimetry assays is routinely performed using primarily orthovoltage irradiators at a conventional dose rate of approximately 1 Gy/min. However, incidental/ accidental exposures caused by nuclear weapons can be more complex. The aim of this work was to simulate the DNA damage effects mimicking those caused by the detonation of a several kilotons improvised nuclear device (IND). For this, we modeled complex exposures to: 1. a mixed (photons + IND-neutrons) field and 2. different dose rates that may come from the blast, nuclear fallout, or ground deposition of radionuclides (gro

An estimate assay for low-level exposure to ionizing radiation based on mass spectrometry quantification of γ-H2AX in human peripheral blood lymphocytes.

Exposure to environmental ionizing radiation (IR) is ubiquitous, and large-dose exposure to IR is known to cause DNA damage and genotoxicity which is associated with an increased risk of cancer. Whether such detrimental effects are caused by exposure to low-dose IR is still debated. Therefore, rapid and early estimation of absorbed doses of IR in individuals, especially at low levels, using radiation response markers is a pivotal step for early triage during radiological incidents to provide adequate and timely clinical interventions. However, there is currently a crucial shortage of methods c

RENEB Inter-Laboratory comparison 2017: limits and pitfalls of ILCs.

PURPOSE: In case of a mass-casualty radiological event, there would be a need for networking to overcome surge limitations and to quickly obtain homogeneous results (reported aberration frequencies or estimated doses) among biodosimetry laboratories. These results must be consistent within such network. Inter-laboratory comparisons (ILCs) are widely accepted to achieve this homogeneity. At the European level, a great effort has been made to harmonize biological dosimetry laboratories, notably during the MULTIBIODOSE and RENEB projects. In order to continue the harmonization efforts, the RENEB

Detection of radiation effects in polymer gel dosimeters using 129Xe NMR.

Polymer gel dosimeters consist of monomers, with or without cross-linking agents, dispersed in a gel. Upon exposure to ionizing radiation, polymerization proceeds within the gel matrix, thereby changing several measurable physical properties that can then be related quantitatively to absorbed dose. Several previous studies have examined how various nuclear magnetic resonance (NMR) properties, such as the relaxation rates of water protons, change with dose, and magnetic resonance imaging (MRI) has been used successfully to measure three-dimensional dose distributions in irradiated polymer gels.

Dicentric chromosomes in monolayers of human lymphocytes produced by monochromatized synchrotron radiation with photon energies from 1.83 keV to 17.4 keV.

A systematic approach was used to examine the induction of dicentric chromosomes in human lymphocytes irradiated in vitro with monochromatized synchrotron radiation produced with photon energies in the range from 1.83 keV to 17.4 keV. To avoid potential confounding factors that could influence the outcome of the experiments, only blood from one individual was used. Since for the irradiation experiments with these low photon energies the local dose variations would become unacceptable, monolayers of lymphocytes attached within 3 h PHA stimulation have been used. The culture conditions ensured t

UV radiation ocular exposure dosimetry.

There is currently some degree of controversy as to the magnitude of cataract and other ocular diseases related to human lifetime exposure to UV radiation (UVR). Concerns about the depletion of stratospheric ozone and the related increase in terrestrial UVR exposure have emphasized the importance of resolving this controversy. A careful study of ocular exposure to environmental sunlight demonstrates that it is not simple to determine accurately the level of solar UVR exposure of the human eye. Past attempts to measure or calculate UVR exposure of the eye have generally relied on the measuremen

The challenges of defining hormesis in epidemiological studies: The case of radiation hormesis.

In the current radiation protection system, preventive measures and occupational exposure limits for controlling occupational exposure to ionizing radiation are based on the linear no-threshold extrapolation model. However, currently an increasing body of evidence indicates that this paradigm predicts very poorly biological responses in the low-dose exposure region. In addition, several in vitro and in vivo studies demonstrated the presence of hormetic dose response curves correlated to ionizing radiation low exposure. In this regard, it is noteworthy that also the findings of different epidem

Identifying radiation responsive exon-regions of genes often used for biodosimetry and acute radiation syndrome prediction.

Gene expression (GE) analysis of FDXR, DDB2, WNT3 and POU2AF1 is a promising approach for identification of clinically relevant groups (unexposed, low- and high exposed) after radiological/nuclear events. However, results from international biodosimetry exercises have shown differences in dose estimates based on radiation-induced GE of the four genes. Also, differences in GE using next-generation-sequening (NGS) and validation with quantitative real-time polymerase chain reaction (qRT-PCR) was reported. These discrepancies could be caused by radiation-responsive differences among exons of the

Application of a semi-automated dicentric scoring system in triage and monitoring occupational radiation exposure.

The dicentric chromosome assay (DCA) is considered the gold standard for radiation biodosimetry, but it is limited by its long dicentric scoring time and need for skilled scorers. The automation of scoring dicentrics has been considered a strategy to overcome the constraints of DCA. However, the studies on automated scoring methods are limited compared to those on conventional manual DCA. Our study aims to assess the performance of a semi-automated scoring method for DCA using ex vivo and in vivo irradiated samples. Dose estimations of 39 blind samples irradiated ex vivo and 35 industrial radi

Direct and indirect tasks on assessment of dose and time distributions and thresholds of acute radiation exposure.

Mathematical methods were developed to construct dose and time distributions and their associated risks and threshold values for lethal and non-lethal effects of acute radiation exposure to include mortality and incidence, prodromal vomiting, and agranulocytosis. A new distribution (T-model) was obtained to describe time parameters of acute radiation syndrome such as the latency period, time to onset of vomiting, and time to initiation of agranulocytosis. Based on the dose and time distributions, the parameter translation method was defined using an orthogonal regression, which allows one to s

Dose-response relationships on the expression profile of cytokeratins and vimentin in rat submandibular glands following fractionated irradiation.

OBJECTIVE: The extent of radiogenic damage in salivary gland (SG) tissue depends on the radiation dose (RD), the fractionation (FN) and the localization of SG in the radiation field (RF). While the functional restriction and the radiogenic SG tissue damage are well documented using histomorphological, electron-microscopic and enzyme-histochemical methods, immunohistochemical analysis (IH) of cytokeratins (CK), epithelial differentiation markers, and vimentin, a marker of mesenchymal cells, are rare. Previous studies have shown stronger immunoreactivities of CK in irradiated glands exposed to 6

Protective effect of hydroferrate fluid, MRN-100, against lethality and hematopoietic tissue damage in γ-radiated Nile tilapia, Oreochromis niloticus.

Hydroferrate fluid, MRN-100, an iron-based compound derived from bivalent and trivalent ferrates, is a potent antioxidant compound. Therefore, we examined the protective effect of MRN-100 against γ-radiation-induced lethality and damage to hematopoietic tissues in fish. A total of 216 Nile tilapia fish (Oreochromis niloticus) were randomly divided into four groups. Group 1 served as a control that was administered no radiation and no MRN-100 treatment. Group 2 was exposed only to γ-radiation (15 Gy). Groups 3 and 4 were pre-treated with MRN-100 at doses of either 1 ml/l or 3 ml/l in water for

Axial cineangiography in congenital heart disease. Section I. Concept, technical and anatomic considerations.

Cineangiographic axial techniques were designed to overcome the limitations of conventional angiography in the diagnosis of congenital heart disease. Two basic patient (or equipment) maneuvers are involved; 1) long axis of the heart is aligned perpendicular to the X-ray beam, and 2) rotation of patient results in the heart being radiographically sectioned at 30 degree angles. To accomplish this with fixed vertical and horizontal X-ray tubes, three positions were developed: 1) "hepato-clavicular," "4 chamber," 2) "long axial oblique," 3) "anterior-posterior axial." A fourth, the "sitting-up" pr

Effects of sub-lethal doses of photo-activated disinfection against Porphyromonas gingivalis for pharmaceutical treatment of periodontal-endodontic lesions.

BACKGROUND: Microorganisms treated by photo-activated disinfection (PAD) in combined periodontal-endodontic (perio-endo) lesions would be exposed to sub-lethal doses of PAD (sPAD). This study evaluated the effect of sPAD using toluidine blue O (TBO) in combination with diode laser irradiation on the growth and biofilm-formation ability of Porphyromonas gingivalis as an endo-periodontal pathogen. METHODS: The antibacterial and antibiofilm potential of sPAD against P. gingivalis was analyzed at sub-lethal doses of TBO and irradiation time of diode laser on a colony-forming unit and crystal viol
