There are abnormalities of haemostasis and rheological properties in uncontrolled diabetics which could well be involved in the pathogenesis of diabetic microangiopathy and macroangiopathy. Erythrocyte deformability was reduced with a negative correlation with the degree of control (blood sugar and glycosylated hemoglobin). In uncontrolled insulin dependent diabetics (IDD), returning blood sugar to normal with a continuous infusion of insulin using an artificial pancreas, improved deformability in several hours; even if the raised blood sugar was maintained artificially, a perfusion of insulin

Glucose-6-phosphate dehydrogenase (G-6-PDH) deficiency is the most common known human genetic polymorphism. This study tested the hypothesis that G-6-PDH deficiency worsens sepsis-induced erythrocyte dysfunction. Sepsis (24 h) was induced by cecal ligation and puncture in wild-type (WT) and G-6-PDH-deficient (G-6-PDH activity 15% of WT) mice. Erythrocyte responses were tested in whole blood as well as in subpopulations of circulating erythrocytes. Whereas erythrocyte deformability was similar in unchallenged deficient and WT animals, sepsis decreased erythrocyte deformability that was more pro

NO is present in the blood at 10(-7) M under physiological conditions, but at concentrations higher than 10(-6) M during inflammatory disease states. The aim of this study was to characterize what are the effects of these different NO concentrations on erythrocyte structural and functional properties. Blood was collected from eleven healthy men and incubated with SpermineNONOate in order to expose it during incubation time to NO concentrations between 10(-7) M and 10(-3) M. We measured erythrocyte aggregation and deformability, membrane lipid peroxidation and fluidity, p50, hemoglobin, oxyhemo

Venous blood from patients with sickle-cell disease in the steady state or in crisis was progressively deoxygenated in vitro to study the effect of BW12C, a new compound designed to stabilize haemoglobin in the oxy-conformation, on the deformability (filterability) of washed erythrocytes. At a final concentration of 1.5 mM, BW12C significantly increased erythrocyte deformability, compared with no added compound, at all levels of deoxygenation below normal arterial PO2. At concentrations of 3.0 and 5.0 mM, BW12C prevented any significant reduction of erythrocyte deformability, or increase in si

Infusions of hydroxyethylstarch (HES) solutions as plasma substitutes and flow improvers play an increasing role in the treatment of various diseases. Little is known about the effect of HES solutions of different molecular weights on the flow properties of blood i.e. blood viscosity, plasma viscosity, erythrocyte aggregation and erythrocyte deformability (filtrability). In the present study the influence of 500 ml-infusions of a 6% solution of HES (MW 450,000) has been compared to that of 6% solutions of HES 40,000. Infusions of 500 ml of HES 450,000 into healthy persons induced a hemodilutio

Hydroxyurea is a drug that is used to treat some patients with sickle cell disease. We have measured the deformability of sickle erythrocytes incubated in hydroxyurea in vitro and found that hydroxyurea acts to decrease the deformability of these cells. The deformability of normal erythrocytes was not significantly affected by hydroxyurea except at very high concentrations. Hydroxyurea also did not consistently reduce the deformability of sickle erythrocyte ghosts. We propose that the decreased deformability, observed in vitro, is due to the formation of methemoglobin and other oxidative proce

Low plasma estrogens, vitamin D deficiency, obesity, diabetes, cardiovascular diseases, thromboembolism, and impaired microcirculation are linked to the severity of covid-19. Studies have suggested that these comorbidities also are related to erythrocyte factors linked to increased blood viscosity in microcirculation such as erythrocyte aggregation and erythrocyte deformability. Increased blood viscosity in microcirculation can lead to a decrease in oxygenation and nutrition of tissues. Therefore erythrocyte aggregation and erythrocyte deformability may be involved in covid-19 severity, leadin

To determine the pathogenesis of neonatal hyperbilirubinaemia after oxytocin-induced labour venous cord blood from 95 healthy newborn infants was examined. Of these, 15 were delivered by elective caesarean section, 40 after spontaneous labour, and 40 after oxytocin-induced labour. There was no significant difference in any haematological or biochemical variable between the first two groups. Infants born after oxytocin-induced labour, however, showed clear evidence of increased haemolysis associated with significantly decreased erythrocyte deformability (P less than 0.001). In-vitro studies sho

Factors causing erythrocyte destruction in patients with congenital erythrocyte membrane disorders include 1) decreased cell deformability, 2) increased cell viscosity, 3) decreased membrane stability, 4) increased intracellular Ca2+ concentrations, 5) changes in membrane permeability, 6) extracellular conditions such as hypoxia and acidic pH, or 7) hypersplenism. erythrocytes with shape changes due to abnormality of membrane skeleton lose deformability to negotiate the interendothelial slits into the splenic venous sinus, and stagnate within the cordal space where metabolic stress reduces mem

Although several studies have been published regarding rheological behaviour of red blood cells in beta and delta-beta thalassaemia traits, little information about erythrocyte deformability in alpha-thalassaemia carriers is available. We aimed to determine erythrocyte deformability in heterozygous (silent, -α/αα) and homozygous (minor alpha-thalassaemia, -α/-α) carriers of the alpha-thalassaemia trait for the alpha 3.7 deletion, the most common in our geographical area. We evaluated erythrocyte deformability by means of the elongation index (EI) in a Rheodyn SSD at 12, 30 and 60 Pa, along wit

We aimed to investigate the effects of vitamin B12 deficiency on hemorheological parameters, and the changes in these parameters following vitamin B12 treatment. 33 patients (mean-age:7 ± 5.7 years) diagnosed as nutritional vitamin B12 deficiency, and 31 age and sex matched controls (mean-age:7.1 ± 5.2 years) were enrolled. Erythrocyte deformability and aggregation were determined by an ectacytometer, plasma and whole blood viscosities by a cone-plate rotational viscometer. The differences between patients and controls were compared. Hemorheological parameters were repeated in the patient grou

BACKGROUND: Erythrocyte deformability, the ability of red blood cells to bend and twist as they pass through capillaries, is es-sential for tissue perfusion. This study investigates the effects and underlying mechanisms of rosmarinic acid treatment on erythrocyte deformability in rats subjected to lower limb ischemia-reperfusion injury.
METHODS: The study was conducted on Wistar albino rats weighing 400-450 g. The rats were randomly divided into five groups: the control group with no treatment (C), the group receiving only the solvent dimethyl sulfoxide (DMSO), the ischemia-reperfusion group

A study of rheological determinants (plasma viscosity, whole-blood viscosity, and erythrocyte deformability) was made in 24 matched pairs of patients with homozygous sickle cell disease, with and without homozygous alpha-thalassaemia 2. Patients with coexisting alpha-thalassaemia showed a significant increase in erythrocyte deformability measured as filtration of washed erythrocytes through 5 micron diameter pores and also as viscosity of whole blood at high shear rate (230s-1) and standard haematocrit (0.45). This rheological advantage may explain the beneficial effect of alpha-thalassaemia 2

Physical activity combined with low temperatures can improve physical fitness, the efficiency of many organ systems, and mental well-being. The study investigated the effects of single sessions of various forms of physical activity (running, walking, lying down) conducted in a thermal climate chamber at a temperature of -5°C on blood rheological properties in young men. The participants (21 men aged 22 years), when performing the activities, were fasting and hydrated with 500 ml of water. Erythrocyte deformability increased at a shear stress of 4.24 Pa in the walking group and in the jogging g

The most important rheologic parameters were examined in six patients with acute occlusion of the central retinal artery. Blood samples were taken as part of the initial clinical examination on the day of the occlusion before the onset of therapy, and the following parameters were studied: (1) yield shear stress; (2) hematocrit; (3) plasma viscosity; (4) extent of erythrocyte aggregation; (5) erythrocyte deformability; (6) plasma fibrinogen concentration. A definite shear stress could be measured in all patients, something not possible in rheologically healthy volunteers. Hematocrit values wer

Although there has been considerable progress in understanding the factors that determine the invasiveness of Plasmodium falciparum merozoites, the collective role of the biophysical characteristics of erythrocyte deformability in the invasion process is poorly understood. Cell shape, cytoplasmic viscosity, and membrane stability are the main determinants of erythrocyte deformability, but it remains unknown how these properties affect the merozoite invasiveness. This study aimed to investigate computationally (i) the role of erythrocyte morphology and merozoite-induced erythrocyte membrane dam

Isouramil is one of the two aglycones derived from broad beans (Vicia faba) with the capacity to autoxidize and generate hydrogen peroxide. During its incubation with erythrocytes in vitro, it causes a marked decrease in cellular deformability at low shear forces, as measured by cone plate viscometry. At the same time the membrane shear modulus of elasticity, as determined by the dimensions of pressure induced pseudopods (PIPs), is increased. These experiments suggest that alterations in the mechanical properties of erythrocyte membranes may be the basis for changes in cellular deformability a
