Salinity and ionic composition drive prokaryotic community assembly in Tuolekule Lake, China.

Saline lakes in arid and semi-arid regions are home to unique ecosystems characterized by high salinity and alkalinity, as well as extreme environmental stresses. Despite these harsh conditions, they support diverse extremophilic microbial communities with distinct structures, offering substantial biomedical and biotechnological potential. Tuolekule Lake is a hypersaline, alkaline lake situated in a closed basin in the northern foothills of the East Tianshan Mountains in Xinjiang, China. This high-altitude extreme environment constitutes a distinct ecosystem. However, the structure of its micr

What Are the Essential Water Molecules in Regulating Lipid Membrane Fluidity?

Living organisms often endure extreme dehydration, which disrupts the cell membrane structure and function. Dehydration can induce a fluid-to-gel phase transition in lipid bilayers, compromising their fluidity and biological role. Lipid bilayer self-assembly is governed by intricate interactions between water and lipid head groups, but which water molecules are crucial for maintaining fluidity? We address this fundamental question using molecular dynamics (MD) simulations of a liquid-ordered (Lo) lipid bilayer over a hydration range of h = 2-30. We have identified a critical threshold at hydra

Oxygen competition induces chlorosis in thermoacidophilic methanotroph-microalgae cocultures.

Methanotroph-microalgae cocultures offer a promising bioprocess platform for methane (CH₄) and carbon dioxide (CO₂) valorisation and have been proposed for sustainable single-cell protein (SCP) production. In coculture, the thermoacidophilic methanotroph Methylacidiphilum sp. RTK17.1 can benefit from oxygen supplied by the extremophilic microalga Galdieria sp. RTK37.1 under low-O₂ conditions, enhancing growth and methane oxidation. However, oxygen availability is a critical factor that can rapidly become limiting and is dependent on the relative abundance of each coculture partner. Here, we in

Pathway(s) and distribution of photosystem II-cyclic electron flow.

Photosystem II is a highly conserved protein complex found across all oxygenic phototrophs and the source of almost all biologically available electrons and atmospheric oxygen. This large, membrane-bound complex is at continuous risk of damage under direct, full sunlight, and those organisms that live under such conditions have developed photoprotective mechanisms to address this stressor. One such mechanism, photosystem II-cyclic electron flow (PSII-CEF), is a poorly understood pathway by which charge-separated electrons are recombined with holes after an extended separation and multiple elec

Microbial and synthetic biology solutions for waste management and biomanufacturing off Earth.

Waste management aboard the International Space Station is a critical challenge due to limited space, microgravity, and continuous waste generation. This manuscript explores biotechnological strategies that convert waste into valuable resources, supporting sustainability and reducing reliance on Earth. Biowaste decomposition transforms organic waste into nutrients and biogas, while regenerative life support systems recycle carbon, water, and minerals. Biomining enables microbes to extract rare elements and upcycle electronic waste into mission-critical metals. Engineered extremophiles demonstr

Production and physicochemical characterization of exopolysaccharides from the salt-tolerant actinobacterial strain Zhihengliuella halotolerans DT-Z1.

Extremophiles often produce exopolysaccharides (EPSs) as a strategy in harsh environments, thus attracting significant interest for their properties and applications. A salt-tolerant EPS-producing Zhihengliuella halotolerans DT-Z1 was acquired and its EPS yield was optimized using response surface methodology, significantly increased from 2.86 to 10.26 g/L. Two fractions (EPS1-0 and EPS1-1) were purified by ethanol precipitation and anion exchange and gel filtration chromatography. Structural characteristics confirmed that both EPS1-0 and EPS1-1 are linear β-(2-6)-linked fructans with high mol

Unveiling hidden microbial diversity in Mars 2020 mission assembly cleanrooms with molecular insights into the persistence and perseverance of novel species defying metagenome sequencing.

NASA cleanrooms, which are critical for assembling space mission components, are maintained under stringent decontamination protocols to minimize biological contamination. These environments are characterized by nutrient-poor and oligotrophic conditions, leading to low microbial loads. Despite extensive cleaning, oligotrophs capable of surviving in such conditions continue to persist, often remaining undetected due to their low abundance, resistance to environmental stresses, and difficulties in biomolecule extraction. Even with shotgun metagenome sequencing technologies, these microbes may go

Purple photosynthetic bacteria (PPB) are versatile microorganisms capable of producing various value-added chemicals, e.g., biopolymers and biofuels. They employ diverse metabolic pathways, allowing them to adapt to various growth conditions and even extreme environments. Thus, they are ideal organisms for the Next Generation Industrial Biotechnology concept of reducing the risk of contamination by using naturally robust extremophiles. Unfortunately, the potential of PPB for use in biotechnology is hampered by missing knowledge on regulations of their metabolism. Although Rhodospirillum rubrum

The extremo-α-carbonic anhydrase from the thermophilic bacterium Sulfurihydrogenibium azorense is highly inhibited by sulfonamides.

The α-carbonic anhydrase (CA, EC 4.2.1.1) from the newly discovered extremophilic bacterium Sulfurihydrogenibium azorense (SazCA) is the most effective CA known to date. Here we investigated the inhibition profile of this enzyme with a series of aromatic and heterocyclic sulfonamides, and one sulfamate. Many clinically used sulfonamides, such as acetazolamide, methazolamide, ethoxzolamide, dichlorophenamide, dorzolamide, brinzolamide, topiramate, celecoxib and sulpiride were low nanomolar/subnanomolar SazCA inhibitors (KIs in the range of 0.9-10.8 nM) whereas simple aromatic derivatives were l

Lipophilic bioactive compounds from thermophilic cyanobacterium Leptolyngbya sp. HNBGU-004: Implications for countering VRSA resistance.

Extremophiles thrive in extreme conditions, showcasing rich and unexplored diversity. This resilience hints at the existence of novel biochemical pathways and unique bioactive compounds. In contrast, the issue of drug resistance and excessive misuse of antibiotics in various settings, such as healthcare, agriculture, and veterinary medicine, has contributed to the emergence and spread of drug-resistant microorganisms. In the present research, Leptolyngbya sp. HNBGU-004, was obtained from an extreme location, a hot water spring in the Garhwal Himalayan region of India. The lipophilic fraction d

Selective survival of anaerobic digester microbes after C. bescii pretreatment of manure.

Extremophilic biological pretreatment (EBP) of dairy manure using Caldicellulosiruptor bescii at 75°C dramatically alters the microbial community structure and activity in anaerobic digesters (ADs). This study evaluated microbial abundance and activity in manure, EBP, and ADs at industrial scales using 16S sequencing of rDNA and rRNA. Results revealed that bacterial communities remained largely active and resilient throughout the system, but acetoclastic methanogens were sensitive to EBP treatment and failed to reestablish in ADs. In contrast, hydrogenotrophic methanogens survived EBP and pers

Genome sequence analysis of Pseudomonas extremaustralis provides new insights into environmental adaptability and extreme conditions resistance.

The genome of the Antarctic bacterium Pseudomonas extremaustralis was analyzed searching for genes involved in environmental adaptability focusing on anaerobic metabolism, osmoregulation, cold adaptation, exopolysaccharide production and degradation of complex compounds. Experimental evidences demonstrated the functionality of several of these pathways, including arginine and pyruvate fermentation, alginate production and growth under cold conditions. Phylogenetic analysis along with genomic island prediction allowed the detection of genes with probable foreign origin such as those coding for

Anion inhibition studies of the fastest carbonic anhydrase (CA) known, the extremo-CA from the bacterium Sulfurihydrogenibium azorense.

The α-carbonic anhydrase (CA, EC 4.2.1.1) from the extremophilic bacterium Sulfurihydrogenibium azorense, SazCA, is the fastest CA known to date as a catalyst for CO(2) hydration to bicarbonate and protons. We report an inhibition study of this enzyme with inorganic anions and several other small molecules known to interact with these metalloenzymes. Bicarbonate, carbonate and sulfate were ineffective SazCA inhibitors whereas most other inorganic anions were submillimolar inhibitors. The best inhibition was observed with trithiocarbonate, diethyldithiocarbamate, sulfamide, sulfamate, phenylbor

Verrucomicrobial methanotrophs: ecophysiology of metabolically versatile acidophiles.

Methanotrophs are an important group of microorganisms that counteract methane emissions to the atmosphere. Methane-oxidising bacteria of the Alpha- and Gammaproteobacteria have been studied for over a century, while methanotrophs of the phylum Verrucomicrobia are a more recent discovery. Verrucomicrobial methanotrophs are extremophiles that live in very acidic geothermal ecosystems. Currently, more than a dozen strains have been isolated, belonging to the genera Methylacidiphilum and Methylacidimicrobium. Initially, these methanotrophs were thought to be metabolically confined. However, genom

Engineering antimicrobial coating of archaeal poly-γ-glutamate-based materials using non-covalent crosslinkages.

We are now entering a new age of intelligent material development using fine, sustainable polymers from extremophiles. Herein we present an innovative (but simple) means of transforming archaeal poly-γ-glutamate (PGA) into extremely durable polyionic complexes with potent antimicrobial performance. This new supra-polymer material (called PGA/DEQ) was subjected to nuclear magnetic resonance and X-ray diffraction spectroscopies to characterize in structural chemistry. Calorimetric measurements revealed its peculiar thermal properties; to the best of our knowledge, it is one of the most heat-resi

A thermophilic-like ene-reductase originating from an acidophilic iron oxidizer.

Ene-reductases originating from extremophiles are gaining importance in the field of biocatalysis due to higher-stability properties. The genome of the acidophilic iron-oxidizing bacterium "Ferrovum" sp. JA12 was found to harbor a thermophilic-like ene-reductase (FOYE-1). The foye-1 gene was ligated into a pET16bp expression vector system, and the enzyme was produced in Escherichia coli BL21 (DE3; pLysS) cells in yields of 10 mg L-1. FOYE-1 showed remarkable activity and rates on N-phenylmaleimide and N-phenyl-2-methylmaleimide (up to 89 U mg-1, >97 % conversion, 95 % (R)-selective) with both

De novo designed bright, hyperstable rhodamine binders for fluorescence microscopy.

De novo protein design has emerged as a powerful strategy with the promise to create new tools. The practical performance of designed fluorophore binders, however, has remained far from meeting fluorescence microscopy demands. Here, we design de novo Rhodamine Binder (Rhobin) tags that combine ideal properties including size, brightness, and now adding hyperstability. Rhobin allows live and fixed cell imaging of a wide range of subcellular targets in mammalian cells. Its reversible fluorophore binding further enables live super-resolution STED microscopy with low photobleaching, as well as PAI
