Use of repetitive sequences and the polymerase chain reaction technique to classify genetically related Bradyrhizobium japonicum serocluster 123 strains.

We have determined that repetitive (repetitive extragenic palindromic [REP] and enterobacterial repetitive intergenic consensus [ERIC]) sequences used in conjunction with the polymerase chain reaction technique (REP and ERIC PCR) provide an effective means of differentiating between and classifying genetically related Bradyrhizobium japonicum serocluster 123 strains. Analysis of REP and ERIC PCR-generated dendrograms indicated that this technique can effectively differentiate between closely related strains which were indistinguishable by using other classification methods. To maximize the gen

Advanced modeling and optimizing for surface sterilization process of grape vine (Vitis vinifera) root stock 3309C through response surface, artificial neural network, and genetic algorithm techniques.

In vitro, sterilization is one of the key components for proceeding with plant tissue cultures. Since the effectiveness of sterilization has a direct impact on the culture's final outcomes, there is a crucial need for optimization of the sterilization process. However, compared with traditional optimizing methods, the use of computational approaches through artificial intelligence-based process modeling and optimization algorithms provides a precise optimal condition for in vitro culturing. This study aimed to optimise in vitro sterilization of grape rootstock 3309C using RSM, ANN, and genetic

Genetic screening of spinal muscular atrophy using a real-time modified COP-PCR technique with dried blood-spot DNA.

BACKGROUND: Spinal muscular atrophy (SMA) is a common neuromuscular disorder caused by mutations in SMN1. More than 95% of SMA patients carry homozygous SMN1 deletion. SMA is the leading genetic cause of infant death, and has been considered an incurable disease. However, a recent clinical trial with an antisense oligonucleotide drug has shown encouraging clinical efficacy. Thus, early and accurate detection of SMN1 deletion may improve prognosis of many infantile SMA patients. METHODS: A total of 88 DNA samples (37 SMA patients, 12 carriers and 39 controls) from dried blood spots (DBS) on fi

Developing cell therapy techniques for respiratory disease: intratracheal delivery of genetically engineered stem cells in a murine model of airway injury.

Interest has increased in the use of exogenous stem cells to optimize lung repair and serve as carriers of a therapeutic gene for genetic airway diseases such as cystic fibrosis. We investigated the survival and engraftment of exogenous stem cells after intratracheal injection, in a murine model of acute epithelial airway injury already used in gene therapy experiments on cystic fibrosis. Embryonic stem cells and mesenchymal stem cells were intratracheally injected 24 hr after 2% polidocanol administration, when epithelial airway injury was maximal. Stem cells were transfected with reporter ge

The applicability of the SERS technique in food contamination testing - The detailed spectroscopic, chemometric, genetic, and comparative analysis of food-borne Cronobacter spp. strains.

Microorganisms assigned as Cronobacter are Gram-negative, facultatively anaerobic, bacteria widely distributed in nature, home environments, and hospitals. They can also be detected in foods, milk powder, and powdered infant formula (PIF). Additionally, as an opportunistic pathogen, Cronobacter may cause serious infections, sometimes leading to the death of neonates and infants. Thus, it is essential to test food products for the presence of Cronobacter spp. The currently used standard described in ISO 22964:2017 is a laborious method that could be easily replaced by surface-enhanced Raman sca

Searching for molecular abnormalities in genetic diseases by the use of a double labeling technique. II. Deficiency of a basic protein in fibroblasts of patients with Pompe's disease.

We used a double labeling technique to search for molecular defects in two fibroblast strains obtained from patients with Pompe's disease. Analysis of the double labeled subcellular fractions by sodium dodecyl sulfate (SDS) electrophoresis did not reveal any abnormalities except in the "mitochondrial-lysosomal" fraction. In this fraction ratio deviations indicated that in Pompe's disease there was a significant decrease in counts of a protein with molecular weight of about 29,000. After solubilization by freeze-thawing this protein was shown to have an isoelectric point of 7.9 in contrast to t

Protocol for a controlled human infection with genetically modified Neisseria lactamica expressing the meningococcal vaccine antigen NadA: a potent new technique for experimental medicine.

INTRODUCTION: Neisseria lactamica is a commensal organism found in the human nasopharynx and is closely related to the pathogen N. meningitidis (meningococcus). Carriage of N. lactamica is associated with reduced meningococcal carriage and disease. We summarise an ethically approved protocol for an experimental human challenge study using a genetically modified strain of N. lactamica that expresses the meningococcal antigen NadA. We aim to develop a model to study the role of specific bacterial antigens in nasopharyngeal carriage and immunity, to evaluate vaccines for their efficacy in prevent

Inactivation of Sebol Induces Male Infertility in Spodoptera exigua via Arrested Meiosis: A Potential Molecular Target for Genetics-Based Sterile Insect Technique.

The beet armyworm, Spodoptera exigua, is a significant agricultural pest managed primarily through chemical insecticides, which present environmental concerns and promote resistance. Genetics-based sterile insect technique (gSIT) provides an alternative by inducing male sterility without irradiation. In this study, we cloned the S. exigua boule gene (Sebol), a 741-nucleotide open reading frame encoding a 246-amino-acid protein with an RNA recognition motif. Phylogenetic analysis confirmed its conservation across Lepidoptera and high similarity to orthologs in Spodoptera litura and Helicoverpa

Common genetic variants of MUTYH are not associated with cutaneous malignant melanoma: application of molecular screening by means of high-resolution melting technique in a pilot case-control study.

MUTYH glycosylase recognizes the 8-oxoG:A mismatch and is able to excise the adenine base using proofreading mechanisms. Some papers have reported a strong association between cancer development or aggressiveness and MUTYH gene mutations. The aim of this study was to find a possible association between the most frequent MUTYH mutations and melanoma in the context of a case-control pilot study. One hundred ninety-five melanoma patients and 195 healthy controls were matched for sex and age. Clinical and laboratory data were collected in a specific database and all individuals were analyzed for M

Genetic diversity of populations of Old World screwworm fly, Chrysomya bezziana, causing traumatic myiasis of livestock in the Gulf region and implications for control by sterile insect technique.

Fly larvae were collected from 181 cases of traumatic myiasis in livestock in 10 regions of four countries in the Middle East Gulf region: Iran, Iraq, Saudi Arabia and Oman. The predominant fly species responsible for cases was the Old World screwworm (OWS) fly, Chrysomya bezziana Villeneuve (Diptera: Calliphoridae). In cases from Iran and Oman, which included non-OWS fly species, OWS fly was found solely responsible for 67.6% of cases and jointly with other fly species for a further 12.7% of cases. The major hosts were sheep and goats, together comprising 84.6% of the total, which reflects th

Identification and evaluation of two testis-specific serine/threonine kinase genes from multi-tissue transcriptomes as potential genetic targets of sterile insect technique in Zeugodacus tau.

BACKGROUND: Zeugodacus tau (Walker) is a notorious agricultural pest causing significant economic losses in vegetable production for many years. Sterile insect technique (SIT) has emerged as an environmentally sustainable pest management strategy. However, discovery of molecular targets applicable for SIT implementation in Z. tau still constitutes a significant research gap. RESULTS: We conducted comparative transcriptome analysis of four male tissues (midgut, Malpighian tubules, fat body, and testis), identifying 9653 differentially expressed genes (DEGs) with predominant testis enrichment.

Current Diagnostic Approaches in the Genetic Diagnosis of Disorders of Sex Development.

Disorders of sex development (DSD) are a clinically and genetically highly heterogeneous group of congenital disorders. The most accurate and rapid diagnosis may be possible with a complementary multidisciplinary diagnostic approach, including comprehensive clinical, hormonal, and genetic investigations. Rapid and accurate diagnosis of DSD requires urgency in terms of gender selection and management of the case. Despite the genetic tests performed in current daily practice, the genetic cause is still not elucidated in a significant proportion of cases. Karyotype analysis can be used as a stand

Reading disability: evidence for a genetic etiology.

A review of evidence for genetic influences on reading disabilities (RD) is presented, with focus on twin study design and sib-pair linkage techniques. DeFries-Fulker multiple regression analyses result in significant estimates of heritability for group deficits on several reading and language measures. Structural equation modeling techniques reveal the presence of significant common and independent genetic effects on individual differences on reading skills. Finally, linkage techniques confirm a candidate locus for RD on chromosome 6.

[Expert consensus on the genetic diagnosis for Dystrophinopathies].

Dystrophinopathies, including Duchenne muscular dystrophy, Becker muscular dystrophy and dilated cardiomyopathy, are X-linked recessive genetic disorders due to variants of the dystrophin gene, which can seriously affect quality of life and health. Genetic diagnosis plays a crucial role in their diagnosis, treatment, and prevention. How to rationally select and standardize the use of various genetic techniques is a skill that clinicians must acquire. By compiling expertise of experts from the relevant areas and guidelines published home and abroad, this consensus has provided a guidance from t

Genetic susceptibility to Alzheimer disease.

Alzheimer disease (AD) is the leading cause of dementia in the elderly. Less than a decade ago, it was questioned as to whether or not genes were even involved in anything but rare early onset AD. Since that time, using a variety of genetic epidemiological and molecular biological techniques, four loci have been identified that play a role in the genetic susceptibility of AD. AD presents as a prototype of the power of genetic techniques in defining the etiology of a complex disease.
