Red Cell Genotyping by Multiplex PCR Identifies Antigen-Matched Blood Units for Transfusion-Dependent Thai Patients.

BACKGROUND: Antigen-negative red cell transfusion is required for transfusion-dependent patients. We developed multiplex PCR for red cell genotyping and calculated the possibility of finding compatible predicted phenotypes in Thai blood donor populations according to red cell alloantibodies found among Thai patients. METHODS: 600 DNA samples obtained from unrelated healthy central and northern Thai blood donors were tested with the newly developed multiplex PCR for FY*A, FY*B, JK*A, JK*B, RHCE*e, RHCE*E, DI*A and GYP*Hut, GYP*Mur, GYP*Hop, GYP*Bun, and GYP*HF allele detections. Additionally,

Genetics of gestational trophoblastic disease.

The abnormal pregnancies complete and partial hydatidiform mole are genetically unusual, being associated with two copies of the paternal genome. Typical complete hydatidiform moles (CHMs) are diploid and androgenetic, while partial hydatidiform moles (PHMs) are diandric triploids. While diagnosis can usually be made on the basis of morphology, ancillary techniques that exploit their unusual genetic origin can be used to facilitate diagnosis. Genotyping and p57 immunostaining are now routinely used in the differential diagnosis of complete and partial hydatidiform moles, for investigating unus

Comparison of multilocus genotyping and a commercial beta-giardin qPCR assay for detection of Giardia duodenalis zoonotic assemblages in cat and dog samples.

BACKGROUND: Giardia duodenalis is a protozoan of dogs and cats that comprises several genotypes (assemblages A-G). The common assemblages affecting small animals (C, D, F, G) are not associated with disease in humans. However, assemblages A and B can be zoonotic. Several assays can be used for detection of G. duodenalis, but there is a need for an accurate and faster detection method for G. duodenalis zoonotic assemblages. The aim of this study was to compare a beta-giardin quantitative polymerase chain reaction (PCR) (bg-qPCR) to a standard multilocus genotyping technique for detection of G.

Skin Swabbing as a Noninvasive Alternative to Fin Clipping for Commercial Automated Genotyping.

Fin clipping remains the most common sampling method for genotyping zebrafish, yet it requires anesthesia and tissue removal, raising animal welfare concerns and potentially impacting behavior in the short term. To address this, we have validated a non-invasive skin swabbing technique that eliminates the need for fin clipping adults while maintaining genotyping accuracy during automated genotyping. This method involves gently swabbing the fish's skin to collect mucus and epidermal cells for DNA extraction. This approach avoids anesthesia, reduces handling stress, and is much less invasive than

Advancing blood group diagnosis with molecular methods: A focus on clinically challenging cases.

Traditional serological methods often struggle with the accurate identification of rare or complex blood groups. This study addresses the challenges in diagnosing clinically difficult blood groups by employing genotyping and sequencing techniques to enhance blood supply accuracy. Two patients with challenging blood groups underwent serological testing and molecular diagnostics using fluorescent PCR typing and Sanger sequencing. Patient 1 showed a discrepancy in serological typing, indicating a B subgroup confirmed by a B/O\*1 genotype and a 721C>T mutation in the ABO gene Exon 7, establishing

Colorimetric loop-mediated isothermal amplification (cLAMP) assay for the genotyping of a thrombophilia genetic risk factor, MTHFR (C677T).

Methylenetetrahydrofolate reductase (MTHFR) is an enzyme involved in regulating serum homocysteine and folate levels. A single nucleotide polymorphism (SNP) of MTHFR (rs1801133; C677T) is strongly associated with an increased risk of thrombophilia, heart attack, stroke, and dementia. Genotyping this SNP is vital; however, techniques need to be simplified. This study developed a colorimetric loop-mediated isothermal amplification (cLAMP) technique to genotype the C677T in thrombophilia-susceptible individuals. The results were compared with those of kompetitive allele-specific PCR (KASP) and po

Rapid microarray-based genotyping of Chlamydia spp. strains from clinical tissue samples.

Pathogenic Chlamydia (C.) psittaci and C. trachomatis strains can be genotyped based on variations in the ompA genomic locus. In the present chapter, we describe rapid genotyping assays for both chlamydial agents using the ArrayStrip™ (AS) microarray platform. The test is targeting multiple discriminatory sites in the variable domains of the ompA gene by using 35 (C. psittaci) and 61 (C. trachomatis) oligonucleotide probes representing genotype-specific polymorphisms. In addition to discrimination among the established genotypes, this approach allows identification of atypical strains that wer

Genotyping of Listeria monocytogenes isolates from poultry carcasses using high resolution melting (HRM) analysis.

An outbreak situation of human listeriosis requires a fast and accurate protocol for typing Listeria monocytogenes. Existing techniques are either characterized by low discriminatory power or are laborious and require several days to give a final result. Polymerase chain reaction (PCR) coupled with high resolution melting (HRM) analysis was investigated in this study as an alternative tool for a rapid and precise genotyping of L. monocytogenes isolates. Fifty-five isolates of L. monocytogenes isolated from poultry carcasses and the environment of four slaughterhouses were typed by HRM analysis

Application of four molecular typing methods for analysis of Mycobacterium fortuitum group strains causing post-mammaplasty infections.

A cluster of cases of post-augmentation mammaplasty surgical site infections occurred between 2002 and 2004 in Campinas, in the southern region of Brazil. Rapidly growing mycobacteria were isolated from samples from 12 patients. Eleven isolates were identified as Mycobacterium fortuitum and one as Mycobacterium porcinum by PCR-restriction digestion of the hsp65 gene. These 12 isolates, plus six additional M. fortuitum isolates from non-related patients, were typed by pulsed-field gel electrophoresis (PFGE) and three PCR-based techniques: 16S-23S rRNA internal transcribed spacer (ITS) genotypin

Updated overview on von Willebrand disease: focus on the interest of genotyping.

Introduction: Von Willebrand disease (VWD) is the most common inherited bleeding disorder, characterized by a quantitative or qualitative defect of von Willebrand factor (VWF), a multimeric glycoprotein crucial for primary hemostasis and coagulation. VWD pathophysiology is heterogeneous as it includes several types and subtypes which therapeutic management is different. The mainstays of VWD treatment are desmopressin and replacement therapy based on both plasma-derived concentrates and a recently developed recombinant VWF. VWD definitive diagnosis is achieved by a battery of phenotypic biologi

[Detection of a clonal complex with Brucella abortus biovar 2 genotype as founder in B. abortus isolates from Argentina].

Brucella abortus is the causative agent of bovine brucellosis, a worldwide zoonosis. Up to date, eight biovars of B. abortus have been described. In Argentina, biovar 1 is the most frequently isolated. However, biovar 2, which is more pathogenic than biovar 1, is also found. Molecular methods for subtyping isolates are necessary for allowing epidemiological surveillance and control of eradication programs. Due to the genetic homogeneity of the genus Brucella, the development of molecular typing tools has been difficult. The publication of microorganism genomes facilitates the design of this ap

Hydatidiform Moles: The Contribution of Ancillary Techniques in Refining Their Histopathological Diagnosis.

A hydatidiform mole (HM) is the most common form of gestational trophoblastic disease (GTD). Differentiating hydatidiform moles (HMs) from non-molar pregnancies and distinguishing complete HMs (CHMs) from partial HMs (PHMs) remains challenging due to overlapping morphological features and a high rate of misclassification. This study aimed to evaluate reliable immunohistochemical markers for improving diagnostic accuracy and addressing the limitations of current molecular techniques. We retrospectively analyzed 64 cases of HMs and hydropic abortions (HAs), diagnosed in women aged 17-36 years be

First identification of zoonotic assemblages A and B of Giardia duodenalis in mules and donkeys in Brazil.

Research on Giardia duodenalis in mules and donkeys is scarce in Brazil, especially in molecular epidemiology. Therefore, we performed molecular detection and genotyping of G. duodenalis in mules and donkeys from São Paulo, Brazil. Through diagnostic techniques, we compared the agreement between the parasitological examination and the Polymerase Chain Reaction (PCR), through diagnostic techniques. A total of 110 animals, 46 mules, and 64 donkeys were investigated in this study. Fecal samples were collected directly from the rectal ampulla of the animals and submitted to the parasitological inv

In-cell PCR method for specific genotyping of genomic DNA from one individual in a mixture of cells from two individuals: a model study with specific relevance to prenatal diagnosis based on fetal cells in maternal blood.

BACKGROUND: During recent years, much attention has been paid to the possibility of using fetal cells circulating in the pregnant woman's blood for prenatal diagnosis of genetic or chromosomal abnormalities. Although successes have been achieved in enrichment procedures for fetal cells from maternal blood samples, the use of such an approach for genotyping by molecular biology techniques in a more routine setting has been hampered by the large contamination of maternal nucleated blood cells in the cell isolates. Therefore, a new method based on in-cell PCR is described, which may overcome this

Self-Interference Digital Optofluidic Genotyping for Integrated and Automated Label-Free Pathogen Detection.

Pathogen, prevalent in both natural and human environments, cause approximately 4.95 million deaths annually, ranking them among the top contributors to global mortality. Traditional pathogen detection methods, reliant on microscopy and cultivation, are slow and labor-intensive and often produce subjective results. While nucleic acid amplification techniques such as polymerase chain reaction offer genetic accuracy, they necessitate costly laboratory equipment and skilled personnel. Consequently, isothermal amplification methods like recombinase polymerase amplification (RPA) have attracted int

A fetus coexisting with a complete hydatidiform mole with trisomy 9 of maternal origin.

A complete hydatidiform mole (CHM) coexisting with a viable fetus is a rare finding in pregnancies. Accurate diagnosis often relies on ultrasonographic, histopathological and molecular techniques in the definite diagnosis. To the best of our knowledge, a liveborn fetus coexisting with CHM with trisomy 9 has not been described. The use of molecular genotyping and immunohistochemical laboratory investigations enabled the CHM to be fully characterized. Postzygotic diploidization of a triploid conception arising from dispermy is the proposed mechanism of its formation.

Giardia and Cryptosporidium in mammalian wildlife--current status and future needs.

Environmental pollution with human and domestic-animal fecal material is recognized as a potential pathogen pathway for wildlife infections with zooanthropomorphic protozoan parasites such as Giardia and Cryptosporidium. In this article, we review current knowledge about the diversity of free-living and captive terrestrial and marine mammalian wildlife species infected with Giardia and Cryptosporidium. The combination of prevalence studies with modern molecular-genotyping techniques is providing valuable insights into the host specificity and possible transmission routes of these two important

Rapid Identification and Differential Detection of Infectious Bursal Disease Virus by an Innovative ARMS-qRT-PCR Assay.

Infectious bursal disease (IBD), caused by IBD virus (IBDV), is a major threat to the poultry industry, with the cocirculation of multiple genotyping strains complicating the diagnosis and control. Since the differences of antigenicity among different genotyping and their differences with the commonly used vaccine strains, rapid and accurate identification and differential detection are crucial for surveillance and control of IBDV. In this study, a novel amplification refractory mutation system quantitative reverse transcription PCR (ARMS-qRT-PCR) assay, for the rapid differential diagnosis of
