Clinical biochemistry of hyperthermia.

Heatstroke is the most severe form of heat-related disorders that include mild heat intolerance, heat exhaustion and heat stress. The incidence of heat-related disorders is increasing due to several factors that include climate change, co-morbidities and drug usage. Patients with heatstroke present with a core body temperature above 40°C, multiorgan dysfunction and central nervous system disorder. The pathogenesis of heatstroke is not fully understood; however, heat-shock proteins, inflammatory cytokines and their modulators have been implicated. The clinical biochemistry laboratory plays an i

Selective recruitment of NAT10-catalyzed ac4C-modified mRNAs into stress granules by poly(A)-binding protein promotes mRNA stability and plant heat-stress tolerance.

Global warming poses a considerable threat to crop production, making heat stress a challenge in agriculture, yet how epitranscriptomic modifications contribute to plant heat-stress responses remains to be explored. This study reveals the role of phase separation in plant heat-stress tolerance and shows that N-acetyltransferase 10 (NAT10), which encodes a cytosine N4 acetyltransferase, contributes to heat resistance. We found that NAT10 interacts with polyadenylate-binding protein (PABP), which contains intrinsically disordered regions, thereby facilitating the selective recruitment of ac4C-mo

MED26 facilitates Mediator-Pol II complex assembly and promotes nuclear stress body formation during heat stress.

Cells adapt to environmental stress by reprogramming gene expression. During heat stress, the transcription factor heat shock factor 1 (HSF1) translocates into the nucleus and activates heat shock-responsive loci, including the long non-coding RNA HSATIII, promoting nuclear stress body (nSB) formation. We show that the Mediator complex subunit MED26 enhances HSATIII transcription and nSB formation through a mechanism distinct from its role at heat-shock response (HSR) genes. Under heat stress, MED26-containing Mediator is recruited to HSR genes and nSBs through interactions with HSF1 and MED15

[Heat-related problems in the elderly].

In particular in the elderly patient, exposure to heat can lead to disturbances of the circulatory system and of the water and electrolyte balance. Provided that certain prophylactic measures are taken, serious problems are unlikely to occur. Food and drink should be matched to the ambient temperature, and permanent medication should be checked. In the case of confused persons, nursing personnel should substitute for any failure to make the necessary acclimatization changes (appropriate clothing). In the case of incontinent patients, it must be remembered that the diapered area is not availabl

[Heat waves: health impacts].

During the summer of 2003, record high temperatures were reported across Europe, causing thousands of casualties. Heat waves are sporadic recurrent events, characterised by intense and prolonged heat, associated with excess mortality and morbidity. The most frequent cause of death directly attributable to heat is heat stroke but heat waves are known to cause increases in all-cause mortality, specially circulatory and respiratory mortality. Epidemiological studies demonstrate excess casualties cluster in specific risk groups. The elderly, those with chronic medical conditions and the socially i

Impact of lycopene supplementation on oxidative balance, heat-shock proteins, and lipid profile in heat-stressed goat kids.

The present study was conducted to assess the effects of lycopene (LP) supplementation on antioxidant status, lipid peroxidation, heat-shock proteins, and lipid profile in heat-stressed goat kids. Twenty-one healthy growing Barbari goat kids (4-8 months old) with an average body weight of 9.87 ± 1.09 kg were randomly allocated into three experimental groups, comprising seven animals per group. The activities of superoxide dismutase, catalase, glutathione peroxidase, and total antioxidant capacity increased linearly (P < 0.023) with increasing dietary LP levels. Conversely, concentrations of my

Heat and chemical stressors: Independent and interactive impacts on the pig ovary.

Seasonal infertility as a phenotypic consequence of heat stress has been recognized in production animals for decades and more recently in women. Systemic impacts of heat stress include compromised intestinal integrity, and elevated prolactin and insulin. These effects alone can compromise female fertility. There are also ovarian endocrine changes and molecular sequelae that have been identified. This review summarizes the current literature with a focus on the pig as a model. In addition to direct thermal effects on fecundity, the influence of heat on the ovarian capacity to respond to chemic

Caffeine attenuates spermatogenic disorders in mice with induced chronic scrotal hyperthermia.

OBJECTIVE: Chronic scrotal hyperthermia (SHT) can lead to serious disorders of the male reproductive system, with oxidative stress playing a key role in the onset of these dysfunctions. Thus, we evaluated the impact of caffeine, a potent antioxidant, on cellular and tissue disorders in mice with chronic SHT. METHODS: In this experimental study, 56 adult male NMRI mice were allocated into seven equal groups. Apart from the non-treated control group, all were exposed to heat stress. Two groups, termed "preventive" and "curative," were orally administered caffeine. The preventive mice began rece

Qingshu Yiqi tea prescription attenuates heat-stressed injury: network pharmacology and experimental validation.

Heat stress (HS) poses a significant threat to health, necessitating effective interventions. This study investigated the therapeutic mechanisms of Qingshu Yiqi Tea Prescription (QSYQTP) against HS-induced organ damage in mice. Utilizing an integrated pharmacology approach combining histopathology, network pharmacology, biochemical assays, and qRT-PCR, we assessed QSYQTP's effects in a heat-stressed murine model. QSYQTP supplementation suppressed serum aminotransferase activities (ALT, AST), reduced MDA concentrations, and enhanced antioxidant enzymes (T-SOD, GSH). It also downregulated heat s

Effects of heat environments on depressive disorders: Neurogenesis-mediated mechanisms.

Heat may induce depressive disorders by affecting hippocampal neurogenesis. Heat stress may activate the hypothalamic-pituitary-adrenal (HPA) axis, leading to elevated glucocorticoid levels, which in turn cause hippocampal damage, suppress the expression of brain-derived neurotrophic factor (BDNF), impair neurogenesis and synaptic plasticity, and ultimately promote the onset of depression. Glucocorticoid receptors play a dual regulatory role in this process: moderate upregulation can counteract stress, but their deficiency or overactivation may impair both the BDNF signaling pathway and the ne

Heat Stress Alters the Intestinal Microbiota and Metabolomic Profiles in Mice.

BACKGROUND: Heat stress has negative effects on the intestinal health of humans and animals. However, the impact of heat stress on intestinal microbial and metabolic changes remains elusive. Here, we investigated the cecal microbial and metabolic profiles in mice in response to heat stress. METHODS: The mouse heat stress model was constructed by simulating a high-temperature environment. Twenty mice were randomly assigned to two groups, the control group (CON, 25°C) and the heat treatment group (HS, 40°C from 13:00 to 15:00 every day for 7 days). Serum and cecal contents were collected from t

Heat intolerance: predisposing factor or residual injury?

Great individual variability exists in the ability to sustain heat stress. Some individuals are more susceptible to heat than others. Those individuals, among the young active population (athletes, military recruits, laborers), are at risk to contract exertional heat stroke. Low tolerance to heat results from either functional factors, or from congenital or acquired disturbances. In most cases heat intolerance can be foreseen. Cases of dehydration, overweight, low physical fitness, lack of acclimatization, febrile or infectious diseases, and skin disorders should be regarded a priori as predis

3-Hydroxykynurenine as a Potential Ligand for Hsp70 Proteins and Its Effects on Drosophila Memory After Heat Shock.

Kynurenine products of tryptophan metabolism are modifiers of the nervous activity and oxidative processes in mammals and invertebrates. 3-Hydroxykynurenine (3HOK) in moderate concentrations is a lipid peroxidation inhibitor. However, its accumulation and oxidative auto-dimerization lead to oxidative stress development manifested in age-related neurodegenerative diseases (NDD) and neurological disorders provoked by acute stress. Different forms of stress, the mostly studied being heat shock response, rely on functioning of heat shock proteins of the Hsp70 superfamily. Since kynurenines are cal

[Preventive effect of quinacrine on the development of heat injury by stabilizing membrane phospholipids metabolism and beta-adrenoceptor].

Previous studies have demonstrated that the activation of phospholipase A2 (PLA2) during heat stress would caused disorder of membrane phospholipids metabolism, accompanied by a decrease in phosphatidylcholine and phosphatidyl-serine and an increase in arachidonic acid. In heat stressed rats, a down-regulation of beta-adrenergic receptor in lung tissue was observed due to activation of PLA2. In the present work, it was demonstrated that pretreatment of rats with PLA2 inhibitor, quinacrine (20 mg/kg, I.P.), 1 h before heat stress could block these membrane lipid alterations, and the tolerance o

Protective effects of epigallocatechin gallate in the mice induced by chronic scrotal hyperthermia.

One of the most common complications of chronic scrotal hyperthermia (SHT) is a serious disorder in the male reproductive system. The most important factor in the occurrence of these disorders is oxidative stress. Currently, we investigated the effects of epigallocatechin gallate (EGCG), as a highly potent antioxidant, against cells and tissue disorders in mice affected by chronic SHT. Fifty-six male adult NMRI mice were allocated into seven equal groups. Except the non-treated (Control) group, six other groups were exposed to heat stress. Two treated groups including Preventive and Curative r

Children in sauna: electrocardiographic abnormalities.

We studied changes in ECG in 81 children and 20 adolescents and young adults when they were exposed to heat stress in a climatic chamber, which resembled an ordinary Finnish sauna. Changes were similar to those caused by increased sympathetic activity in 57% of the subjects during the bath and in 23% during the recovery period. Three children had extrasystoles during and four after sauna. One previously healthy 5-year-old girl with wandering pacemaker suffered a 3.3 sec sinus arrest and had reversible junctional rhythm during the heat exposure. We conclude that, although these changes were tra

Seasonal heat stress: Clinical implications and hormone treatments for the fertility of dairy cows.

Heat stress has consequences on both the physiology and reproductive performance of cows, but the most dramatic effect for dairy producers is the decrease produced in fertility. The effects of heat stress on fertility include an increased number of days open, reduced conception rate, and larger number of cows suffering different types of anestrus. Once becomes pregnant, heat stress affects also the reproductive success of the cow through its direct effects on the ovary, uterus, gametes, embryo, and early fetus. This article reviews current knowledge of the effects of heat stress on fertility i

Nitric oxide: Potential therapeutic target in Heat Stress-induced Multiple Organ Dysfunction.

As climate change intensifies, urgent action is needed to address global warming and its associated health risks, particularly in vulnerable regions. Rising global temperature and increasing frequency of heatwaves present a hidden health risk, disrupting the body's temperature regulation and leading to severe consequences such as heat stress-induced multiple organ dysfunction (HS-MOD). Multiple organ injury triggered by heat stress involves complex molecular pathways such as nitric oxide dysregulation, inflammation, oxidative stress, mitochondrial dysfunction, calcium homeostasis disruption, a
