Clodronate Liposomes Untangle the Role of Hemocytes in Apis mellifera Response to Temperature Variation and Microbial Infection.

INTRODUCTION: The cellular immune response, mediated by hemocytes, is a fundamental component of honey bee (Apis mellifera) health. However, the specific contribution of hemocyte subtypes to resilience against combined stressors remains poorly characterized. This study investigated the effect of temperature and bacterial challenge on hemocyte abundance. We employed clodronate liposomes (CLD) for the first time in honey bees as a novel tool for the selective depletion of hemocytes to investigate this gap. METHODS: Five-day-old (nurses) and fifteen-day-old (foragers) honey bees were treated wit

Molecular responses of Mytilus coruscus hemocytes to lipopolysaccharide and peptidoglycan as revealed by 4D-DIA based quantitative proteomics analysis.

Mytilus are sessile filter feeders that live in close contact with numerous marine microorganisms. Hemocytes, the immunocompetent cells of Mytilus, participate in the immune response in a very efficient manner. Lipopolysaccharide (LPS) and peptidoglycan (PGN) follow specific microbe/pathogen-associated molecular patterns (MAMPs or PAMPs) and are involved in immune stimulation in host cells. This study evaluated the molecular profiles and reactions at protein level of Mytilus hemocytes after stimulation with LPS and PGN. Mytilus coruscus was challenged in vivo with LPS and PGN. The hemocytes we

Effects of copper pyrithione (CuPT) on apoptosis, ROS production, and gene expression in hemocytes of white shrimp Litopenaeus vannamei.

Copper pyrithione (CuPT) is used globally to prevent biofouling. However, it poses certain risks to aquatic ecosystems. To understand the effects of CuPT on Litopenaeus vannamei after exposure to different concentrations of CuPT (0, 64, and 128 μg/L), the apoptotic cell ratio, production of reactive oxygen species (ROS), and gene expression in the hemocytes were studied at 0, 3, 12, 24, and 48 h. The results revealed that ROS production was induced significantly at 3-48 h only in the 128 μg/L groups. The apoptotic cell ratio was increased significantly at 12 and 24 h in the 64 μg/L groups, and

Production of reactive oxygen species by hemocytes from the marine mussel, Mytilus edulis: lysosomal localization and effect of xenobiotics.

Hemolymph of M. Edulis is rich in phagocytic hemocytes. Hemocytes contain numerous lysosomes which, in turn, contain various hydrolytic enzymes. Phagocytic activity of M. edulis hemocytes is thought to be associated with NAD(P)H-oxidase activity of the plasma membrane. The laser dye, dihydrorhodamine 123 (DHR), was used for cytochemical and biochemical detection of the generation of reactive oxygen species (ROS) by isolated M. edulis hemocytes. Hemocytes readily take up DHR from the suspension medium and selectively concentrate it in the lysosomes, wherein DHR is oxidized to fluorescent rhodam

Schistosoma mansoni in susceptible and resistant snail strains Biomphalaria tenagophila: in vivo tissue response and in vitro hemocyte interactions.

Schistosomiasis is a parasitic disease that is highly prevalent, especially in developing countries. Biomphalaria tenagophila is an important invertebrate host of Schistosoma mansoni in Brazil, with some strains (e.g. Cabo Frio) being highly susceptible to the parasite, whereas others (e.g. Taim) are completely resistant to infection. Therefore, B. tenagophila is an important research model for studying immune defense mechanisms against S. mansoni. The internal defense system (IDS) of the snail comprises hemocytes and hemolymph factors acting together to recognize self from non-self molecular

[A morphofunctional analysis of the hemocytes in the cricket Gryllus bimaculatus (Orthoptera: Gryllidae) normally and in acute microsporidiosis due to Nosema grylli].

Microsporidians (M) are supposed to be ancient eukaryotic parasites with a broad range of animal hosts, being especially abundant in Arthropoda. They are supposed to pass a long way of adaptation to parasitism, that usually means inhibiting or avoiding host immune reactions alongside with the reduction of pathogenicity. However M, unlike other eukaryotic obligate parasites, preserved a high pathogenicity, comparable with one of viruses, and thus they could be expected to possess a unique mode of interactions with their hosts. The goal of the present work is to assess how M influence the cellul

Effects of parasitism and pesticide exposure on characteristics and functions of hemocyte populations in the freshwater snail Lymnaea palustris (Gastropoda, Pulmonata).

Morphological characteristics and functions of hemocytes were used to compare the immunological effects of biological and chemical stress in the freshwater snail Lymnaea palustris. Animals were either infected by a trematode parasite (Metaleptocephalus sp.), or exposed to environmental contaminants, namely atrazine and hexachlorobenzene (HCB). Three populations of circulating hemocytes, morphologically and cytochemically distinct (round cells, hyalinocytes, granulocytes), were identified in both control and parasitized or pesticide-exposed snails. After 6 h of exposure, HCB and atrazine result

Specific inhibitors of mitogen-activated protein kinase and PI3-K pathways impair immune responses by hemocytes of trematode intermediate host snails.

To characterize molecular mechanisms regulating snail cellular immune responses, the contributions of mitogen-activated protein kinases (MAPKs) and phosphatidylinositol 3-kinase (PI3-K) were examined in hemocytes of the trematode intermediate host snails Biomphalaria glabrata and Lymnaea stagnalis. Simultaneous measurement of phagocytosis/encapsulation and H2O2 production by hemocytes in the presence or absence of specific signal transduction inhibitors was used to assess the role of extracellular-signal regulated kinases 1 and 2 (ERK1/2), p38, JNK and PI3-K. Hemocyte spreading was significant

Life-Stage-Specific Hematological Profiles in Yellow Mealworm Beetles (Tenebrio molitor): Reference Intervals and Hemocyte Characterization.

BACKGROUND: The edible insect industry, driven by the nutritional and environmental benefits of mealworms, faces increasing challenges from pathogens affecting Tenebrio molitor farming systems. Hemocyte-based health assessments could provide valuable diagnostic insights, yet standardized hemolymph processing protocols and hematological reference intervals (RIs) are currently lacking for this species. OBJECTIVES: This study aimed to develop practical protocols for hemolymph sampling, refine hemocyte classification, and establish hematological RIs for both larval and adult stages of T. molitor.

A type III heat shock protein 40 regulates the expression of antimicrobial peptides and hemocyte phagocytosis in Portunus trituberculatus.

Heat shock protein 40 (HSP40) is molecular chaperone that participates in various abiotic and biotic stress. In this study, a type III HSP40 (PtHSP40-III) that mainly expresses in the thoracic ganglion of Portunus trituberculatus is identified. Despite PtHSP40-III shows relatively lower transcriptional level in hemocyte, its transcripts could be significantly induced by Vibrio alginolyticus. The recombinant protein of PtHSP40-III displays binding activity toward both Gram-negative bacteria and Gram-positive bacteria as well as pathogen-associated molecular patterns (PAMPs) such as lipopolysacc

Diversity of Toll-like Receptor Gene Transcripts in Hemocytes of Planorbarius corneus Mollusks (Gastropoda, Pulmonata) Uninfected and Infected with Bilharziella polonica.

The hemocyte transcriptome was for the first time obtained and analyzed in uninfected Planorbarius corneus snails and snails naturally infected with Bilharziella polonica parthenites (Schistosomatidae). Transcripts encoding all groups of immune factors previously described in other gastropods were found in assemblies. Pathogen recognition molecules were the most diverse group of transcripts encoding immunity factors. Toll-like receptors (TLRs) of 11 types were identified. The TLR set did not differ between infected and uninfected snails. The data expand the knowledge of snail immune responses

Jack bean (Canavalia ensiformis) urease induces eicosanoid-modulated hemocyte aggregation in the Chagas' disease vector Rhodnius prolixus.

Ureases are multifunctional proteins that display biological activities independently of their enzymatic function, such as induction of exocytosis and insecticidal effects. Rhodnius prolixus, a major vector of Chagas' disease, is a model for studies on the entomotoxicity of jack bean urease (JBU). We have previously shown that JBU induces the production of eicosanoids in isolated tissues of R. prolixus. In insects, the immune response comprises cellular and humoral reactions, and is centrally modulated by eicosanoids. Cyclooxygenase products signal immunity in insects, mainly cellular reaction

Male and Female Cat Fleas (Ctenocephalides felis) Differ in Bacterial Control, Survival, and Circulating Hemocyte Responses Following Systemic Challenge.

Although both male and female cat fleas (Ctenocephalides felis) consume vertebrate blood throughout adulthood, previous studies of flea immunity have focused primarily on females, leaving whether systemic immune responses differ between the sexes largely unknown. To address this knowledge gap, we compared bacterial persistence, survival, circulating hemocyte abundance, and phagocytic activity in male and female C. felis following microinjection of Escherichia coli into the hemocoel. Females exhibited significantly lower infection prevalence and bacterial burdens than males 24 h post-challenge.

Laboratory blood test profiling reveals distinct biochemical and hemocyte features of KRAS mutated non-small cell lung cancer.

BACKGROUND: The testing for capability of some routine blood test parameters to reflect the biology of non-small cell lung carcinoma with different driver mutations is of great interest and practice significance. We aim to screen these variables and, if allowed, develop a novel predictive model based on results of these routine blood tests commonly performed in clinical practice to inform which can help doctors assess the patient's genetic mutation status as early as possible before surgery. METHODS: For the exploration cohort, we included 1,595 patients who were diagnosed with non-small cell

Down Syndrome Cell Adhesion Molecule Triggers Membrane-to-Nucleus Signaling-Regulated Hemocyte Proliferation against Bacterial Infection in Invertebrates.

Down syndrome cell adhesion molecule (Dscam) generates tens of thousands of isoforms by alternative splicing, thereby providing crucial functions during immune responses. In this study, a novel Dscam signaling pathway was investigated in crab, which remains poorly characterized in invertebrates. Bacterial infection induced the cytoplasmic cleavage of Dscam intracellular domains (ICDs) by γ-secretase, and then the released ICDs carrying specific alternatively spliced exons could directly interact with IPO5 to facilitate nuclear translocation. Nuclear imported ICDs thus promoted hemocyte prolife

Ultrastructural localization of a sialic acid-specific hemolymph lectin in the hemocytes and other tissues of the hard tick Ixodes ricinus (Acari; Chelicerata).

Lectins have been suggested to function as pattern-recognition molecules in invertebrate immune mechanisms. A lectin from the hemolymph of the tick Ixodes ricinus with main specificity for sialic acid was characterized and antibodies directed against this lectin were prepared. In this study, these antibodies were used to localize the lectin in the tissues of I. ricinus. Immunoreactivity with poly- and monoclonal antibodies was detected in the granules of both types of granular hemocytes, at the membrane of hemocytes, and at the basal laminae surrounding the hemocoel. Furthermore, cells attache

Cytoplasmic tail of transmembrane dscam controls antibacterial responses by regulating cell proliferation-related genes in hemocytes of Chinese mitten crab (Eriocheir sinensis).

In arthropods, the involvement of Dscam (Down syndrome cell adhesion molecule) in innate immunity has been extensively demonstrated. Its cytoplasmic tail contains multiple conserved functional sites, which indicates its involvement in different intracellular signaling pathways. In this study, we focused on the role of the cytoplasmic tail of Dscam in the Chinese mitten crab (Eriocheir sinensis) immune defense. In the group with cytoplasmic tail knockdown (the site was located on constant exons 37 and 38), 3885 differentially expressed genes (DEGs) were identified. The DEGs were enriched in sma

Innate immunity in the edible ascidian Halocynthia roretzi developing soft tunic syndrome: Hemolymph can eliminate the causative flagellates and discriminate allogeneic hemocytes.

The infection of the kinetoplastid flagellate Azumiobodo hoyamushi causes soft tunic syndrome that often results in mass mortality in the aquaculture of the edible ascidian Halocynthia roretzi. In the diseased ascidian individuals, the flagellates are exclusively found in the tunic matrix that entirely cover the epidermis, and never invade into internal tissues, such as a mantle. The present study for the first time demonstrated that the ascidian blood plasma and hemolymph have an activity to agglutinate and disintegrate the flagellates, suggesting the innate immunity protects the internal tis

Eriocheir sinensis 14-3-3ζ regulates hemocyte phagocytosis, apoptosis and antimicrobial peptide expression during Spiroplasma eriocheiris infection.

Tremor disease, caused by Spiroplasma eriocheiris in the Chinese mitten crab (Eriocheir sinensis), is highly destructive, leading to mass mortality and substantial economic losses in crab aquaculture. Our previous studies have confirmed that E. sinensis 14-3-3ζ (Es14-3-3ζ) contributes to host defense against S. eriocheiris infection in crabs. However, the detailed mechanism by which Es14-3-3ζ regulates host resistance to S. eriocheiris requires further investigation.
