Data on LC-MS profile of Brucea javanica (L.) Merr. and the NMR data of its major indole alkaloids.

This article presents two types of phytochemical data obtained from Brucea javanica (L.) Merr. roots, a medicinal plant belonging to the Simaroubaceae family. The high-resolution LC-MS dataset comprised the chemical profile of dichloromethane extract, which was utilised to annotate 35 chemical constituents. For annotations, the measured spectral data were compared with the in-silico spectral data generated from 920 molecular structures previously reported in Simaroubaceae. Indole alkaloids, quassinoids, aliphatics and lignan were the chemical groups identified in the root extract. The second d

Single-nucleus RNA Sequencing and multi-omics reveal Uncaria-derived indole alkaloids induce hepatocyte injury by mediating CAR/PPARα axis.

BACKGROUND: Risk assessment and management of endogenous potentially toxic components are critical for promoting the rational clinical use of herbal medicines. However, most herbal medicines lack sufficient safety data in clinical, especially for those with multiple botanical sources. As a commonly used multi-botanical source herbal medicine, Uncariae Ramulus Cum Uncis-derived indole alkaloids (IA-URCU) are primarily responsible for its potential hepatotoxicity in clinical practice. Nevertheless, the underlying mechanism of URCU-induced hepatocyte injury remains unclear. PURPOSE: This study a

Discovery of Indole Alkaloids Crienamides A and B from Penicillium citrinum by a Simulated MS/MS-Guided Molecular Network Strategy.

The building blocks-based molecular network (BBMN) strategy is effective for targeted natural product identification but depends on existing MS/MS data. To address this, we propose the simulated MS/MS-guided molecular network (SMMN) strategy, which combines virtual molecule design and MS/MS simulation to identify novel compounds containing target building blocks. We developed the MZmol Analyzer, a user-friendly web tool, to automate the SMMN strategy. Using this approach and the target 6-methyl-l-pipecolate (6-MeP), we identified two new indole alkaloid hybrids, crienamides A and B. In vitro i

A Novel Series of Indole Alkaloid Derivatives Inhibit Dengue and Zika Virus Infection by Interference with the Viral Replication Complex.

Here, we identified a novel class of compounds which demonstrated good antiviral activity against dengue and Zika virus infection. These derivatives constitute intermediates in the synthesis of indole (ervatamine-silicine) alkaloids and share a tetracyclic structure, with an indole and a piperidine fused to a seven-membered carbocyclic ring. Structure-activity relationship studies indicated the importance of substituent at position C-6 and especially the presence of a benzyl ester for the activity and cytotoxicity of the molecules. In addition, the stereochemistry at C-7 and C-8, as well as th

Genome-wide association studies reveal genetic variants associated with antineoplastic monoterpenoid indole alkaloid accumulation in Catharanthus roseus.

Catharanthus roseus produces pharmacologically important monoterpenoid indole alkaloids (MIAs), yet their natural accumulation is low, limiting therapeutic exploitation. To dissect the genetic basis of natural variation in MIA accumulation, we integrated phenotypic, chemotypic, and genomic analyses of 93 C. roseus accessions sampled from six locations across India, including New Delhi, Lucknow, Jodhpur, Bangalore, and two locations in Gujarat: Navsari and Bardoli. Morphological characterization showed limited differentiation among locations, whereas accessions from Gujarat tended to be taller
