The feedback phase of type I interferon induction in dendritic cells requires interferon regulatory factor 8.

Dendritic cells (DCs) produce type I interferons (IFNs) in greater amounts than other cells, but the mechanisms remain elusive. Here we studied the role of a transcription factor, IRF8, in DC induction of type I IFNs. Upon newcastle disease virus (NDV) infection, bone marrow-derived plasmacytoid and conventional DCs induced IFN transcripts, exhibiting two-phase kinetics. The second, amplifying phase represented an IFN feedback response that accounted for much of IFN protein production. Induction of second phase transcription required IRF8. Mouse cytomegalovirus (MCMV) and Toll-like receptor-me

Striking the right balance with type I interferon signalling in cancer.

Type I interferons (IFNs), particularly IFNα and IFNβ, have an important role in cancer therapy, enhancing antitumour immunity and improving the efficacy of both conventional treatments and immunotherapies. However, despite considerable investment and research in IFN-based treatments, clinical success in solid malignancies has been hampered by toxicity and limited therapeutic efficacy. Recent studies show that type I IFNs can exert both immune-stimulatory and immune-suppressive effects within tumours, with their activity shaped by oncogenic signalling, chromatin state, the tumour microenvironm

Anti-inflammatory roles of type I interferon signaling in the lung.

Excessive or persistent inflammation may have detrimental effects on lung structure and function. Currently, our understanding of conserved host mechanisms that control the inflammatory response remains incompletely understood. In this study, we investigated the role of type I interferon signaling in the inflammatory response against diverse clinically relevant stimuli. Using mice deficient in type I interferon signaling (IFNAR1-/-), we demonstrate that the absence of interferon signaling resulted in a robust and persistent inflammatory response against Pseudomonas aeruginosa, lipopolysacchari

[Potential role of type I interferon in the genetic progression of melanoma].

We have followed the genomic progression of cutaneous melanoma in visceral metastases using genome-wide copy number analysis. We have detected an increased chromosomal instability due to the loss of several DNA repair genes. Furthermore, we found co-amplifications of HGF and MET genes in metastases. The most interesting finding was gene amplifications of several, mostly IFN-regulated immune cell genes in lung metastases. Next we have defined a type I IFN resistance gene expression signature (GES) using human cell lines, several elements of which were proved to be stable in vitro and in vivo as

Acute activation of bronchopulmonary vagal nociceptors by type I interferons.

Type I interferon receptors are expressed by the majority of vagal C-fibre neurons innervating the respiratory tract Interferon alpha and beta acutely and directly activate vagal C-fibers in the airways. The interferon-induced activation of C-fibers occurs secondary to stimulation of type 1 interferon receptors Type 1 interferons may contribute to the symptoms as well as the spread of respiratory viral infections by causing coughing and other defensive reflexes associated with vagal C-fibre activation ABSTRACT: We evaluated the ability of type I interferons to acutely activate airway vagal aff

Differential Induction of Type I and III Interferons by Staphylococcus aureus.

Staphylococcus aureus is a leading cause of bacterial pneumonia, and we have shown previously that type I interferon (IFN) contributes to the pathogenesis of this disease. In this study, we screened 75 S. aureus strains for their ability to induce type I and III IFN. Both cytokine pathways were differentially stimulated by various S. aureus strains independently of their isolation sites or methicillin resistance profiles. These induction patterns persisted over time, and type I and III IFN generation differentially correlated with tumor necrosis factor alpha production. Investigation of one is

Evasion mechanisms of the type I interferons responses by influenza A virus.

The type I interferons (IFNs) represent the first line of host defense against influenza virus infection, and the precisely control of the type I IFNs responses is a central event of the immune defense against influenza viral infection. Influenza viruses are one of the leading causes of respiratory tract infections in human and are responsible for seasonal epidemics and occasional pandemics, leading to a serious threat to global human health due to their antigenic variation and interspecies transmission. Although the host cells have evolved sophisticated antiviral mechanisms based on sensing i

Distinct Effects of Type I and III Interferons on Enteric Viruses.

Interferons (IFNs) are key host cytokines in the innate immune response to viral infection, and recent work has identified unique roles for IFN subtypes in regulating different aspects of infection. Currently emerging is a common theme that type III IFNs are critical in localized control of infection at mucosal barrier sites, while type I IFNs are important for broad systemic control of infections. The intestine is a particular site of interest for exploring these effects, as in addition to being the port of entry for a multitude of pathogens, it is a complex tissue with a variety of cell type

Divergent effects of type-I interferons on regulatory T cells.

Regulatory T cells (Treg) exert a dominant role in the protection of unwanted immune responses and in the resolution of inflammation. To ensure the proper mounting of protective immune responses, Treg should be finely modulated by microenvironmental signals, mostly conveyed by cytokines. Type-I interferons are pleiotropic cytokines, best known for their anti-viral activities but also playing relevant immunostimulatory as well as immunomodulatory functions. The impact of type-I interferons on Treg homeostasis and functions is quite controversial, as some studies indicate that interferons sustai

Type I and III interferon production in response to RNA viruses.

The biology of RNA viruses is closely linked to the type I and type III interferon (IFN) response of the host. These viruses display a range of molecular patterns that may be detected by host cells resulting in the induction of IFNs. Consequently, there are many examples of mechanisms employed by RNA viruses to block or delay IFN induction and reduce the expression of IFN-stimulated genes (ISGs), a necessary step in the virus lifecycle because of the capacity of IFNs to block virus replication. Efficient transmission of viruses depends, in part, on maintaining a balance between virus replicati

Characterization of the human type I interferon receptor by ligand blotting.

The human type I interferon (IFN) receptor was characterized by ligand blotting. In this method, plasmalemma proteins or detergent-lysed whole-cell extracts from human Burkitt lymphoma cell lines were separated on polyacrylamide gels and subsequently transferred onto nitrocellulose sheets. Probing the blots with 3 x 10(-10) M 125I-labeled recombinant IFN-alpha A (125I-rIFN-alpha A) revealed an IFN-alpha-binding protein with an apparent molecular mass of 95 kDa (p95). Performing the electrophoretic run under reducing conditions completely abrogated the signal on the blot, indicating that the ty

TRPM2 couples cell-autonomous type-I interferon signaling to pigmentation homeostasis.

Transient Receptor Potential Melastatin 2 (TRPM2), a Ca2+-permeable cation channel, regulates innate and adaptive immunity and has recently been implicated in vitiligo, an autoimmune pigmentary disorder. However, whether TRPM2 exerts cell-autonomous immunoregulatory functions and how such signaling intersects with pigmentation remain unknown. Here, we reveal an unexpected role for TRPM2 as an intrinsic suppressor of pigmentation through type-I interferon (IFN) signaling in melanocytes. Pharmacological inhibition, genetic silencing, and gain-of-function approaches demonstrate that TRPM2 negativ

Prevalence of neutralizing autoantibodies against type I interferon in dengue patients.

The presence of neutralizing autoantibodies (NAbs) against type I IFN (IFN-I) has been associated with severe complications arising from various viral infections. However, the prevalence of IFN-I NAbs in dengue virus (DENV) infection remains unclear. Here, we enrolled 216 hospitalized dengue patients in 2024, including 7 individuals with dengue shock syndrome (DSS) and 42 individuals with dengue hemorrhagic fever (DHF). Utilizing a sensitive luciferase reporter assay, IFN-I NAbs were detected in 7 patients (14.3% of DSS/DHF cases), and their neutralizing ability was further validated in DENV-i

Type-I interferon-activated NK cells control polycythemia vera in vivo.

Polycythemia vera (PV) is a clonal hematopoietic stem cell disorder resulting in overproduction of erythrocytes. Although interferon alfa (IFN-α) has shown therapeutic efficacy in PV and other myeloproliferative neoplasms (MPN), its precise mechanism of action remains poorly understood. In this study, we identify natural killer (NK) cells as primary immune effectors responsive to IFN-α treatment in PV essential for disease control in vivo. Using a transgenic mouse model of PV, we demonstrate that IFN-α induces the expansion of CD27+ NK cells in the bone marrow. In patients with PV or essential

Systemic inflammation impairs myelopoiesis and interferon type I responses in humans.

Systemic inflammatory conditions are classically characterized by an acute hyperinflammatory phase, followed by a late immunosuppressive phase that elevates the susceptibility to secondary infections. Comprehensive mechanistic understanding of these phases is largely lacking. To address this gap, we leveraged a controlled, human in vivo model of lipopolysaccharide (LPS)-induced systemic inflammation encompassing both phases. Single-cell RNA sequencing during the acute hyperinflammatory phase identified an inflammatory CD163+SLC39A8+CALR+ monocyte-like subset (infMono) at 4 h post-LPS administr

Keratin 16 spatially inhibits type I interferon responses in stressed skin.

The stress-induced keratin intermediate filament gene/protein KRT16 (K16) is spatially restricted to the suprabasal compartment of the epidermis and extensively used as a biomarker for psoriasis, hidradenitis suppurativa, atopic dermatitis and other inflammatory disorders. However, its role in these conditions remains poorly defined. Here we show that K16 negatively regulates type-I interferon (IFN) signaling and innate immune responses. In mouse skin in vivo, loss of Krt16 leads to exacerbation of imiquimod-induced psoriasiform disease and heightened recruitment of neutrophils in a phorbol es
