Characterization of Jk(a+(weak)): a new blood group phenotype associated with an altered JK*01 allele.

BACKGROUND: The clinically important Kidd (JK) blood group system is considered to be relatively uncomplicated, both serologically and genetically. The JK*01 and JK*02 alleles give rise to Jk(a) and Jk(b) antigens, respectively, and silenced alleles result in Jk(a-b-). Other inherited variants analogous to Fy(x) and weak D phenotypes have not been characterized for JK, although recent abstracts indicate their presence. STUDY DESIGN AND METHODS: Six index samples from individuals whose RBCs reacted variably or weakly with different sources of anti-Jk(a) and 300 controls of the four known JK ph

Hematopoietic Chimera in a Male Blood Donor and His Dizygotic Twin Sister.

Twin hematopoietic chimera in humans is a phenomenon that was discovered accidentally and the prevalence of which remains unclear. The resolution of chimera cases requires studying family medical records, data analysis, and investigations of hematopoietic cells and cells from other tissues. The interactions among ABO, Lewis, and secretor histo-blood group systems are explored to resolve cases of hematopoietic chimera. Here we report a rare case of hematopoietic chimera where twins present a mixed field reaction in the ABO, Rh, and Kidd red blood cell phenotyping. Using red blood cells separate

Terminal and interstitial deletions of the long arm of chromosome 7: a review with five new cases.

Sixteen cases of terminal deletions and 17 cases of interstitial deletions of the long arm of chromosome 7 have been reported to date. We present two new cases of the former and three of the latter. The somatic changes in these patients are tabulated and an update on the anomalies associated with the various cytogenetic entities is presented. Changes found in over one-third of patients with 7q terminal deletion syndrome include: developmental delay, pre- and postnatal growth retardation, generalized hypotonia, abnormal electroencephalograms with or without seizures, feeding problems in infancy

Neonatal and Obstetrical Outcomes of Pregnancies Complicated by Alloimmunization.

BACKGROUND AND OBJECTIVES: Despite advances in the prevention of rhesus (Rh)(D) alloimmunization, alloantibodies to Rh(D) and non-Rh(D) red blood cell antigens continue to be detected in ∼4% of US pregnancies and can result in hemolytic disease of the fetus and newborn (HDFN). Recent reports on HDFN lack granularity and are unable to provide antibody-specific outcomes. The objective of this study was to calculate the frequency of alloimmunization in our large hospital system and summarize the outcomes based on antibody specificity, titer, and other clinical factors. METHODS: We identified all

A uniform method for the simultaneous blood group phenotyping of Fya , Fyb , Jka , Jkb , S, s̅, P1, k applying lateral-flow technique.

BACKGROUND AND OBJECTIVES: A lateral flow assay for simultaneous blood group typing of ABO, RhD, C, E, c, e, Cw and K with stable end-point and without centrifugation is in routine use since several years (MDmulticard® ). The typing of extended phenotype parameters belonging to the Duffy, Kidd, MNSs blood group systems and others, however, has not yet been demonstrated for this technique. Reliable detection of Fyx , a weak Fyb phenotype with a pronounced quantitative reduction of the number of Fyb antigens on the erythrocyte surface, remains a weakness of current serological blood grouping tec

Blood transfusion in beta thalassaemia major.

Conventional treatment of beta thalassaemia major is based on regular blood transfusion from early childhood. Maximum effectiveness of transfusion therapy depends on the following. (1) Availability of safe blood. Donation programmes should aim at retaining repeat donors, who carry decreased risk of transmitting blood-borne infections. Donors should be screened with laboratory tests performed to the highest possible standard of quality. Selection of safe donors can be improved by the adoption of questionnaires containing direct questions on risk factors for transfusion transmissible infections.

Red Blood Cell Alloimmunization in Korean Patients With Myelodysplastic Syndrome and Liver Cirrhosis.

Red blood cell (RBC) alloimmunization varies across human populations and ethnic groups. We evaluated the characteristics of RBC alloimmunization and compared the risk of alloimmunization in Korean patients with myelodysplastic syndrome (MDS) and liver cirrhosis (LC), two representative diseases in which chronic transfusion is required. In total, 115 MDS patients and 202 LC patients transfused with RBCs between 2013 and 2015 were retrospectively included. Twenty patients (6.3%) were newly alloimmunized (five MDS patients, 4.3%; 15 LC patients, 7.4%). The median number of RBC units transfused i

Autologous Infant and Allogeneic Adult Red Cells Demonstrate Similar Concurrent Post-Transfusion Survival in Very Low Birth Weight Neonates.

OBJECTIVE: Based on the hypothesis that neonatal autologous red blood cell (RBC) survival (RCS) is substantially shorter than adult RBC, we concurrently tracked the survival of transfused biotin-labeled autologous neonatal and allogeneic adult RBC into ventilated, very low birth weight infants. STUDY DESIGN: RBC aliquots from the first clinically ordered, allogeneic adult RBC transfusion and from autologous infant blood were labeled at separate biotin densities (biotin-labeled RBC [BioRBC]) and transfused. Survival of these BioRBCs populations were concurrently followed over weeks by flow cyt

Analysis of haemovigilance reports reveals 12.5% of acute haemolytic transfusion reactions are attributed to antibodies to low-incidence antigens.

BACKGROUND AND OBJECTIVES: Many transfusion centres no longer perform an antiglobulin crossmatch on patients without clinically significant red cell alloantibodies, but instead use an abbreviated crossmatch. This policy brings many benefits but has an acknowledged risk of haemolytic transfusion reactions (HTRs). Historical estimates of reactions caused by undetected antibodies to low-incidence antigens (LIAs) were in the region of 1 reaction per 500,000 red cell transfusions. This meta-analysis of haemovigilance reports compares historical estimates with recorded events. MATERIALS AND METHODS

Extended red cell antigen frequencies in a Southwestern Colombian donor population: Implications for phenotype-matched transfusion strategies.

BACKGROUND AND AIMS: The complex history of genetic admixture in Latin American populations creates unique erythrocyte antigen profiles. This study aimed to determine the extended red blood cell (RBC) antigen frequencies in a blood donor population from Southwestern Colombia, a region with significant African ancestral heritage, to support phenotype-matched transfusion strategies and optimize donor selection. MATERIALS AND METHODS: A cross-sectional study was conducted on 501 voluntary blood donors at a highly complex hospital blood bank in Cali, Colombia. Extended serological phenotyping for

A Gly565-->Ala substitution in human erythroid band 3 accounts for the Wu blood group polymorphism.

BACKGROUND: Reports published in 1976 and 1980 described the low-incidence red cell antigen Wu. Distinction of Wu from all other known low-incidence antigens and from the ABO, Rh, Lutheran, Duffy, Kidd, P, and X-linked blood group systems allowed Wu to be placed in the International Society of Blood Transfusion's 700 series, designated as 700013. Recently, a blood donor apparently homozygous for Wu has been identified. This report documents the serologic and molecular findings of samples from this individual and the members of his family. STUDY DESIGN AND METHODS: Blood samples from 26 member

Red cell alloimmunization in multitransfused hepatobiliary patients at hospital Selayang.

BACKGROUND: Transfusion support is vital for the management of patients with hepatobiliary disease. Repeated blood transfusions increase the risk of alloimmunization, i.e., the development of alloantibodies, which might lead to difficulties in blood crossmatching. AIMS: This study aims to: (1) determine the incidence of red blood cell (RBC) alloimmunization and (2) evaluate the associations between antibody development and demographic factors among hepatobiliary patients. METHOD: ABO blood grouping, antibody screening, antibody identification and crossmatch were done on all patients samples

Red blood cell alloimmunization in transfused oncology patients at a high-complexity center in Colombia.

BACKGROUND: Red blood cell transfusion is essential for managing anemia in cancer patients but can induce erythrocyte alloimmunization, particularly against Rh (anti-E) and Kell (anti-K) antigens, which complicates transfusion compatibility and increases the risk of hemolytic events. This study assessed the prevalence and profile of irregular antibodies in transfused oncology patients at a high-complexity center. METHODS: A retrospective descriptive study analyzed medical records of 179 adult patients with solid tumors or hematologic neoplasms who received red blood cell transfusions. Demogra

Urea transport and Kidd blood groups.

The gene encoding for the human erythrocyte urea transporter (HUT11 clone) has recently been cloned (Olives et al., J. Biol. Chem., 269, 3/649, 1994). It has been localized at 18q12-q21, like the Kidd blood group gene. In vitro translation experiments indicated that cDNA HUT11 could induce the synthesis of a 36 kDa protein which can be immunoprecipitated by an anti-Jk3 antibody (a human antibody produced by Jk(a-b-) individuals). This antibody also precipitates a 46-60 kDa protein from human red blood cells, except from those with Jk(a-b-) phenotype. After N-glycanase digestion, the length of

The search for heterogeneity in insulin-dependent diabetes mellitus (IDDM): linkage studies, two-locus models, and genetic heterogeneity.

One hundred families with insulin-dependent diabetes mellitus (IDDM) were analyzed for linkage with 27 genetic markers, including HLA, properdin factor B (BF), and glyoxalase 1(GLO) on chromosome 6, and Kidd blood group (Jk) on chromosome 2. The linkage analyses were performed under several different genetic models. An approximate correction for two-locus linkage analysis was developed and applied to four markers. Two different heterogeneity tests were implemented and applied to all the markers. One, the Predivided-Sample Test, utilizes various criteria thought to be relevant to genetic hetero

Red blood cell extended antigen typing in Omani patients with sickle cell disease to enhance daily transfusion practice.

Many Omani patients with sickle cell disease (SCD) undergo red blood cell (RBC) transfusions that are only matched for ABO and D, making RBC alloimmunization a significant concern in this population. Currently, the integration of molecular assays and hemagglutination testing helps to determine RBC phenotypes and genotypes, facilitating the provision of compatible blood and minimizing additional alloimmunization risks in patients with SCD. Based on this finding, our objective was to use molecular methods to predict the extended antigen profile of Omani patients with SCD across various blood gro

Red blood cell alloantibodies in transfused patients with haematological malignancies at Mbarara Regional Referral Hospital and the Uganda Cancer Institute: Prevalence, specificities and associated factors.

BACKGROUND AND OBJECTIVES: Patients with haematological malignancies often require blood transfusion support. Multiple allogeneic blood transfusions may result in alloimmunization, complicating future transfusions. This study determined alloantibody prevalence, specificities and factors associated with the presence of red blood cell (RBC) alloantibodies among transfused patients with haematological malignancies at Mbarara Regional Referral Hospital (MRRH) and the Uganda Cancer Institute (UCI).
