Manganese deficiency and toxicity: effects on carbohydrate metabolism in the rat.

Although manganese deficiency and manganese toxicity both have pathological consequences, the underlying biochemical lesions have not been well defined. Manganese is involved in carbohydrate metabolism; either deficiency or excess results in abnormal carbohydrate metabolism. Clinical studies have shown that patients with chronic manganism have hypoglycemia following a glucose load. One report has been published of a diabetic patient who responded to oral manganese with a consistent drop in blood glucose. Rats fed a manganese deficient diet respond to an oral glucose load with a diabetic type o

Toxic-metabolic, nutritional, and medicinal-induced disorders of cerebellum.

The human cerebellum is composed of 2 hemispheres and a narrow medial section (vermis). Three pairs of dense fiber bundles (peduncles) connect the cerebellum to the brain. The cerebellum possesses widespread outgoing connections. Insult can result in neurologic deficits, including ataxia, hypotonia, dysarthria, and ocular motility problems. It is particularly susceptible to toxic effects of metabolic and medicinal insults. The cerebellum is potentially sensitive to alcohol, drug exposure, illicit drugs, and environmental poisons (mercury, lead, manganese, and toluene/benzene derivatives). The

Neurotoxicity of Vanadium.

Vanadium (V) is a transition metal that presents in multiple oxidation states and numerous inorganic compounds and is also an ultra-trace element considered to be essential for most living organisms. Despite being one of the lightest metals, V offers high structural strength and good corrosion resistance and thus has been widely adopted for high-strength steel manufacturing. High doses of V exposure are toxic, and inhalation exposure to V adversely affects the respiratory system. The neurotoxicological properties of V are just beginning to be identified. Recent studies by our group and others

The genetic toxicology of metal compounds: I. Induction of lambda prophage in E coli WP2s(lambda).

A number of metal compounds have been shown to be human carcinogens. Others, while not proven human carcinogens, are able to cause tumors in laboratory animals. Short-term bacterial assays for genotoxic effects have not been successful in predicting the carcinogenicity of metal compounds. We report here the ability of some metal compounds to cause the induction of lambda prophage in E coli WP2s(lambda). By far the strongest inducing ability was observed with K2CrO4, followed by Pb(NO3)2 greater than MnCl2 greater than Ni(OOCCH3)2 greater than CrCl2 greater than NaWO4 greater than Na2MoO4 great

Metal-induced neurotoxicity in a RAGE-expressing C. elegans model.

Environmental and occupational metal exposure poses serious global concerns. Metal exposure have severally been associated with neurotoxicity and brain damage. Furthermore, receptor for advanced glycation end products (RAGE) is also implicated in neurological disorders, particularly those with altered glucose metabolism. Here, we examine potential compounding effect of metal exposure and RAGE expression on dopamine (DA) and serotonin (SER) neurons in C. elegans. In addition, we evaluate the effect of RAGE expression on DA and SER neurons in hyperglycemic conditions. Newly generated RAGE-expres

Manganese toxicity in children receiving long-term parenteral nutrition.

BACKGROUND: In patients receiving long-term parenteral nutrition (PN), cholestatic disease and nervous system disorders have been associated with high blood concentrations of manganese. In such patients, the normal homoeostatic mechanisms of the liver and gut are bypassed and the requirement for this trace element is not known; nor has it been certain whether hypermanganesaemia causes the cholestasis or vice versa. We explored the direction of effect by serial tests of liver function after withdrawal of manganese supplements from children receiving long-term PN. We also examined the relation b

Hypermanganesemia Induced Chorea and Cognitive Decline in a Tea Seller.

BACKGROUND: Manganese associated neurotoxicity and neurodegeneration is quite rare yet established neurological disorder. This neurotoxic element has predilection for depositing in basal ganglia structures, manifesting mainly as parkinsonian and dystonic movement disorders with behavioral abnormalities. CASE REPORT: We report a 40-year-old man who presented with a subacute onset bilateral, asymmetric hyperkinetic movement disorder (predominantly left sided chorea) with multi-domain cognitive impairment, dysarthria, and generalized rigidity. L-tyrosine hydroxylase activity in the rat brain after chronic oral administration of manganese chloride.

In rats chronically treated with a high oral load of MnCl2 a significant increase in the activity of L-tyrosine hydroxylase was observed in neostriatum, midbrain and hippocampus one month after the beginning of the experiment. The augmented enzymatic activity persisted in neostriatum, midbrain and hypothalamus on the third month and remained elevated only in neostriatum on the sixth month. After eight months a significant decrease in the activity of the enzyme was found in neostriatum with no changes in the remaining regions studied. These findings are interesting since human manganese intoxic

Mechanism of SO2 effect on the simultaneous removal of NOx and propane over bifunctional NiMn2O4-CeO2 catalysts.

Nitrogen oxides (NOx) and volatile organic compounds (VOCs) are the main pollutants in flue gas, and the synergistic removal of NOx and VOCs in the presence of SO2 is still a challenge. In this work, the microstructure of NiMn2O4-CeO2 catalysts and the distribution of sulfur-containing substances were studied to reveal the inactivation mechanism of sulfur poisoning. NOx conversion could reach more than 80 % at 100-250 °C, and C3H8 conversion could achieve 90 % at 210 °C in the synergistic reaction on NiMn2O4-CeO2 catalyst, which has dual active sites with propane oxidation and SCR reactions ca

B/L Basal Ganglia Lesions in a Child Leading to a Diagnosis of Glucose-6-Phosphate Dehydrogenase Deficiency.

Bilateral basal ganglia lesions are a common non-specific finding seen in many diseases. One of the differential diagnoses for it, in a child, is kernicterus occurring due to hyperbilirubinemia. Glucose-6-phosphate dehydrogenase (G6PD) deficiency is a common cause of severe hyperbilirubinemia. A 1-year old child presented to the hospital with history of generalized dystonia in the previous 3 days. MRI showed evidence of symmetrical lesions in bilateral globus pallidus, which were hyperintense on T2/FLAIR and isointense on T1. Patient's blood test revealed G6PD deficiency. Hence, a diagnosis of

A severe case of multiple metal poisoning in a child treated with a traditional medicine.

A child who had been treated orally with a traditional medicine was admitted to hospital with diarrhoea, vomiting and severe metabolic acidosis. Following treatment for renal failure and supportive therapy, the child was discharged well. Analysis of the traditional remedy, a black powder, using inductively coupled plasma-optical emission revealed very high concentrations of iron and manganese. Other metals were also present at lesser, but still high concentrations. The composition suggested that this was a sample obtained from a surface fracture in Transvaal dolomite. Metal poisoning is not un

Effects of Typical Underground Coal Mine Environmental Factors on CO Oxidation Performance of Sn-Containing Catalyst.

One of the primary causes of casualties as a result of underground coal mine disasters is the generation of high concentrations of carbon monoxide (CO). In this study, a copper (Cu)-manganese (Mn)-tin (Sn) composite oxide catalyst was prepared using the co-precipitation method, and the effects of CO concentration (1-7%), reaction temperature (25-300 °C), and water poisoning degree (0-100%) on CO catalytic oxidation performance were systematically investigated using a dynamic activity testing system. The results demonstrated that within the CO concentration range of 1-7%, the catalyst was able

Neurotoxicity of Metal Mixtures.

Metals are the oldest toxins known to humans. Metals differ from other toxic substances in that they are neither created nor destroyed by humans (Casarett and Doull's, Toxicology: the basic science of poisons, 8th edn. McGraw-Hill, London, 2013). Metals are of great importance in our daily life and their frequent use makes their omnipresence and a constant source of human exposure. Metals such as arsenic [As], lead [Pb], mercury [Hg], aluminum [Al] and cadmium [Cd] do not have any specific role in an organism and can be toxic even at low levels. The Substance Priority List of Agency for Toxic

Association of exposure to manganese and iron with relaxation rates R1 and R2*- magnetic resonance imaging results from the WELDOX II study.

OBJECTIVE: Magnetic resonance imaging is a non-invasive method that allows the indirect quantification of manganese (Mn) and iron (Fe) accumulation in the brain due to their paramagnetic features. The WELDOX II study aimed to explore the influence of airborne and systemic exposure to Mn and Fe on the brain deposition using the relaxation rates R1 and R2* as biomarkers of metal accumulation in regions of interest in 161 men, including active and former welders. MATERIAL AND METHODS: We obtained data on the relaxation rates R1 and R2* in regions that included structures within the globus pallid

Manganese and Parkinson's disease: a critical review and new findings.

BACKGROUND: Excess accumulation of manganese (Mn) in the brain results in a neurological syndrome with cognitive, psychiatric, and movement abnormalities. The highest concentrations of Mn in the brain are achieved in the basal ganglia, which may precipitate a form of parkinsonism with some clinical features that are similar and some that are different to those in Parkinson's disease (PD). Recently, scientists have debated the possibility that Mn may have an etiological role in PD or that it may accelerate the expression of PD. OBJECTIVE: The goal of this review was to examine whether chronic

The association of bone and blood manganese with motor function in Chinese workers.

Manganese (Mn) is an essential element. However, Mn overexposure is associated with motor dysfunction. This cross-sectional study assessed the association between bone Mn (BnMn) and whole blood Mn (BMn) with motor function in 59 Chinese workers. BnMn and BMn were measured using a transportable in vivo neutron activation analysis system and inductively coupled plasma mass spectrometry, respectively. Motor function (manual coordination, postural sway, postural hand tremor, and fine motor function) was assessed using the Coordination Ability Test System (CATSYS) and the Purdue Pegboard. Relations

Copper, Iron, and Manganese Toxicity in Neuropsychiatric Conditions.

Copper, manganese, and iron are vital elements required for the appropriate development and the general preservation of good health. Additionally, these essential metals play key roles in ensuring proper brain development and function. They also play vital roles in the central nervous system as significant cofactors for several enzymes, including the antioxidant enzyme superoxide dismutase (SOD) and other enzymes that take part in the creation and breakdown of neurotransmitters in the brain. An imbalance in the levels of these metals weakens the structural, regulatory, and catalytic roles of d

Differential deposition of manganese in the rat brain following subchronic exposure to manganese: a T1-weighted magnetic resonance imaging study.

BACKGROUND: Manganism is a central nervous system disorder caused by toxic exposure to manganese. Manganism has been related to occupational exposures, liver diseases, prolonged parenteral nutrition, and abuse of illicit drugs. Initially manifested by a reversible neuropsychiatric syndrome (locura manganica), the main symptoms and signs of manganism are emotional lability, compulsive behavior and visual hallucinations. Locura manganica is followed by an irreversible extrapyramidal syndrome, the onset of which occurs years after chronic exposure. OBJECTIVES: To characterize the regional distri
