Antifungal activity and killing kinetics of anidulafungin, caspofungin and amphotericin B against Candida auris.

BACKGROUND: Candida auris is an emerging MDR pathogen. It shows reduced susceptibility to azole drugs and, in some strains, high amphotericin B MICs have been described. For these reasons, echinocandins were proposed as first-line treatment for C. auris infections. However, information on how echinocandins and amphotericin B act against this species is lacking. OBJECTIVES: Our aim was to establish the killing kinetics of anidulafungin, caspofungin and amphotericin B against C. auris by time-kill methodology and to determine if these antifungals behave as fungicidal or fungistatic agents again

Synergistic bacterial inactivation by combining antibiotics with nanosecond electric pulses.

Antibiotic resistance mechanisms render current antibiotics ineffective, requiring higher concentrations of existing drugs or the development of more powerful drugs for infection treatment. This study demonstrates the synergistic inactivation of a gram-positive (Staphylococcus aureus) and a gram-negative (Escherichia coli) bacteria by combining either tobramycin or rifampicin with 300-ns electric pulses (EPs). For EPs depositing the same total energy density into the sample with no drug, higher electric fields induced greater inactivation, indicating a threshold for irreversible electroporatio

Activity of moxifloxacin and linezolid against Mycobacterium tuberculosis in combination with potentiator drugs verapamil, timcodar, colistin and SQ109.

Current treatment for tuberculosis (TB) is complicated by the emergence of multidrug resistant TB (MDR-TB). As a result, there is an urgent need for new powerful anti-TB regimens and novel strategies. In this study, we aimed to potentiate a moxifloxacin + linezolid backbone as treatment for MDR-TB with the efflux pump inhibitors verapamil and timcodar as well as with drugs that act on mycobacterial cell wall stability such as colistin and SQ109. Using a time-kill kinetics assay, the activities of moxifloxacin, linezolid, verapamil, timcodar, colistin and SQ109 as single drugs against Mycobacte

Assessing the potential of four cathelicidins for the management of mouse candidiasis and Candida albicans biofilms.

As the most common fungal pathogen of humans, severe drug resistance has emerged in the clinically isolated Candida albicans, which lead to the urgency to develop novel antifungal agents. Here, four our previously characterized cathelicidins (cathelicidin-BF, Pc-CATH1, Cc-CATH2, Cc-CATH3) were selected and their antifungal activities against C. albicans were evaluated in vitro and in vivo using amphotericin B and LL-37 as control. Results showed that all four cathelicidins could eradicate standard and clinically isolated C. albicans strains with most MIC values ranging from 1 to 16 μg/ml, in l

Bis-indolic compounds as potential new therapeutic alternatives for tularaemia.

Francisella tularensis is the etiological agent of tularaemia and a CDC class A biological threat agent. Few antibiotic classes are currently useful in treating tularaemia, including the aminoglycosides gentamicin and streptomycin, fluoroquinolones, and tetracyclines. However, treatment failures and relapses remain frequent and F. tularensis strains resistant to antibiotics have been easily selected in vitro. In this study, we evaluated the activity of new synthetic bis-indole derivatives against this pathogen. Minimum inhibitory concentrations (MICs) of four compounds (dcm01 to dcm04) were de

An optimized SYBR Green I/PI assay for rapid viability assessment and antibiotic susceptibility testing for Borrelia burgdorferi.

Lyme disease caused by Borrelia burgdorferi is the most common tick-borne disease in the US and Europe. Unlike most bacteria, measurements of growth and viability of B. burgdorferi are challenging. The current B. burgdorferi viability assays based on microscopic counting and PCR are cumbersome and tedious and cannot be used in a high throughput format. Here, we evaluated several commonly used viability assays including MTT and XTT assays, fluorescein diacetate assay, Sytox Green/Hoechst 33342 assay, the commercially available LIVE/DEAD BacLight assay, and SYBR Green I/PI assay by microscopic c

Advances in Spatial Heterogeneity and Cellular Localization Specificity of Intratumoral Bacteria.

Bacteria in tumors are increasingly recognized to be a part of the tumor microenvironment and their biological function depends on the location and relative abundance of a given microorganism or microbial community. Genetic sequencing data directly from tissue samples to detection of active metabolic signals from microorganisms, cell-microbiome interactions, ultrastructural localization of microorganisms; these are all different kinds of inferences about the functional role of the microbiome, and the evidence for "microbial presence and function" grows in a stepwise fashion. Furthermore, most

Enhancement of heat resistance of Bacillus thermoamylovorans drives enhanced PET degradation.

Microbial polyethylene terephthalate (PET) degradation has emerged as an environmentally friendly approach to reducing plastic pollution, but microbial activity is often limited by the high temperatures (60-70 °C) required for efficient degradation. In this study, six endogenous thermotolerant genes were identified in the PET-degrading strain Bacillus thermoamylovorans JQ3. Overexpression of the genes hrcA, hsp20A, and hsp20B significantly enhanced thermotolerance in Escherichia coli, which increased cell viability by >24.2% at temperatures above 40 °C. Similarly, overexpression of hsp20A in B

Mechanisms and efficacy of wet heat, dry heat, and steam-based treatments for bacterial spore inactivation.

Dry and wet heat treatments are widely used physical methods for microbial inactivation, and their effectiveness in destroying bacterial spores has been extensively demonstrated. This review systematically discusses the bactericidal effects and underlying mechanisms of dry heat, wet heat, and steam-based treatments on bacterial spores. Wet heat inactivates spores through heat-induced protein denaturation and dipicolinic acid release, resulting in multi-level structural damage, including spore coat collapse, cortical peptidoglycan hydrolysis, and core hydration. On the other hand, dry heat trea

Assessing the persistence of Sporothrix schenckii and Sporothrix brasiliensis yeasts on hard, non-porous surfaces and the efficacy of disinfectants.

Sporotrichosis is a serious fungal disease of humans and animals worldwide. The ongoing epidemic of cat-transmitted sporotrichosis caused by Sporothrix brasiliensis in South America raises major public health concerns, yet data on environmental persistence and disinfectant efficacy are scarce. Using the quantitative disk carrier method (ASTM 2197), we evaluated persistence and disinfectant susceptibility of Sporothrix schenckii and Sporothrix brasiliensis yeasts on hard, non-porous stainless-steel surfaces. Both species remained viable for at least 6 weeks, supporting the potential for fomite-

Klebsiella pneumoniae biofilm formation predicts its survival in human serum.

Klebsiella pneumoniae is a prominent pathogen causing life-threatening bloodstream infections. Although biofilm formation and resistance to human serum are well-recognized virulence traits, their interrelatedness during K. pneumoniae bloodstream infections remains unclear. Here, we hypothesize that biofilm production is related to K. pneumoniae's ability to thrive in human serum and, therefore, may predict the strains' ability for serum survival. We analyzed 57 clinical, genetically diverse classical K. pneumoniae strains and characterized their survival and biofilm-producing ability in human

Lactic acid bacteria, potentially probiotic, in whole-grain pet biscuits: survival and technological potential of millet-based formulations (Pennisetum glaucum).

The relationship between humans and pets has driven the development of functional foods. This study aimed to develop pet biscuits containing Lacticaseibacillus paracasei GV17 and Lactococcus lactis GV103 and to evaluate the effects of wheat flour, millet flour, and a combination of both on the stability of lactic acid bacteria (LAB), resistance to simulated gastrointestinal tract (GIT) conditions, technological characteristics, and viability during storage. Three formulations were prepared using wheat flour (WF), millet flour (MF), and a mixture of flours (WMF), in which the GV17 and GV103 str

Lyophilization and Field Survival of Epiphytic Yeasts from the Phyllosphere of Native Cacao in the Peruvian Amazon.

Epiphytic yeasts are promising biocontrol agents of plant diseases but preserving and transferring them to the field is challenging. Here, we studied six cost-effective lyophilization protective agents to preserve seven strains of Amazonian yeast species isolated from the phyllosphere of native cacao (Theobroma cacao) in Peru. We evaluated the viability of yeasts at 30 and 90 days post-lyophilization in vitro, and their survival after controlled inoculation on cacao fruits in the field. The best protective agents were maltodextrin, honey + skim milk, and honey. Wickerhamomyces anomalus KLG-014

Validation of prototype virus inactivation from seven virus families of pandemic potential with a novel low-cost, field-deployable RNA extraction and storage method.

The Centers for Research in Emerging Infectious Diseases (CREID) was established to enhance pandemic preparedness by studying emerging/reemerging pathogens, especially in resource-limited regions. To overcome infrastructure challenges, a low-cost, field-deployable method for extracting total nucleic acids is essential, eliminating reliance on expensive equipment, power, and cold chain systems used in traditional extraction techniques. To address this challenge, we developed an RNA extraction and storage method (RNAES) that meets these criteria. Herein, we report RNAES inactivation efficacy aga

Lactoferrin Affects the Viability of Bacteria in a Biofilm and the Formation of a New Biofilm Cycle of Mannheimia haemolytica A2.

Respiratory diseases in ruminants are responsible for enormous economic losses for the dairy and meat industry. The main causative bacterial agent of pneumonia in ovine is Mannheimia haemolytica A2. Due to the impact of this disease, the effect of the antimicrobial protein, bovine lactoferrin (bLf), against virulence factors of this bacterium has been studied. However, its effect on biofilm formation has not been reported. In this work, we evaluated the effect on different stages of the biofilm. Our results reveal a decrease in biofilm formation when bacteria were pre-incubated with bLf. Howev

Klebsiella pneumoniae survives on surfaces as a dry biofilm.

BACKGROUND: Dry surface biofilms (DSB) are widespread in healthcare settings presenting a challenge to cleaning and disinfection. Klebsiella pneumoniae has been a focus of attention due to antibiotic resistance and the emergence of hypervirulent strains. Few studies have demonstrated K pneumoniae survival on surfaces following desiccation. METHODS: DSB were formed over 12 days. Bacterial culturability and transfer were investigated following DSB incubation up to 4 weeks. Bacterial viability in DSB was investigated with live/dead staining using flow cytometry. RESULTS: K pneumoniae formed mat
