Structure of Amyloid Peptide Ribbons Characterized by Electron Microscopy, Atomic Force Microscopy, and Solid-State Nuclear Magnetic Resonance.

Polypeptides often self-assemble to form amyloid fibrils, which contain cross-β structural motifs and are typically 5-15 nm in width and micrometers in length. In many cases, short segments of longer amyloid-forming protein or peptide sequences also form cross-β assemblies but with distinctive ribbon-like morphologies that are characterized by a well-defined thickness (on the order of 5 nm) in one lateral dimension and a variable width (typically 10-100 nm) in the other. Here, we use a novel combination of data from solid-state nuclear magnetic resonance (ssNMR), dark-field transmission electr

Osteocytic Osteolysis in PTH-treated Wild-type and Rankl-/- Mice Examined by Transmission Electron Microscopy, Atomic Force Microscopy, and Isotope Microscopy.

To demonstrate the ultrastructure of osteocytic osteolysis and clarify whether osteocytic osteolysis occurs independently of osteoclastic activities, we examined osteocytes and their lacunae in the femora and tibiae of 11-week-old male wild-type and Rankl-/- mice after injection of human parathyroid hormone (PTH) [1-34] (80 µg/kg/dose). Serum calcium concentration rose temporarily 1 hr after PTH administration in wild-type and Rankl-/- mice, when renal arteries and veins were ligated. After 6 hr, enlargement of osteocytic lacunae was evident in the cortical bones of wild-type and Rankl-/- mice

Statistical models to characterize colon tumor stiffness heterogeneity through representative atomic force microscopy maps.

The study of the impact of physical forces and on cells has emerged as a fertile field of investigation. Applications in oncology are especially transformative since tumor stiffness was found associated with fundamental processes such as tumorigenicity, disease progression, and resistance to therapy. We present an integrated atomic force microscopy-statistical modeling and machine learning approach in colon cancer to link clinical and phenotypical parameters with local maps of tissue stiffness. Statistical modeling found both known associations, such as with age or tumor stage, but also novel

Co2+-mediated adsorption facilitates atomic force microscopy of DNA molecules at double-helix resolution.

Atomic force microscopy (AFM) has demonstrated the ability to resolve single DNA molecules in liquid at a spatial resolution that is sufficient to visualize the double helix structure and variations therein. Such variations can be due to inherent configurational flexibility and may be related to, e.g., DNA sequence, ionic screening, supercoiling, or protein binding. These AFM experiments require DNA to be adhered to a solid and preferably flat support. For high-resolution, in-liquid AFM studies so far, such adhesion has commonly been achieved using Ni2+ ions to electrostatically bridge between

Detection of Human Bladder Epithelial Cancerous Cells with Atomic Force Microscopy and Machine Learning.

The development of noninvasive methods for bladder cancer identification remains a critical clinical need. Recent studies have shown that atomic force microscopy (AFM), combined with pattern recognition machine learning, can detect bladder cancer by analyzing cells extracted from urine. However, these promising findings were limited by a relatively small patient cohort, resulting in modest statistical significance. In this study, we corroborated the AFM technique's capability to identify bladder cancer cells with high accuracy using a controlled model system of genetically purified human bladd

Effect of antibiotics on mechanical properties of Bordetella pertussis examined by atomic force microscopy.

In recent years, the coevolution of microorganisms with current antibiotics has increased the mechanisms of bacterial resistance, generating a major health problem worldwide. Bordetella pertussis is a bacterium that causes whooping cough and is capable of adopting different states of virulence, i.e. virulent or avirulent states. In this study, we explored the nanomechanical properties of both virulent and avirulent B. pertussis as exposed to various antibiotics. The nanomechanical studies highlighted that only virulent B. pertussis cells undergo a decrease in their cell elastic modulus and hei

Unraveling Interaction Mechanisms between Molybdenite and a Dodecane Oil Droplet Using Atomic Force Microscopy.

Molybdenite (MoS2) is a mineral that has drawn great interest because of its potential application in various fields. To facilitate the flotation of molybdenite, the mineral pulp is commonly treated with nonpolar oil additives to promote hydrophobicity and to form an oil bridge between ultrafine molybdenite particles for agglomeration. In this study, dodecane was chosen as a model oil to investigate the flotation mechanisms of molybdenite with nonpolar oil. The interaction forces between a micrometer-sized dodecane droplet and the molybdenite basal plane in various electrolyte solutions were d

Quantitative assessment of intermolecular interactions by atomic force microscopy imaging using copper oxide tips.

Atomic force microscopy is an impressive tool with which to directly resolve the bonding structure of organic compounds1-5. The methodology usually involves chemical passivation of the probe-tip termination by attaching single molecules or atoms such as CO or Xe (refs 1,6-9). However, these probe particles are only weakly connected to the metallic apex, which results in considerable dynamic deflection. This probe particle deflection leads to pronounced image distortions, systematic overestimation of bond lengths, and in some cases even spurious bond-like contrast features, thus inhibiting reli

Microfluidic platform for the elastic characterization of mouse submandibular glands by atomic force microscopy.

The ability to characterize the microscale mechanical properties of biological materials has the potential for great utility in the field of tissue engineering. The development and morphogenesis of mammalian tissues are known to be guided in part by mechanical stimuli received from the local environment, and tissues frequently develop to match the physical characteristics (i.e., elasticity) of their environment. Quantification of these material properties at the microscale may provide valuable information to guide researchers. Presented here is a microfluidic platform for the non-destructive e

A novel technique for quantifying mouse heart valve leaflet stiffness with atomic force microscopy.

BACKGROUND AND AIM OF THE STUDY: The use of genetically altered small animal models is a powerful strategy for elucidating the mechanisms of heart valve disease. However, while the ability to manipulate genes in rodent models is well established, there remains a significant obstacle in determining the functional mechanical properties of the genetically mutated leaflets. Hence, a feasibility study was conducted using micromechanical analysis via atomic force microscopy (AFM) to determine the stiffness of mouse heart valve leaflets in the context of age and disease states. METHODS: A novel AFM

Three-Dimensional Visualization of Conductive Filaments in TiO2 via Conductive Atomic Force Microscopy Tomography.

Direct three-dimensional visualization of conductive filaments (CFs) in valence change memory is essential for resolving whether their spatial evolution is governed by stochastic defect generation or by electrode geometry. Here, we apply conductive atomic force microscopy tomography to a TiO2 (10 nm)/Pt structure, extending this technique to TiO2-based CFs previously inaccessible to tomographic characterization. The tomogram reveals a reverse-cone morphology whose terminus preferentially connects to a Pt bottom electrode (BE) protrusion, evidencing dual field enhancement at both interfaces. Cu

Atomic Force Microscopy of Poliovirus Particles After Inactivation by Chemical Methods and Accelerated Electrons.

The traditional method used in the production of inactivated vaccines is chemical inactivation using beta-propiolactone or formaldehyde. An alternative method is inactivation by irradiation. Virus inactivation is often accompanied by a change in particle shape, which can negatively affect the preservation of antigens and immunogenicity. Therefore, determining the shape and structure of the viral particle after inactivation is an important step in the development of antiviral vaccines. The poliovirus strain Sabin 2 was inactivated with a dose of 30.5 ± 0.5 kGy. in a pulsed linear electron accel

Analyzing Morphological Properties of Early-Stage Toxic Amyloid β Oligomers by Atomic Force Microscopy.

Protein misfolding diseases, like Alzheimer's, Parkinson's, and Huntington's disease, are associated with misfolded protein aggregation. Alzheimer's disease is related to a progressive neuronal death induced by small amyloid β oligomers. Here, we describe the procedure to prepare and identify different types of small toxic amyloid β oligomers by atomic force microscopy (AFM).

High-resolution atomic force microscopy as a tool for topographical mapping of surface budding.

Extracellular vesicles (EVs) are membranous nanoparticles secreted by almost all cell types. Reflecting the physiopathological state of the parental cell, EVs circulate in all body fluids, reaching distant cell targets and delivering different bioactive cargoes. As biological carriers, EVs influence their microenvironment altering cellular responses, being considered promising biomarkers for both physiological and pathological conditions. EVs are heterogeneous in terms of size and composition, depending on cell type and exposure to stimuli, and different methods have been developed to characte
