Neospora caninum, is an obligate intracellular parasite, can infect domestic and wild dogs, equines and ruminants. It was described in 1988, and its economic importance is attributed to costs of abortion, the value of the fetuses, artificial insemination and coverage, the decrease in milk production, the increase in the disposal and replacement of animals. In four rural settlements of the municipality of Corumba, MS (Mato Grande, Paiolzinho, Taquaral and Tamarineiro) was used a combination of qualitative methodologies in 37 semi-structured interviews and group dynamics for analysis with partic

Lesions suggestive of protozoal infection such as nonsuppurative myositis, nonsuppurative myocarditis, nonsuppurative pneumonia associated with focal cellular necrosis and multifocal necrotizing nonsuppurative encephalitis were observed in 89 (34%) out of 258 aborted bovine fetuses. Immunohistochemistry (IHC) revealed tachyzoites and cysts of Neospora caninum in 55 (61.8%) of these 89 fetuses. Several fetal variables were tested by multivariate logistic regression, in which increasing odds for positivity in IHC were associated with the presence of lesion in the lung (OR=12.11) and less autolyt

An immunosuppressed mouse model was used to determine the effects of amprolium and sulfadiazine on experimental Neospora caninum infections. Both drugs were given in the drinking water. Neither drug was effective in treating infections when given 7 days after inoculation of tachyzoites, when clinical signs of disease had developed. Amprolium did not prevent deaths or development of clinical signs when given in the drinking water at 1 mg/ml or 5 mg/ml 3 days after inoculation of tachyzoites. Sulfadiazine in drinking water was not effective when given at 0.5 mg/ml but was effective in preventing

We used immunohistochemistry and PCR to identify Neospora caninum in 6 infected marine mammal species, including 2 pups, that stranded in the northeastern Pacific Ocean. Our findings suggest the expansion of this parasite's host range to marine mammals, underscoring the effect of terrestrial pathogens that flow from land to sea.

Neospora caninum is one of the main agents that causes abortions in cattle worldwide. However, little is known about the pathogenesis of neosporosis in small ruminants, especially goats. Gerbils (Meriones unguiculatus) have been used as a model for neosporosis, and this species is highly susceptible to infection by bovine N. caninum strains. The present study aimed to evaluate the susceptibility of gerbils to a N. caninum isolate from goats. The placentas were obtained from naturally infected goats, that presented with mild to severe lymphoplasmacytic and histiocytic infiltrate, foci of necros

BACKGROUND: Neospora caninum (N. caninum), an obligate intracellular apicomplexan parasite, infects nucleated cells across diverse mammalian hosts, resulting in severe clinical outcomes, including reproductive failure (abortion, stillbirth) and neurological impairment. Ferroptosis, an iron-dependent, non-apoptotic cell death pathway characterized by excessive lipid peroxidation, is closely associated with disrupted iron metabolism and oxidative stress. Deferiprone (DFP), a clinically approved iron chelator, is widely used in the treatment of iron overload disorders such as β-thalassemia, Alzhe

Neospora caninum is a heteroxenous parasitic Apicomplexan protozoan, able to infect a variety of domestic and wild animals, mainly associated with reproductive disorders and abortions in susceptible species. Considering the importance of Romania in the European buffalo industry and the severe economic losses caused by N. caninum infection in livestock, the aim of the present study was to assess the occurrence of neosporosis in buffaloes raised in household and farming systems in northwestern Romania. Overall, 197 serum samples were tested for the presence of N. caninum-specific antibodies by E

Neospora caninum is an apicomplexan protozoan responsible for abortion in ruminants. The present study aimed to diagnose an abortion from an Anglo Nubian goat from a dairy herd located in Buenos Aires province, Argentina. The goat delivered a fetus of approximately 3 months gestation that was studied by indirect fluorescent antibody test (IFAT), Immunoblot (IB), histopathology (HP), immunohistochemistry (IHC), and molecular assays (PCR, sequencing and microsatellite genotyping). Interferon gamma knock-out mice were inoculated with a pool of tissues for isolation attempts. The mother had IFAT t

Neospora caninum is an intracellular protozoan parasite that has drawn increasing interest due to its association with worldwide repetitive bovine abortions, which cause billionaire losses to the meat and dairy industries annually. Innate immunity plays an important role in infection control, and N. caninum activates the production of inflammatory mediators through toll-like receptors, NOD-like receptors, and mitogen-activated protein kinase signaling pathways. Advances in the knowledge of initial host-parasite interactions are desirable for the design of control measures against the infection

Due to the high prevalence and economic impact of neosporosis, the development of safe and effective vaccines and therapies against this parasite has been a priority in the field and is crucial to limit horizontal and vertical transmission in natural hosts. Limited data is available regarding factors that regulate the immune response against this parasite and such knowledge is essential in order to understand Neospora caninum induced pathogenesis. Mitogen-activated protein kinases (MAPKs) govern diverse cellular processes, including growth, differentiation, apoptosis, and immune-mediated respo

We compared the transplacental-transmission ability of Toxoplasma gondii and Neospora caninum in cattle. One uninfected pregnant heifer served as control, while three were inoculated with N. caninum K9WA strain and four with T. gondii RH strain at their midgestational period. Both infected groups showed clinical signs and antibodies either to N. caninum or T. gondii, while the control animal was normal. Two (50%) Toxoplasma dams aborted on days 6 and 11 postinoculation. T. gondii tachyzoites were found in various organs of those dams that had abortions but not in their fetuses. Two Neospora da

Neospora caninum is a major cause of reproductive losses in ruminants, but information on antibody dynamics in naturally infected goats remains limited. This study provides the first longitudinal assessment of anti-N. caninum antibody dynamics and seroconversion during gestation, parturition, and lactation in a semi-intensive goat herd in Argentina. A total of 262 serum samples were collected from 45 goats at six physiological stages, from early gestation to 100 days of lactation, and analyzed using the indirect fluorescent antibody test (IFAT). Overall seropositivity was high (88.9%), with an

Neospora caninum is a leading cause of abortion in cattle worldwide, yet the role of host genetic background in early immune responses remains incompletely understood. This study compared early transcriptional responses to N. caninum infection in two cattle breeds with contrasting selection histories using a controlled in vivo model. Fourteen-month-old, non-pregnant Argentine Creole and Holstein heifers (n = 8 per breed) were intravenously inoculated with live N. caninum tachyzoites or mock-treated. Peripheral blood mononuclear cells were collected at 7 days post-infection and analyzed by bulk

Infection with Neospora caninum leads to reproductive failure in ruminants, such as cattle and goats; however, no effective vaccines or treatments are currently available to control this infection. Carefully regulating the glycolysis of host cells is essential for the intracellular survival of pathogens. Nonetheless, the impact of N. caninum infection on host cell glycolysis and the effects and mechanisms of host cell glycolysis on the intracellular survival of this parasite remains unclear. In this study, the analysis of metabolomics and transcriptomics revealed that N. caninum infection incr

Neospora caninum is an intracellular protozoan that is the causative agent of neosporosis, a disease relevant to the veterinary field for causing neuromuscular disease in dogs and repeated abortions in ruminants. In the present moment, there are no therapies that are able to induce sterile cure against the infection and prevention strategies rely only in environmental management. In this context, we aimed to determine the protective role of the Interferon alpha and beta receptor (IFNAR) during an experimental infection with N. caninum. We performed in vivo experiments with genetically deficien

Toxoplasma gondii and Neospora caninum infections in dogs are predominantly manifest asymptomatic. However, these infections can also present highly varied and potentially severe clinical signs. This is due to the parasites' ability to replicate in a number of cell types within the host organism, with N. caninum exhibiting a particular tropism for the central and peripheral nervous systems, and T. gondii targeting the central nervous system and musculature. In clinical practice, toxoplasmosis and neosporosis are often considered to be closely related diseases, despite their distinct epidemiolo

This study aimed to evaluate the N. caninum associated abortions in cattle in the state of Santa Catarina, in the southern Brazil. Aborted bovine fetuses were necropsied, submitting organ samples for histopathological evaluation. Brain fragments were analyzed by polymerase chain reaction (PCR). The diagnosis of abortion due to N. caninum was established through histopathology and molecular analysis in 53.84% (28/52) of the cases, with PCR detection in 71.42% (20/28). The histopathological evaluation showed lesions in 75% of the cases, characterized by mononuclear necrotizing encephalitis, mono

Neospora caninum is an important pathogen commonly causing spontaneous abortion in livestock. The parasite is known to remain in cysts in an inactive state; or it can undergo expansive development within an intermediate host. However, the mechanisms that trigger the proliferation of N. caninum have not been thoroughly elucidated. For various organisms, it has been demonstrated that microRNAs (miRNAs) can act as important endogenous regulatory factors in gene regulation during cell differentiation and development. However, miRNAs and their function have not been studied in N. caninum. In this s

There are not any records on the detection of Toxoplasma gondii and Neospora caninum in tissues of wild birds in the African continent. The aim of the study was to investigate the occurrence of DNA from these protozoan parasites in brain tissue samples collected in years 2014-2015 from 110 wild and domestic birds of 15 orders. Birds came mainly from the province of Limpopo (n=103); the other seven birds came from other five provinces of South Africa. Parasite DNAs were detected by PCR in animal brains. While all samples were negative for N. caninum, T. gondii DNA was detected in three (2.7%) b
