Microfluidic-assisted self-assembly of information-bearing oligomers.

Increasing the complexity of self-assembled supramolecular nanostructures necessitates considered design and synthesis of the precursor molecular components as well as precise control over the reaction microenvironment. Conventional batch techniques are susceptible to kinetic trapping of intermediate species, resulting in reduced target yields for progressively complex assemblies. In contrast, the microenvironmental control offered by microfluidics enables local thermodynamic minima to be circumvented, enabling self-assembly processes to proceed to completion. Here, the dynamic covalent assemb

Lipid composition greatly affects the in vitro surface activity of lung surfactant protein mimics.

A crucial aspect of developing a functional, biomimetic lung surfactant (LS) replacement is the selection of the synthetic lipid mixture and surfactant proteins (SPs) or suitable mimics thereof. Studies elucidating the roles of different lipids and surfactant proteins in natural LS have provided critical information necessary for the development of synthetic LS replacements that offer performance comparable to the natural material. In this study, the in vitro surface-active behaviors of peptide- and peptoid-based mimics of the lung surfactant proteins, SP-B and SP-C, were investigated using th

Solid-Phase Insertion of N-mercaptoalkylglycine Residues into Peptides.

N-mercaptoalkylglycine residues were inserted into peptides by reacting N-free amino groups of peptides, which were initially synthesized on 2-chlorotrityl resin (Cltr) using the Fmoc/tBu method, with bromoacetic acid and subsequent nucleophilic replacement of the bromide by reacting with S-4-methoxytrityl- (Mmt)/S-trityl- (Trt) protected aminothiols. The synthesized thiols containing peptide-peptoid hybrids were cleaved from the resin, either protected by treatment with dichloromethane (DCM)/trifluoroethanol (TFE)/acetic acid (AcOH) (7:2:1), or deprotected (fully or partially) by treatment wi

Microarray-based technologies for the discovery of selective, RNA-binding molecules.

The identification of small molecules that bind specifically to RNA is a challenge. However, the recent explosion in knowledge about the role RNA plays in a number of physiological processes apart from coding for protein sequences makes it a highly interesting target for chemical probes and therapeutics. One technology that has played an important role in the discovery of RNA-binding molecules is microarrays. Microarrays have been broadly employed to screen, profile, and quantify RNA interactions, and will likely play an important role in the discovery of new classes of ligands going forward.

Development of New Peptidomimetic NADPH Oxidase Inhibitors with Antithrombotic Properties.

The ability of synthetic peptides inhibitors of NOX1 to effectively block the production of ROS by the enzyme was studied with different methodologies. Specifically, taking advantage of our understanding of the active epitope of the regulatory NOX1 subunit NOXA1 as a potent inhibitor of NOX1-derived O2⋅- formation, a panel of peptidomimetic derivatives of this peptide were designed and synthesized with the aim of improving their activity and increasing their stability in plasma. The results highlighted that improved efficacy and potency was found for both the peptide-peptoid hybrid GS2, wherea

Combining Elements from Two Antagonists of Formyl Peptide Receptor 2 Generates More Potent Peptidomimetic Antagonists.

Structural optimization of a peptidomimetic antagonist of formyl peptide receptor 2 (FPR2) was explored by an approach involving combination of elements from the two most potent FPR2 antagonists described: a Rhodamine B-conjugated 10-residue gelsonin-derived peptide (i.e., PBP10, RhB-QRLFQVKGRR-OH) and the palmitoylated α-peptide/β-peptoid hybrid Pam-(Lys-βNspe)6-NH2. This generated an array of hybrid compounds from which a new subclass of receptor-selective antagonists was identified. The most potent representatives displayed activity in the low nanomolar range. Synthesis of Heterotelechelic Poly(N-ethylglycine) Polymers: from Polypeptoid Lipid Conjugates to Protein-Polymer Conjugates.

The need for alternative hydrophilic polymers to polyethylene glycol (PEG) has intensified due to increasing concerns about immunogenicity and hypersensitivity reactions. In this work, we report the synthesis of well-defined heterotelechelic poly-N-ethylglycine (pNEtGly) via acid-catalyzed ring-opening polymerization of N-substituted N-carboxyanhydrides with a degree of polymerization from 25 to 400. The Leuchs synthesis was modified and optimized for the preparation of high-purity N-ethylglycine NCA monomers, enabling controlled polymerization with the use of organic acid catalysts. The resul

Complexation of Polypeptides and Polypeptoids with Nucleic Acids: Does Chirality Matter for Salt Stability and Morphology?

Liquid-liquid phase separation (LLPS), especially via coacervation, offers a novel drug delivery strategy by encapsulating therapeutic agents within phase-separated droplets, thereby improving stability, solubility, and controlled release. Polypeptides and polypeptoids are ideal biomaterials for these systems due to their versatility and tunable properties. Polypeptoids are particularly advantageous, offering enhanced enzymatic resistance and greater control over molecular interactions, making them suitable for complexation studies. This research explores the binary complexation of L-, D,L-, a

"Molecular Masks" for ACE2 to Effectively and Safely Block SARS-CoV-2 Virus Entry.

Coronavirus Disease 2019 (COVID-19) remains a global health crisis, despite the development and success of vaccines in certain countries. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus that causes COVID-19, uses its spike protein to bind to the human cell surface receptor angiotensin-converting enzyme 2 (ACE2), which allows the virus to enter the human body. Using our unique cell screening technology, we identified two ACE2-binding peptoid compounds and developed dimeric derivatives (ACE2P1D1 and ACE2P2D1) that effectively blocked spike protein-ACE2 interaction, result

Exploring Alternative Zinc-Binding Groups in Histone Deacetylase (HDAC) Inhibitors Uncovers DS-103 as a Potent Ethylhydrazide-Based HDAC Inhibitor with Chemosensitizing Properties.

In this work, we synthesized a series of peptoid-based histone deacetylase (HDAC) inhibitors with variations in the linker region and zinc-binding groups. All compounds were investigated for their HDAC inhibition, antiplasmodial activity, and cytotoxicity against native and cisplatin-resistant carcinoma cell lines. The ethylhydrazide 20 (DS-103) proved to be the most effective compound in these primary screenings. DS-103 showed nanomolar inhibition of class I HDACs and of HDAC6 (class IIb). To further investigate the binding mode of DS-103, a crystal structure of DS-103 in complex with HDAC6 w

Optimization of the Linker Domain in a Dimeric Compound that Degrades an r(CUG) Repeat Expansion in Cells.

RNA repeat expansions are responsible for more than 30 incurable diseases. Among them is myotonic dystrophy type 1 (DM1), the most common form of adult on-set muscular dystrophy. DM1 is caused by an r(CUG) repeat expansion [r(CUG)exp] located in the 3' untranslated region (UTR) of the dystrophia myotonica protein kinase gene. This repeat expansion is highly structured, forming a periodic array of 5'CUG/3'GUC internal loop motifs. We therefore designed dimeric compounds that simultaneously bind two of these motifs by connecting two RNA-binding modules with peptoid linkers of different geometrie

Antibacterial peptidomimetics via fragment display on small-molecule scaffolds.

In response to the growing threat of antimicrobial resistance, scaffold-based peptidomimetics were explored as potential antibacterial agents. This innovative design approach involved fragment display on small-molecule scaffolds of ultrashort peptides and peptidomimetics with alternating cationic/hydrophobic motifs. Compounds, based on ultrashort peptides or peptide/β-peptoid hybrids attached to small-molecule scaffolds comprising bis-, tris-, and tetrakis(bromomethyl)benzene derivatives as well as triamines, were tested for antibacterial activity against a panel of Gram-negative and Gram-posi

Hydrazide-Based Class I Selective HDAC Inhibitors Completely Reverse Chemoresistance Synergistically in Platinum-Resistant Solid Cancer Cells.

In this work, we have synthesized a set of peptoid-based histone deacetylase inhibitors (HDACi) with a substituted hydrazide moiety as zinc-binding group. Subsequently, all compounds were evaluated in biochemical HDAC inhibition assays and for their antiproliferative activity against native and cisplatin-resistant cancer cell lines. The hydrazide derivatives with a propyl or butyl substituent (compounds 5 and 6) emerged as the most potent class I HDAC selective inhibitors (HDAC1-3). Further, compounds 5 and 6 outperformed entinostat in cytotoxicity assays and were able to reverse chemoresistan

Potential Applications for Growth Hormone Secretagogues Treatment of Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) arises from neuronal death due to complex interactions of genetic, molecular, and environmental factors. Currently, only two drugs, riluzole and edaravone, have been approved to slow the progression of this disease. However, ghrelin and other ligands of the GHS-R1a receptor have demonstrated interesting neuroprotective activities that could be exploited in this pathology. Ghrelin, a 28-amino acid hormone, primarily synthesized and secreted by oxyntic cells in the stomach wall, binds to the pituitary GHS-R1a and stimulates GH secretion; in addition, ghrelin i

Rapid Two-Step Multicomponent Synthesis and Structure-degradation Relationships of Selective HDAC6 PROTAC Degraders.

This work reports on the development of selective histone deacetylase 6 (HDAC6) degraders based on a peptoid scaffold. Structure-based design identified the isocyanide-derived cap region as suitable exit vector for linker attachment, enabling rapid generation of CRBN-recruiting PROTACs via the Ugi four-component reaction. A focused library of 12 degraders revealed a strong dependence of activity on linker composition, with octyl-linked compounds (9e-h) showing the strongest HDAC6 degradation with half-maximal degradation values of 17-36 nM in kinetic HDAC6 degradation assays. The lead compound

Kinetic Frustration of Collagen Folding Drives Condensate-Mediated Fibrous Assembly.

Collagen assembly is generally assumed to proceed hierarchically with rapid triple-helix folding preceding higher-order organization. Here, we show that this paradigm can break down when triple helix formation becomes kinetically frustrated. Using collagen-mimetic peptides containing cis-trans-isomerizing cationic and anionic peptoid residues, we selectively delay folding kinetics and demonstrate that liquid-liquid phase separation (LLPS) can occur prior to fibrous assembly. Delayed folding enables weak multivalent charge-pair interactions to become kinetically relevant, generating a phase-sep
