Potential of entomopathogenic fungi for Glossina austeni control: insights into microbiome alterations and implications on sustainable management of the pest.

Glossina austeni Newstead (Diptera: Glossinidae) is a competent vector of the trypanosomes causing human African trypanosomiasis and the African animal trypanosomosis. Management of this pest has primarily involved trapping methods, Sterile Insect Technique, and research into vector competence-symbiotic interactions. Nevertheless, the use of entomopathogenic fungi (EPF) in integrated pest management programs for G. austeni control remains limited. Moreover, different tsetse fly species exhibit varying susceptibility to different EPF strains, indicating that no single strain is universally effe

Oral delivery of double-stranded RNA in larvae of the yellow fever mosquito, Aedes aegypti: implications for pest mosquito control.

RNA interference has already proven itself to be a highly versatile molecular biology tool for understanding gene function in a limited number of insect species, but its widespread use in other species will be dependent on the development of easier methods of double-stranded RNA (dsRNA) delivery. This study demonstrates that RNA interference can be induced in the mosquito Aedes aegypti L. (Diptera: Culicidae) simply by soaking larvae in a solution of dsRNA for two hours. The mRNA transcripts for β-tubulin, chitin synthase-1 and -2, and heat shock protein 83 were reduced between 30 and 50% thre

Efficacy of biological control agents against Popillia japonica: A strategy to reduce the risk of pest spread via potted plant substrates.

The international trade of potted plants represents a pathway for the unintentional spread of harmful organisms, leading to new infestations in pest-free areas. The Japanese beetle Popillia japonica, which is considered one of the most threatening insect pests in Europe, can evade detection at the larval stage by hiding within the substrates of potted plants. To reduce the risk of P. japonica dispersal via potted plants, we tested the efficacy of entomopathogenic nematodes (EPNs) and fungi (EPF) as biological control agents (BCAs). Field trials with potted cherry trees were conducted in an Ita

Factors influencing willingness to pay for biological pest control products: the significance of farmers' perceived health risks and knowledge in Bangladesh.

BACKGROUND: The increasing use of chemical pesticides and insecticides poses serious risks to ecosystems, soil quality, and human health. In Bangladesh, many farmers have limited awareness of the safe use, disposal, and health implications of these chemicals. This study aimed to assess farmers' knowledge of biological pest control products, examine their willingness to pay (WTP) for such products, and identify the factors influencing their adoption. Data were collected from 220 farmers through a multistage random sampling technique from two upazilas in the Mymensingh district of Bangladesh. Th

Integrated Pest Management of Fruit Mites in Apple Orchards: Biological and Chemical Control, Resistance Management, and Regional Perspectives from Southeastern Kazakhstan.

Fruit mites, particularly Panonychus ulmi, Tetranychus urticae, and eriophyid mites, are among the most economically important arthropod pests in apple orchards, causing significant reductions in photosynthetic efficiency, fruit quality, and yield. Their management has traditionally relied on chemical acaricides; however, the widespread development of resistance, adverse effects on beneficial arthropods, and increasing environmental concerns have highlighted the limitations of pesticide-dependent control strategies. This review critically evaluates current advances in the management of major f

RNAi assay in the 28-spotted ladybeetle Henosepilachna vigintioctopunctata suggests nuclear receptor HR3 as a potential molecular target for pest control.

BACKGROUND: Nuclear hormone receptors play crucial roles in embryonic development, neural regulation, metabolism, and organogenesis in insects. This study aimed to investigate the expression and function of the nuclear hormone receptor 3 (HR3) in the 28-spotted ladybeetle, Henosepilachna vigintioctopunctata. RESULTS: Our findings revealed that HvHR3 was expressed at all developmental stages, with the highest expression levels in the 1st instar larvae, 3rd instar larvae, and pupa. HvHR3 silencing in both the 1st and 3rd instar larvae resulted in high mortality rates. Reduced HvHR3 expression i

The genome of the stable fly, Stomoxys calcitrans, reveals potential mechanisms underlying reproduction, host interactions, and novel targets for pest control.

BACKGROUND: The stable fly, Stomoxys calcitrans, is a major blood-feeding pest of livestock that has near worldwide distribution, causing an annual cost of over $2 billion for control and product loss in the USA alone. Control of these flies has been limited to increased sanitary management practices and insecticide application for suppressing larval stages. Few genetic and molecular resources are available to help in developing novel methods for controlling stable flies. RESULTS: This study examines stable fly biology by utilizing a combination of high-quality genome sequencing and RNA-Seq a

Optimizing pesticide delivery in cotton: impact of unmanned aerial spraying system configurations on canopy deposition and implications for pest control efficacy.

BACKGROUND: Effective management of insect pests and diseases in cotton, such as aphids, whiteflies and bollworms, is critically dependent on achieving adequate pesticide coverage throughout the plant canopy. Conventional ground spraying methods are often inefficient and can damage crops, whereas the adoption of unmanned aerial spraying systems (UASS) presents a promising alternative. However, the efficacy of UASS applications is highly dependent on system configuration, and a lack of data on spray deposition within dense cotton canopies hinders the development of effective application protoco

Nanomaterial-encapsulated dsRNA of ecdysone-induced early gene E75, a potential RNAi-based SIT strategy for pest control against Bactrocera dorsalis.

Bactrocera dorsalis is a notorious pest widely distributed across most Asian countries. With the rapid development of pesticide resistance, new pest control methods are urgently needed. RNAi-based sterile insect technique (SIT) is a species-specific pest management strategy for B. dorsalis control. It is of great significance to identify more target genes from B. dorsalis, and improve the RNAi efficiency. In this study, microinjection-based RNAi results showed that six 20E response genes were necessary for male fertility of B. dorsalis, of which E75 was identified as the key target according t

A new approach to insect-pest control--combination of neurotoxins interacting with voltage sensitive sodium channels to increase selectivity and specificity.

Voltage-sensitive sodium channels are responsible for the generation of electrical signals in most excitable tissues and serve as specific targets for many neurotoxins. At least seven distinct classes of neurotoxins have been designated on the basis of physiological activity and competitive binding studies. Although the characterization of the neurotoxin receptor sites was predominantly performed using vertebrate excitable preparations, insect neuronal membranes were shown to possess similar receptor sites. We have demonstrated that the two mutually competing anti-insect excitatory and depress

Enhancing Biological Control of Maize Seedling-Stage Pests Through Combined Application of Cnidium monnieri and Endophytes.

Agricultural intensification has weakened natural pest regulation by reducing habitat resources for natural enemies and limiting plant-mediated resistance. Here, we conducted field experiments during 2024-2025 to evaluate the combined effects of Cnidium monnieri field-margin planting and maize seeds coated with the endophytic fungal strains YC (Beauveria bassiana) and PL (Purpureocillium lilacinum) on predator conservation, pest suppression, and biological control in maize agroecosystems. Compared with natural grass margins, C. monnieri significantly increased predator abundance, species richn

Developing CRISPR-based sex-ratio distorters for the genetic control of fruit fly pests: A how to manual.

Agricultural pest control using genetic-based methods provides a species-specific and environmentally harmless way for population suppression of fruit flies. One way to improve the efficiency of such methods is through self-limiting, female-eliminating approaches that can alter an insect populations' sex ratio toward males. In this microreview, we summarize recent advances in synthetic sex ratio distorters based on X-chromosome shredding that can induce male-biased progeny. We outline the basic principles to guide the efficient design of an X-shredding system in an XY heterogametic fruit fly s

Pou2f3 Modulates Mosquito Host-Seeking Behavior by Regulating Skin Microbiota and Their Production of Volatiles: Implications for Eco-Friendly Pest Control Strategies.

Skin microbiota-derived volatile metabolites are critical for mosquito host-seeking and disease transmission. Similar volatile organic compounds (VOCs)-mediated mechanisms may apply to agricultural pests (e.g., aphids, whiteflies). We demonstrate that Pou2f3, a skin-enriched transcription factor, regulates β-defensin1 expression, shaping microbiota composition and volatile production. Pou2f3-deficient mice exhibited increased Staphylococcus abundance and elevated 1-methoxy-2-propanol levels, enhancing attraction in Culex and Aedes mosquitoes. This VOC, alongside other microbiota-derived compou

Knockdown of Clavesin family genes NlClvs1l, NlClvs1t, and NlClvs2l in Nilaparvata lugens reveals their potential as novel targets for pest control strategies.

The brown planthopper (BPH) Nilaparvata lugens is one of the most destructive pests of rice, and its management has primarily relied on chemical insecticides. Currently, the chemical management of BPH is facing challenges due to the development of pesticide resistance. RNA interference (RNAi) provides attractive alternative to chemical insecticides, provided that suitable target genes are identified. Clavesin (clathrin vesicle-associated Sec14 protein) proteins are involved in multiple biological processes, including endocytosis, oxidative stress response, and the regulation of lysosome morpho

Assessing Birds of Prey as Biological Pest Control: A Comparative Study with Hunting Perches and Rodenticides on Rodent Activity and Crop Health.

Rodent damage significantly affects agriculture around the world. Rodenticides can sometimes control pests, but they are costly, may cause secondary poisoning to nontarget wildlife, and can become less efficient over time due to bait shyness and resistance. Using wildlife as biological pest control agents, particularly barn owls (Tyto spp.), has been suggested as an alternative. Barn owl nest boxes and hunting perches have been added to increase predator pressure, yet few studies have examined their effectiveness. We conducted a field study in forty-five 10 × 10 m2 plots to compare three treat

Integrating phototaxis research with pest control: laboratory-based wavelength preference screening and field efficacy evaluation of sticky traps for Monolepta hieroglyphica management.

BACKGROUND: Monolepta hieroglyphica (Motschulsky) is a significant agricultural pest, primarily infesting crops such as maize, soybean and cotton. Recently, with the commercialization of Bacillus thuringiensis (Bt) transgenic maize, this nontarget insect has emerged as a potential primary pest. Current management relies heavily on chemical control, but prolonged and improper insecticide use can promote resistance development and negatively affect natural enemies. Consequently, based on the visual system of insects, developing comprehensive physical trapping strategies is of the utmost importan

Two heat shock cognate 70 genes involved in spermatogenesis regulate the male fertility of Zeugodacus cucurbitae, as potential targets for pest control.

The melon fly Zeugodacus cucurbitae Coquillett (Diptera: Tephritidae) is an agricultural quarantine pest threatening fruit and vegetable production. Heat shock cognate 70 (Hsc70), which is a homolog of the heat shock protein 70 (Hsp70), was first discovered in mice testes and plays an important role in spermatogenesis. In this study, we identified and cloned five Hsc70 genes from melon fly, namely ZcHsc70_1/2/3/4/5. Phylogenetic analysis showed that these proteins are closely related to Hsc70s from other Diptera insects. Spatiotemporal expression analysis showed that ZcHsc70_1 and ZcHsc70_2 ar
