[Pharmacoepidemiological analysis of outpatient-based practice of antibacterial therapy of ENT organ infections].

The objective of the present study was to estimate the effectiveness of the antibacterial therapeutic regimen based at an outpatient facility. The data for statistical analysis were collected by examining 1542 patients presenting with infections of the upper pulmonary tract and ENT organs in 10 cities of the Russian Federation. It was shown that macrolides are most commonly used to treat the above infections whereas beta-lactams are prescribed significantly less frequently. The main pharmacodynamic and pharmacokinetic features of pharmaceuticals from the group of protected penicillins are desc

Use and overuse of sumatriptan. Pharmacoepidemiological studies based on prescription register and interview data.

The serotonin agonist sumatriptan was marketed in Denmark in 1992 for the treatment of acute attacks of migraine and cluster headache. The clinical development program of the drug was impressive. However, knowledge of the long-term use of sumatriptan in unselected patients was lacking. Misuse of sumatriptan was reported in single patients shortly after the introduction of the drug. The aim of the present thesis was, therefore, to provide an epidemiological description of sumatriptan use with particular emphasis on overuse. The author conducted three studies, two of which were exclusively based

Pharmacoepidemiology of Oxycodone in the USA: An Observational Study of ARCOS, Medicaid, and Medicare Drug Databases.

The US continues to battle an opioid crisis, substantially influenced by prescription opioids, with nearly 80 000 opioid-related deaths in 2023. This study aims to characterize oxycodone's distribution in the United States (US) from 2000 to 2023 using three data sources. Morphine Milligram Equivalents (MME) of oxycodone were calculated from the Drug Enforcement Administration annual summary reports from the Automation of Reports and Consolidated Orders System (ARCOS); the number of units and prescriptions of oxycodone per enrollee were calculated using the Medicaid State Drug Utilization Data

Comparative ranking of marginal confounding impact of natural language processing-derived versus structured features in pharmacoepidemiology.

OBJECTIVE: To explore the ability of natural language processing (NLP) methods to identify confounder information beyond what can be identified using claims codes alone for pharmacoepidemiology. METHODS: We developed a retrospective cohort for high vs low dose proton pump inhibitors from linked Medicare claims (2008-2017) and electronic health record data for patients with a history of peptic ulcer disease or osteoarthritis. Clinical notes authored one year before first dispensing date were processed by off-the-shelf tools: bag-of-n-grams, latent Dirichlet allocation, a linguistics-focused to

Glucagon-Like Peptide-1 Receptor Agonists in Psychiatry: A Pharmacoepidemiological Scoping Review.

Pharmacoepidemiological studies have expanded the capacity to detect therapeutic benefits and adverse drug effects of medications. In the case of glucagon-like peptide-1 receptor agonists (GLP-1RAs), their fast uptake for treating diabetes and obesity has generated substantial real-world experience, facilitating exploration of benefits for additional indications. In the context of psychiatric disorders, studies based on pharmacovigilance data and large electronic health record (EHR) databases have shown that initial concerns regarding an association between suicidality and GLP-1RA use were not

Pharmacoepidemiological Analysis of Oral Contraceptive Use in Adolescents in a German Longitudinal Cohort Study.

To assess the use of oral contraceptives (OC) in adolescents, using data from a longitudinal, population-based pediatric cohort study (LIFE Child). We also investigated associations between OC use and socioeconomic status (SES), and associations between OC use and potential adverse drug reactions such as effects on blood pressure. We included 609 female participants of the LIFE Child cohort, aged ≥13 to <21 years, who visited the study center between 2012 and 2019. Data collection compromised drug use in the past 14 days, SES, and anthropometric data such as blood pressure. An analysis of cova

Use of Pregabalin - A Nationwide Pharmacoepidemiological Drug Utilization Study with Focus on Abuse Potential.

INTRODUCTION: Pregabalin is currently approved for the treatment of epilepsy, generalized anxiety disorder and neuropathic pain with a licensed dosage range of 150 mg to 600 mg/day. Growing concern about the abuse potential of pregabalin is partly based on reports of pregabalin being used in dosages that exceed the approved therapeutic range. METHODS: To identify predictors of pregabalin use above recommended dosage, we conducted a pharmacoepidemological drug utilization study using the Danish nationwide registers. We deployed 4 measures of abuse: high use (≥600 mg/day) or very high use (≥1 2

Drug prescribing patterns in Parkinson's disease: a pharmacoepidemiological survey in a cohort of ambulatory patients.

PURPOSE: Drug treatment of idiopathic Parkinson's disease (IPD) is a difficult task, and comorbidity and comedication add to its complexity. Since in IPD there is little information about drug use, this study investigated drug prescribing and indications in IPD patients. METHODS: From June 1997 to April 1998, a cross-sectional survey of IPD outpatients was performed and demographic and clinical data and information about drug treatments was collected and analysed. RESULTS: In the 130 IPD patients included in the study, anti-Parkinson drug (APD) prescriptions increased with disease duration a

Overall safety evaluation of the risk signals of sacubitril/valsartan: A postmarketing pharmacoepidemiology study from 2015 to 2024.

BACKGROUND: Sacubitril/valsartan is a widely used cardiovascular agent characterized by its dual inhibition of the renin-angiotensin-aldosterone system and neprilysin. However, existing evidence on the safety of sacubitril/valsartan is primarily limited to clinical studies; this results in an inability to provide a timely update on associated adverse events. AIM: To mine and systematically describe adverse events related to sacubitril/valsartan from September 2015 to June 2024 using the Food and Drug Administration Adverse Event Reporting System (FAERS) database. METHODS: The reporting odds

Antimicrobial use quantification in adult dairy cows - Part 2 - Developing a foundation for pharmacoepidemiology by comparing measurement methods.

As stewardship programmes seek to influence therapeutic choices and reduce resistance selection pressure, it is essential to understand the multiple factors which may influence any given antimicrobial use measure. This article compares 9 measures of antimicrobial use pertaining to adult cows in US dairy systems based on treatment records standardized to a single standardized regimen (SReg) format. There are 3 fundamental parameters underlying the nine measures: amount of active substance (AMOUNT), number of SRegs (ANIMALS) and treatment time (TIME). Additionally, three use categories were cons

Analgesic use, blood dyscrasias, and case-control pharmacoepidemiology. A critique of the International Agranulocytosis and Aplastic Anemia Study.

The International Agranulocytosis and Aplastic Anemia Study (IAAAS) of analgesic-induced risks of blood dyscrasias represents the current "state of the art" in case-control pharmacoepidemiology. We present a conceptual framework for examining the goal, methods, and analysis of an epidemiologic study of drug risks and review the IAAAS within this framework. In our view, the new risk estimates reported by the IAAAS are not inherently more accurate than existing ones, nor have they been measured in clinically and sociodemographically relevant groups of patients over the anticipated course of ther

Review of Recent Pharmacoepidemiologic Post-Market Safety Studies Through the Lens of the Estimand Framework.

The ICH E9(R1) estimand framework provides a systematic approach to ensure alignment among clinical trial objectives, trial conduct, statistical analyses, and interpretation of results, however, whether it can be readily utilized for the pharmacoepidemiologic safety studies has not been established. We selected articles on drug safety published in the Journal Pharmacoepidemiology and Drug Safety (PDS), during 2020 to investigate whether estimand attributes were well defined in the study design and reporting. We found that among twenty-five articles selected, nineteen were cohort studies and si

The first pharmacoepidemiologic investigations: national drug safety policy in the United States, 1901-1902.

The pharmaceutical industry developed in the late 19th century as a consequence of both scientific and commercial innovations, such as the development of diphtheria antitoxin and the commercialization of smallpox vaccine. Two tetanus outbreaks in 1901 -- from contaminated diphtheria antitoxin in St. Louis, Missouri, and contaminated smallpox vaccine in Camden, New Jersey -- raised public concern about pharmaceutical safety. In St. Louis, errant manufacturing processes were found to be the source of the outbreak. In Camden, investigation identified contaminated vaccine from one manufacturer as
