Proteus mirabilis harboring carbapenemase NDM-5 and ESBL VEB-6 detected in Austria.

We describe a case of carbapenemase-harboring Proteus mirabilis together with detection of NDM-5 in Austria accompanied by other bacterial strains with a wide range of beta-lactamases including OXA-181 and VEB-6. Isolates were obtained from a subphrenic abscess from one patient who was previously treated with broad-spectrum antibiotics in Bangladesh.

Regional outbreak of CTX-M-2 β-lactamase-producing Proteus mirabilis in Japan.

Proteus mirabilis is a common cause of urinary tract infection. Wild-type P. mirabilis strains are usually susceptible to penicillins and cephalosporins, but occurrences of P. mirabilis producing extended-spectrum β-lactamases (ESBLs) have been recently reported. Here, we surveyed the prevalence of cefotaxime resistance among P. mirabilis strains at seven different hospitals in Kanagawa Prefecture, Japan, and investigated their molecular epidemiology to explain the mechanism of their spread. The prevalence of cefotaxime resistance among P. mirabilis increased annually, from 10.1 % in 1998 to 2

Complicated catheter-associated urinary tract infections due to Escherichia coli and Proteus mirabilis.

Catheter-associated urinary tract infections (CAUTIs) represent the most common type of nosocomial infection and are a major health concern due to the complications and frequent recurrence. These infections are often caused by Escherichia coli and Proteus mirabilis. Gram-negative bacterial species that cause CAUTIs express a number of virulence factors associated with adhesion, motility, biofilm formation, immunoavoidance, and nutrient acquisition as well as factors that cause damage to the host. These infections can be reduced by limiting catheter usage and ensuring that health care professio

[WITHDRAWN: [Clinical strain of Proteus mirabilis able to produce a plasmid mediated ceftazidimase.]].

Resistance to third generation cephalosporins due to the acquisition and expression of extended spectrum beta-lactamases (ESBLs) enzymes among Gram negative bacteria continue to pose a challenge to infection management worldwide. The aim of this study was to determine the implication of the enzymatic mechanism and to describe the properties of ESBLs from a clinical strain of Proteus mirabilis NC4150 isolated in the intensive care unit at the Military Hospital in Tunisia during the period 2004. The isolate was identified, tested for antimicrobial susceptibility and analysed for presence of ESBL

Encrustation of indwelling urethral catheters by Proteus mirabilis biofilms growing in human urine.

The encrustation and blockage of four types of urinary catheters was studied in a simple laboratory model of the catheterized bladder. Pooled human urine was supplied to the bladder chamber at 0.5 mL/min. The bladder urine was inoculated with a clinical strain of Proteus mirabilis that had been isolated from an encrusted catheter. The models were operated until the catheters blocked and atomic absorption spectrometry was used to assess the amounts of calcium and magnesium deposited on the catheters. Scanning electron microscopy was also used to locate and assess the degree of encrustation. All

[Alkaline encrusted cystitis secondary to Proteus mirabilis infection in a HIV infected patient].

We described for the first time an alkaline encrusting cystitis in a patient with the acquired immune deficiency. This is an entity characterized by severe dysuria and long standing urinary frequency, alkaline urine and radiographically visible calcification of the urinary bladder. It has been observed in patients with permanent Foley catheter, persistent infections due to microorganisms with urolytic activity, urinary bilharziosis or tuberculosis, or malignant bladder tumors. None of these entities were described and the patient had all conditions for this syndrome.

Genomic and Biological Characterization of a Novel Proteus mirabilis Phage with Anti-Biofilm Activity.

The emergence of multidrug-resistant (MDR) Proteus mirabilis poses a significant threat in porcine farming and public health, highlighting the need for alternative biocontrol agents. This study aimed to isolate and characterize a lytic bacteriophage with therapeutic potential against MDR P. mirabilis. Using the clinical MDR P. mirabilis strain Pm 07 as host, a bacteriophage, vB_Pmc_P-07 (P-07), was successfully isolated from fecal and sewage samples via an enrichment protocol. Phage P-07 forms plaques surrounded by a distinct translucent "halo," suggesting the production of depolymerase. It ac

Clonal Dissemination of NDM-Producing Proteus mirabilis in a Teaching Hospital in Sousse, Tunisia.

Proteus mirabilis (P. mirabilis) is an opportunistic pathogen involved in urinary tract infections as well as various nosocomial infections. Emerging resistances to beta-lactams in this species complicates potential treatment since it is intrinsically resistant to colistin. Eleven isolates of carbapenem-non-susceptible P. mirabilis were identified in Sousse Hospital, Tunisia, from January 2018 to December 2022. MICs were determined and isolates were sequenced to determine their resistomes, sequence types, virulence factors, and their clonal relationships. Susceptibility testing showed that all

Use of quorum sensing antagonists to deter the formation of crystalline Proteus mirabilis biofilms.

Proteus mirabilis biofilms are a major cause of urinary catheter blockage. Antibiotic-impregnated catheters used to prevent catheter blockage have achieved limited success. Research has examined the efficacy of quorum sensing inhibitors against Pseudomonas aeruginosa biofilms, but there are few reports of the effects of these compounds against crystalline P. mirabilis biofilms. This study examined the effect of two quorum sensing antagonists, p-nitrophenyl glycerol (PNPG) and tannic acid, against crystalline P. mirabilis biofilms. Tannic acid and PNPG were observed to inhibit the quorum sensin

Comparison of PCR-based molecular markers for the characterization of Proteus mirabilis clinical isolates.

Proteus mirabilis is one of the most important pathogens associated with complicated urinary tract infections (acute pyelonephritis, bladder infections, kidney stones) and bacteremia, affecting patients with anatomical abnormalities, immunodeficiency, and long-term urinary catheterization. For epidemiological purposes, various molecular typing methods, such as pulse-field gel electrophoresis (PFGE) or ribotyping, have been developed for this pathogen. However, these methods are labor intensive and time-consuming. We evaluated the discriminatory power of several PCR-based fingerprinting methods

Biotransformation of Allium Sulfur Metabolites by Proteus mirabilis Yields Cepathiosoxide, a Trithiabicyclic Sulfoxide.

Exposing onion-derived sulfur metabolites to Proteus mirabilis yielded cepathiosoxide, a structurally distinct trithiabicyclic sulfoxide formed through microbial biotransformation. LC-MS revealed the loss of native Allium tetrasulfides and the emergence of a unique cepathiosoxide. NMR spectroscopy and ECD established the existence of a rigid [4.2.1] framework with an embedded sulfoxide. Cepathiosoxide affected colorectal cancer cell viability without impacting normal human colon epithelial cells. These findings indicate that microbial sulfur metabolism provides structurally distinct and potent

Isolation and genomic characterization of a lytic bacteriophage against multidrug-resistant Proteus mirabilis.

Multidrug-resistant (MDR) Proteus mirabilis is a significant opportunistic pathogen, largely due to its ability to form robust biofilms that are recalcitrant to conventional antibiotic therapy. Bacteriophages represent a promising alternative, yet their therapeutic potential is contingent on a thorough characterization of their biological functions and genomic architecture. This study details the isolation and comprehensive analysis of a novel bacteriophage targeting MDR P. mirabilis. A novel lytic bacteriophage, designated Proteus phage ram_arti_1324, was isolated from sewage. Transmission el

Interaction of the SXT/R391 element ICEPmiJpn1 with its natural host Proteus mirabilis.

Integrative and conjugative elements (ICEs) of the SXT/R391 family are mobile genetic elements that integrate into the bacterial host chromosome and can be transferred horizontally, spreading antimicrobial resistance genes. Our study aimed to evaluate aspects of the relationship between ICEPmiJpn1, one of the most widespread SXT/R391 variants, with its natural host Proteus mirabilis. For this investigation, we used isogenic strains (containing or not the ICEPmiJpn1) that enabled us to evaluate the influence of this element on several physiological aspects of P. mirabilis as well as the effect

Plasma activated water pre-treatment substantially enhances phage activity against Proteus mirabilis biofilms.

The ongoing antimicrobial resistance crisis has incentivised research into alternative antibacterial and antibiofilm agents. One of them is plasma-activated water (PAW), which is produced by exposing water to a cold plasma discharge. This process generates a diverse array of reactive oxygen and nitrogen species (ROS/RNS) with antimicrobial properties. Another intensively studied class of alternative antimicrobials are bacteriophages, attracting attention due to their specificity and strong antibacterial activity. As combinations of different types of antimicrobials are known to often exhibit s

Fluoxetine and thioridazine inhibit efflux and attenuate crystalline biofilm formation by Proteus mirabilis.

Proteus mirabilis forms extensive crystalline biofilms on indwelling urethral catheters that block urine flow and lead to serious clinical complications. The Bcr/CflA efflux system has previously been identified as important for development of P. mirabilis crystalline biofilms, highlighting the potential for efflux pump inhibitors (EPIs) to control catheter blockage. Here we evaluate the potential for drugs already used in human medicine (fluoxetine and thioridazine) to act as EPIs in P. mirabilis, and control crystalline biofilm formation. Both fluoxetine and thioridazine inhibited efflux in

Epidemiology of extended-spectrum β-lactamase, AmpC, and carbapenemase production in Proteus mirabilis.

Proteus mirabilis strains that produce extended-spectrum β-lactamase (ESBL), AmpC β-lactamase, and carbapenemase pose potential threats to patient care because most clinical diagnostic laboratories may not attempt to detect these three major groups of enzymes. Therefore, the objective of this study was to ascertain if P. mirabilis isolates collected from our heathcare facility possess various mechanisms of resistance to β-lactams (i.e., ESBL, AmpC, and carbapenemases) and to additionally arrive at conclusions regarding concurrent testing for these three mechanism of drug resistance in order to
