Cyclin D1 overexpression in non-Hodgkin's lymphoma with chromosome 11 bcl-1 rearrangement.

BACKGROUND: Approximately 70% of centrocytic (mantle-cell) lymphomas have the chromosomal translocation t(11;14) (q13; q32) and associated rearrangements at the bcl-1 breakpoint locus and at the cyclin D1 (PRAD1, CCND1) gene, thus implicating this gene in the pathogenesis of centrocytic lymphoma. In order to determine cyclin D1 expression in hematopoietic neoplasms with and without bcl-1 or cyclin D1 rearrangements, northern blot analysis was performed. METHODS: Total RNA was isolated from peripheral blood cells of patients with hematopoietic neoplasms in leukemic phase, including three patie

Non-coding RNAs in gynecologic cancer.

The term "gynecologic cancer" pertains to neoplasms impacting the reproductive tissues and organs of women encompassing the endometrium, vagina, cervix, uterus, vulva, and ovaries. The progression of gynecologic cancer is linked to various molecular mechanisms. Historically, cancer research primarily focused on protein-coding genes. However, recent years have unveiled the involvement of non-coding RNAs (ncRNAs), including microRNAs, long non-coding RNAs (LncRNAs), and circular RNAs, in modulating cellular functions within gynecological cancer. Substantial evidence suggests that ncRNAs may wiel

Novel BMP2K::PDGFRA fusion in an unusual myeloid/lymphoid neoplasm with eosinophilia.

BACKGROUND: Myeloid and/or lymphoid neoplasms with eosinophilia PDGFRA gene fusions usually occur with FIP1L1 as the partner gene; however, novel partners have been described. These novel partners are sometimes responsive to tyrosine kinase inhibitor therapy. PURPOSE: We describe here a patient with a PDGFRA fusion with a previously undescribed partner gene, BMP2K. METHODS: RNA sequencing with the Archer Fusion Plex Pan Solid Tumor Panel (IDT) was used to detect fusions. DNA sequencing using a custom IDT panel was also performed. RESULTS: An in-frame BMP2K::PDGFRA fusion was detected in a p

Transcriptomic classification and clinicopathologic correlation of hepatocellular neoplasms with steatotic features in non-cirrhotic livers.

Differentiating early-stage hepatocellular carcinoma (HCC) from hepatocellular adenoma (HCA), particularly in the context of steatosis, remains a diagnostic challenge. Accurate distinction is essential for appropriate clinical management, yet reliable ancillary assays are limited. This study focuses on hepatocellular neoplasms with steatotic change in non-cirrhotic livers, aiming to minimise confounding factors from cirrhosis and to identify genetic alterations specific to tumour biology. Using total RNA sequencing, we analysed the gene expression profiles of 12 HCA cases, ten HCC cases (inclu

Myeloproliferative Neoplasm Driven by ETV6-ABL1 in an Adolescent with Recent History of Burkitt Leukemia.

ETV6-ABL1 gene fusion is a rare genetic rearrangement in a variety of malignancies, including myeloproliferative neoplasms (MPN), acute lymphoblastic leukemia (ALL), and acute myeloid leukemia (AML). Here, we report the case of a 16-year-old male diagnosed with a MPN, 7 months post-completion of treatment for Burkitt leukaemia. RNA sequencing analysis confirmed the presence of an ETV6-ABL1 fusion transcript, with an intact, in-frame ABL tyrosine-kinase domain. Of note, secondary ETV6-ABL1-rearranged neoplastic diseases have not been reported to date. The patient was started on a tyrosine kinas

Is H19 RNA a Useful Marker of Acromegaly and Its Complications? A Preliminary Study.

Acromegaly is a rare endocrine disorder caused by somatotroph pituitary adenoma. Besides its typical symptoms, it contributes to the development of cardiovascular, metabolic, and bone comorbidities. H19 RNA is a long non-coding RNA and it is suspected to be involved in tumorigenesis, cancer progression, and metastasis. H19 RNA is a novel biomarker for the diagnosis and monitoring of neoplasms. Moreover, there might be an association between H19 and cardiovascular and metabolic diseases. We enrolled 32 acromegaly patients and 25 controls. We investigated whether whole blood H19 RNA expression i

Aberrant EVI1 splicing contributes to EVI1-rearranged leukemia.

Detailed genomic and epigenomic analyses of MECOM (the MDS1 and EVI1 complex locus) have revealed that inversion or translocation of chromosome 3 drives inv(3)/t(3;3) myeloid leukemias via structural rearrangement of an enhancer that upregulates transcription of EVI1. Here, we identify a novel, previously unannotated oncogenic RNA-splicing derived isoform of EVI1 that is frequently present in inv(3)/t(3;3) acute myeloid leukemia (AML) and directly contributes to leukemic transformation. This EVI1 isoform is generated by oncogenic mutations in the core RNA splicing factor SF3B1, which is mutate

The modification of ferroptosis and abnormal lipometabolism through overexpression and knockdown of potential prognostic biomarker perilipin2 in gastric carcinoma.

BACKGROUND: To investigate the biological relationship, mechanism between perilipin2 and the occurrence, advancement of gastric carcinoma, and explore the mechanism of lipid metabolism disorder leading to gastric neoplasm, and propose that perilipin2 is presumably considered as a potential molecular biomarker of gastric carcinoma. METHODS: RNA-seq was applied to analyze perilipin2 and differentially expressed genes modulated by perilipin2 in neoplastic tissues of both perilipin2 overexpression and knockdown groups in vivo. The mechanism was discovered and confirmed by Rt-qPCR, immunoblotting,

Development and Validation of an m6A RNA Methylation Regulators-Based Signature for Predicting the Prognosis of Adrenocortical Carcinoma.

BACKGROUND: Adrenocortical carcinoma (ACC) is an aggressive and rare neoplasm that originates from the cortex of the adrenal gland. N6-methyladenosine (m6A) RNA methylation, the most common form of mRNA modification, has been reported to be correlated with the occurrence and development of the malignant tumor. This study aims to identify the significance of m6A RNA methylation regulators in ACC and construct a m6A based signature to predict the prognosis of ACC patients. MATERIALS AND METHODS: RNA-seq data from The Cancer Genome Atlas (TCGA) database was used to identify the expression level

Sclerosing epithelioid mesenchymal neoplasm of the pancreas - a proposed new entity.

We have encountered pancreatic tumors with unique histologic features, which do not conform to any of the known tumors of the pancreas or other anatomical sites. We aimed to define their clinicopathologic features and whether they are characterized by recurrent molecular signatures. Eight cases were identified; studied histologically and by immunohistochemistry. Selected cases were also subjected to whole-exome sequencing (WES; n = 4), RNA-sequencing (n = 6), Archer FusionPlex assay (n = 5), methylation profiling using the Illumina MethylationEPIC (850k) array platform (n = 6), and TERT promot

Targeted RNA sequencing in the routine clinical detection of fusion genes in salivary gland tumors.

Salivary gland tumors represent a diverse group of neoplasms that occasionally pose a diagnostic challenge for pathologists, particularly with limited sampling. Gene fusions, which may reflect genetic drivers, are increasingly recognized in a subset of these neoplasms, and can be leveraged for diagnostic purposes. We performed a retrospective analysis on a cohort of 80 benign and malignant salivary gland tumors, enriched for subtypes known to harbor recurrent fusion events, to validate the diagnostic use of a targeted RNA sequencing assay to detect fusion transcripts. Testing identified fusion

Pediatric Erythroid Sarcoma Diagnostically Confirmed by Identification of a Recurrent NFIA::CBFA2T3 Fusion.

Erythroid sarcoma (ES) is exceedingly rare in the pediatric population with only a handful of reports of de novo cases, mostly occurring in the central nervous system (CNS) or orbit. It is clinically and pathologically challenging and can masquerade as a nonhematopoietic small round blue cell tumor. Clinical presentation of ES without bone marrow involvement makes diagnosis particularly difficult. We describe a 22-month-old female with ES who presented with a 2-cm mass involving the left parotid region and CNS. The presence of crush/fixation artifact from the initial biopsy made definitive cla

Clinical and biological implications of driver mutations in myelodysplastic syndromes.

Myelodysplastic syndromes (MDS) are a heterogeneous group of chronic hematological malignancies characterized by dysplasia, ineffective hematopoiesis and a variable risk of progression to acute myeloid leukemia. Sequencing of MDS genomes has identified mutations in genes implicated in RNA splicing, DNA modification, chromatin regulation, and cell signaling. We sequenced 111 genes across 738 patients with MDS or closely related neoplasms (including chronic myelomonocytic leukemia and MDS-myeloproliferative neoplasms) to explore the role of acquired mutations in MDS biology and clinical phenotyp

Myeloid somatic mutation panel testing in myeloproliferative neoplasms.

Myeloproliferative neoplasms are characterised by somatic mutations in pathways that regulate cell proliferation, epigenetic modifications, RNA splicing or DNA repair. Assessment of the mutational profile assists diagnosis and classification, but also aids assessment of prognosis, and may guide the use of emerging targeted therapies. The most practical way to provide information on numerous genetic variants is by using massively parallel sequencing, commonly in the form of disease specific next generation sequencing (NGS) panels. This review summarises the diagnostic and prognostic value of so

In situ hybridization with single-stranded RNA probes to demonstrate infrequently elevated gli mRNA and no increased ras mRNA levels in meningiomas and astrocytomas.

Using computer-assisted digital image-analysis, the level of ras and gli messenger RNA (mRNA) in neoplasms of meningeal and glial origin was determined in an attempt to correlate these parameters with histological tumor severity. We used single-stranded, asymmetric, radiolabelled RNA probes to detect the amount of ras and gli mRNA present in formalin-fixed, paraffin-embedded tumor material. Such archival material can provide an immediate, larger sample base than with fresh samples. The extent of ras mRNA expression in 25 brain tumors was not significantly different than normal cerebellum. Howe

Epstein-Barr virus-associated smooth muscle tumour in an immunosuppressed patient: a rare clinical scenario.

Epstein-Barr virus-associated smooth muscle tumours (EBV-SMTs) are rare neoplasms typically affecting immunosuppressed individuals such as organ transplant recipients. This case follows a woman in her 30s who presented with multi-organ masses 5 years after a kidney transplant. While initial suspicion focused on common post-transplant infections or malignancy, a liver biopsy revealed a smooth muscle neoplasm positive for EBV by EBV-encoded RNA in-situ hybridisation, consistent with EBV-SMT. Managing EBV-SMT is challenging as no standardised treatment regimens exist. When reducing immunosuppress

Crosstalk between alternative splicing and non-coding RNAs in hepatocellular carcinoma: from regulatory mechanism to therapeutic implications.

Hepatocellular carcinoma (HCC) is a malignant neoplasm characterized by high incidence and mortality rates globally. Its pathogenesis and progression are intricate and multifaceted, necessitating comprehensive analysis and elucidation. Recent studies have demonstrated that non-coding RNAs (such as long non-coding RNAs, circular RNAs, and microRNAs) and alternative splicing, which are central to post-transcriptional regulation, engage in extensive regulatory interactions. These interactions form a complex regulatory network that significantly influences the malignant properties such as stemness

The expression of MALAT1 long non-coding RNA is associated with good prognosis in mantle cell lymphoma.

MALAT1 is a long non-coding RNA (lncRNA) with altered expression in many cancers but poorly studied in B-cell neoplasms like mantle cell lymphoma (MCL), a very aggressive B-cell lymphoma. To elucidate the role of MALAT1 in MCL we have investigated its expression in 219 primary tumors using microarray data. We also analyzed the expression of its natural antisense transcript (TALAM1), which is key for the processing of MALAT1 RNA to its active isoform. High expression of MALAT1/TALAM1 lncRNAs was consistently linked to favorable clinical behavior in MCL, independently of other alterations previo
