Mice tolerant to nitrous oxide are not tolerant to barbiturates.

Mice continuously exposed to 50% nitrous oxide for 2 weeks or more become tolerant to nitrous oxide, as measured by an 8% to 19% increase in their nitrous oxide ED50 (the dose required to abolish the righting reflex in 50% of the animals). Control mice tolerant to nitrous oxide were given intraperitoneal injections of secobarbital (50 mg/kg), phenobarbital (120 mg/kg), or pentobarbital (55 or 65 mg/kg). Sleep onset times, sleep times, and serum levels of barbiturates upon awakening did not differ between the two groups. These results suggest that nitrous oxide and barbiturates do not share a c

Barbiturates and lung cancer in humans.

In a hypothesis-generating study looking for possible carcinogenic effects of drugs in humans, each of three barbiturates (pentobarbital sodium, phenobarbital, and secobarbital sodium) showed a statistically significant association with the subsequent development of lung cancer, with relative risks ranging from 1.5 to 2.8. Further analysis showed that the association was not explained by the increased prescription of barbiturate drugs shortly before the diagnosis of lung cancer or by an association of barbiturate use with cigarette smoking. Much of the data did not support a causal relationshi

A comparison of the potencies of a series of barbiturates at the neuromuscular junction and on the central nervous system.

The ability of a series of barbiturates to depress the depolarizing action of carbachol at the end-plate of guinea-pig lumbrical muscle was studied. The compounds studied were: amorbarbital, aprobarbital, barbital, barbituric acid, butabarbital, butalbital, dimethylbutylethyl barbituric acid, hexobarbital, mephobarbitak, secobarbital, thiamylal, and thiopental. The depressant activity was sensitive to small changes in structure of the compounds strongly suggesting that a specific receptor site was involved in the interaction of the drug with the tissue. The observed relative potencies on the m

A plausible causal relationship between the increased use of fentanyl as an obstetric analgesic and the current opioid epidemic in the US.

Drug poisoning deaths have more than doubled in the United States since 2000 with fentanyl and fentanyl analogues primarily responsible for the jump in opioid deaths. Robust data indicate a convincing correlation between the exposure of the fetus to other labor medications (morphine, pethidine hydrochloride, barbiturates, phenobarbitone, meperidine, and secobarbital) and the later addiction of young adults to the same category of drug. We present the hypothesis that this effect is also true of the opioid, fentanyl: there is a causal relationship between the increased popularity of fentanyl as

Treatment of the barbiturate abstinence syndrome.

Patients with the barbiturate abstinence syndrome require pharmacotherapy (and hospitalization) when there is a history of the ingestion of greater than 0.4 g of secobarbital or its equivalent for 90 or more days, or of 0.6 g for 30 or more days, or a previous history of barbiturate withdrawal seizures or delirium. The pharmacological management of withdrawal, which is rational, safe, efficacious and relatively simple, is described and recommended. This treatment relies on a phenobarbital loading-dose technique which is titrated to clinical end-points. The prolonged elimination of phenobarbita

Peculiar pharmacokinetics of brallobarbital as a source of complications in Vesparax intoxication.

1. The rate of elimination of brallobarbital, a component of Vesparax depends on the route of administration. The half-life following oral administration is considerably longer than after intravenous injection. This corresponds with longer sleeping times. 2. The half-life of brallobarbital administered intravenously could be increased by administration of its metabolites. This suggests that a metabolite, formed especially after oral administration, is responsible for the retarded elimination of brallobarbital. 3. The presence of the other components of Vesparax, namely secobarbital and hydroxy

[Rapid identification and quantitation of barbiturates in plasma using solid-phase extraction combined with GC-FID and GC-MS method].

A rapid screening method for the quantitation and identification of 7 barbiturates in plasma is presented. The method employs solid-phase extraction at pH 3 using X-5 resin as absorbent. Identification of each barbiturate is achieved by retention time and GC-MS. Detection threshold (0.3-0.5 micrograms.ml-1 in a 0.5 ml plasma sample and 1 microliter injection volume), precision (RSD 3.46-6.41%), and linearity (r > 0.99) were satisfactory for blood level monitoring and clinical toxicological applications.

Routine identification and determination of 11 barbiturates in biological samples.

A very easy, reliable and specific gas-chromatographic method for identification and dosage of 11 barbiturates in plasma is presented. Methylene unit values determined on two types of column (one polar, one non-polar) allows very accurate identification with a minimum risk of error due to fluctuations in operating conditions. Sensitivity of the method is 0.5 microgram/ml plasma with a flame ionization detector. Derivatization procedure is complete, without any degradation phenomenon as tested by mass spectrometry. Such a method can be easily used as routine procedure for toxicological or pharm

Barbiturates inhibit progesterone synthesis in cultured Leydig tumor cells and human granulosa cells.

Screening drugs used in obstetrical practice for effects on steroid hormone synthesis revealed that phenobarbital inhibited progesterone synthesis in MA-10 Leydig tumor cells. The inhibition was apparently a drug class effect since it could be reproduced by other barbiturates. Barbiturate blockade was reversible and could be bypassed in the MA-10 cells by using 22-hydroxycholesterol. Human granulosa cell progesterone synthesis was also inhibited in a dose dependent fashion by phenobarbital, secobarbital and barbituric acid. Significant inhibition occurred in dose ranges that would be therapeut

Validation of an automated microplate enzyme immunoassay for screening of postmortem blood for drugs of abuse.

The objective of this study was to compare the sensitivity and specificity of an enzyme immunoassay employing antibodies bound to a microtiter plate (MPEIA) with those of two radioimmunoassays for screening postmortem blood from selected coroner's cases for drugs of abuse. The radioimmunoassays were a coated-tube radioimmunoassay (CTRIA) and a double antibody radioimmunoassay (DARIA). Specimens consisted of 260 postmortem blood specimens from coroner's cases. Immunoassay results (positive or negative) were compared with confirmed results on those cases by gas chromatography-mass spectrometry,

Detection of drugs using XAD-2 resin. I:Choice of resin, chromatographic conditions, and recovery studies.

Amberlite XAD-2, a nonionic polystyrene divinylbenzene resin, was first used for the analysis of drugs in urine and a number of reports have described the development at optimal conditions for extraction, including type of resin columns, pH conditions, and eluting solvents. XAD-4 and XAD-7 resins were compared to the similarly structured XAD-2 resin and no significant advantage over the XAD-2 resin for drug screening was observed. A quantity of 5 to 6 g of resin was found to have sufficient capacity for the extraction of 200 ml of pentobarbital solution (1 mg/100ml). A column flow rate of appr

Application of a vidicon spectrometer for simultaneous multicomponent drug determinations.

A rapid scanning vidicon spectrometer has been evaluated for the simultaneous determination of drugs in mixtures, without a separation step. Spectral data in the ultraviolet region are collected on-line with a small computer at repetition rates of 100 scans per second. Absorbance data at several wavelengths are processed by matrix equations to resolve them into the concentration of each component in each mixture. Results are reported for two-component mixtures of procainamide and N-acetylprocainamide in serum, for two-component aqueous mixtures of butabarbital and seconbarbital, and for seven-

Modulation of phencyclidine-sensitive ethidium binding to the Torpedo acetylcholine receptor: interaction of noncompetitive inhibitors with carbamylcholine and cobra alpha-toxin.

Ethidium is one of two fluorescent ligands known to bind to the noncompetitive inhibitor (NCI) site in the central ion channel of the Torpedo acetylcholine receptor with a micromolar dissociation constant. To further characterize heterotropic allosteric regulation of ligand binding in general, and of ethidium binding in particular, to the Torpedo receptor, we measured the effects of three liquid anesthetics (diethyl ether, halothane, and butanol), two barbiturates (secobarbital and thiamylal), and urethane. The phencyclidine-sensitive chromatic shift and the quantum yield increase associated w

Pentobarbital quantification in the presence of phenobarbital by fluorescence polarization immunoassay.

We developed a method for measuring pentobarbital in samples that also contain phenobarbital. The phenobarbital is destroyed by adding sodium hydroxide and then heating at 95 degrees C for 60 min. Pentobarbital is not affected by this pretreatment and can then be measured with an Abbott TDx barbiturates kit. Using this method, we obtained an average analytical recovery of 98% of added pentobarbital and a correlation of y = 0.953x + 3.4 vs HPLC. The intra-assay CV was 3.7% at 25.8 mg/L; the interassay CV was 6.2% at 16.1 mg/L. Other long-acting barbiturates, e.g., hexobarbital, are also effecti

Solid-phase extraction and GC/MS confirmation of barbiturates from human urine.

A highly selective and sensitive procedure has been developed for isolating and identifying barbiturates in human urine. With a new disposable bonded silica gel solid-phase extraction (SPE) column and hexobarbital as an internal standard (IS), amobarbital, butabarbital, pentobarbital, phenobarbital, secobarbital, and methaqualone were selectively isolated from endogenous urine components. Capillary gas chromatography/ion trap mass spectrometry (GC/MS) analysis of the extracts generated a full mass spectrum for the detection, identification, and quantitation of barbiturates. Linear quantitative

Interaction of stereoisomers of barbiturates with [3H]alpha-dihydropicrotoxinin binding sites.

Racemic depressant barbiturates inhibit the binding of gamma-aminobutyric acid antagonist [3H]alpha-dihydropicrotoxinin (DHP) to rat brain membranes with IC50 values ranging from 5 to 50 microM. The (-) isomers of pentobarbital and secobarbital were three to four-fold more potent that their (+) isomers in inhibiting [3H]DHP binding. In contrast, the (+) isomer of hexobarbital was a better inhibitor than (-) hexobarbital. The stereoisomers of 1-methyl-5-phenyl-5-propylbarbituric acid (MPPB), which show opposite pharmacological activity, inhibited [3H]DHP binding in a biphasic manner with a plat

Isotopic analogs as internal standards for quantitative GC/MS analysis--molecular abundance and retention time difference as interference factors.

The following analyte/isotope-labeled internal standard (IS) systems are adapted to further study the interference phenomenon previously reported from our laboratory--the intensity ratio of the ion-pair designated for a specific analyte/2H-analog system increases as the solvent used to reconstitute the extraction/derivatization residue is increased: (a) Three analyte/2H-analog pairs with 2H-atoms positioned at allylic sites (butalbital, secobarbital, methohexital); (b) Two analyte/2H-analog pairs without these structural features (pentobarbital, phenobarbital); and (c) Two analyte/13C-analog p
