Longitudinal changes of iron in the substantia nigra of early-stage Parkinson's disease.

BACKGROUND: Higher iron occurs in the substantia nigra in Parkinson's disease (PD), but its behavior over time is poorly understood. OBJECTIVE: To determine if nigral iron measured by quantitative susceptibility MRI can track progression of early stages of PD. METHODS: Thirty-four early-stage PD patients (disease duration < 4 yrs) and 35 matched controls were recruited. Iron content in the substantia nigra and related regions was obtained using quantitative susceptibility mapping with zero-referencing at both baseline and one-year follow-up, along with clinical metrics. RESULTS: Of five cli

A population of dopaminergic neurons in the substantia nigra is required for food-anticipatory activity.

Anticipating daily food availability is a conserved circadian behavior that persists even in animals lacking the suprachiasmatic nucleus, yet its neural substrates remain poorly defined. Previous studies implicated dopamine signaling in food-anticipatory activity (FAA) but lacked the resolution to identify the responsible neuronal population. Here, we conditionally deleted tyrosine hydroxylase (Th) from molecularly defined dopamine neuron populations in mice. Broad deletion of Th in dopamine transporter-expressing neurons nearly abolished FAA, whereas the restoration of Th in substantia nigra

Astrocytes mediate the dopaminergic modulation of tonic GABAergic signaling in substantia nigra.

Recent studies have implicated dopaminergic signaling within the substantia nigra pars reticulata (SNr) in the emergence of Parkinsonian motor deficits. To better understand the mechanisms underlying this dependence, the ability of dopamine to modulate SNr neuron activity was studied in ex vivo mouse brain slices. In addition to presynaptically inhibiting phasic GABA release, D2 dopamine receptor (D2R) signaling unexpectedly suppressed a tonic, GABAA receptor-mediated inhibition of SNr neuron spiking. Our studies demonstrated that this tonic modulation was largely mediated by GAT-1-dependent G

Regional neuromelanin reduction in the substantia nigra in different subtype of Parkinson's disease.

BACKGROUND: This study aimed to investigate neuromelanin-sensitive MRI (NM-MRI) characteristics in the substantia nigra pars compacta (SNc) across Parkinson's disease (PD) subtypes and their association with motor and cognitive performance. METHODS: Four groups were included: PD patients with mild motor predominant (PD-MMP, n = 37), intermediate (PD-IM, n = 41), diffuse malignant (PD-DM, n = 12), and healthy controls (HC, n = 51). Clinical evaluations and MRI scans were performed. Signal values from NM-MRI were extracted using SNc and background masks. Contrast-to-noise ratio (CNR) values for

A Spatially Resolved View on the Aging Substantia nigra: An Exploratory Proteomic Study.

Physiological aging is accompanied by structural and molecular changes in the brain, with varying degrees in different brain areas, and is considered one of the major risk factors for neurodegenerative diseases. Thus, the present study focuses on elucidating age-related changes in the substantia nigra pars compacta (SNpc), a brain region particularly vulnerable in Parkinson's disease. Here, the aim is to gain a spatially resolved view of aging-dependent alterations to conclude early processes potentially involved in neurodegeneration. Neuromelanin granules and SNpc tissue are isolated from tis

Metal dyshomeostasis in the substantia nigra of patients with Parkinson's disease or multiple sclerosis.

Abnormal metal distribution in vulnerable brain regions is involved in the pathogenesis of most neurodegenerative diseases, suggesting common molecular mechanisms of metal dyshomeostasis. This study aimed to compare the intra- and extra-neuronal metal content and the expression of proteins related to metal homeostasis in the substantia nigra (SN) from patients with Parkinson's disease (PD), multiple sclerosis (MS), and control subjects. Metal quantification was performed via ion-beam micro-analysis in neuromelanin-positive neurons and the surrounding tissue. For proteomic analysis, SN tissue l

Diagnostic utility of 7T neuromelanin imaging of the substantia nigra in Parkinson's disease.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder that presents a diagnostic challenge due to symptom overlap with other disorders. Neuromelanin (NM) imaging is a promising biomarker for PD, but adoption has been limited, in part due to subpar performance at standard MRI field strengths. We aimed to evaluate the diagnostic utility of ultra-high field 7T NM-sensitive imaging in the diagnosis of PD versus controls and essential tremor (ET), as well as NM differences among PD subtypes. A retrospective case-control study was conducted including PD patients, ET patients, and contro

Local iron deficiency in the substantia nigra directly contributes to hyperlocomotion phenotypes.

Brain iron is precisely regulated, and disrupted brain iron homeostasis is implicated in neuropsychological disease. Mounting evidence connects the iron status of the substantia nigra (SN) with locomotion-related neural symptomatology. Researchers in this field have long speculated that iron deficiency in the SN directly causes the high-locomotion symptoms observed in neuropsychiatric disorders. However, no direct experimental evidence of a causal relationship has been presented. To explore the relationship between iron deficiency in the SN and locomotion-related phenotypes, we stereotaxically

Angiotensin type 2 receptors: Role in aging and neuroinflammation in the substantia nigra.

Overactivity of the angiotensin-type-1 receptor (AT1)/NADPH-oxidase axis enhances aging processes, neuroinflammation and neurodegeneration. The role of AT2 receptors in the above-mentioned AT1-related effects in the aged brain, particularly substantia nigra, was investigated in this study. In the nigra, we observed a progressive decrease in AT2 mRNA expression with aging, and AT2 deletion led to changes in spontaneous motor behavior, dopamine receptors, renin-angiotensin system, and pro-oxidative and pro-inflammatory markers similar to those observed in aged wild type (WT) mice. Both aged WT m

Transient glutathione depletion in the substantia nigra compacta is associated with neuroinflammation in rats.

Glutathione (GSH) deficiency has been identified as an early event in the progression of Parkinson's disease. However, the role of GSH in the etiology and pathogenesis of this neurodegenerative disorder is not well established. The aim of this study is to assess the effect of transient GSH depletion in the substantia nigra pars compacta (SNpc) on neuroinflammation after the injection of a single dose of l-buthionine sulfoximine (BSO) into the SNpc of male Sprague-Dawley rats. The results showed that BSO treatment stimulates microglia (p<0.01) and astroglial response (p<0.01), c-Jun N-terminal

Substantia nigra hyperechogenicity and Parkinson's disease risk in patients with essential tremor.

BACKGROUND: Several studies have reported an increased risk for patients with essential tremor to develop Parkinson's disease. In addition, hyperechogenicity in the area of the substantia nigra has been associated with a markedly increased risk for Parkinson's disease. The objective of this study was to evaluate the validity of substantia nigra hyperechogenicity in patients with essential tremor as a risk marker for Parkinson's disease. METHODS: Transcranial sonography was performed in 70 patients suffering from essential tremor. Fifty-four of these patients were available for follow-up after

Oxygen and glucose deprivation (OGD)-induced spreading depression in the Substantia Nigra.

Spreading depression (SD) is a profound depolarization of neurons and glia that propagates in a wave-like manner across susceptible brain regions, and can develop during periods of compromised cellular energy such as ischemia, when it influences the severity of acute neuronal damage. Although SD has been well characterized in the cerebral cortex and hippocampus, little is known of this event in the Substantia Nigra (SN), a brainstem nucleus engaged in motor control and reward-related behavior. Transverse brain slices (250 μm; P21-23 rats) containing the SN were subject to oxygen and glucose de

Topography of dyskinesias and torticollis evoked by inhibition of substantia nigra pars reticulata.

GABAergic neurons of the substantia nigra pars reticulata (SNpr) and globus pallidus pars interna (GPi) constitute the output pathways of the basal ganglia. In monkeys, choreiform limb dyskinesias have been described after inhibition of the GPi, but not the SNpr. Given the anatomical and functional similarities between these structures, we hypothesized that choreiform dyskinesias could be evoked by inhibition of an appropriate region within the SNpr. The GABAA receptor agonist, muscimol, was infused into various sites within the SNpr and the adjacent STN of freely moving macaques. The effect o

Loss of Mecp2 in substantia nigra dopamine neurons compromises the nigrostriatal pathway.

Mutations in the methyl-CpG-binding protein 2 (MeCP2) result in Rett syndrome (RTT), an X-linked disorder that disrupts neurodevelopment. Girls with RTT exhibit motor deficits similar to those in Parkinson's disease, suggesting defects in the nigrostriatal pathway. This study examined age-dependent changes in dopamine neurons of the substantia nigra (SN) from wild-type, presymptomatic, and symptomatic Mecp2(+/-) mice. Mecp2(+) neurons in the SN in Mecp2(+/-) mice were indistinguishable in morphology, resting conductance, and dopamine current density from neurons in wild-type mice. However, the

Restless legs and substantia nigra hypoechogenicity are common features in Friedreich's ataxia.

Friedreich's ataxia (FA) is a multisystemic degenerative disease, but the prevalence of restless legs syndrome (RLS) is unknown. FA patients might be particularly susceptible to develop RLS as FA presents with features commonly associated with RLS, e.g. multisystemic network dysfunction, peripheral neuropathy and disturbances in subcellular brain iron homeostasis. In this work, we assessed the following: (1) the prevalence of RLS; (2) the prevalence of sonographic hypoechogenicity of the substantia nigra (SN), which is known to be associated with idiopathic RLS; and (3) the relation between bo

Pseudotype-dependent lentiviral transduction of astrocytes or neurons in the rat substantia nigra.

Gene transfer to the central nervous system provides powerful methodology for the study of gene function and gene-environment interactions in vivo, in addition to a vehicle for the delivery of therapeutic transgenes for gene therapy. The aim of the present study was to determine patterns of tropism exhibited by pseudotyped lentiviral vectors in the rat substantia nigra, in order to evaluate their utility for gene transfer in experimental models of Parkinson's disease. Isogenic lentiviral vector particles encoding a GFP reporter were pseudotyped with envelope glycoproteins derived from vesicula

Substantia nigra and Parkinson's disease: a brief history of their long and intimate relationship.

The substantia nigra was discovered in 1786 by Félix Vicq d'Azyr, but it took more than a century before Paul Blocq and Georges Marinesco alluded to a possible link between this structure and Parkinson's disease. The insight came from the study of a tuberculosis patient admitted in Charcot's neurology ward at la Salpêtrière because he was suffering from unilateral parkinsonian tremor. At autopsy, Blocq and Marinesco discovered an encapsulated tumor confined to the substantia nigra, contralateral to the affected side, and concluded that tremor in that particular case resulted from a midbrain le

Vascular Parkinsonism: a case of lacunar infarction localized to mesencephalic substantia nigra.

An 80-year-old caucasian man with acute onset parkinsonism is reported here. Lower limbs were predominantly involved and he did not show remarkable improvement with dopaminergic therapy. His MRI investigation showed an isolated lesion on the right side of the mesencephalon, consistent the localization of substantia nigra. The association of acute onset parkinsonism and this radiological finding is very rare in the literature.

Effects of barbiturates on ATP-sensitive K channels in rat substantia nigra.

ATP-sensitive K channels are widely expressed in cytoplasmic membranes of neurons, and they couple cell metabolism to excitability. They are thought to be involved in neuroprotection against cell damage during hypoxia, ischemia and excitotoxicity by hyperpolarizing neurons and reducing excitability. Although barbiturates are often used in patients with brain ischemia, the effects of these agents on neuronal ATP-sensitive K channels have not been clarified. We studied the effects of thiopental and pentobarbital on surface ATP-sensitive K channels in principal neurons of rat substantia nigra par
