The HTLV-I oncoprotein Tax inactivates the tumor suppressor FBXW7.

UNLABELLED: Human T-cell leukemia virus type 1 (HTLV-I) is the etiological agent of adult T-cell leukemia (ATL). Mutational analysis has demonstrated that the tumor suppressor, F-box and WD repeat domain containing 7 (FBXW7/FBW7/CDC4), is mutated in primary ATL patients. However, even in the absence of genetic mutations, FBXW7 substrates are stabilized in ATL cells, suggesting additional mechanisms can prevent FBXW7 functions. Here, we report that the viral oncoprotein Tax represses FBXW7 activity, resulting in the stabilization of activated Notch intracellular domain, c-MYC, Cyclin E, and mye

Response of legal and illegal cigarette prices to a tax increase in Ethiopia.

BACKGROUND: In 2020, Ethiopia passed a landmark tax proclamation implementing an evidence-based mixed excise system aimed at curbing tobacco use. This study evaluates the impact of the tax increase of more than 600% on both legal and illegal cigarette prices in order to gauge the impact of the tax reform in the presence of a sizeable illicit cigarette market. METHODS: Data on 1774 cigarette prices were obtained from retailers during Empty Cigarette Pack Surveys in the capital and major regional cities conducted in 2018 and 2022. Packs were categorised as 'legal' or 'illicit' using criteria fr

The Pink Tax: Sex and Gender Disparities in Peripheral Artery Disease.

Peripheral artery disease (PAD) is an atherosclerotic disease associated with significant functional impairment, morbidity, and mortality. Among women, PAD remains poorly recognized and undermanaged. Compared with men, women with PAD tend to be underdiagnosed or misdiagnosed, have poorer quality of life, and experience higher rates of PAD-related morbidity and cardiovascular mortality. In this review, we describe the sex- and gender-related differences in the epidemiology, presentation, diagnosis, and management of PAD. We provide specific recommendations to overcome these factors, including g

Differential price responses for tobacco consumption: implications for tax incidence.

Increasing tobacco taxes is considered the most effective an cost-effective policy to reduce tobacco consumption. However, a common objection to tobacco taxes is that they tend to rely disproportionately on the poorest individuals since less affluent smokers incur proportionately greater expenditures on cigarettes compared with more affluent smokers. Such objections usually assume that all smokers throughout the income distribution react similarly to an increase in tobacco prices. But, if less affluent smokers are more sensitive to price changes (ie, they have a higher demand price elasticity),

'Tax-otsubo': stress cardiomyopathy following an encounter with the Inland Revenue.

An 89-year-old man developed chest pain and palpitations shortly after finishing a stressful 40 min phone call to HM Revenue and Customs. After admission to the emergency department, he had a cardiovascular collapse followed soon after by a cardiac arrest due to ventricular fibrillation (VF). The troponin T was elevated and his ECG showed extensive deep T wave inversion with prolongation of the QT interval. A portable hand-held ultrasound device (VScan; GE Healthcare) was used to demonstrate classical apical ballooning of the left ventricular apex indicating a diagnosis of takotsubo stress car

The Effect of the Earned Income Tax Credit on Housing and Living Arrangements.

As rents have risen and wages have not kept pace, housing affordability in the United States has declined over the last 15 years, impacting the housing and living arrangements of low-income families. Housing subsidies improve the housing situations of low-income families, but less than one in four eligible families receive a voucher. In this article, we analyze whether one of the largest anti-poverty programs in the United States-the Earned Income Tax Credit (EITC)-affects the housing (eviction, homelessness, and affordability) and living arrangements (doubling up, number of people in the hous

In-work tax credits for families and their impact on health status in adults.

BACKGROUND: By improving two social determinants of health (poverty and unemployment) in low- and middle-income families on or at risk of welfare, in-work tax credit for families (IWTC) interventions could impact health status and outcomes in adults. OBJECTIVES: To assess the effects of IWTCs on health outcomes in working-age adults (18 to 64 years). SEARCH METHODS: We searched 16 electronic academic databases, including the Cochrane Public Health Group Specialised Register, Cochrane Database of Systematic Reviews (The Cochrane Library 2012, Issue 7), MEDLINE and EMBASE, as well as six grey

Public opinions on tax and retail-based tobacco control strategies.

BACKGROUND: While tobacco taxes and smoke-free air regulations have significantly decreased tobacco use, tobacco-related illness accounts for hundreds of thousands of annual deaths. Experts are considering additional strategies to further reduce tobacco consumption. METHODS: We investigated smokers' (n=2118) and non-smokers' (n=2210) opinions on existing and theoretical strategies, including tax and retailer-based strategies in New York City, across three cross-sectional surveys. RESULTS: Compared with smokers, non-smokers were significantly more likely (p<0.05) to favour all tobacco control

Highlights on distinctive structural and functional properties of HTLV Tax proteins.

Human T cell leukemia viruses (HTLVs) are complex human retroviruses of the Deltaretrovirus genus. Four types have been identified thus far, with HTLV-1 and HTLV-2 much more prevalent than HTLV-3 or HTLV-4. HTLV-1 and HTLV-2 possess strictly related genomic structures, but differ significantly in pathogenicity, as HTLV-1 is the causative agent of adult T cell leukemia and of HTLV-associated myelopathy/tropical spastic paraparesis, whereas HTLV-2 is not associated with neoplasia. HTLVs code for a protein named Tax that is responsible for enhancing viral expression and drives cell transformation

Wip1 and p53 contribute to HTLV-1 Tax-induced tumorigenesis.

BACKGROUND: Human T-cell Leukemia Virus type 1 (HTLV-1) infects 20 million individuals world-wide and causes Adult T-cell Leukemia/Lymphoma (ATLL), a highly aggressive T-cell cancer. ATLL is refractory to treatment with conventional chemotherapy and fewer than 10% of afflicted individuals survive more than 5 years after diagnosis. HTLV-1 encodes a viral oncoprotein, Tax, that functions in transforming virus-infected T-cells into leukemic cells. All ATLL cases are believed to have reduced p53 activity although only a minority of ATLLs have genetic mutations in their p53 gene. It has been sugges

Mechanisms of persistent NF-kappaB activation by HTLV-I tax.

Human T cell leukemia virus type I (HTLV-I) is the causative agent of a fatal malignancy known as adult T cell leukemia (ATL). The HTLV-I Tax protein is thought to play a significant role in the initiation and pathogenesis of HTLV-I-mediated disease. Tax is a potent oncogene that deregulates cellular gene expression by persistently activating signaling pathways such as NF-kappaB. Tax activation of NF-kappaB is critical for the immortalization and survival of HTLV-I-infected T cells. In this review, we describe recent insights into the mechanisms employed by Tax to activate the canonical and no

A tax on luxury: HTLV-I infection of CD4+CD25+ Tregs.

Almost a quarter of a century ago, Oldstone and colleagues proposed that infection of cells by noncytopathic viruses may lead to an alteration of the cells' ability to produce certain products or perform certain tasks, i.e., inhibition of "luxury function." In this issue of the JCI, this topic has been revisited by Yamano et al., who demonstrate that human T cell lymphotropic virus type I (HTLV-I) infection of CD4(+)CD25(+) Tregs in patients with HTLV-I-associated myelopathy/tropical spastic paraparesis (HAM/TSP) results in a decrease in FOXP3 mRNA and protein expression. This leads to the ina

[Pathomechanism of HTLV-I associated arthropathy and the role of tax gene].

HTLV-I is known to be a causative agent for adult T cell leukemia. Recent studies revealed this virus is also related to several autoimmune disorders, such as arthropathy, myelopathy and Sjögren's syndrome. We studied etiology of HTLV-I associated arthropathy (HAAP), and found that tax is a causative gene for synovial proliferation and induction of immunogenicity. HTLV-I transgenic mice supported the etiopathological role of tax gene. Our results suggested that HAAP is considered to be a prototype of rheumatoid arthritis, and tax is a best tool for recognizing pathomechanism of rheumatoid arth

Can health taxes help sustain Africa's HIV response and research agenda? Potential fiscal space, implementation risks, and equity considerations.

Sub-Saharan Africa's HIV response is under acute financing stress as external assistance becomes less predictable, debt service costs displace public expenditure, and governments face growing demands on health and community systems. Health taxes on tobacco, alcohol, and sugar-sweetened beverages are increasingly proposed as one instrument of domestic resource mobilisation, but their contribution to HIV sustainability is contested, and their design space in African contexts is imperfectly characterised. The present paper asks under what conditions health taxes can contribute to sustaining HIV s

The association between excise taxes and smoking and vaping transitions-Findings from the 2016-2020 ITC United States surveys.

BACKGROUND: While a growing number of studies examined the effect of e-cigarette (EC) excise taxes on tobacco use behaviors using cross-sectional surveys or sales data, there are currently no studies that evaluate the impact of EC taxes on smoking and vaping transitions. METHODS: Using data from the US arm of the 2016-2020 International Tobacco Control Four Country Smoking and Vaping Survey (ITC 4CV), we employed a multinomial logit model with two-way fixed effects to simultaneously estimate the impacts of cigarette/EC taxes on the change in smoking and vaping frequencies. RESULTS: Our bench

From shadow to sustainability: How informality, environmental taxes, and green innovation reshape carbon and biodiversity futures in the G7 countries.

The shadow economy remains a blind spot in climate-and-biodiversity policy. However, its interaction with fiscal and technological forces can significantly affect the success or failure of sustainability transitions. We propose a novel integrated framework that combines econometric models with deep learning to examine the role of the shadow economy, environmental taxes and green innovation on consumption-based CO2 emissions and biodiversity in the G7 countries. Using data from 1994 to 2020, the study employs Cross-sectionally Autoregressive Distributed-lag (CS-ARDL) and Fully Modified Ordinary

The alcohol industry, the tobacco industry, and excise taxes in the US 1986-89: new insights from the tobacco documents.

BACKGROUND: The UCSF Industry Documents Library has provided public health researchers with key insights into the organization of political activities in the tobacco industry. Much less is known about the alcohol industry. In the US, there is some existing evidence of cooperation between the two industries, particularly in areas where there are mutual interests and/or policy goals at stake. Efforts to raise excise taxes on tobacco and alcohol products are one such example. METHODS: We systematically searched the UCSF Industry Documents Library for data on alcohol industry actors and their pol

Tobacco Control in Botswana: Effectiveness of Tax Changes in Reducing Tobacco Demand.

INTRODUCTION: Botswana has one of the highest adult tobacco-use rates in the WHO African Region. Despite measures and policies put in place to control tobacco consumption, there is still a widespread use of tobacco products in the country. This is because demand for tobacco and tobacco products is mostly affected by changes in tobacco prices and related taxes. This study therefore seeks to analyze the responsiveness of cigarette demand to price changes in Botswana. METHODS: This study analyses Botswana's conditional price elasticity of cigarette demand using Deaton's model. Deaton's method us

Effects of a Sugar-Sweetened Beverages Tax on Caries in Italy: A Modelling Study.

UNLABELLED: <p>Introduction: Despite efforts to reduce sugar consumption, Italy continues to experience high levels of caries, particularly among 12-year-old children. The introduction of a sugar tax is one strategy that gained traction as a mean of curbing excessive sugar intake. This study aimed to assess the potential impact of a sugar tax on reducing caries prevalence through the analysis of two scenarios: a hypothetical implementation of the tax in 2008 (Scen1), compared against actual 2017 data, and a projection of the effects of the planned 2025 tax extending to 2034 (Scen2). METHODS:
