Targeting neutral sphingomyelinase 2 by cambinol decreases cell proliferation and migration of metastatic castration-resistant prostate cancer cells.

Metastatic castration-resistant prostate cancer (mCRPC) remains a major clinical challenge in the treatment of advanced-stage prostate cancer, particularly among American men. Despite current therapeutic options, disease progression and resistance continue to limit patient outcomes. Therefore, the exploration of novel therapeutic strategies is urgently needed. This study investigates the anticancer potential of cambinol, a selective inhibitor of neutral sphingomyelinase 2, alone and in combination with apalutamide, in mCRPC cells. mCRPC cells were treated with different concentrations of cambi

Dermatological adverse events in Chinese prostate cancer patients treated with the androgen receptor inhibitor apalutamide.

BACKGROUND: Apalutamide, an androgen receptor inhibitor, has shown good efficacy in treating prostate cancer (PCa). However, dermatological adverse events (dAEs) are common and threatening, and relevant studies in China are limited. METHODS: This was a retrospective, single-center analysis. We included PCa patients who were hospitalized and received apalutamide treatment at one comprehensive hospital in eastern China from August 2020 to March 2023. These patients were categorized into two groups for comparative analysis: those with (dAEs+) and without dAEs (dAEs-) based on the presence or abs

Early favorable prostate-specific antigen response prediction in metastatic hormone sensitive prostate cancer.

There is an unmet need for a tool that could predict early favorable prostate-specific antigen (PSA) response in metastatic hormone sensitive prostate cancer (mHSPC) patients receiving androgen receptor pathway inhibitor (ARPI). Here, we train and validate a multivariable logistic regression model to predict early favorable PSA response (≤0.2 ng/mL by 6 months) in these patients. Patients randomly allocated to the ARPI arms of the LATITUDE (abiraterone), TITAN (apalutamide), and ARASENS (darolutamide) trials, are split 60:40 into training (n = 1030) and internal validation (n = 688) cohorts. Real-world comparison of enzalutamide and apalutamide in metastatic hormone-sensitive prostate cancer: a multi-institutional propensity score-matched analysis.

BACKGROUND: Androgen receptor signaling inhibitors (ARSIs) such as enzalutamide (Enz) and apalutamide (Apa), in combination with androgen deprivation therapy (ADT), are standard for metastatic hormone-sensitive prostate cancer (mHSPC). However, direct real-world comparisons, particularly in Japanese populations, are limited. METHODS: We retrospectively reviewed 227 mHSPC patients treated with Enz (n = 112) or Apa (n = 115) plus ADT. Propensity score matching (PSM) adjusted for baseline differences. Primary endpoints were prostate-specific antigen (PSA) response, time to castration resistance

Cancer-control outcomes of patients with metastatic hormone-sensitive prostate cancer and ≥ 10 bone metastases receiving apalutamide: a real-world cohort.

PURPOSE: Currently available post-hoc TITAN study data indicate favorable cancer-control in metastatic hormone-sensitive prostate cancer (mHSPC) patients treated with apalutamide, even in patients with high metastatic burden, such as ≥ 10 bone metastases. However, these findings have never been validated in real-world setting. PATIENTS AND METHODS: We relied on the FRAMCAP (FRAnkfurt Metastatic Cancer database of the Prostate) and stratified apalutamide-treated mHSPC patients according to number of bone metastases (≥ 10 vs. < 10). Primary endpoints were time to metastatic castration-resistant

The impact of concomitant therapies on the efficacy and safety of relugolix in advanced prostate cancer.

Relugolix is a once-daily oral gonadotropin-releasing hormone antagonist that was approved by the U.S. Food and Drug Administration in 2020 for the treatment of advanced prostate cancer. Relugolix is commonly co-prescribed with complementary oncologic therapies or agents targeting associated comorbidities (e.g. cardiovascular disease). In this podcast, we review studies which evaluate relugolix with concomitant next-generation hormonal therapies (abiraterone, apalutamide, and enzalutamide), chemotherapy (docetaxel), radiotherapy, and common cardiovascular medications (antihypertensives, antith
