Evolution of thyroid hormone signaling in animals: Non-genomic and genomic modes of action.

Much research has focused on vertebrate thyroid hormone (TH) synthesis and their function in development and metabolism. While important differences in TH synthesis and signaling exist, comparative studies between vertebrates fail to explain the evolutionary origins of this important regulatory axis. For that, one needs to make sense out of the diverse TH effects which have been described in invertebrate phyla but for which a mechanistic understanding is largely missing. Almost every major group of non-vertebrate animals possesses the capability to synthesize and metabolize thyroid hormones an

Rubrolide R: a new furanone metabolite from the ascidian Synoicum of the Indian Ocean.

A new furanone metabolite of the rubrolide family, rubrolide R as diacetate (1), was isolated from a new species of the ascidian Synoicum, besides the known compounds rubrolide A (as diacetate), cadiolide B and prunolide A. The structure of the new rubrolide was elucidated by a study of spectral data. The crude extract and isolated compounds (prunolide A and cadiolide B) showed antiviral activity against the Japanese encephalitis virus. Prunolide A showed cytotoxic activity against breast cancer cell lines at a concentration of < 1 μM.

Effects of tributyltin and other metals on the phenoloxidase activating system of the tunicate, Styela plicata.

Toxic metals, such as tributyltin (TBT), contribute substantially to anthropogenic pollution in many estuarine environments. Animals that live in those environments, particularly invertebrate filter feeders like tunicates, are likely to be exposed to substantial metal contamination. This study investigates the effects of TBT and other metals on the phenoloxidase activity of the estuarine tunicate, Styela plicata, in an effort to identify a biochemical marker of metal pollution. Hemocytes harvested from S. plicata that were exposed to tributyltin or copper in aquaria had significantly enhanced

Evaluating the impact of an increase in sea water temperature on the palmerolides of the Antarctic benthic tunicate Synoicum adareanum.

Global change is producing significant ecological impacts in Antarctic marine organisms, although most of them are still understudied. Here, we report the changes in the chemical profile of the colonial tunicate Synoicum adareanum in response to gradual temperature increases in controlled aquarium experiments as a proxy to determine the effects of global warming. This is the first study on the effects of increasing seawater temperature on the natural products of an Antarctic tunicate. From specimens collected at Livingston Island in the South Shetland Islands in January 2022, we isolated the k

Screening the Irish Marine Biorepository Identifies a New Bryostatin Analog as Potent Inhibitor of Activated B-Cells Diffuse Large B-Cell Lymphoma.

Activated B-cell diffuse large B-cell lymphoma (ABC-DLBCL) is an aggressive cancer with poor response to standard chemotherapy. In search of new therapeutic leads, a library of 435 fractions prepared from the Irish marine biorepository was screened against 2 ABC-DLBCL cell lines (TMD8 and OCI-Ly10) and a non-cancerous control cell line (CB33). Active fractions are prioritized based on potency and selectivity. Among them, fractions from the nudibranch Antiopella cristata and the ascidian Diplosoma listerianum exhibit the most potent and selective cytotoxicity. Bioassay-guided fractionation of D

Evolution and origin of merlin, the product of the Neurofibromatosis type 2 (NF2) tumor-suppressor gene.

BACKGROUND: Merlin, the product of the Neurofibromatosis type 2 (NF2) tumor suppressor gene, belongs to the ezrin-radixin-moesin (ERM) subgroup of the protein 4.1 superfamily, which links cell surface glycoproteins to the actin cytoskeleton. While merlin's functional activity has been examined in mammalian and Drosophila models, little is understood about its evolution, diversity, and overall distribution among different taxa. RESULTS: By combining bioinformatic and phylogenetic approaches, we demonstrate that merlin homologs are present across a wide range of metazoan lineages. While the phy

Targeting microglial dysfunction: The antioxidant potential of and the regulation of microglial responsiveness by dermatan sulfate from a marine invertebrate.

Microglia are the main immune cells of the central nervous system (CNS) and are responsible for maintaining tissue homeostasis. Their chronic activation contributes to neuroinflammation and the progression of neurodegenerative diseases (NDs). Dermatan sulfate (DS) obtained from the ascidian Phallusia nigra (PnDS) exhibits the same sulfation pattern (IdoUA(2S)-GalNAc(6S)) as DS found in neurogenic regions of adult mammals. In the present study, the effects of PnDS on the modulation of murine BV-2 microglial cells subjected to paraquat (PQ)-induced oxidative stress were investigated. Cells were

Developing natural product drugs: Supply problems and how they have been overcome.

The development of natural product-derived drugs has some unique problems associated with the process, which can be best described as the "problem of supply". In this short review, four examples are given demonstrating how the "supply problem" was overcome using as examples the development of Picato® from a plant, Kyprolis® modified from a microbial metabolite, Halaven® a totally synthetic compound based on a marine sponge metabolite and Yondelis® isolated from a marine tunicate and now known to be from an as yet uncultured microbe in the tunicate. The methods used are described in each case a

Metabolic enhancers supporting 1-carbon cycle affect sperm functionality: an in vitro comparative study.

The sperm plasma membrane is a sensitive target to oxidative stress. The most representative reactive oxygen species (ROS) scavengers in the genital tract, hypotaurine and glutathione, require, for their synthesis, cysteine whose availability is associated with the 1-carbon cycle (1-CC). Human, bovine and ascidian spermatozoa were incubated with compounds supporting the 1-CC (Vitamin B6, Methylcobalamin, 5 Methyl Tetrahydrofolate, Zinc Bisglycinate and N-acetyl-cysteine) (TRT) and compared to the effects induced solely by N-acetyl-cysteine (NAC). In control groups (CNTRL), spermatozoa were inc

Localization of symbiotic cyanobacteria in the colonial ascidian Trididemnum miniatum (Didemnidae, Ascidiacea).

Trididemnum miniatum is a colonial ascidian harboring the photosymbiotic prokaryote Prochloron sp. These bacterial cells are located in the tunic of the host animal. The present study revealed, by ultrastructural analysis, that the Prochloron cells were exclusively distributed and proliferated in the tunic. They were shown to be embedded in the tunic matrix and to have no direct contact with ascidian cells. Some tunic cells of the ascidians, however, did phagocytize and digest the symbiont. Round cell masses were sometimes found in the tunic and appeared to consist of disintegrating cyanobacte

The potent anticancer compound ecteinascidin-743 (ET-743) as its 2-propanol disolvate.

Single crystals of the title compound, C(39)H(43)N(3)O(11)S.2C(3)H(8)O, have been obtained from 2-propanol/water solutions. ET-743 belongs to the group of ecteinascidins (ETs), which is a family of novel marine tetrahydroisoquinoline derivatives characterized by a monobridged pentacyclic skeleton. Three large principal planar groups are observed in the three-dimensional structure of the ET-743 molecule, corresponding to three aromatic units which are nearly perpendicular to each other. In the crystal, the methoxy group on the large fused ring system adopts an anti conformation with respect to

Microbial Distortion? Impacts of Delayed Preservation on Microbiome Diversity and Composition in a Marine Invertebrate.

Field collections of marine invertebrates are often accompanied by delays in preservation, which may impact microbiome composition. Here, we tested the effects of delayed preservation and relaxation methods on microbiome diversity and composition in the colonial ascidian Trididemnum solidum using 16S rRNA amplicon sequencing. Replicate samples collected from Belizean reefs were either (1) immediately preserved in ethanol ("control"), (2) held in ambient seawater for 3 h before preservation ("SW"), or (3) held in ambient seawater with menthol (a common pre-preservation relaxation technique for

Marine invertebrates cross phyla comparisons reveal highly conserved immune machinery.

Naturally occurring histocompatibility responses, following tissue-to-tissue allogeneic contacts, are common among numerous colonial marine invertebrate taxa, including sponges, cnidarians, bryozoans and ascidians. These responses, often culminating in either tissue fusions or rejections, activate a wide array of innate immune components. By comparing two allorejection EST libraries, developed from alloincompatible challenged colonies of the stony coral Stylophora pistillata and the ascidian Botryllus schlosseri, we revealed a common basis for innate immunity in these two evolutionary distant

Plant-like mating in an animal: sexual compatibility and allocation trade-offs in a simultaneous hermaphrodite with remote transfer of sperm.

The importance of sexual compatibility between mates has only recently been realized in zoological research into sexual selection, yet its study has been central to botanical research for many decades. The reproductive characteristics of remote mating, an absence of precopulatory mate screening, internal fertilization and embryonic brooding are shared between passively pollinated plants and a phylogenetically diverse group of sessile aquatic invertebrates. Here, we further characterize the sexual compatibility system of one such invertebrate, the colonial ascidian Diplosoma listerianum. All 66

Marine Organisms from the Yucatan Peninsula (Mexico) as a Potential Natural Source of Antibacterial Compounds.

A total of 51 sponges (Porifera) and 13 ascidians (Chordata) were collected on the coast of the Yucatan Peninsula (Mexico) and extracted with organic solvents. The resulting extracts were screened for antibacterial activity against four multidrug-resistant (MDR) bacterial pathogens: the Gram-negative Acinetobacter baumannii, Klebsiella pneumoniae, and Pseudomonas aeruginosa and the Gram-positive Staphylococcus aureus. The minimum inhibitory concentrations (MICs) of the organic extracts of each marine organism were determined using a broth microdilution assay. Extracts of eight of the species,

Multi-stage single-cell atlas of a chordate non-embryonic development unveils dedifferentiating bud founder cells.

Colonial tunicates are the only chordates capable of forming fully functional bodies from somatic tissues through non-embryonic development, or budding. In Botryllus schlosseri, budding produces new individuals (zooids) from a portion of the peribranchial epithelium of a preexisting zooid. While the anatomy of this process has been described in detail, the cellular and molecular mechanisms underpinning bud initiation remain elusive. Here, we present a multi-stage single-cell transcriptomic atlas of a non-embryonic development in a chordate, capturing initiation and development of zooids. We id

Commensal oerstediid nemerteans from deep-sea styelid ascidians.

We report a new commensal relationship between oerstediid nemerteans and deep-sea ascidians. A total of six nemertean individuals were collected from the peribranchial cavities of several Styelidae ascidians attached to small rocks at bathyal depths (1,400-1,409 m) off northeastern Japan in the northwestern Pacific Ocean. Phylogenetic analyses based on combined mitochondrial cytochrome c oxidase subunit I (COI), 16S rRNA, histone H3, 18S rRNA, and 28S rRNA gene sequences placed this species within the 'Clade Gononemertes' of the hoplonemertean family Oerstediidae.
