Implications of Modest Reductions in ABI Measurements for Risk of Major Adverse Limb Events Among 223,350 Veterans.

BACKGROUND: Low resting ankle-brachial index (ABI) is a marker of poor cardiovascular outcomes. Whether modest decrements in ABI translate into higher risks of major adverse limb events (MALE) is unclear because prior studies are small or lack racial diversity and incident outcomes. OBJECTIVES: The purpose of this study was to assess the association between the full range of ABI measures and incident MALE in a large, diverse cohort. METHODS: Using data from the Veterans Aging Cohort Study-National Cohort, we analyzed a prospective, longitudinal cohort of veterans free of prevalent peripheral

Infant gut microbiome composition correlated with type 1 diabetes acquisition in the general population: the ABIS study.

AIMS/HYPOTHESIS: While autoantibodies are traditional markers for type 1 diabetes development, we identified gut microbial biomarkers in 1-year-old infants associated with future type 1 diabetes up to 20 years before diagnosis. METHODS: Infants enrolled in the longitudinal general population cohort All Babies In Southeast Sweden (ABIS) provided a stool sample at a mean age of 12.5 months. Samples (future type 1 diabetes, n=16; healthy controls, n=268) were subjected to 16S ribosomal RNA (rRNA) sequencing and quantitative PCR. Microbial differences at the taxonomic and core microbiome levels w

Associations of ankle-brachial index (ABI) with cerebral arterial disease and vascular events following ischemic stroke.

OBJECTIVES: Low ankle-brachial index (ABI), indicative of peripheral arterial disease (PAD), is a risk factor for stroke. ABI has been shown to be associated with cerebral arterial disease and prognosis following stroke. We studied the associations of the degree of ABI lowering with extracranial carotid disease (ECD), intracranial large artery disease (ICLAD), and subsequent vascular events in a prospective cohort of acute ischemic stroke patients. METHODS: ABI, extracranial and intracranial cerebral arteries were assessed in a blinded manner. ABI was categorized into 0.9-1.3 (normal), 0.8-0.

The golgi comprises a paired stack that is separated at G2 by modulation of the actin cytoskeleton through Abi and Scar/WAVE.

During the cell cycle, the Golgi, like other organelles, has to be duplicated in mass and number to ensure its correct segregation between the two daughter cells. It remains unclear, however, when and how this occurs. Here we show that in Drosophila S2 cells, the Golgi likely duplicates in mass to form a paired structure during G1/S phase and remains so until G2 when the two stacks separate, ready for entry into mitosis. We show that pairing requires an intact actin cytoskeleton which in turn depends on Abi/Scar but not WASP. This actin-dependent pairing is not limited to flies but also occurs

Abi, Sra1, and Kette control the stability and localization of SCAR/WAVE to regulate the formation of actin-based protrusions.

BACKGROUND: In animal cells, GTPase signaling pathways are thought to generate cellular protrusions by modulating the activity of downstream actin-regulatory proteins. Although the molecular events linking activation of a GTPase to the formation of an actin-based process with a characteristic morphology are incompletely understood, Rac-GTP is thought to promote the activation of SCAR/WAVE, whereas Cdc42 is thought to initiate the formation of filopodia through WASP. SCAR and WASP then activate the Arp2/3 complex to nucleate the formation of new actin filaments, which through polymerization exe

Assessment of Liver and Kidney Safety of Silver Fir (Abies alba) Branch Extract: An Open-Label Human Study of a Dietary Supplement.

Extracts of silver fir (Abies alba) branches are rich in polyphenolic compounds and are increasingly used as dietary supplements. Despite their growing consumption, systematic human data on their safety remain limited. This study evaluated the effects of a standardized silver fir branch extract on biochemical markers of liver and kidney function in healthy adults. An open-label, single-arm human study was conducted in 15 healthy volunteers (eight men and seven women; mean age 48 years). Following a two-week comparison period, participants consumed 900 mg/day of a standardized silver fir branch

Sprouts and Needles of Norway Spruce (Picea abies (L.) Karst.) as Nordic Specialty-Consumer Acceptance, Stability of Nutrients, and Bioactivities during Storage.

Developing shoots, i.e., sprouts, and older needles of Norway spruce (Picea abies (L.) Karst.) have traditionally been used for medicinal purposes due to the high content of vitamins and antioxidants. Currently, sprouts are available as, for example, superfood and supplements. However, end-product quality and nutritive value may decline in the value-chain from raw material sourcing to processing and storage. We studied (1) impacts of different drying and extraction methods on nutritional composition and antioxidative properties of sprouts and needles, (2) differences between sprouts and needle

Manganese deficiency of silver fir trees (Abies alba) at a reforested site in the Jura mountains, Switzerland: aspects of cause and effect.

We examined manganese uptake and translocation in 30-year-old silver fir trees (Abies alba Mill.) showing severe symptoms of needle chlorosis by analyzing both xylem and phloem sap of shoots and an extract of root sap originating from both xylem and phloem elements. Manganese concentrations in shoot xylem and phloem sap were significantly lower in chlorotic trees than in neighboring healthy trees. The Mn concentration of root sap was also lower in these Mn-deficient trees than in healthy trees, indicating reduced Mn uptake by Mn-deficient trees. Although Mn deficiency had no effect on the Mn c

The influence of ozone and acid mist on the amount and wettability of the surface waxes in Norway spruce [Picea abies (L.) Karst.].

Three-year old Norway spruce [Picea abies (L.) Karst.] trees from two clones (4076 and 4102) of a medium altitude southern German provenance were transplanted in May 1986 into an acid brown earth soil. During the summers of 1987 and 1988 the trees were exposed in large-scale fumigation chambers (solardomes) to ozone at 100 or 20 nl 1-1 (control). Acid (pH 3.6) or control mist (pH 5.5) was applied daily to trees. The wettability of current and 1-year-old needles was assessed in November 1988 by measuring the contact angle of water droplets placed on needles, and the amount of surface wax was de

Risk Factors Associated With Diabetic Foot Ulcers and Its Relationship With ABI and Brachial-Ankle Pulse Wave Velocity.

To evaluate the risk factors associated with diabetic foot ulcers (DFUs) and to analyze the relationship of DFUs and the value of Ankle-Brachial Index (ABI) and brachial-ankle pulse wave velocity (baPWV). In this retrospective study, the risk factors associated with DFUs were analyzed, and the value of ABI and baPWV were measured to find its relationship with DFUs. Binary logistic regression analysis indicated that neuropathy and ABI were independent risk factors for DFUs. The patients were divided into 2 groups according to the value of ABI. For patients with DFUs, the value of baPWV decrease

Rac interacts with Abi-1 and WAVE2 to promote an Arp2/3-dependent actin recruitment during chlamydial invasion.

Chlamydiae are Gram-negative obligate intracellular pathogens to which access to an intracellular environment is fundamental to their development. Chlamydial attachment to host cells induces the activation of the Rac GTPase, which is required for the localization of WAVE2 at the sites of chlamydial entry. Co-immunoprecipitation experiments demonstrated that Chlamydia trachomatis infection promoted the interaction of Rac with WAVE2 and Abi-1, but not with IRSp53. siRNA depletion of WAVE2 and Abi-1 abrogated chlamydia-induced actin recruitment and significantly reduced the uptake of the pathogen

Shifts in resource allocation and aggravation of foliage development restrict the growth rate of Picea abies under increasing atmospheric humidity at high latitudes.

Global warming is accompanied by rising precipitation, atmospheric water vapour content, and specific humidity at high latitudes. The rising amount and frequency of rainfall increase the air relative humidity (RH) on a local scale, especially within forest canopies. We studied the effects of artificially elevated environmental humidity (RH and soil moisture) on leaf gas exchange, stomatal responses and growth of young Picea abies trees at the Free Air Humidity Manipulation site in eastern Estonia. Manipulation did not affect the net assimilation rate (An) but affected the stomatal responses, n

Screening of the antioxidant and antibacterial effects of extracted essential oils from Thunbergia coccinea, Acacia polyacantha, Polygonum micrpcephallum, Abies spectabilis and Clerodendrum colebrookianum.

During the previous few decades, it has been seen that there is a rapid emergence of pathogens resistant to multiple antibiotics. This has now become a global crisis. Some unexplored or less explored plants also provide some antibacterial, bactericidal and antioxidant properties. The antibacterial, bactericidal effects of extracted essential oils (EEOs) of Thunbergia coccinea, Acacia polyacantha, Polygonum micrpcephallum, Abies spectabilis and Clerodendrum colebrookianum was tested in comparison with standard antibiotics. The methods chosen were disc diffusion and deduction of minimum inhibito
