The emergence of multidrug-resistant (MDR) strains is a major health problem worldwide. There is an urgent need for novel strategies to combat bacterial infections caused by MDR strains like Pseudomonas aeruginosa and Acinetobacter baumannii. Quorum sensing (QS) is a critical communication system in bacterial community controlling survival and virulence. The awareness of the importance of QS in bacterial infections has stimulated research to identify QS inhibitors (QSIs) to defeat microbes. In this study, four FDA-approved drugs (besides azithromycin as positive QSI) were tested for potential

The rise and global dissemination of multidrug-resistant (MDR) Acinetobacter baumannii has become a major public health threat. However, the constant emergence of drug-resistant mutants has resulted in increasingly limited therapeutic options. Tigecycline (TGC) continues to be one of the few antibiotics still effective against many A. baumannii strains. Herein, we discovered that quercetin (Qu) synergizes with TGC, enhancing its therapeutic efficacy against MDR A. baumannii by reducing the minimum inhibitory concentration (MIC) by 4- to 32-fold. Mechanistic analysis revealed that the synergist

Carbapenem-resistant Acinetobacter baumannii (CRAB), an important opportunistic pathogen, is a major cause of healthcare-associated infections. The polymyxins (colistin and polymyxin B) are the last line of defense in the treatment of CRAB infections, and there is an urgent need to develop novel alternative therapeutic strategies. In this study, we found that the antimicrobial peptide DvAMP exhibited satisfactory antibacterial and antibiofilm activity against CRAB. In addition, DvAMP showed tolerable stability in salt ions and serum and exhibited low toxicity in vivo. Investigation of the unde

Acinetobacter baumannii is an opportunistic Gram-negative bacterium representing one of the leading causes of ventilator-associated pneumonia. The development of pneumonia results from a complex interplay between pathogens and pulmonary innate mucosal immunity. Therefore, the knowledge of the host immune responses is pivotal for the development of effective therapeutics to treat A. baumannii infections. Previous studies were conducted using cell lines and animal models, but a comprehensive understanding of the interaction between A. baumannii and primary human immune cells is still lacking. To

In recent years, the bloodstream infection rate of Gram-negative bacilli has continued to increase. Among them, drug-resistant bacteria have a higher mortality rate and longer hospital stay, especially the bloodstream infection of carbapenem-resistant Acinetobacter baumannii (CRAB). Polymyxin began to be used clinically in the 1950s and has antibacterial activity against multidrug resistant and poly drug-resistant Gram-negative bacilli. It can also be used as an effective permeation agent for the cell envelope of Gram-negative bacilli. Polymyxin is reserved for microbiologically clear drug-res

Colonization of wounds with multidrug-resistant organisms is a difficult orthopedic problem. Acinetobacter infections are especially difficult because they are resistant to all currently available antibiotics. We present the use of a novel skin sanitizer, Stay Byotrol Clean (Byotrol Inc, Spartanburg, South Carolina), to treat a multidrug-resistant wound infection. A 31-year-old T10 paraplegic man presented with chronic bilateral stage IV decubitus trochanteric ulcers. Cultures grew methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococcus. The ulcers were initially trea

BACKGROUND: Acinetobacter baumannii is usually associated with nosocomial pneumonia or bacteremia. Reports of A. baumannii skin and soft-tissue infection (SSTI) are uncommon.
METHODS: We performed a retrospective review of 57 inpatients admitted to a naval hospital ship and identified 8 patients with A. baumannii-associated SSTI. Demographic and clinical characteristics were compared between these patients and 49 patients with A. baumannii infections that were not SSTIs. We also reviewed 18 cases of A. baumannii-associated SSTI from the literature.
RESULTS: Our 8 cases of A. baumannii-associ

The overuse of antibiotics has led to antibiotic-resistant Acinetobacter baumannii. This study evaluated the protective effect of a recombinant protein, derived from Omp34's loop 3 repeats, as a potential vaccine candidate against A. baumannii in a murine sepsis model. Recombinant Omp34 and L3 × 5 proteins were expressed in E. coli BL21 using the autoinduction method. The proteins were then purified and verified. To assess their protective efficacy, the proteins were injected into BALB/c mice, and the vaccine's effectiveness in both prevention and treatment stages was evaluated in a murine sep

INTRODUCTION: International guidelines recommend definitive combination antibiotic therapy for the management of serious infections involving carbapenem-resistant Acinetobacter (CRAB) species. The commonly available combination options include high-dose sulbactam, polymyxins, tetracyclines, and cefiderocol. Scanty prospective data exist to support this approach.
METHODS: Patients with CRAB bacteraemia, ventilator-associated pneumonia (VAP), or both were categorized based on whether they received combination therapy or monotherapy. The 30-day mortality was compared between the two groups. Inve
