Dearomative [2 + 1] Spiroannulation of Bromophenols with Electron-Deficient Alkenes.

A base-assisted dearomative [2 + 1] spiroannulation of p/o-bromophenols with activated olefins (methylenemalonates) to construct various cyclopropyl spirocyclohexadienone skeletons is reported. Furthermore, several other halophenols (X = Cl, I) were also tolerated in this process. Control experiments reveal a dearomative Michael addition of phenols at their halogenated positions to methylenemalonates, followed by intramolecular radical-based SRN1 dehalogenative cyclopropanation. However, according to the density functional theory (DFT) calculations, an SN2 dehalogenative cyclopropanation with

Nickel-Catalyzed Reductive Cascade Arylalkylation of Alkenes with Alkylpyridinium Salts.

Herein, we describe a nickel-catalyzed reductive deaminative arylalkylation of tethered alkenes with pyridinium salts as C(sp3) electrophiles. This two-component dicarbofunctionalization reaction enables the efficient synthesis of various benzene-fused cyclic compounds bearing all-carbon quaternary centers. The approach presented in this paper proceeds under mild conditions, tolerating a wide variety of functional groups and heterocycles. It has been used to functionalize complicated molecules at a late stage.

Electrochemical Oxidative Difunctionalization of Alkenes to Access α-Oxygenated Ketones.

Dioxygenation of alkenes was developed by the combination of electrochemical synthesis and aerobic oxidation, leading to easy accessibility of α-oxygenated ketones in an eco-friendly fashion. Using air as the oxygen source and the absence of transition metals were the critical features of this protocol. A wide range of alkenes and N-hydroxyimides were found to be compatible and provided α-oxygenated ketones in moderate to high yields.

Copper-Catalyzed 1,2-Aminocyanation of Unactivated Alkenes via Cyano Migration.

A copper-catalyzed aminocyanation of alkenes has been achieved through distal cyano migration using O-benzoylhydroxylamines and N-fluorobenzenesulfonimides. This method offers a rapid approach to generate diverse β-amino and β-sulfonimido nitriles. These reactions feature mild conditions, tolerance of sensitive functional groups, and excellent regioselectivity. Mechanistic studies suggest that these transformations are initiated by a copper-catalyzed amination step followed by a cyano migration step.

BCl3-mediated ene reaction of sulfur dioxide and unfunctionalized alkenes.

The first ene reactions of SO(2) and unfunctionalized alkenes are reported. Calculations suggest that the endergonic ene reactions of SO(2) with alkenes can be used to generate beta,gamma-unsaturated sulfinyl and sulfonyl compounds. Indeed, in the presence of one equivalent of BCl(3), the unstable sulfinic acid form stable sulfinic acid.BCl(3) complexes that can be reacted in situ with NCS to generate corresponding sulfonyl chlorides, or with a base to generate corresponding sulfinates. The latter can be reacted with electrophiles to generate sulfones, or with silyl chloride to form beta, gamma

Visible-Light-Induced Difunctionalization of Alkenes with Masked Trifluoroacetyl Reagents.

Herein, a trifluoroacetylation difunctionalization reaction of electron-deficient alkenes has been developed via a visible-light catalytic strategy. Initially, the trifluoroacetylation allylation of alkenes was realized by synergistic photoredox and palladium catalysis. Mechanistic investigations revealed that this transformation proceeds sequentially through radical addition, reduction, and substitution processes. Furthermore, trifluoroacetylation amination of alkenes was further achieved via an energy-transfer pathway. Both protocols feature mild reaction conditions, broad substrate scope, a

Electrochemical Cobalt/Vanadium Relay Catalysis: Enantioselective Markovnikov Hydrooxygenation of Alkenes.

The direct enantioselective intermolecular Markovnikov hydrooxygenation of alkenes remains a fundamental challenge, as it requires simultaneous control over regioselectivity, stereochemistry, and reactivity. Here we report an electrochemical cobalt/vanadium relay catalytic strategy that overcomes these constraints by decoupling regio- and stereochemical control across two distinct catalytic events. A cobalt hydride selectively engages alkenes through metal-hydride hydrogen atom transfer to generate a Markovnikov alkyl radical, while a vanadium catalyst mediates a subsequent stereodetermining b

Copper-Catalyzed Selective Amino-alkoxycarbonylation of Unactivated Alkenes with CO.

1,2-Amino-difunctionalization reactions of alkenes allow the efficient introduction of different functional groups and the rapid construction of valuable functionalized amines. In this respect, we report a copper-catalyzed 1,2-amino-alkoxycarbonylation of unactivated alkenes with CO and alkylamine precursors in the presence of a Lewis acid additive. The novel protocol allows direct access to valuable β-amino acid derivatives from easily available starting materials. The presented methods feature high chemo- and regioselectivities, good functional group tolerance, and substrate scope including

Cu-catalyzed photoredox chlorotrifluoromethylation of polysubstituted alkenes and pharmacological evaluation.

A visible-light-mediated chlorotrifluoromethylation catalyzed by a copper-based photo-redox catalyst of internal alkenes is reported. The reaction proceeds with complete regioselectivity under mild reaction conditions using commercially available F3CSO2Cl as both the trifluoromethyl and chlorine source, leading to the synthesis of added-value chemicals with atom-economy. A vast array of internal alkenes were functionalized in decent to good yields, highlighting a great tolerance to various functional groups. Copper-catalyzed radical-mediated intermolecular cyanomonofluoroalkylation of alkenes and alkynes.

A novel copper-catalyzed three-component reaction of monofluoroalkylation agents, alkenes and TMSCN is described. In addition, alkynes could also be compatible with the reaction system to obtain three-component products for the first time with moderate yield and excellent E/Z stereoselectivity. This reaction provides a facile method for the synthesis of cyanomonofluoroalkyl compounds, which may serve as potentially useful organic intermediates for further transformations. Preliminary mechanistic investigation indicated that monofluoroalkyl free radicals were involved in the cyanomonofluoroalky

Photoredox cooperative N-heterocyclic carbene/palladium-catalysed alkylacylation of alkenes.

Three-component carboacylation of simple alkenes with readily available reagents is challenging. Transition metal-catalysed intermolecular carboacylation works for alkenes with strained ring or directing groups. Herein, we develop a photoredox cooperative N-heterocyclic carbene/Pd-catalysed alkylacylation of simple alkenes with aldehydes and unactivated alkyl halides to provide ketones in good yields. This multicomponent coupling reaction features a wide scope of alkenes, broad functional group compatibility and free of exogenous photosensitizer or external reductant. In addition, a series of

Reduction of Activated Alkenes by PIII /PV Redox Cycling Catalysis.

The carbon-carbon double bond of unsaturated carbonyl compounds was readily reduced by using a phosphetane oxide catalyst in the presence of a simple organosilane as the terminal reductant and water as the hydrogen source. Quantitative hydrogenation was observed when 1.0 mol % of a methyl-substituted phosphetane oxide was employed as the catalyst. The procedure is highly selective towards activated double bonds, tolerating a variety of functional groups that are usually prone to reduction. In total, 25 alkenes and two alkynes were hydrogenated to the corresponding alkanes in excellent yields o

Controllable, Sequential, and Stereoselective C-H Allylic Alkylation of Alkenes.

The direct conversion of C-H bonds into new C-C bonds represents a powerful approach to generate complex molecules from simple starting materials. However, a general and controllable method for the sequential conversion of a methyl group into a fully substituted carbon center remains a challenge. We report a new method for the selective and sequential replacement of three C-H bonds at the allylic position of propylene and other simple terminal alkenes with different carbon groups derived from Grignard reagents. A copper catalyst and electron-rich biaryl phosphine ligand facilitate the formatio

Diastereo- and Enantioselective CuH-Catalyzed Hydroamination of Strained Trisubstituted Alkenes.

Amine-substituted cyclobutanes and cyclopropanes are important substructures in biologically active compounds. Moreover, many of the cycloalkane units bear multiple substituents and stereocenters. Therefore, synthetic methods that produce polysubstituted aminocyclobutanes and aminocyclopropanes in a highly diastereo- and enantioselective manner are of importance. Herein, we describe the diastereo- and enantioselective synthesis of various types of polysubstituted aminocyclobutanes and aminocyclopropanes through CuH-catalyzed hydroamination of 1-substituted cyclobutenes and cyclopropenes. These

Diastereoselective intramolecular oxidopyrylium-alkene [5 + 2]-cycloaddition of substrates with a β-chiral center on alkene tethers: synthesis of 8-oxabicyclo-[3.2.1]-octenone heterocycles.

We investigated the asymmetric intramolecular oxidopyrylium-alkene [5 + 2]-cycloaddition reaction of substrates bearing tethered alkenes containing β-chiral centers. The effect on the cycloadduct diastereoselectivity was examined for carbon chain tethered alkenes or alkoxy tethered alkenes with varying chain lengths that can form 5-, 6- and 7-membered rings. We also probed the effect of varying steric bulk on the β-chiral center substituents on cycloadduct diastereoselectivity. Various substrates with carbon chain tethered alkenes were synthesized using the Lewis acid mediated conjugate additi

Oximophosphinothiolation of Alkenes with Sodium Nitrite, Elemental Sulfur, and Secondary Arylphosphines.

A simple and efficient oximophosphinothiolation of alkenes with sodium nitrite, elemental sulfur, and secondary arylphosphines has been developed. This transformation can be carried out under mild and additive-free conditions, employing NaNO2 and S8 as the NO source and sulfur source, respectively. A wide variety of α-dithiophosphinate oximes could be accessed via this method in moderate to good yields, and the reaction showed good tolerance to diverse functional groups.

Visible-Light Photoredox-Catalyzed Reductive Coupling of Heteroaryl Chlorides with Alkenes.

We have developed a photocatalytic reductive system for the coupling of heteroaryl chlorides with alkenes, wherein the alkenes act as alkylating reagents. The mechanism involves two key single-electron reduction steps: first, activation of the C-Cl bond; and second, reduction of the resulting benzylic radical to a benzylic carbanion. This strategy enables efficient activation and alkylation of aryl C-Cl bonds.

Organophotoredox-Catalyzed Carbamoylative Carboxylation of Alkenes at Ambient Pressure of CO2.

Owing to the inert nature and limited mass transfer of gaseous CO2 into the biphasic reaction medium, the direct incorporation of CO2 into complex molecular architectures at ambient temperature and pressure remains a daunting challenge. Here, the study reports a transition metal-free organophotoredox-catalyzed three-component difunctionalization of alkenes that integrates carbamoyl radicals with CO2 fixation under visible light irradiation via a reductive radical-polar crossover (RRPCO) mechanism. Remarkably, oxamic acids serve as bifunctional reagents, and the protocol operates under mild con

PhSO2CF2H as a difluoromethylene bridge connecting electrophiles and electronically diverse alkenes.

The difluoromethylene (-CF2-) motif is highly valuable in pharmaceuticals, agrochemicals, and materials science due to its unique physicochemical properties. Herein, we present a broadly applicable strategy for the modular synthesis of CF2-containing compounds by using difluoromethylene to bridge electrophiles and electronically diverse alkenes, with difluoromethyl phenyl sulfone (PhSO2CF2H) as the difluoromethylene source. Central to this approach is the activation of (phenylsulfonyl)difluoromethylated derivatives (PhSO2CF2R) by the excited-state Hantzsch ester anion (HE-), which enables effi

Ligand-Promoted Copper-Catalyzed Radical Relay for Alkyl-Alkynylation of Alkenes.

The alkynylation of alkenes provides an efficient platform for synthesizing compounds containing C≡C bonds. However, the intermolecular alkynylation of alkenes has been largely restricted to precious metal palladium-catalyzed systems, limiting reaction versatility. Herein, we report a copper-catalyzed radical relay strategy for intermolecular alkyl-alkynylation of alkenes and construction of challenging C(sp3)-C(sp3) bonds and C(sp3)-C(sp) bonds in one pot. This method utilizes alkene-tethered α-carbonyl bromides as the alkyl source, which undergo single-electron transfer to generate alkyl rad
