Whole-Genome sequencing and phylogenomic analyses of Alternaria japonica and A. telliensis (Section Japonicae).

Species of Alternaria are necrotrophic fungi responsible for leaf spot diseases on a wide range of cultivated and wild plants. Species delimitation within this genus, however, often relies on morphological features and a limited set of molecular markers, which can constrain phylogenetic resolution at finer taxonomic scales. This is the case for Alternaria telliensis, a species recently described from Algeria and subsequently reported in Iran, whose relationships within section Japonicae require clarification. To provide a higher resolution framework for this species, we performed whole-genome

Critical role of AaTgl1 lipase/hydrolase in virulence and lesion formation in Alternaria alternata.

Lipases and hydrolases play important roles in diverse biological processes in fungi. Their roles in virulence in the phytopathogenic fungus Alternaria alternata remain poorly understood. The AaTgl1 protein of A. alternata is a putative lipase/hydrolase characterized by a triacylglycerol lipase domain, an α/β-hydrolase superfamily domain, and a catalytic triad (Ser353, Asp522, His548) within the conserved GxSxG motif. Sequence analysis revealed only 64-68% similarity to orthologs from other filamentous fungi, suggesting a specialized role in lipid metabolism that may contribute to the ecologic

Invasive phaeohyphomycosis co-infection with Alternaria spp. and Curvularia spp. in a neutropenic host.

Phaeohyphomycoses are infections caused by dark-walled dematiaceous fungi. Alternaria and Curvularia are two genera of dematiaceous molds known to cause invasive fungal rhinosinusitis, particularly in immunocompromised patients. Co-infection with two dematiaceous fungi is rarely reported in the literature. This report describes a case of biopsy proven invasive fungal rhinosinusitis with Alternaria spp. and Curvularia spp. co-infection in a neutropenic host. The infection characteristics, microbiologic findings, and treatment are described.

Alternaria fungus growing on top of cyanoacrylate glue in a patient with perforated corneal ulcer.

PURPOSE: To demonstrate a case where Alternaria fungus grew on top of cyanoacrylate glue used to seal a perforated corneal ulcer. OBSERVATIONS: We document the clinical course of a rare case of Alternaria keratitis over the course of 6 months. Despite the purported antifungal properties of cyanoacrylate glue demonstrated in vitro, this case provides in vivo evidence that this substance can serve as a scaffold on which pathogenic fungi may grow. CONCLUSION: This report demonstrates the importance of close follow up of patients with corneal glue patches in place. Ophthalmologists should contin

Morphological, Molecular, and Pathogenic Characterization of Alternaria Associated with Apple Fruit Rot in China.

Alternaria fruit rot is one of the most prevalent postharvest diseases of apples in China. To clarify the species associated with this disease, we characterized Alternaria isolates from symptomatic fruit using morphological traits and multi-gene phylogenetic analyses, and further assessed their pathogenicity on apple. A total of 373 isolates were obtained from decayed apples collected in major production and storage regions, including Shandong, Shaanxi, Gansu, and Beijing. Based on colony characteristics and sporulation patterns, isolates were assigned to two groups resembling A. alternata and

In silico evaluation of isochlorogenic acid as a potential AbPf2 inhibitor for managing Alternaria pathogenicity.

BACKGROUND: Alternaria species cause significant agricultural losses through necrotrophic pathogenesis, regulated by the AbPf2 transcription factor during early infection stages. This pivotal regulator controls virulence initiation and host-pathogen interactions, presenting an ideal antifungal target. Bambusa balcooa leaf extract, rich in bioactive flavonoids, offers untapped potential for discovering natural inhibitors against such fungal transcription factors. OBJECTIVE: This study employs an integrated in silico approach to screen and characterize LC-MS profiled phytocompounds of Bambusa b

An overview of possible pathogenesis mechanisms of Alternaria alternata in chronic rhinosinusitis and nasal polyposis.

Chronic Rhinosinusitis (CRS) is a multifactorial disease, and different etiologies like metabolism and immunity disorders, bacterial superantigens, biofilms, and fungal allergens are known to develop this disease, especially the CRS with nasal polyps. Alternaria alternata (Alternaria) is one of the most prevalent airborne fungal species in the nasal discharge, which might have vigorous immunologic activities in nasal epithelial cells and play an essential role in the pathogenesis of CRS. Moreover, the interaction between this fungus and the innate and adaptive immune systems leads to the devel

Tenosynovitis caused by Scedosporium apiospermum infection misdiagnosed as an Alternaria species: a case report.

BACKGROUND: Scedosporium apiospermum, which can usually be isolated from soil, polluted stream water and decaying vegetation, is increasingly recognized as an opportunistic dematiaceous fungus. The mortality rate of infection in immunocompromised hosts is over 50%. S. apiospermum is commonly responsible for dermal and epidermal infections (i.e., mycetoma) after traumatic penetration. CASE PRESENTATION: A 73-year-old woman was admitted to our hospital complaining of painful swelling and tenderness on the dorsum of the proximal left wrist and hand. The symptoms had persisted for approximately 2

Expression of Alt a 1 allergen from Alternaria alternata in the yeast Yarrowia lipolytica.

Allergies affect almost 25% of the population in industrialized countries. Alternaria alternata is known to be a significant source of aeroallergens and sensitization to this mold is a risk factor for the development of wheezing, asthma, and atopic dermatitis. Diagnosis and treatment of allergies requires the production of large amounts of pure and well defined protein. Yarrowia lipolytica, a non-pathogenic ascomycete able to secrete high levels of enzymes that can grow in inexpensive substrates, has been considered a useful host for heterologous gene expression. In the present work, we have d

A role for measurement of nasal IgE antibodies in diagnosis of Alternaria-induced rhinitis in children.

BACKGROUND: Rhinitis is a very common disease, frequently caused by sensitisation to inhalant allergens. Negative results from skin prick tests (SPT) and in vitro IgE tests generally lead to a diagnosis of non-allergic rhinitis. However, it is possible, as indicated by studies addressed with dust mites or pollens that the production of specific IgE occurs exclusively at nasal level. METHODS: We measured specific nasal IgE in children suffering from rhinitis in the periods when Alternaria spores were present in the air. All subjects underwent SPT with a standard panel of aeroallergens (Staller
