Free-living amoebae in an oil refinery wastewater treatment facility.

Free Living Amoebae (FLA) are ubiquitous microorganisms reported from harsh environmental conditions. Oil refinery facilities consume vast volumes of water during their processes, generating a large amount of wastewater. The present study aimed to evaluate the wastewater treatment process in an oil refinery wastewater treatment facility (ORWWTF) for the presence of FLA. Water samples were collected from an oil refinery wastewater (ORWW) for nine months. After recording physical-chemical features, samples were cultivated onto non-nutrient agar (NNA). The discriminative fragments of the ribosoma

Novel Azoles as Antiparasitic Remedies against Brain-Eating Amoebae.

Balamuthia mandrillaris and Naegleria fowleri are opportunistic protozoan pathogens capable of producing infection of the central nervous system with more than 95% mortality rate. Previously, we have synthesized several compounds with antiamoebic properties; however, synthesis of compounds that are analogues of clinically used drugs is a highly desirable approach that can lead to effective drug development against these devastating infections. In this regard, compounds belonging to the azole class possess wide range of antimicrobial properties and used clinically. In this study, six novel benz

Lectins as virulence factors in Entamoeba histolytica and free-living amoebae.

Currently, there is growing interest in the identification and purification of microbial lectins due to their involvement in the pathogenicity mechanisms of pathogens, such as Entamoeba histolytica and free-living amoebae. The Gal/GalNAc lectin from E. histolytica participates in adhesion, cytotoxicity and regulation of immune responses. Furthermore, mannose- and galactose-binding protein have been described in Acanthamoeba castellanii and Balamuthia mandrillaris, respectively and they also contribute to host damage. Finally, in Naegleria fowleri, molecules containing mannose and fucose are im

A hidden battle in the dirt: Soil amoebae interactions with Paracoccidioides spp.

Paracoccidioides spp. are thermodimorphic fungi that cause a neglected tropical disease (paracoccidioidomycosis) that is endemic to Latin America. These fungi inhabit the soil, where they live as saprophytes with no need for a mammalian host to complete their life cycle. Despite this, they developed sophisticated virulence attributes allowing them not only to survive in host tissues but also to cause disease. A hypothesis for selective pressures driving the emergence or maintenance of virulence of soil fungi is their interaction with soil predators such as amoebae and helminths. We evaluated t

Brain-Eating Amoebae: Predilection Sites in the Brain and Disease Outcome.

Acanthamoeba spp. and Balamuthia mandrillaris are causative agents of granulomatous amoebic encephalitis (GAE), while Naegleria fowleri causes primary amoebic meningoencephalitis (PAM). PAM is an acute infection that lasts a few days, while GAE is a chronic to subacute infection that can last up to several months. Here, we present a literature review of 86 case reports from 1968 to 2016, in order to explore the affinity of these amoebae for particular sites of the brain, diagnostic modalities, treatment options, and disease outcomes in a comparative manner.

Identification of Pif1 helicases with novel accessory domains in various amoebae.

Pif1 helicases are a conserved family of eukaryotic proteins involved in the maintenance of both nuclear and mitochondrial DNA. These enzymes possess a number of known and putative functions, which facilitate overall genome integrity. Here we have identified multiple subtypes of Pif1 proteins in various pathogenic and non-pathogenic amoeboid species which possess additional domains not present in other Pif1 helicases. These helicases each possess one of five different accessory domains, which have roles in ubiquitination, origin of DNA replication recognition or single-stranded nucleic acid bi

Legionella pneumophila pathogesesis: a fateful journey from amoebae to macrophages.

Legionella pneumophila first commanded attention in 1976, when investigators from the Centers for Disease Control and Prevention identified it as the culprit in a massive outbreak of pneumonia that struck individuals attending an American Legion convention (). It is now clear that this gram-negative bacterium flourishes naturally in fresh water as a parasite of amoebae, but it can also replicate within alveolar macrophages. L. pneumophila pathogenesis is discussed using the following model as a framework. When ingested by phagocytes, stationary-phase L. pneumophila bacteria establish phagosome

Free-living amoebae protecting Legionella in water: the tip of an iceberg?

Bacteria are a main food source for free-living amoebae inhabiting aquatic systems. Some bacteria however, have the ability to prevent intracellular destruction and can survive and grow in amoebic cells as endosymbionts. Free-living amoebae are well adapted to their hostile environmental conditions and are resistant to both desiccation, elevated temperatures and various disinfectants. For their endosymbionts, amoebae represent perfect vectors, providing both protection against adverse environmental conditions and transportation. There is increasing interest in the potential role of free-living

Contribution to the problem of the so-called nonpathogenic amoebae in the intestine of man.

Among 10,418 patients of a Prague hospital, a plain infection with intestinal parasitic protozoans was identified in 1,319 persons (12.7%). Of these, 3.5% were infested with Giardia intestinalis, 0.3% with Entamoeba histolytica forma minuta, 5.7% with Endolimax nana. We evaluated the frequency of findings of protozoans in various clinical diagnoses. A statistically significant increase in frequency was recorded for E. nana in diagnoses of eosinophilia, giardiasis, amoebiasis and helminthiasis. A slight increase above the average was found for Entamoeba coli in diagnoses of giardiasis and helmi

Synthesis and Evaluation of Benzylamine Inhibitors of Neuropathogenic Naegleria fowleri "Brain-Eating" Amoeba.

Current therapy for primary amoebic meningoencephalitis (PAM), a highly lethal brain infection in humans caused by Naegleria fowleri amoeba, is restricted to repurposed drugs with limited efficacy and success. Discovery of an antiamoebic benzylamine scaffold 2 precipitated a medicinal chemistry effort to improve potency, cytotoxicity profile, and drug-like properties. Thirty-four compounds were prepared, leading to compound 28 with significant gains in potency (EC50 = 0.92 μM), solubility, and microsomal stability and a demonstrated absence of cytotoxicity in SH-SY5Y human neuroblastoma cells

Bacterial killing in macrophages and amoeba: do they all use a brass dagger?

Macrophages are immune cells that are known to engulf pathogens and destroy them by employing several mechanisms, including oxidative burst, induction of Fe(II) and Mn(II) efflux, and through elevation of Cu(I) and Zn(II) concentrations in the phagosome ('brass dagger'). The importance of the latter mechanism is supported by the presence of multiple counteracting efflux systems in bacteria, responsible for the efflux of toxic metals. We hypothesize that similar bacteria-killing mechanisms are found in predatory protozoa/amoeba species. Here, we present a brief summary of soft metal-related mec

Preparation and evaluation of standardized amoeba antigen from axenic cultures of Entamoeba histolytica.

The successful mass cultivation of Entamoeba histolytica strains in pure culture has made it possible to produce pure amoeba antigens, which should improve the prospects for immunodiagnostic research and simplify the interpretation of the results obtained in tests for amoeba antibodies. The authors have mass-produced immunodiagnostic antigens for amoebiasis serology from 2 axenic strains of E. histolytica and standardized them as dry powders, which upon reconstitution contained approximately 1.8 mg N/ml, or roughly the equivalent of 10 x 10(6) amoebae per ml.The antigens were evaluated with 121

Multiple evolutionary routes to cytoskeletal arborization revealed by the rhizarian amoeba Filoreta ramosa.

The eukaryotic cytoskeleton generates remarkable diversity in cellular architecture despite being built from conserved actin and tubulin polymers. Diversification of cytoskeletal regulators, motors, and filament-organizing proteins produces varied morphologies, yet certain architectures repeatedly emerge in distantly related lineages. One notable example is cytoskeletal arborization, occurring in metazoan neurons and amoeboid lineages throughout the eukaryotic tree. Whether these similar branched architectures arise through conserved cytoskeletal organization, shared molecular reuse, or conver

Refining AMOEBA Electrostatics for d-Glucose and 1→4-Linked d-Glucose Oligosaccharides.

Carbohydrate simulations are highly sensitive to the treatment of hydroxyl electrostatics because hydration, conformational sampling, and reactivity are governed by dense hydrogen-bonding networks. Here, we examine how oxygen quadrupole scaling affects AMOEBA d-glucose models generated with Poltype2 by comparing the standard 0.7 scaling with full 1.0 scaling for α- and β-d-glucose monosaccharides, 1→4-linked oligomers, and concentrated solutions. Increasing the oxygen quadrupole scaling from 0.7 to 1.0 improves monosaccharide hydration free energies, increases d-glucose-water hydrogen-bond cou

Cyclic Peptide Natural Product Inspired Inhibitors of the Free-Living Amoeba Balamuthia mandrillaris.

Balamuthia mandrillaris is a pathogenic free-living amoeba (pFLA) that can cause infection of the central nervous system (CNS), called Balamuthia amoebic encephalitis (BAE), as well as cutaneous and systemic diseases. Patients infected with B. mandrillaris have a high mortality rate due to a lack of effective treatments. A nonoptimized antimicrobial drug regimen is typically recommended; however, it has poor antiparasitic activity and can cause various and severe side effects. Cyclic peptides exhibit a broad spectrum of antimicrobial activities but are underexplored for their antiamoebic activ

Nanopore sequencing improves the draft genome of the human pathogenic amoeba Naegleria fowleri.

Naegleria fowleri is an environmental protist found in soil and warm freshwater sources worldwide and is known for its ability to infect humans and causing a rapid and mostly fatal primary amoebic meningoencephalitis. When contaminated water enters the nose, the facultative parasite follows the olfactory nerve and enters the brain by crossing the cribriform plate where it causes tissue damage and haemorrhagic necrosis. Although N. fowleri has been studied for several years, the mechanisms of pathogenicity are still poorly understood. Furthermore, there is a lack of knowledge on the genomic lev

The interaction between the amoeba Balamuthia mandrillaris and extracellular matrix glycoproteins in vitro.

Balamuthia mandrillaris, a soil amoeba, is the causative agent of Balamuthia granulomatous amoebic encephalitis, a life-threatening brain infection. This amoeba is acquired from contaminated soil and may enter the host through cutaneous lesions or through nasal passages, migrating to the lungs or brain. During invasion, B. mandrillaris has access to components of the extracellular matrix (ECM) of the host. Therefore, we investigated the interaction of B. mandrillaris with 3 ECM glycoproteins (collagen-I, fibronectin and laminin-1) that are encountered in host connective tissues and at the basa
