[H-NMR spectroscopy. Specificity of microbial sialidases against complex substrates].

The specificities of one viral and five bacterial sialidases were investigated by 1H-NMR-spectroscopy with substrates or substrate mixtures containing two sialic acid residues of different linkage types. This technique allows - in contrast to the methods used before - the simultaneous determination of the rates of hydrolysis of both NeuAc linkages in a single experiment. The substrate specificities of the enzymes are discussed on the basis of the relation of the rate constants k/k'. The data obtained are more exact and more informative than those of separate experiments as reported previously.

The specificity of viral and bacterial sialidases for alpha(2-3)- and alpha(2-6)-linked sialic acids in glycoproteins.

The anomeric specificity of six sialidases (Vibrio cholerae, Arthrobacter ureafaciens, Clostridium perfringens, Newcastle disease virus, fowl plague virus and influenza A2 virus sialidases) was assessed with sialylated antifreeze glycoprotein, ovine submandibular gland glycoprotein and alpha 1-acid glycoprotein, resialylated specifically in alpha(2-3) or alpha(2-6) linkage with N-acetylneuraminic acid or N-glycolylneuraminic acid using highly purified sialyltransferases. The rate of release of sialic acid from these substrates was found to correlate well with the specificity observed earlier w

Development of a novel anti-inflammatory recombinant uricase with extended half-life for gout therapy.

Gout is a form of inflammatory arthritis that results from elevated serum uric acid levels and the deposition of urate crystals in multiple joints. The inflammatory response during an acute gout attack is mediated by the activation of the NLRP3 inflammasome, leading to the release of IL-1β and inducing a localized tissue inflammatory response. Urate lowering therapies such as Pegloticase effectively reduce serum uric acid levels but are generally associated with an increase in acute gout flares. In this study, we developed a long-acting anti-inflammatory recombinant uricase by sequential fusin

Reutilization of Cr-Y zeolite obtained by biosorption in the catalytic oxidation of volatile organic compounds.

This work aims at the reutilization of a Cr-loaded NaY zeolite obtained by biorecovery of chromium from water as catalyst in the oxidation of volatile organic compounds (VOC). Cr-NaY catalysts were obtained after biosorption of Cr(VI) using a bacterium, Arthrobacter viscosus, supported on the zeolite. The biosorption experiments were conducted at different pH values in the range 1-4. The catalysts were characterized by several techniques, namely ICP-AES, SEM-EDS, XRD, XPS, Raman, H(2)-TPR and N(2) adsorption. The zeolite obtained at pH 4 has the highest content of chromium, 0.9%, and was selec

Mineralization of carbofuran by a soil bacterium.

A bacterium, tentatively identified as an Arthrobacter sp., was isolated from flooded soil that was incubated at 35 degrees C and repeatedly treated with carbofuran (2,3-dihydro-2,2-dimethyl-7-benzofuranyl N-methylcarbamate). This bacterium exhibited an exceptional capacity to completely mineralize the ring-labeled C in carbofuran to CO(2) within 72 to 120 h in a mineral salts medium as a sole source of carbon and nitrogen under aerobic conditions. Mineralization was more rapid at 35 degrees C than at 20 degrees C. No degradation of carbofuran occurred even after prolonged incubation under ana

Investigating the effect of copper and magnesium ions on nitrogen removal capacity of pure cultures by modified non-competitive inhibition model.

Copper, a common heavy metal, may be beneficial for or poisonous to microbial activity. The objective of this study was to determine the effect of different copper ion concentrations on the nitrogen removal performance of Arthrobacter arilaitensis strain Y-10 and Pseudomonas taiwanensis strain J488. The non-competitive inhibition model was employed to evaluate the 50% inhibition concentrations (IC50 values) of copper ions toward the pure strains. In the absence of magnesium ions, a low concentration of copper (0.1 mg/L) significantly enhanced the ammonium removal ability of strain Y-10 and its

Genetic diversity of phthalic acid esters-degrading bacteria isolated from different geographical regions of China.

Thirty-two strains of phthalic acid ester (PAEs)-degrading bacteria were isolated from thirteen geographically diverse sites by enrichment using mixtures of PAEs as the sole source of carbon and energy. Sequence analyses of the 16S rRNA gene indicated that these isolates were from six genera (Arthrobacter, Gordonia, Rhodococcus, Acinetobacter, Pseudomonas, and Delftia). To evaluate the genetic diversity among them, the molecular typing method rep-PCR with primers based on enterobacterial repetitive intergenic consensus, repetitive extragenic palindromes, and BOXAIR sequences was performed. Psychrophilic Bacteria and Enzyme Activity in Antarctic Ice Environments.

Psychrophilic organisms exhibit many adaptation mechanisms that enable them to survive at low temperatures ranging from -20 to 20 °C. Among other characteristics, these organisms produce cold-active enzymes with considerable biotechnological potential. The purpose of this study was to isolate and characterize bacteria from ice core samples collected on Horseshoe, Hovgaard, and Nansen Islands. Microorganisms from the latter two locations have been investigated only to a limited extent to date. Based on the phylogenetic analysis of 16S rRNA nucleotide sequences, eight bacterial isolates, includi

Antarctic Salt-Cones: An Oasis of Microbial Life? The Example of Boulder Clay Glacier (Northern Victoria Land).

The evaporation of a localized, highly saline water body of the Boulder Clay debris-covered glacier, in the Northern Victoria Land, probably generated the accumulation of mirabilite (Na2SO4 × 10H2O) and thenardite (Na2SO4) in a glacier salt-cone. Such an extremely cold and salty environment resembles the conditions on Mars, so it can be considered a terrestrial analog. The study was aimed at gaining a first glimpse at the prokaryotic community associated with Antarctic mirabilite and thenardite minerals and also to find clues about the origin of the salts. For this purpose, samples were analyz

Specific Rhizobacteria Responsible in the Rhizosheath System of Kengyilia hirsuta.

The rhizosheath is a critical interface supporting the exchange of resources between plants and their associated environment of soil. Favorable microenvironment of rhizosphere soil provides the rhizosheath formed and then promotes desert plant survival. However, it remains unclear how rhizosheath benefits the colonization of pioneer plants in alpine desert under changing environment. In this study, we investigated the effect of different soil moisture and sterilization treatments (three moisture levels and unsterilized or sterilized soil) on rhizosheath forming process of Kengyilia hirsuta (K.

Creation of a robust and R-selective ω-amine transaminase for the asymmetric synthesis of sitagliptin intermediate on a kilogram scale.

The creation of an R-selective ω-amine transaminase (ω-ATA) as biocatalyst is crucial for the asymmetric amination of prochiral ketones to produce sitagliptin intermediates because rare ω-ATAs are R-selective in nature and most of them suffer from poor stability and low activity toward bulky prochiral ketones. Here, the gene of an R-selective ω-ATA was cloned from Arthrobacter cumminsii ZJUT212 (AcATA) and expressed in Escherichia coli. The best variants (M1 + M122H and M1+T134 G) were obtained using a semi-rational protein design after screening. These variants not only exhibited improved act

Isolation and characterization marine bacteria capable of degrading lignin-derived compounds.

Lignin, a characteristic component of terrestrial plants. Rivers transport large amounts of vascular plant organic matter into the oceans where lignin can degrade over time; however, microorganisms involved in this degradation have not been identified. In this study, several bacterial strains were isolated from marine samples using the lignin-derived compound vanillic acid (4-hydroxy-3-methoxybenzoic acid) as the sole carbon and energy source. The optimum growth temperature for all isolates ranged from 30 to 35°C. All isolates grew well in a wide NaCl concentration range of 0 to over 50 g/L, w

Antibacterial Effect of Zinc Oxide-Based Nanomaterials on Environmental Biodeteriogens Affecting Historical Buildings.

The colonization of microorganisms and their subsequent interaction with stone substrates under different environmental conditions encourage deterioration of materials by multiple mechanisms resulting in changes in the original color, appearance and durability. One of the emerging alternatives to remedy biodeterioration is nanotechnology, thanks to nanoparticle properties such as small size, no-toxicity, high photo-reactivity, and low impact on the environment. This study highlighted the effects of ZnO-based nanomaterials of two bacteria genera isolated from the Temple of Concordia (Agrigento'

Epigenetics and Behçet's Disease: DNA Methylation Specially Highlighted.

Behçet's disease (BD) is a multisystem inflammatory disease with unknown etiology. Although evidence about the pathogenesis of BD is growing, the actual cause of this disease is unclear. Both genetic and epigenetic factors are claimed to play significant roles in BD. Epigenetic factors such as age, gender, smoking as well as exogenous factors like diet, infection, stress are related to the onset and clinical manifestations of BD. DNA methylation refers to a major epigenetic element which influences gene activities with catalyzing DNA using a set of DNA methyltransferases (Dnmts). DNA methylati
